Biorthogonal labelling with fluorescent small molecules is an indispensable tool for diagnostic and biomedical applications. In dye‐based 5‐ethynyl‐2′‐deoxyuridine (EdU) cell proliferation assays, augmentation of the fluorescent signal entails an overall enhancement in the sensitivity and quality of the method. To this end, a rapid, divergent synthetic procedure that provides ready‐to‐click pH‐insensitive rhodamine dyes exhibiting outstanding brightness was established. Compared to the shortest available synthesis of related high quantum‐yielding rhodamines, two fewer synthetic steps are required. In a head‐to‐head imaging comparison involving copper(I)‐catalyzed azide alkyne cycloaddition reactions with in vitro administered EdU, our new 3,3‐difluoroazetidine rhodamine azide outperformed the popular 5‐TAMRA‐azide, making it among the best available choices when it comes to fluorescent imaging of DNA. In a further exploration of the fluorescence properties of these dyes, a set of bis‐MPA dendrons carrying multiple fluorescein or rhodamine units was prepared by branching click chemistry. Fluorescence self‐quenching of fluorescein‐ and rhodamine‐functionalized dendrons limited the suitability of the dyes as labels in EdU‐based experiments but provided new insights into these effects. This review presents precisely defined amphiphilic dendrons, their self‐association properties, and their different uses. Dendrons, also named dendritic wedges, are composed of a core having two different types of functions, of which one type is used for growing or grafting branched arms, generally multiplied by 2 at each layer by using 1→2 branching motifs. A large diversity of structures has been already synthesized. In practically all cases, their synthesis is based on the synthesis of known dendrimers, such as poly(aryl ether), poly(amidoamine) (in particular PAMAM), poly(amide) (in particular poly(L‐lysine)), 1→3 branching motifs (instead of 1→2), poly(alkyl ether) (poly(glycerol) and poly(ethylene glycol)), poly(ester), and those containing main group elements (poly(carbosilane) and poly(phosphorhydrazone)). In most cases, the hydrophilic functions are on the surface of the dendrons, whereas one or two hydrophobic tails are linked to the core. Depending on the structure of the dendrons, and on the experimental conditions used, the amphiphilic dendrons can self‐associate at the air‐water interface, or form micelles (eventually tubular, but most generally spherical), or form vesicles. These associated dendrons are suitable for the encapsulation of low‐molecular or macromolecular bioactive entities to be delivered in cells. This review is organized depending on the nature of the internal structure of the amphiphilic dendrons (aryl ether, amidoamine, amide, quaternary carbon atom, alkyl ether, ester, main group element). Dendrons are an important class of macromolecules that can be used for a broad range of applications. Recent studies have indicated that mixtures of oligoglycerol detergent (OGD) regioisomers are superior to individual regioisomers for protein extraction. The origin of this phenomenon remains puzzling. Here we discuss the synthesis and characterization of dendritic oligoglycerol regioisomer mixtures and their implementation into detergents. We provide experimental benchmarks to support quality control after synthesis and investigate the unusual utility of OGD regioisomer mixtures for extracting large protein quantities from biological membranes. Specifically amplifying the emission signals of optical probes in tumors is an effective way to improve the tumor‐imaging sensitivity and contrast. In this paper, the first case of dendron‐based fluorescence turn‐on probes mediated by a Förster resonance energy transfer (FRET) mechanism is reported. Dendrons up to the fourth generation with a hydrophilic oligo(ethylene glycol) scaffold are synthesized by a solid‐phase synthesis strategy, and show precise and defect‐free chemical structures. To construct the fluorescence turn‐on probe, one Cy5.5 molecule is conjugated to the focal of a G3 dendron through a robust linkage and eight Black Hole Quencher 3 (BHQ‐3) molecules are conjugated to its periphery through a PEG chain bearing a reductively cleavable disulfide linkage. By in vitro and in vivo experiments, it is demonstrated that the fluorescence of the dendron‐based probe can be activated effectively and rapidly in the reductive environments of tumor cells and tissues, and the probe thus exhibits amplified tumor signals and weak normal tissue signals. Compared with the reported nanoscale turn‐on probes, the dendron‐based probe has several significant advantages, such as well‐defined chemical structure, precisely controllable fluorophore/quencher conjugation sites and ratio, desirable