The replacement time of the human stratum corneum of fifteen body regions was determined by measuring the days required for a strongly substantive fluorescent marker, Dansyl chloride, to disappear. The horny layer was stained by a 24-h patch exposure to 5% Dansyl chloride (5-dimethylamino-I-naphthalene-sulphonyl chloride) in petrolatum. In most body regions the renewal time for the horny layer was about 2 weeks, in agreement with the findings of other investigators. The advantage of Dansyl chloride over another compound, tetrachlorosalicylanilide, which has been used for this purpose, is that it is not an irritant. [ABSTRACT FROM AUTHOR]

The guinea-pig was used as a model for the analysis of flare and rash reactions observed after intravenous injections of DNBSO3 Na in animals with a contact allergy to dinitrochlorbenzene. Histological and pharmacological studies could not confirm the mechanism of an Arthus type of reaction. It is suggested that in addition to allergic mechanisms non-specific and non-allergic phenomena may also play a role in provoking the clinical symptoms of flare and rash in hypersensitive animals. The determination of free formaldehyde in pieces of clothing brought by patients with a formaldehyde contact dermatitis, together with patch tests with these samples has revealed that such patch tests are of very little clinical value. The level of free formaldehyde in almost all samples investigated was shown to be lower than the clinically required concentration. Withdrawal of clothes containing formaldehyde from the patient's wardrobe nevertheless is always followed by disappearance of symptoms. The discrepancy between these findings is discussed.
