Physiological studies have shown that afferents from the nucleus paragigantocellularis (PGi) in the rostral ventral medulla underlie the modulation of locus coeruleus (LC) activity by a variety of stimuli. However, there have been no anatomical demonstrations of a monosynaptic projection from neurons in the PGi to the LC. Thus, biotinylated dextran amine (BDA) was iontophoretically injected into the ventral medulla and single-tissue sections were processed for peroxidase localization of BDA and gold-silver labeling of tyrosine hydroxylase (TH). Discrete microinjections of BDA were placed into either the medial or lateral aspects of the ventral medulla. For medially placed injections, a medio-dorsal pathway to the LC was observed. This trajectory resulted in a predominant innervation of the ventral LC. Lateral injection placements yielded a fiber pathway that coursed more laterally within the medullo-pontine reticular formation and primarily innervated the dorsolateral LC. These light microscopic data suggested that neurons in the PGi use distinct pathways to innervate the LC and are topographically organized within this structure. Electron microscopic analyses of the LC region indicated that axon terminals originating from either subregion were equally likely to contact noradrenergic neurons in the LC. Approximately 57% and 62% of BDA-labeled terminals originating from the medial (n=150) or lateral (n=150) aspects of the ventral medulla, respectively, formed heterogeneous synaptic contacts (i.e., inhibitory- and excitatory-type) with dendrites containing TH. It is well known that the PGi is a functionally diverse region that is involved in sensory integration, autonomic regulation and pain modulation. It is also known that LC efferents are spatially organized with respect to their postsynaptic targets. Taken together, our findings that subdivisions of the ventral medulla topographically and monosynaptically innervate the LC suggest that regionally specific PGi neurons target subsets of LC neurons with efferent targets that may possess analogous functional correlates., (Copyright 1998 Elsevier Science B.V.)

The caudal ventrolateral medulla (CVL) contains neurons that are vasodepressor and are a critical component of the baroreceptor reflex pathway. While electrophysiological studies suggest that CVL neurons are intercalated in the baroreceptor pathway between the nucleus tractus solitarius (NTS) and the rostral ventrolateral medulla (RVL), there is no direct evidence for this projection. Therefore, we identified CVL neurons that project to RVL by retrogradely labelling them with wheat germ agglutinin-apo-horseradish peroxidase conjugated to colloidal gold (WAHG) injected into the RVL. Retrogradely labelled neurons were seen in previously identified vasodepressor areas of the rostral CVL that are critical for the baroreceptor reflex. Double labelling for WAHG and tyrosine hydroxylase (TH) immunocytochemistry indicated that CVL neurons that project to the RVL (CVL --> RVL neurons) are distinct from the noradrenergic neurons of the A1 cell group. To establish the presence of a direct projection from the NTS to CVL --> RVL neurons, the retrograde tracer WAHG was pressure injected into the RVL and the anterograde tracer biocytin was iontophoresed into the NTS of anesthetized rats. After 4-6 h, anesthetized rats were perfused transcardially with 3.75% acrolein in 2% paraformaldehyde and sections through the CVL were processed for both markers. By light microscopy, numerous biocytin-labelled varicose processes overlapped neurons containing WAHG in the CVL. By electron microscopy, biocytin was found in myelinated and unmyelinated axons and in axon terminals (0.9 + 0.02 microns) that contained primarily small clear vesicles. These terminals formed predominantly asymmetric synapses on large (1.5-6.0 microns in diameter) dendrites within the CVL. Some of the post-synaptic perikarya and large dendrites contained WAHG associated with lysosomes and multivesicular bodies, indicating that they belong to neurons which project to the RVL. We conclude that CVL --> RVL neurons are (a) distinct from A1 noradrenergic cells; (b) receive direct synaptic contacts from NTS efferent terminals; (c) are potently and monosynaptically excited (asymmetric synapses) by NTS efferent terminals. These data support the hypothesis that CVL neurons are intercalated between the NTS and the RVL in the baroreceptor reflex pathway.

Expt. 1 showed that electrical stimulation of either the main dorsal or ventral branch of the subdiaphragmatic vagus could produce inhibition of the nociceptive tail-flick reflex in lightly anesthetized rats. The antinociception produced by electrical stimulation of the dorsal subdiaphragmatic vagus was eliminated by resection of the right cervical vagus, but relatively unaffected by resection of the left cervical vagus. The opposite effects for cervical vagal resection were obtained with electrical stimulation of the ventral branch of the subdiaphragmatic vagus. These results indicate that the antinociceptive effects of subdiaphragmatic vagal stimulation are mediated via uncrossed afferents traveling in the cervical vagus to activate an inhibitory spinopetal system. These findings are consistent with the established anatomy of vagal afferents. Expt. 2 showed that degeneration of the dorsal subdiaphragmatic vagus did not alter the threshold intensity of right cervical vagal stimulation necessary to produce inhibition of the tail-flick reflex. These results demonstrate that the antinociceptive effects of cervical vagal stimulation are primarily due to activation of the cardiopulmonary component of the nerve, rather than the subdiaphragmatic component. The second experiment also demonstrated that the subdiaphragmatic branch of the vagus can be selectively degenerated with ricin while leaving the cervical branch intact, even though the cell bodies of both sets of afferents are located within the nodose ganglion. These data are discussed in terms of vagal afferents and their role in the modulation of nociceptive transmission.

Electrical stimulation of afferents of the right cervical vagus inhibited the tail-flick reflex elicited by noxious heat in barbiturate-anesthetized rats. This inhibitory effect was eliminated in rats receiving local anesthetic blockade of either the nucleus tractus solitarii (NTS), the lateral reticular nuclei, the nucleus raphe magnus-medullary reticular formation, or nucleus raphe obscurus regions of the medulla. Similarly, the vasodepressor and bradycardic effects of vagal stimulation were either attenuated or eliminated by local anesthetic blockade of these regions. Microinjection of the non-specific glutamate antagonist gamma-D-glutamylglycine (DGG) into the NTS region also eliminated vagally evoked inhibition of the tail-flick reflex, hypotension, and bradycardia. Conversely, microinjection of glutamate into the NTS region resulted in inhibition of the tail-flick reflex, hypotension, and bradycardia. These findings with DGG and glutamate are consistent with the view that glutamate serves as a neurotransmitter of the primary vagal afferents mediating these antinociceptive and cardiovascular responses. These results are discussed in terms of vagal afferent influences on somatosensory, somatomotor, and cardiovascular function.
