Fig fruit is widely accepted in warm and tropical regions due to the sweet taste and health benefits. The polysaccharides in fig fruit are responsible for the health benefits. However, information regarding polysaccharide structure remains limited. In this study, the water-soluble polysaccharides (FFP) were extracted from fig fruit with a yield of 15.3 mg/g. Anion exchange chromatography and gel permeation chromatography were used for purification. A leading polysaccharide was purified and characterized as below. The monosaccharide composition of FFP included arabinose (Ara), galactose (Gal), galacturonic acid (GalA) and rhamnose (Rha). The glycosidic linkages were revealed to be L-Ara-(1→, →5)-L-Ara-(1→, →2,4)-L-Rha-(1→, →2)-L-Rha-(1→, →4)-D-Gal-(1→ and D-Gal-(1 → . Nuclear magnetic resonnance (NMR) spectra revealed that FFP had →4)-α-D-GalpA-(1 → 2)-α-L-Rhap-(1→ as backbone. The side chains included Galp-β-D-(1 → 4)-Galp-β-D-(1 → 4)-Galp-β-D-(1 → 4)-Galp-β-D-(1→ and α-L-Araf-(1 → 5)-α-L-Araf-(1 → . They were linked to C-4 of Rhap. FFP inhibited the production of NO, IL-6 and IL-1β levels induced by LPS. IL-6 and IL-1β levels were returned to normal. Chitin oligosaccharides have garnered significant attention due to their biological activities, particularly their immunomodulatory properties. However, O-acetylation in chemically preparing chitin oligosaccharides seems inevitable and leads to some uncertainty on the bioactivity of chitin oligosaccharides. In this study, an O-acetyl-free chitin oligosaccharides and three different O-acetylated chitin oligosaccharides with degree of polymerization ranging from 2 to 6 were prepared using ammonia hydrolysis, and their structures and detailed components were further characterized with FTIR, NMR and MS. Subsequently, the effects of O-acetylation on the immunomodulatory activity of chitin oligosaccharides were investigated in vitro and in vivo. The results suggested that the chitin oligosaccharides with O-acetylation exhibited better inflammatory inhibition than pure chitin oligosaccharides, significantly reducing the expression of inflammatory factors, such as IL-6 and iNOS, in the LPS-induced RAW264.7 macrophage. A series of derivatives of salidroside with mirror isomer glucose and different phenyl moieties were synthesized by Schmidt glycosylation in satisfactory yields, and their antioxidant and anti-inflammatory activities were evaluated by using LPS-induced RAW264.7 cells. One of the synthesized derivatives ʟ-Sal-4, bearing ʟ-glycosyl and -OMe modification at the phenyl ring, exhibited high activity in inhibiting the production of pro-inflammatory cytokines and oxidative stress biomarker MDA as well as in enhancing the activity of SOD enzyme, compared with the natural product and its corresponding ᴅ-enantiomer. Further proteomic analysis suggested that ʟ-Sal-4 exerted its anti-inflammatory activity through metabolic reprogramming. The in vitro activity showed that ʟ-Sal-4 is a potent antioxidant and anti-inflammatory agent. The polymeric nanoparticles (PNPs) loaded with prednisolone were developed to exhibit pH-responsive properties owing to the attachment of a hydrazone linkage between the copolymer chitosan and mPEG. In the diseased cellular environment, the hydrazone bond tends to break due to reduced pH, leading to the release of the drug from the PNPs at the required site of action. The fabricated PNPs exhibit spherical morphology, optimum size (∼200 nm), negative surface charge, and monodispersed particle size distribution. The encapsulation efficiency of the PNPs was determined to be 71.1 ± 0.79 % and two experiments (polymer weight loss and drug release) confirmed the pH-responsive properties of the PNPs. The cellular study cytotoxicity assay showed biocompatibility of PNPs and drug molecule-mediated toxicity to A549 cells. The ligand atrial natriuretic peptide-attached PNPs internalized into A549 cells via natriuretic peptide receptor-A to achieve target specificity. The