Two genes encoding rat cystatin-related prostate protein (Cstrp), previously called CRP (Devos et al., 1993), were mapped to chromosome 3q41 by fluorescent in situ hybridization. The results were confirmed using a panel of mouse-rat hybrids that segregate rat chromosomes. Analysis of genomic DNA indicates that the Cstrp locus comprises probably more than three very similar genes.

To map human chromosome 12p aberrations by fluorescence in situ hybridization (FISH), cosmids were isolated or obtained for 14 known 12p loci (D12S119, D12S158, D12S178, D12S370, D12S380E, A2M, CACNLlAl, FGF6, GAPD, KRAS2, PRB1, PZP, TPI1, and VWF). Using two-color FISH with three labeled probes to interphase nuclei, and to prometaphase chromosomes where possible, the order of these loci was sequentially determined to be pter-D12S158- D12S380E-CACNL1A1-FGF6-D12S370-VWF-GAPD-TPIl-A2M-PZP-PRBl-D12S178-D12S119-KRAS2-cen. Two cell lines were analyzed with this set of cosmids. The EBV-transformed cell line TA carries a der(12) with a deletion of bands 12p13.1→p11.2. D12S178, D12S119, and KRAS2 were absent in the der(12), whereas the other loci were present. The second cell line, GM01203A, exhibits a balanced t(4;12)(4q25; 12p13.3) with a breakpoint between FGF6 and D12S370.

We have constructed a physical map of a 4.6-cM region of human chromosome band 12p13.3 that contains a translocation breakpoint from a mesothelioma with a t(X;12)(q22;p13). The map contains a contig of 22 yeast artificial chromosomes (YACs), onto which we have placed 18 sequence tagged site (STS) markers, including seven genes: D12S370, FGF6, KCNA1, KCNA5, KCNA6, NTF3, and VWF. A second YAC contig, comprised of 22 YAC clones, was located distal to the mesothelioma breakpoint and contained 12 STS markers, including four genes (CACNL1A1, D12S380E, D12S381E, and D12S382E). Based on STS content and fluorescence in situ hybridization experiments, two stable, nonchimeric YAC clones were found that span the mesothelioma breakpoint. A long-range restriction map of an 800-kb region was constructed and used to refine the mesothelioma breakpoint to a region of approximately 100 kb, flanked by the potassium channel genes KCNA1 and KCNA5. The latter was confirmed by direct visual hybridization (DIRVISH) experiments, using cosmids isolated for markers flanking the breakpoint as probes.

Intrachromosomal telomeric sequences (TTAGGG)n were analyzed in the two members of the family Giraffidae, the giraffe and the okapi. The giraffe has a diploid chromosome number of 2n = 30, whereas the okapi chromosome number varies from 2n = 46 to 2n = 45 and 2n = 44 due to a “recent” Robertsonian fusion event. The interstitial telomeres that we detected in these species are of two types: (1) In the okapi, a long interstitial telomeric element is present at the fusion site of the rob(4;26). The nature of this interstitial telomeric element suggests that it is a remnant of the telomeres of the ancestral chromosomes that participated in the fusion event. (2) In the giraffe, short stretches or degenerate telomeric sequences which are part of the satellite DNA are present at intrachromosomal sites. The results of this study provide insights into the origin of interstitial telomeric sequences in the Giraffidae.

Structural rearrangements including deletions of the short arm of chromosome 12 are frequent cytogenetic findings in various hematologic malignant disorders. Using FISH with a panel of DNA probes we detected loss of a common region of 12p in 22 patients with different hematologic disorders. Nine of them were characterized cytogenetically by a del(12p), seven by unbalanced translocations, and in the remaining cases the loss of the 12p region was masked by translocations and insertions, adding extra material to the short arm of chromosome 12. The smallest commonly deleted region found in all cases analyzed included ETV6, the gene for p27kip1 (CDKN1B), and the D12S178 marker.

Rat prostatic binding protein genes C1, C2, and C3 were mapped on rat chromosome 5 by in situ hybridization on rat peripheral blood chromosome preparations using three different cDNA probes. Of the grains detected, 15.9%, 25.2%, and 19.6%, respectively, mapped to chromosome 5. For each probe, the label was predominantly located on 5q31, but for C2 and C3 an additional site on 5q21 was found. The results suggest that three genes coding for the different polypeptide chains of rat prostatic binding protein map to the same chromosome and presumably to the same chromosome band.

Genetic sequences of the novel NSP gene, which encodes neuroendocrine-specific proteins, were isolated from cDNA libraries constructed with mRNA isolated from human lung carcinoma cells. Hybridization analysis of a panel of human × mouse cell hybrids with an 0.8-kb NSP cDNA probe indicated that the human NSP gene is probably located on chromosome 14. Fluorescence in situ hybridization analysis of metaphase chromosomes using overlapping genomic clones of NSP as a probe localized the NSP gene to chromosome region 14q21→q22.
