Abstract

In some locations, artisanal and small-scale gold-mining (ASGM) represents a significant source of anthropogenic Hg to freshwater environments. The Hg released from ASGM can contaminate aquatic fauna and pose health risks to downstream populations. Total Hg (THg) concentrations, speciation, and isotopic compositions were analyzed in water, suspended particulate matter, soil, and bottom sediment samples from pristine areas and in places of active and legacy gold mining along the Oyapock River (French Guiana) and its tributaries. Mass-independent fractionation (MIF) of even Hg isotopes in top soils (Delta Hg-200 = -0.06 +/- 0.02 parts per thousand, n = 10) implied the uptake of gaseous Hg(0) by plants, rather than wet deposition, as the primary Hg source. Odd isotope MIF was lower in deep soils (Delta Hg-199 = -0.75 +/- 0.03 parts per thousand, n = 7) than in top soils (Delta Hg-199 = -0.55 +/- 0.15 parts per thousand, n = 3). This variation could be attributed to differences between the isotopic signatures of modern and pre-industrial atmospheric Hg. Combining a Hg-isotope binary mixing model with a multiple linear regression based on physicochemical parameters measured in the sediment samples, we determined that active mined creek sediments are contaminated by ASGM activities, with up to 78% of THg being anthropogenic. Of this anthropogenic Hg, more than half (66-74%) originates from liquid Hg(0) that is released during ASGM. The remaining anthropogenic Hg comes from the ASGM-driven erosion of Hg-rich soils into the river. The isotope signatures of anthropogenic Hg in bottom sediments were no longer traceable in formerly mined rivers and creeks.

Abstract

Whether animals can actively avoid food contaminated with harmful compounds through taste is key to understand their ecotoxicological risks. Here, we investigated the ability of honey bees to perceive and avoid food resources contaminated with common metal pollutants known to impair their cognition at low concentrations (lead, zinc and arsenic). In behavioural assays, bees did not discriminate food contaminated with field-realistic concentrations of these metals. Bees only reduced their food consumption and displayed aversive behaviours at very high, unrealistic concentrations of lead and zinc that they perceived through their antennae and proboscis. Electrophysiological analyses confirmed that high concentrations of the three metals in sucrose solution induced a reduced neural response to sucrose in their antennae. Our results thus show that honey bees can avoid metal pollutants in their food, but only at very high concentrations above regulatory levels. Their inability to detect lower, yet harmful, concentrations in a field-realistic range suggests that metal pollution is a major threat for pollinators.

Abstract

Over the course of history, mining and metallurgical activities have influenced the socioeconomic development of human populations. However, these past and current activities can also lead to substantial environmental contamination by various metals. Here, we used an interdisciplinary approach (incorporating archaeology, mineralogy, environmental chemistry and ecotoxicology) to investigate the origin, fate and potential ecotoxicity of anomalous manganese (Mn) concentrations detected in the ancient mining district of Berthelange (medieval period, eastern France). Mineralogical investigations of slag samples showed that smelting temperature conditions in medieval bloomeries led to the production of slags mainly composed of Fe- and Mn-rich olivine, i.e., fayalites. Further mineralogical analyses of bulk soil and clay fractions allowed us to identify the presence of serpentine. This evidence of olivine weathering can account for the release of Mn from slags into the soil. In addition, chemical analyses of total and available (exchangeable and reducible) Mn concentrations in soil samples clearly showed the contribution of slags deposited 1000 years ago to soil contamination. A complementary ecotoxicity bioassay performed on soils from a slag heap using the land snail Cantareus aspersus confirmed that a significant fraction of the Mn detected in soils remains available for partitioning with the soil solution and transfer to soil organisms. Although no growth inhibition of snails was observed after 28 days of exposure, the animals accumulated quite elevated Mn concentrations in their tissues. Our study emphasizes the environmental availability and bioavailability of Mn from ancient metallurgical wastes to soil-dwelling invertebrates, i.e., snails, even one millennium after their deposition. Hence, as for more recent industrial sites, past mining ecosystems must be a cause of concern for the scientific community and public authorities.