PNPs cytotoxicity and pH-response medicated inflammation reduction functionality was studied in inflammation-induced RAW264.7 cell lines. The study observed the PNPs effectively reduced the inflammatory mediators NO and ROS levels in RAW264.7. Feruloylated oligosaccharides (FOs) generated by decomposing plant hemicellulose, offer a wide range of potential applications in both the food and biomedical areas. As a graminaceous plant, bamboo is rich in hemicellulose. However, the structural composition and activity studies of FOs from it were rarely reported. In this study, FOs from Phyllostachys acuta (pFOs) obtained by enzymatic hydrolysis were isolated by AmberliteXAD-2 and C18 SPE columns. Then, pFOs were qualitatively and quantitatively analyzed by UPLC-ESI-MS/MS after labeled by 3-Amino-9-ethyl-carbazole (AEC), and the chemical antioxidant activity of pFOs and effects of pFOs on H 2 O 2 -induced oxidative damage were investigated. Finally, 14 of pFOs isomers were distinguished and identified, of which 10 did not contain hexoses and 4 did, and the three most abundant pFO structures were 12 (Iso 7, F1A1X2H2-AEC, 29.04 %), 11 (Iso 6, F1A1X1H2-AEC, 17.96 %), and 4 (Iso 3-1, F1A1X3-AEC, 15.57 %). The results of antioxidant studies showed that pFOs possessed certain reducing power, scavenging DPPH radicals, scavenging superoxide anion radicals, and scavenging hydroxyl radicals. Among them, the ability to clear DPPH radicals was particularly significant. pFOs significantly reduced the viability of RAW264.7 cells after H 2 O 2 induction, whereas pFOs had a significant protective effect (p < 0.001). pFOs increased the viability of T-AOC and SOD enzymes in oxidatively damaged cells, as well as had a significant inhibition effect on ROS elevation (p < 0.001). Armillaria luteo-virens (A. luteo-virens) is a kind of edible fungus mainly exists in Qinghai-Tibet of China, but at present only very few studies focus on the bioactivities of its polysaccharides. This study aimed to purify and characterize the structure features of a novel intracellular polysaccharide (ALP-A) derived from A. luteo-virens and explore its potential anti-tumor and immunomodulatory activities. Through systematic separation and purification, we obtained a homogeneous ALP-A with an average molecular weight of 23693Da. Structural analysis indicated that ALP-A was mainly composed of glucose and mannose with a molar ratio of 6.02:1. The repeating unit of ALP-A was →4) -α-D-Glcp-(1→ backbone with α-Glcp-(1→ and α-Manp-(6→ side chains which branched at O-2 position. The anti-tumor assays in vivo suggested that ALP-A could effectively restrain S180 solid tumor growth, protect immune organs and promote the secretion of cytokines (IL2, IL6 and TNF-α) in serum. Besides, in vitro immunomodulatory assays indicated that ALP-A could improve proliferation, phagocytic capacity and raise the level of NO and cytokines in Raw264.7 cells. The peel of Citrus reticulate 'Chachiennsis' (Chachi) is widely accepted as a functional food. It is generally recognized that its health benefits are dependent on storage time. However, the chemicals responsible for this phenomonon remain unclear. As bioactive polysaccharides are dominant in Chachi, Chachi polysaccharides with various storage times (5-20 years) were prepared. The monosaccharide composition was analyzed by GC-MS. NMR data revealed that Chachi polysaccharides were mainly consisted of arabinogalacturonan and another pectin with side chain of →4)-β-D-Galp-(1 → . α-L-Araf-(1→, →5)-α-L-Araf-(1→, →4)-α-D-GalpA-(1→, →4)-α-D-GalpAMe-(1→, →4)-β-D-Galp-(1→ and →4)-β-D-Glcp-(1→ were detected. The molecular weight of Chachi polysaccharides decreased along with the extension of storage time. However, the basic structure characteristics remained stable. The immumomodulatory activities of Chachi polysaccharides were improved as the storage time extended. The change of molecular weight was responsible for the improved immunomodulatory activity.
