The Drosophila genome contains at least three loci for the Na,K-ATPase beta-subunit; however, only the protein products of nrv1 and nrv2 have been characterized hitherto. Here, we provide evidence that nrv3 also encodes for a functional Na,K-ATPase beta-subunit, as its protein product co-precipitates with the Na,K-ATPase alpha-subunit. Nrv3 expression in adult flies is restricted to the nervous system in which Nrv3 is enriched in selective types of sensory cells. Because Nrv3 expression is especially prominent in the compound eye, we have analyzed the subcellular and developmental distribution of Nrv3 within the visual cells and related this distribution to those of the alpha-subunit and of the beta-subunits Nrv1 and Nrv2. Prospective visual cells express Nrv2 in the third larval instar stage and during the first half of pupal development. During the last third of pupal life, Nrv3 gradually replaces Nrv2 as the Na,K-ATPase beta-subunit in the photoreceptor cells. Adult photoreceptors express Nrv3 as their major beta-subunit; the visual cells R1-R6 co-express Nrv2 at a low level, whereas R7 and R8 co-express Nrv1. Notably, beta-subunits do not co-distribute exactly with the alpha-subunit at some developmental stages, supporting the concept that the alpha-subunit and beta-subunit can exist in the plasma membrane without being engaged in alpha/beta heterodimers. The non-visual cells within the compound eye express almost exclusively Nrv2, which segregates together with the alpha-subunit to septate junctions throughout development.

The differentiation of copper cells, which secrete stomach acid in Drosophila larvae, has been shown previously to be sensitive to the labialk3 mutation. Here we found that stomach acid secretion in adults was insensitive to labk3. The basis for this stage-specific effect was elucidated by characterizing the development, structure, and function of the adult midgut. First, we demonstrated by copper-dependent fluorescence and morphology that copper cells were present in the adult stomach. Fine-structure analysis of adult copper cells led to the identification of a previously unrecognized plasma membrane domain: apicolateral contacts between copper cells and their neighbors consisted of smooth septate junctions that were enriched in alphabeta-spectrin and ankyrin. Second, we demonstrated that adult copper cells were present in labk3/labvd1 (conditional/null) adults. The labial protein was expressed in adult labk3/labvd1 copper cells, but not in larvae. Thus the labk3 mutation had a stage-specific effect on midgut labial expression, but did not appear to affect protein function. Surprisingly, stomach acidification was dispensable during larval development, since labk3/labvd1 mutant larvae that lacked midgut acidification developed into fertile adults.

Immunohistochemical and physiological studies on various insect photoreceptors have demonstrated that the Na,K-ATPase (sodium pump) is restricted to the nonreceptive nonmicrovillar area of the plasma membrane. Here, we examined the distribution of the Na,K-ATPase in photoreceptor cells of the superposition-type compound eye in the moth Manduca sexta. Using immunofluorescent and immunogold cytochemistry, we show that the Na,K-ATPase is localized to both the nonmicrovillar and the microvillar parts of the plasma membrane. Manduca photoreceptors thus deviate from the common concept that the sodium pump and the molecular components of the photoreceptive machinery reside on different domains of the plasma membrane.

Drosophila melanogaster photoreceptors are highly polarized cells and their plasma membrane is organized into distinct domains. Zonula adherens junctions separate a smooth peripheral surface, the equivalent of the basolateral surface in other epithelial cells, from the central surface (approximately equal to apical surface). The latter consists of the microvillar rhabdomere and the juxtarhabdomeric domain, a nonmicrovillar area between the rhabdomere and the zonulae adherens. The distribution of Na/K-ATPase over these domains was examined by immunocytochemical, developmental, and genetic approaches. Immunofluorescence and immunogold labeling of adult compound eyes reveal that the distribution of Na/K-ATPase is concentrated at the peripheral surface in the photoreceptors R1-R6, but extends over the juxtarhabdomeric domain to the rhabdomere in the photoreceptors R7/R8. Developmental analysis demonstrates further that Na/K-ATPase is localized over the entire plasma membrane in all photoreceptors in early pupal eyes. Redistribution of Na/K-ATPase in R1-R6 occurs at about 78% of pupal life, coinciding with the onset of Rh1-rhodopsin expression on the central surface of these cells. Despite the essential role of Rh1 in structural development and intracellular trafficking, Rh1 mutations do not affect the distribution of Na/K-ATPase. These results suggest that Na/K-ATPase and rhodopsin are involved in distinct intracellular localization mechanisms, which are maintained independent of each other.