Abstract

International audience; Di (2-ethylhexyl) phthalate (DEHP) is recognized in vertebrates as an Endocrine Disrupting Chemical (EDC). DEHP can alter steroid hormones production, development, reproduction and behavior in vertebrates. Only few studies investigated DEHP effects on insects. However, some recent studies on aquatic insects showed that DEHP could also act as an EDC by interfering with the signaling pathways of ecdysteroids, the main hormones involved in the control of insect post-embryonic development and physiology. The aim of the study was to investigate (1) the fate of DEHP within a terrestrial insect species by exposing larvae to food containing a wide range of DEHP concentrations and (2) the effects of this chemical on their post-embryonic development and metamorphosis, by using a multi-level approach. DEHP was shown to be present both in larvae and resulting stages, with higher concentrations in chrysalises and adults than in larvae. DEHP concentrations also decreased at the end of the last larval instar, suggesting the metabolic transformation or excretion of this chemical during this time. Only the two highest DEHP doses induced higher insect mortality, whereas low and intermediate concentrations increased larval food consumption without affecting body weight. Metabolic profiles showed that in control insects, the last three days before metamorphosis correspond to a metabolic transition, but with time-dependent changes in treated insects. Interestingly, DEHP treatments also alter both hemolymphatic ecdysteroid titers and expression levels of ecdysteroid response genes. These results confirm that DEHP can alter insect post-embryonic development and metamorphosis, by interfering with ecdysteroid pathways.

Abstract

International audience; We studied the individual and joint acute toxicity of S-metolachlor (SMOC) and deethylatrazine (DEA - a metabolite of atrazine) on different non-target freshwater crustaceans. We used animals from different ecological groups: two amphipods from surface running water (Gammarus pulex and Gammarus cf. orinos), an isopod from surface stagnant water (Asellus aquaticus) and an amphipod living in groundwater (Niphargus rhenorhodanensis). Organisms were exposed to different levels of SMOC and DEA, alone or in binary mixture. Temperature effect on SMOC toxicity was assessed by exposing G. pulex and N. rhenorhodanensis to SMOC at 11 degrees C and 15 degrees C. Studying mortality as the biological endpoint, N. rhenorhodanensis was more resistant than surface water species towards SMOC and DEA. Among surface water species, G. pulex was the most sensitive while Gammarus cf. orinos and A. aquaticus showed similar responses to both compounds. Temperature increase did not change SMOC toxicity but modify the shape and steepness of the dose-response curve. We used a Model Deviation Ratio (MDR) approach to evaluate the predictability of Concentration Addition (CA) and Independent Action (IA) models to mixture toxicity. Results indicated either an additive or an antagonistic or a synergistic interaction depending on the concentrations combination and the test species. Our finding conclusively show the suitability of CA and IA in predicting mixture toxicities but results should be interpreted with caution according to ecological group of exposed species in risk assessment procedures.

Abstract

International audience; A combined mass-balance and stable isotope approach was set up to identify and quantify dissolved organic carbon (DOC) sources in a DOC-rich (9 mg L-1) eutrophic reservoir located in Western France and used for drinking water supply (so-called Rophemel reservoir). The mass-balance approach consisted in measuring the flux of allochthonous DOC on a daily basis, and in comparing it with the effective (measured) DOC concentration of the reservoir. The isotopic approach consisted, for its part, in measuring the carbon isotope ratios (d13C values) of both allochthonous and autochthonous DOC sources, and comparing these values with the d13C values of the reservoir DOC. Results from both approaches were consistent pointing out for a DOC of 100% allochthonous origin. In particular, the d13C values of the DOC recovered in the reservoir (-28.5 ± 0.2‰; n = 22) during the algal bloom season (May-September) showed no trace of an autochthonous contribution (d13C in algae = -30.1 ± 0.3‰; n = 2) being indistinguishable from the d13C values of allochthonous DOC from inflowing rivers (-28.6 ± 0.1‰; n = 8). These results demonstrate that eutrophication is not responsible for the high DOC concentrations observed in the Rophemel reservoir and that limiting eutrophication of this reservoir will not reduce the potential formation of disinfection by-products during water treatment. The methodology developed in this study based on a complementary isotopic and mass-balance approach provides a powerful tool, suitable to identify and quantify DOC sources in eutrophic, DOC-contaminated reservoirs.