chemical stability, and reproducible pharmacokinetic and pharmacological profiles, and is very promising in tumor detection. First‐in‐class CuII and AuIII metaled phosphorus dendrons were synthesized and showed significant antiproliferative activity against several aggressive breast cancer cell lines. The data suggest that the cytotoxicity increases with reducing length of the alkyl chains, whereas the replacement of CuII with AuIII considerably increases the antiproliferative activity of metaled phosphorus dendrons. Very interestingly, we found that the cell death pathway is related to the nature of the metal complexed by the plain dendrons. CuII metaled dendrons showed a potent caspase‐independent cell death pathway, whereas AuIII metaled dendrons displayed a caspase‐dependent apoptotic pathway. The complexation of plain dendrons with AuIII increased the cellular lethality versus dendrons with CuII and promoted the translocation of Bax into the mitochondria and the release of Cytochrome C (Cyto C). An interesting halogen-substituent effect on the organogelation properties of poly(benzyl ether) dendrons is reported. A new class of poly(benzyl ether) dendrons with halo substituents decorating their periphery was synthesized and fully characterized. A systematic study on the gelation abilities, thermotropic behaviors, aggregated microstructures, and mechanical properties of self-assembled organogels was performed to elucidate the halogen-substituent effects on their organogelation propensity. It was found that the exact halogen substitutions on the periphery of dendrons exert a profound effect on the organogelation propensity, and dendrons G n -Cl ( n=2, 3) and G2-I proved to be highly efficient organogelators. The cooperation of multiple π-π, dispersive halogen, CH-π, and weak C−H ⋅⋅⋅X hydrogen-bonding interactions were found to be the key contributor to forming the self-assembled gels. Dendritic organogels formed from G n -Cl ( n=2, 3) in 1,2-dichloroethane exhibited thixotropic-responsive properties, and such thixotropic organogels are promising materials for future research and applications. A series of poly(aryl ether) dendrons with a monopyrrolo-tetrathiafulvalene unit linked through an acyl hydrazone linkage were designed and synthesized as low molecular mass organogelators (LMOGs). Two of the dendrons could gelate the aromatic solvents and some solvent mixtures, but the others could not gel all solvents tested except for n-pentanol. A subtle change on the molecular structure produces a great influence on the gelation behavior. Note that the dendrons could form the stable gel in the DMSO/water mixture without thermal treatment and could also form the binary gel with fullerene (C60) in toluene. The formed gels undergo a reversible gel-sol phase transition upon exposure to external stimuli, such as temperature and chemical oxidation/reduction. A number of experiments (SEM, FTIR spectroscopy, 1H NMR spectroscopy, and UV/Vis absorption spectroscopy, and XRD) revealed that these dendritic molecules self-assembled into elastically interpenetrating one-dimensional fibrillar aggregates and maintain rectangular molecular-packing mode in organogels. The hydrogen bonding, π-π, and donor-acceptor interactions were found to be the main driving forces for formation of the gels. Moreover, the gel system exhibited gel-induced enhanced emission (GIEE) property in the visible region in spite of the absence of a conventional fluorophore unit and the fluorescence was effectively quenched by introduction of C60. Three triazine-based dendrons ( 1 a- c) were successfully prepared in 70-83 % yields. These newly prepared dendrons are found to be liquid crystalline (LC). Computational investigations on molecular conformations and dipoles of triazine-based dendrons reveal that the substituent on the central triazine unit interrupts strong dipole or H-bond interactions to avoid dimeric formation. The obtained dendrons, not favouring self-assembly into dimers but showing LC behaviours, provides evidence for an approach contrary to the conventional method of inducing LC behaviours of dendrons by dimer or trimer formation, mostly through H-bond interactions. o-Carborane (C2B10H12) was adapted to perform as the core of globular macromolecules, dendrons or dendrimers. To meet this objective, precisely defined substitution patterns of terminal olefin groups on the carborane framework were subjected to Heck cross-coupling reactions or hydroboration leading to hydroxyl terminated arms. These led to new terminal groups (chloro, bromo, and tosyl leaving groups, organic acid, and azide) that permitted ester production, click chemistry, and oxonium ring opening to be performed as examples of reactions that demonstrate the wide possibilities