Microencapsulation is one of the most important methods to enhance the survival of bacteria when exposed to various harsh conditions. The present study evaluated the viability of L. reuteri ATCC 23272 microencapsulated in polysaccharide-based bionanocomposite. Inulin, polydextrose, and pectin were utilized as prebiotics, and magnesium oxide nanoparticles (MgO NPs) as reinforcing agent in the microgel structure. The composition of bionanocomposite was optimized using the simplex-lattice mixture method. Bionanocomposite optimal formulation was achieved by combining 91.6 % inulin and 8.4 % pectin in the presence of MgO NPs. L. reuteri prebiotic score (1.33) and E. coli (1.08), extrusion efficiency (97.57 %), viability after drying (99.37 %), and viability in simulated gastrointestinal conditions (SGI) (91.74 %) were obtained. Not using MgO NPs in the optimal composite structure caused a decrease of 2.14 log CFU/g in SGI. During 28 days of storage of bacteria at 4 and 25 °C, respectively, a reduction of 2.56 and 3.04 log CFU/g was observed for free cells compared to encapsulated cells. SEM, FTIR, and XRD analyses were performed on ingredients and microcapsules with and without bacteria. Nanocomposite hydrogels based on tyramine conjugated gum tragacanth, poly (vinyl alcohol) (PVA), and halloysite nanotubes (HNTs) were prepared by electron beam irradiation and characterized. The FTIR, 1 H NMR, and TGA results confirmed the chemical incorporation of HNTs into gum tragacanth. Gel content and swelling of hydrogels decreased with HNTs loading up to 20 % wt. The mechanical strength of hydrogels increased by increasing HNTs content up to 10 % with 371 kPa fracture stress at 0.95 fracture strain, compared to 312 kPa stress at 0.79 strain for gum tragacanth/PVA hydrogel. Hydrogel's biocompatibility and osteogenic activity were tested by seeding rabbit bone marrow mesenchymal stem cells. The cell viability was >85 % after 7 days of culture. Low mechanical strength and untargeted osteoinduction of chitosan hydrogel limit its application for bone regeneration. This study aimed to develop an injectable chitosan hydrogel with enhanced mechanical strength and improved osteoinductivity for bone tissue engineering. For this purpose, chitosan-modified halloysite nanotubes (mHNTs) were synthesized first. Then, icariin as a bone inducer was loaded into mHNTs (IC@mHNTs), resulting in a sustained drug release system. Further, nanocomposite chitosan/mHNTs hydrogels were prepared by the sol-gel transition, leading to decreased gelation time and temperature and enhanced mechanical strength of the resulting scaffolds. The mesenchymal stem cells were encapsulated into the hydrogels, and in vitro viability assays showed scaffold biocompatibility. Moreover, embedded mHNTs or IC@mHNTs in the scaffold resulted in enhanced proliferation and bone differentiation of encapsulated cells. It was collectively demonstrated that the injectable in situ forming nanocomposite chitosan hydrogel loaded with IC@mHNTs is a promising candidate for bone regeneration., (Copyright © 2021 Elsevier Ltd. In the present study, electron beam irradiation was applied to prepare a chemically crosslinked hydrogel based on tyramine conjugated gum tragacanth. Then, the gel content, swelling behavior and cytotoxicity of the hydrogels were evaluated. The gel content of the hydrogels was in the range of 75-85%. Equilibrium swelling degree of the hydrogels decreased from 51 to 14 with increasing polymer concentration and irradiation dose. Moisture retention capability of the hydrogels after 5h incubation at 37°C was in the range of 45-52 that is comparable with of commercial hydrogels. The cytotoxicity analysis showed the good biocompatibility of hydrogels. These results indicated that electron beam irradiation is a promising method to prepare chemically crosslinked tyramine conjugated gum tragacanth hydrogels for biomedical applications.