Abstract

International audience; Phytoremediation appears to be a promising technique for metal soil clean up, although its successful application on a large scale still remains a challenge. Field experiments for six scented Pelargonium cultivars, conducted on two Pb-contaminated calcareous and acidic soils, revealed vigorous plant growth, with no symptoms of morpho-phytotoxicity in spite of high Pb accumulation levels. Lead contents in the harvestable parts of all plants grown on the acidic and more contaminated soil were significantly higher than those grown on the calcareous soil. Three cultivars (Attar of Roses, Clorinda and Atomic Snowflake) are Pb-hyperaccumulator plants: they accumulated more than 1000 mg Pb kg1 DW, with high biomass produced.

Abstract

Chromium has two main oxidation states, Cr(III) and Cr(VI), that can occur simultaneously in natural waters. Current consensus holds that Cr(VI) is of high ecotoxicological concern, but regards Cr(III) as poorly bioavailable and relatively non-toxic. In this work, the effects and bioaccumulation of Cr(III), Cr(VI) and their mixture were studied using the freshwater clam Corbicula fluminea as a model organism. Mixture exposures were carried out using solutions isotopically enriched in 50Cr(III) or 53Cr(VI), allowing to quantify the contribution of each redox form to total Cr accumulation in the clams. Following exposure to individual redox forms, Cr(III) accumulated preferentially in the digestive glands and Cr(VI) in the gills of C. fluminea. In mixture exposures, both redox forms accumulated mainly in the gills; the concentration of Cr(III) in the digestive glands being much lowered compared with individual exposures. Both oxidation states affected the expression of biomarkers related to energy reserves, cellular damage and mitochondrial functioning, as well as the expression of mRNA for detoxification genes. The observed effects differed between gills and digestive glands. The present study suggests that Cr(III) is a bioavailable and biologically active elemental species deserving more consideration by the ecotoxicological community.

Abstract

International audience; This study sought to use concentration-time-response surfaces to show the effects of exposure to toxic (semi-)metals on peroxidase activity in higher plants as a function of exposure-concentration and exposure-time. Maize (Zea mays L.) seedlings (i.e., leaves and roots) were exposed to arsenic (as As3+) or aluminium (as Al3+) under hydroponic conditions, and their biomass and peroxidase enzyme responses were assessed at different concentration-time-exposures. The 3D ecotoxi-profile generated with these data showed two distinct regions: the first region is formed by exposures (i.e., points for time-concentration pairings) that were not statistically different from the results of the control points (i.e., zero toxicant concentration and all exposure-times), whereas the second region is formed by exposure pairings with results that were statistically different to those obtained from control pairings. Overall, the data show that enzyme activity increased over a shorter exposure-time when there was an increase in the exposure-concentration of the toxicant, which can be seen on a 3-D toxicity profile. We propose that quantitative relationship ratios from different assessed endpoints (e.g., biomass and enzyme activity) and enzymatic concentration-time-response surfaces could be helpful in the field of environmental-policy management.

Abstract

The toxicity test using freshwater ostracods of the species Heterocypris incongruens is a sub-chronic static test that exposes individuals to whole sediments over a period of 6 d, the endpoints being mortality and growth. We tested the hypothesis that endpoints of the sediment bioassay using Heterocypris incongruens are affected by the presence of fine sediment particles by testing control sediment supplied with the commercial test kit with increasing proportions of kaolin clay as a proxy for fines. While mortality was not affected, the results showed that increasing the presence of clay reduced ostracod growth. Based on the variability in growth, a sublethal toxicity threshold of 35% is proposed to distinguish effects due to sediment properties from those due to toxicity. The relevance of this threshold was verified using data from toxicity tests of ambient sediment samples with low levels of contamination. (C) 2016 Elsevier Ltd. All rights reserved.

