• The strategies of co-cultivation for metabolite production have been discovered. • The recent advancements and underlying mechanisms of co-cultivation were proposed. • The interactive cues and metabolic potential of co-cultivation were investigated. Increasing evidence shows that microbial synthesis plays an important role in producing high value-added products. However, microbial monoculture generally hampers metabolites production and limits scalability due to the increased metabolic burden on the host strain. In contrast, co-culture is a more flexible approach to improve the environmental adaptability and reduce the overall metabolic burden. The well-defined co-culturing microbial consortia can tap their metabolic potential to obtain yet-to-be discovered and pre-existing metabolites. This review focuses on the use of a co-culture strategy and its underlying mechanisms to enhance the production of products. Notably, the significance of comprehending the microbial interactions, diverse communication modes, genetic information, and modular co-culture involved in co-culture systems were highlighted. Furthermore, it addresses the current challenges and outlines potential future directions for microbial co-culture. Simultaneous biodegradation of multiple micropollutantslike polycyclic aromatic hydrocarbons (PAHs) and phthalates (PAEs) by microbial consortia remain unclear. Here, four distinct bacterial consortia capable of degrading PAHs and PAEs were domesticated from sludge and its composts. PAH-degrading consortium HS and PAE-degrading consortium EC2 displayed the highest degradation efficiencies for PAHs (37 %-99 %) and PAEs (98 %-99 %), respectively, being significantly higher than those of individual member strains. Consortia HS and EC2 could simultaneously degrade both PAHs and PAEs. Remarkably, a synthetic consortium Syn by co-culturing consortia HS and EC2 demonstrated proficient simultaneous biodegradation for both PAHs (65 %-98 %) and PAEs (91 %-97 %). These consortia changed their community structure with enriching pollutant-degrading genera and extracellular polymeric substance contents to promote simultaneous biodegradation of multiple pollutants. Moreover, consortium Syn significantly enhanced degradation of both PAHs and PAEs in soil and sludge. Bioconversion of methane into bacterioruberin is a promising way to address biomanufacturing substrate costs and greenhouse gas emissions but has not been achieved yet. Herein, this study aimed to upcycle methane to bacterioruberin by microbial consortia. The microbial consortia consist of Methylomonas and Methylophilus capable of synthesizing carotenoids from methane was firstly enriched from paddy soil. Through this microbial community, methane was successfully converted into C50 bacterioruberin for the first time. The bioconversion process was then optimized by the response surface methodology. Finally, the methane-derived bacterioruberin reached a record yield of 280.88 ± 2.94 μg/g dry cell weight. This study presents a cost-effective and eco-friendly approach for producing long-chain carotenoids from methane, offering a significant advancement in the direct conversion of greenhouse gases into value-added products. The substantial discharge of ferroferric oxide nanoparticles (Fe 3 O 4 NPs) into sewage threatens the survival of functional microorganisms in wastewater treatment. This study elucidated responses of anaerobic ammonium oxidation (anammox) consortia to inhibition from high Fe 3 O 4 NPs concentration and recovery mechanisms. The nitrogen removal efficiency decreased by 20.3 % and recovered after 55 days under 1000 mg/L Fe 3 O 4 NPs concentration. Toxicity was attributed to reactive oxygen species (ROS) production. The excessive ROS damaged membrane integrity, nitrogen metabolism, and DNA synthesis, resulting in the inhibition of anammox bacteria activity. However, recovery mechanisms of anammox consortia activity were activated in response to 1000 mg/L Fe 3 O 4 NPs. The increase of heme oxygenase-1, thioredoxin, and nicotinamide adenine dinucleotide-quinone oxidoreductase genes alleviated oxidative stress. Furthermore, the activation of metabolic processes associated with membrane and DNA repair promoted recovery of anammox bacteria activity. The potential of hydrolytic enzyme cocktail obtained from designed bacterial consortium WSh-1 comprising Bacillus subtilis CRN 16, Paenibacillus dendritiformis CRN 18, Niallia circulans CRN 24, Serratia marscens CRN 29, and Streptomyces