Abstract

The factors affecting the accuracy of the measurement of androgens, in particular testosterone, by displacement analysis were investigated. Solvents and supports used in thin-layer chromatography contribute significantly to the apparent testosterone measured in the assay. Values ranged from undetectable to greater than 2 ng for solvents depending on their purity. Evidence is presented that when sex hormone binding globulin from pregnancy plasma is used as a binding system, bound labelled hormone is sensitive to the quantity of charcoal adsorbant used to separate free labelled hormone from protein bound hormone. With an incubation volume of 1 ml 0.5–1.0 mg of charcoal was optimal for the separation of protein bound label from free label. Changes in colloid concentration can affect the reliability of measurements as shown by changes in precision observed at different concentrations of added bovine serum albumin (BSA); 25 mg of added BSA per 100 ml of phosphate buffer was found to give the greatest precision.