of the globular icosahedral carboranes to produce new dendritic or dendrimer-like structures. Polyanionic species were obtained in high yield through the ring-opening reaction of cyclic oxonium compound [3,3′-Co(8-C4H8O2-1,2-C2B9H10)(1′,2′-C2B9H11)] by using terminal hydroxyl groups as nucleophiles. These new polyanionic compounds that contain multiple metallacarborane clusters at their periphery may prove useful as new classes of compounds for boron neutron capture therapy with enhanced water solubility and as cores to make a new class of high-boron globular macromolecules. Flexible, linked dendritic tetraphenylporphyrin (TPP)-fullerene hybrids were synthesized. They were designed to gain insight into and mimic the primary events in the natural photosynthetic reaction center. These multiporphyrin moieties are based on a light-harvesting concept. Moreover, they incorporate multiple redox components aligned along a redox gradient. Newkome-type dendrons were added to these TPP-fullerene hybrids. In principle they can mediate pH-dependent water solubility, which, however, could not be observed in this case. A protecting-group strategy using tert-butyldiphenylsilyl groups allows convergent synthesis of the dendritic compounds. The dendritic multiporphyrins were synthesized separately and can be used as individual building blocks. Atropisomerism was observed in the dendritic compounds, and single atropisomers could be assigned to the corresponding peaks of a characteristic pattern in the NMR spectra. Deprotection of the Newkome-type dendrons was shown to be feasible under mild conditions that leave the redox gradient intact. [ABSTRACT FROM AUTHOR]

An intelligent molecular hydrogel with a volume phase transition was constructed to regulate the chiral packing of a well-known cyanine dye on a dynamically self-assembled chiral nanofiber by using a pH trigger. During the shrinkage of the gel, the chiral nanofiber hierarchically assembled into a superhelix and simultaneously drove the dye molecules to stack, from a predominantly monomer form, in an unexpected helical H-aggregation manner. Through such a transformation, the supramolecular chirality of the system was significantly enhanced and a new property of visual discrimination for chiral amines emerged. [ABSTRACT FROM AUTHOR]

A shrinkable supramolecular metallo-hydrogel based on the L-glutamic acid dendron and magnesium showed reversible volume-phase transition depending on pH changes. The hydrogel further showed selective shrinkage upon addition of positively charged species, while it remained in the gel state when negatively charged species were incorporated. Based on this property, the gel could be used as the matrix to efficiently separate ionic dye mixtures, in which the cationic dye was incorporated predominantly in the shrunken gel, while the negatively charged dye was released into the aqueous solution. More interestingly, the shrinkable gel can be used as a model drug-delivery vehicle for the stepwise release of a two-component drug system, in which the negatively charged drug is released first and then the second component is released with a pH trigger. We describe the design, synthesis, and 'stimuli-responsive' study of ferrocene-linked Fréchet-type [poly(aryl ether)]-dendron-based organometallic gels, in which the ferrocene moiety is attached to the dendron framework through an acyl hydrazone linkage. The low-molecular-weight gelators (LMWGs) form robust gels in both polar and non-polar solvent/solvent mixtures. The organometallic gels undergo stimuli-responsive behavior through 1) thermal, 2) chemical, and 3) electrochemical methods. Among them, conditions 1 and 3 lead to seamlessly reversible with repeated cycles of identical efficiency. Results indicate that the flexible nature of the poly(aryl ether) dendron framework plays a key role in retaining the reversible electrochemical behavior of ferrocene moiety in the LMWGs. Further, the organometallic gelators have exhibited unique selectivity towards Pb2+ ions (detection limit ≈10−8 M). The metal ion-sensing results in a gel-sol phase transition associated with a color change visible to the naked eye. Most importantly, decomplexing the metal ion from the system leads to the regeneration of the initial gel morphology, indicating the restoring ability of the organometallic gel. Chiral Hydrogel Shrinkage A cyanine dye has been incorporated into chiral nanofibers through co ‐ gelation with an amphiphilic dendron and magnesium ions. The resultant intelligent molecular hydrogel undergoes a volume phase transition with a pH trigger. During the gel shrinkage, the helical nanofibers were transformed to a superhelix and the dye molecules were driven to a chiral H ‐ aggregation, a rare aggregation mode for the dye.