Light stimulation of locust (Schistocerca gregaria) photoreceptors results in an actin-dependent translocation of mitochondria towards the photoreceptive microvilli and an antagonistic movement of endoplasmic reticulum towards the cell body. Using immunocytochemical techniques, we have tried to identify myosin-like motors that may drive the light-induced organelle motility. A monoclonal antibody against the motor domain of Acanthamoeba myosin identifies a prominent 110-kDa protein on Western blots of locust retina. Cross-reactivity with two polyclonal anti-myosin antibodies and a monoclonal anti-myosin-I-antibody, together with ATP-dependent binding to actin filaments, provides evidence that the 110-kDa protein is an unconventional myosin. By indirect immunofluorescence, the 110-kDa protein has been localized to both photoreceptors and pigment cells within the retina. In the photoreceptor cells, the 110-kDa protein is bound to the surface of mitochondria. This putative unconventional myosin may thus be a motor protein involved in the light-induced translocation of mitochondria in photoreceptors.

The architecture of the Golgi complex in honeybee photoreceptors has been analyzed by electron-microscopic techniques. The Golgi apparatus consists of several hundred individual stacks of cisternae dispersed throughout the soma of the photoreceptor cell. Two distinct subpopulations of Golgi stacks are distinguishable by their topographic features: (1) a dense row of Golgi stacks is aligned along the palisade-like cisternae of smooth endoplasmic reticulum backing the photoreceptive microvilli; (2) other Golgi stacks are scattered in the remainder of the cell body. The spatial relationship of Golgi stacks to microtubules and actin filaments has also been determined. Electron-microscopic examination of high-pressure-frozen freeze-substituted retinae reveals that Golgi stacks backing the submicrovillar endoplasmic reticulum reside in a cell area without microtubules, whereas the second subpopulation of Golgi stacks is embedded amidst microtubules. Labeling studies with several actin-specific probes, viz., rhodamine phalloidin, monoclonal anti-actin antibodies, and myosin fragments, provide evidence for a juxtaposition of the submicrovillar Golgi stacks to actin filaments. The Golgi membranes are thus ideally positioned to facilitate the transport of Golgi-derived material toward the microvilli along actin filaments.

Light stimulation of locust photoreceptors causes a translocation of submicrovillar cisternae of the endoplasmic reticulum away from the rhabdome, and a movement of mitochondria towards the rhabdome. To examine whether the microtubule cytoskeleton could be involved in these organelle movements, we have analysed the distribution of the microtubule-dependent motor proteins kinesin and cytoplasmic dynein in the retina of the locust Schistocerca gregaria. Both kinesin and cytoplasmic dynein are associated with vesicular structures that are distinct from mitochondria and the submicrovillar endoplasmic reticulum. These results, together with the previous demonstration of a lack of microtubules in the cell area of light-dependent organelle movements, provide evidence that the microtubule cytoskeleton is not involved with light-induced organelle translocations.

The Na,K-ATPase (sodium pump) plays a central role in the physiology of arthropod photoreceptors as it re-establishes gradients for Na+ and K+ after light stimulation. We have mapped the distribution of the Na,K-ATPase in the photoreceptors of the blowfly (Calliphora erythrocephala) by immunofluorescent and immunogold cytochemistry, and demonstrate that the distribution pattern is more complex than previously presumed. High levels of sodium pumps have been detected consistently in all photoreceptors R1-8 on the nonreceptive surface, but no sodium pumps are found on the microvillar rhabdomere. Within the nonreceptive surface of the cells R1-6, however, the sodium pumps are confined to sites juxtaposed to neighboring photoreceptor or glial cells; no sodium pumps have been detected on the parts of the nonreceptive surface exposed to the intra-ommatidial space. In R7 and R8, the sodium pumps are found over the entire nonreceptive surface. The cytoskeletal protein spectrin colocalizes with the sodium pumps suggesting that linkage of the pump molecules to the spectrin-based submembrane cytoskeleton contributes to the maintenance of the complex pattern of pump distribution.

The endoplasmic reticulum (ER) in the photoreceptors of the honeybee drone, Apis mellifera, is highly differentiated and morphologically more complex than suggested by previous studies. In addition to the prominent voluminous submicrovillar cisternae we describe a submitochondrial ER. It separates the mitochondria-containing periphery from the core of the cell. The cell core contains many fenestrated ER cisternae that are horizontally and periodically arranged. We show that all parts of the ER, except for a tubulovesicular portion but including the nuclear envelope, are continuous; all parts appear to accumulate Ca2+ actively and with high affinity. Our results confirm previous suggestions that the submicrovillar ER is the major intracellular Ca2+ -store in the photoreceptors. The submitochondrial ER is thought to participate in Ca2+-regulation in the mitochondrial microenvironment. Moreover, we describe for the first time an extensive, morphologically complex Ca2+-sequestering ER in the pigmented glial cells; it might participate in the regulation of the glycogen metabolism.