sp. CRN 30, was investigated for maximum saccharification. Activity was further enhanced to 1.01 U/ml from 0.82 U/ml by supplementing growth medium with biotin and cellobiose as a cofactor and inducer. Through kinetic analysis, the enzyme cocktail showed a high wheat straw affinity with Michaelis-Menten constant (Km) of 0.68 µmol/L and a deconstruction rate (Vmax) of 4.5 U/ml/min. The statistical optimization of critical parameters increased saccharification to 89 %. The optimized process in a 5-L lab-scale bioreactor yielded 501 mg/g of reducing sugar from NaOH-pretreated wheat straw. Lastly, genomic insights revealed unique abundant oligosaccharide deconstruction enzymes with the most diverse CAZyme profile. The impact and mechanism of fluoranthene (Flr), a typical polycyclic aromatic hydrocarbon highly detected in sludge, on alkaline fermentation for volatile fatty acids (VFAs) recovery and antibiotic resistance genes (ARGs) transfer were studied. The results demonstrated that VFAs production increased from 2189 to 4272 mg COD/L with a simultaneous reduction of ARGs with Flr. The hydrolytic enzymes and genes related to glucose and amino acid metabolism were provoked. Also, Flr benefited for the enrichment of hydrolytic-acidifying consortia (i.e., Parabacteroides and Alkalibaculum) while reduced VFAs consumers (i.e., Rubrivivax) and ARGs potential hosts (i.e., Rubrivivax and Pseudomonas). Metagenomic analysis indicated that the genes related to cell wall synthesis, biofilm formation and substrate transporters to maintain high VFAs-producer activities were upregulated. Moreover, cell functions of efflux pump and Type IV secretion system were suppressed to inhibit ARGs proliferation. In response to the persistent expansion of global resource demands, considerable attention has been directed toward the synthetic microbial consortia (SMC) within the domain of microbial engineering, aiming to address the sustainable management and valorization of biomass wastes. This comprehensive review systematically encapsulates the most recent advancements in research and technological applications concerning the utilization of SMC for biomass waste treatment. The construction strategies of SMC are briefly outlined, and the diverse applications of SMC in biomass wastes treatment are explored, with particular emphasis on its potential advantages in waste degradation, hazardous substances control, and high value-added products conversion. Bioaugmentation shows promise in mitigating ammonia-induced microbial inhibition in anaerobic digestion processes. However, the advanced technical requirements and high costs associated with pure strain cultivation, as well as the time-consuming and labor-intensive process of domesticating consortia, present challenges for industrial applications. Herein, the efficacy of bioaugmentation with nondomesticated mixed microbial consortia was evaluated, which resulted in a significant methane production improvement of 5.6%-11.7% and 10.3%-13.5% under total ammonia nitrogen concentrations of 2.0 and 4.9 g-N/L, respectively. Microbial analysis revealed that at high ammonium levels, the bioaugmented culture facilitated a transition in the methanogenic pathway from acetoclastic to hydrogenotrophic by regulating symbiotic relationships between propionate- and acetate-oxidizing bacteria and methanogens. Consortium type and dose applied were identified as crucial factors determining bioaugmentation effectiveness. Chinese distillers' grains (CDGs) have low fermentation efficiency due to the presence of lignocellulosic components, such as rice husk. In this study, a microbial consortium synthesized was used based on the "functional complementarity" principle to produce lignocellulolytic crude enzyme. The crude enzyme was used to hydrolyze CDGs. After enzymatic hydrolysis, lignocellulose was damaged to varying degrees and the crystallinity decreased. Subsequently, the feed protein was produced using yeast through two pathways. The results showed that the crude enzyme produced by the microbial consortium (comprising Trichoderma reesei, Aspergillus niger, and Penicillium) exhibited excellent enzymatic efficiency, yielding 27.88%, 19.64%, and 10.88% of reducing sugar, cellulose, and hemicellulose. The true protein content of CDGs increased by 53.49% and 48.35% through the first and second pathways, respectively. Notably, the second pathway demonstrated higher economic benefits to produce feed protein. Composting generates odorous gases, including