Abstract

Development of organisms that live on contaminated soils depends on toxicity as well as several physical and chemical soil properties. We aimed to identify plant bioassays most responsive to contaminants and not to confounding factors due to soil type differences. We implemented a multi-site approach in seven contaminated sites and used different ordinary plant bioassays (fourteen-day-shoot biomass and five-day-root and shoot elongation). Most of the sites were contaminated with polycyclic aromatic hydrocarbons (PAHs), and soils were sampled from areas of both high and low contamination. Bioassays were performed on ninety soil samples and were carried out with six model species. We performed analyses of regulatory PAHs and their derivatives content in the samples. Fourteen-day-shoot biomass responses depended on the site's origin, with an intricate response of plants that faced contrasted soil pH and organic matter content and various contaminant levels. Five-day-shoot and root lengths were informative when considering the most heavily PAH-contaminated site, since both measures exhibited a close dose-dependent response to PAHs but not to soil pH or organic matter content. For the other sites, elongation tests revealed tenuous effects somehow related to the presence of PAHs or their derivatives. We propose that tests based on plant development during their autotrophic phase (the fourteen-day-shoot biomass test in this study) are likely more sensitive to environmental stressors but less specific for contaminant-induced effects. Comparatively, tests based on early and heterotrophic plant development could be particularly more specific for soil contaminants, but the associated responses may be of low sensitivity.

HIGHLIGHTS  KMnO4 and Fenton-like treatments were applied to PAH contaminated soils.  The impact of two oxidant doses on PAC contamination were studied.  Treatments were less efficient on soil with low PAC availability.  KMnO4 was more efficient to degrade the PAH than Fenton-like.  Low dose of KMnO4 induced the formation of toxic oxygenated by-products. Abstract Potassium permanganate and Fenton-like oxidations were applied on two PAH-contaminated soils collected on former coking plant and gas plant sites. The impact of oxidant dose on the polycyclic aromatic compound (PAC) evolution, including 16 US-EPA PAHs, 11 oxygenated-and 4 nitrogen heterocyclic-PACs (O-and N-PACs) was studied for both treatments. The content of extractable organic matter and PACs was determined prior and after oxidation. Overall, permanganate treatment was more efficient than Fenton-like to decrease the PAH content, this latter being limited by the contamination availability. However, permanganate treatment resulted in incomplete PAH degradation, leading to the formation of O-PACs, that was limited with the application of higher dose. It underlines the importance of the dose and the oxidant type in the selection of oxidation parameters for remediation purpose, as improper use of oxidant can lead to the accumulation of oxidation by-products that could be as toxic as the parent compounds.

Abstract

International audience; In the present study, we explored the possibility of using the acetylcholinesterase (AChE) as a biomarker after deltamethrin (pyrethroid insecticide) exposure with three strains of the cladoceran Daphnia magna. Four calculated time-weighted deltamethrin concentrations (20.1, 40.3, 80.6 and 161.3 ng L-1) were compared against control acetylcholinesterase activity. Our results showed that after 48 h of deltamethrin exposure, all treatments induced a significant decrease of AChE activities whatever the three considered strains. However, diverse responses were registered in terms of lowest observed effect concentrations (LOEC: 80.6 ng L-1 for strain 1 and 20.1 ng L-1 for strains 2 and 3) revealing differences in sensitivity among the three tested strains of D. magna. Our results suggest that after deltamethrin exposure, the AChE activity responses can be also used as a biomarker of susceptibility (i.e., variation of strain specific response). Moreover, our results show that strain 1 is the less sensitive in terms of IC50-48 h of AChE, whereas it became the most sensitive when considering the EC50-48 h estimated in the standard ecotoxicity test.

Abstract

International audience; The impact of goethite on air-oxidation of PAH-contaminated soils was studied through two sets of experiments. (i) Soil extractable organic matter (EOM) and (ii) whole coking plant soils were oxidized at 60 and 100 °C for 160 d, with/without goethite. Organic matter (OM) mineralization was monitored via CO2 production and polycyclic aromatic compounds (PACs) oxidation was investigated by GC–MS analyses. The decrease in EOM and PAH contents, and the oxygenated-PAC production observed during EOM oxidation, were enhanced by the presence of goethite. PACs were likely transformed at the goethite surface through electron transfer process. Mass carbon balance revealed a transfer from EOM to the insoluble organic fraction indicating condensation/polymerization of organics. Soil oxidation induced a decrease in EOM, PAH but also in oxygenated-PAC contents, underscoring different oxidation or polymerization behavior in soil. The goethite addition had a lesser impact suggesting that indigenous minerals played an important role in PAC oxidation.

Abstract

International audience; The LuminoTox solid phase assay (Lum-SPA), developed to assess the potential ecotoxicity of natural polluted sediments, employs stabilized thylakoids isolated from spinach plant extracts. When thylakoids are exposed to polluted samples, the action of pollutants can interfere with transmission of chlorophyll fluorescence linked to Photosystems I and II, causing a decrease in fluorescence emission. To differentiate between bulk and leachate effects, we developed a complementary assay of the Lum-SPA named the LuminoTox leachate phase assay (Lum-LPA). Twelve natural sediments were selected on the basis of their potential ecotoxicity and different quantities of fines. Results showed that in the Lum-LPA, the IC50s were always lower than those observed in the Lum-SPA. Significant inverse correlations were found between the IC50s obtained with the Lum-SPA and the percentage of fines (silt + clay fraction). In order to identify the cause of the higher ecotoxicity found in the Lum-LPA, we tested the supernatant of the liquid phase recovered after centrifugation (3000g), and it appeared far less toxic than the Lum-LPA filtrates. To confirm the hypothesis that sensitivity in the Lum-LPA could be due to the presence of very fine particles, Photosynthetic Enzyme Complexes (PECs) were exposed to two kinds of clay materials with and without copper (0.6 mg L-1). Similar results were obtained in the Lum-LPA for both cases, revealing the sensitivity of this test to very fine particles.

Abstract

International audience; Trace metals (TM: Co, Ni, Cu, Zn, Cd, and Pb) as well as Al, Mn, and Fe content was measured in needles of a remote silver fir stand in the south of France. TM localisation and behaviour in needles was evaluated by measuring total and internal content of needles of different ages. Measured concentrations fell within background values. Al, Fe, Co, and Pb were trapped in wax following atmospheric particulate deposition. Contrasting accumulation and migration behaviours of the different elements studied were observed. The wax contained less than 10% Mn, Al, Ni, Co, and Zn and 15–45% Fe, Cu, and Cd in the young needles. Lead was mostly located in the wax (50–80%), and this proportion decreased with needle age. Only the internal content of Pb and Fe increased significantly with needle age. Finally, due to atmospheric deposition accumulation, higher input fluxes of Fe, Cu, Cd, and Pb can be expected in forest soil.

Abstract

International audience; The direct and indirect interaction mechanisms of pyrene with (i) various molecular weight fractions of a synthetic humic-like substance (SyHA) and (ii) extracts of natural humic acids (NHA) from Moselle River suspended matter, were investigated using quenching fluorescence and surface tension measurements. Humic materials were characterized in a previous study. The Stern-Volmer associative constants were determined from the quenching technique. Surface tension measurements revealed an increase in surface activity as a function of concentration for each humic fraction independently of the pyrene presence in solution, even during the formation of humic micelles. The results obtained suggest the possibility of specific intermolecular interactions occurring during pyrene entrapment within humic acids. In addition, we show that molecular weight, aliphatic chains (especially those containing nitrogen groups) and acidity are determinant characteristics for pollutant entrapment capacity at concentrations below the critical micellar concentration (CMC) of humic substances.

Abstract

The use of activated sludge as inoculum source in ready biodegradability tests (RBT) suffers from several drawbacks related to the heterogeneity of these communities. In this work, the ability of a 7-day aeration period in a mineral medium to homogenize the characteristics of various activated sludges, as suggested by some RBT, was studied. The biodegradation potential of three activated sludge supernatants obtained from different wastewater treatment plants was assessed in terms of cultivable cell density, dehydrogenasic activity and a profile of hydrolytic enzymes. After the preconditioning, the homogenization of these characteristics in the supernatants was observed, as well as a decrease. When preconditioned inocula were used in acetate RBT, the biodegradation kinetics were homogenized. However, some preconditioned supernatants lost their ability to degrade an easily-assimilable xenobiotic compound (aniline) during the observation period, showing the effect of inoculum preconditioning on the behavior of complex bacterial communities, specialist populations (e.g. aniline degraders) being more sensitive than generalist populations (e.g. acetate degraders). These results show that preconditioning cannot be an optional inoculum pretreatment in RBT, and emphasize the importance of further studies focusing on inoculum homogenization.