ammonia (NH 3 ), hydrogen sulfide (H 2 S), and volatile organic compounds (VOCs). The Biological Trickling Filter (BTF) is effective for odor treatment, but it may have limitations with hydrophobic VOCs. In this study, a strain of Bacillus subtilis with ammonia-reducing ability, a strain of Bacillus cereus with desulfurization ability and a strain of Schizophyllum commune with the ability to degrade dimethyl disulfide were isolated and screened. The three strains were combined to create synthetic microbial consortia for enhancing odor treatment in the BTF. Compared to the activated sludge control, the BTF with synthetic microbial consortia removed 92.43% ammonia, 92.75% hydrogen sulfide. Furthermore, it demonstrated a significant improvement in the removal rates of p-methyl mercaptan, methyl sulfide, and dimethyl disulfide. Highlight • Successful enrichment of a novel microbial consortium for keratin valorization. • The consortium displays a high keratinolytic activity and reproducibility. • Residual keratin has low nutrient content, limiting further bioconversion. • The bioconversion process favors releasing of low sulphur-containing protein. Abstract Keratin refers to a group of insoluble and recalcitrant protein materials. Slaughterhouses produce large amount of keratinous byproducts, which are either disposed or poorly valorized through costly thermochemical processes for animal feed formulation. Learning from nature, keratinolytic microbial consortia stand as a cost-efficient and environmental friendly way to valorize this recalcitrant resource. Directed selection was applied to enrich soil-born microbial consortia, using sequential batch cultivations in keratin medium, while measuring enzymes activity and monitoring consortia compositions via 16S rRNA gene amplicon sequencing. A promising microbial consortium KMCG6, featuring mainly members of Bacteroidetes and Proteobacteria, was obtained. It possessed keratinolytic activity with <25% residual substrate remaining, which also displayed a high degradation reproducibility level after long-term cryopreservation. The presence of 2,4-dichlorophenoxyacetic acid (2,4-D), an organochlorine herbicide, in the environment has raised public concern as it poses hazard to both humans and the ecosystem. Three potential strains having the capability to degrade 2,4-D were isolated from on site agricultural soil and identified as Arthrobacter sp. SVMIICT25, Sphingomonas sp. SVMIICT11 and Stenotrophomonas sp. SVMIICT13. Over 12 days of incubation, 81-90% of 100 mg/L of 2,4-D degradation was observed at 2% inoculum. A shorter lag phase with 80% of degradation efficiency was observed within 5 days when the inoculum size was increased to 10%. Six microbial consortia were prepared by combining the isolates along with in-house strains, Bacillus sp. and Pseudomonas sp. Consortia R3 (Arthrobacter sp. + Sphingomonas sp.), operated with 10% of inoculum, showed 85-90% degradation within 4 days and 98-100% in 9 days. In the biorefinery concept renewable feedstocks are converted to a multitude of value-added compounds irrespective of seasonal or other variations of the complex biomass substrates. Conceptionally, this can be realized by specialized single microbial strains or by co-culturing various strain combinations. In the latter approach strains for substrate conversion and for product formation can be combined. This study addressed the construction of binary microbial consortia based on starch- and sucrose-based production of l- lysine and derived value-added compounds. A commensalism-based synthetic consortium for l- lysine production from sucrose was developed combining an l- lysine auxotrophic, naturally sucrose-negative E. coli strain with a C. glutamicum strain able to produce l -lysine that secretes fructose when grown with sucrose due to deletion of the fructose importer gene ptsF . Mutualistic synthetic consortia with an l- lysine auxotrophic, α-amylase secreting E. coli strain and naturally amylase-negative C. glutamicum strains was implemented for production of valuable fine chemicals from starch. To acquire microbial consortia with effectively precedent degradation of lignin, samples obtained from rotten trunks, rotten stumps and soil near it were screened and isolated after generations of subculture. The dynamic change illustrated that their community structures were affected by pH and tended to be stable after 6 days’ cultivation. The desired one, named DM-1, was gained through successive cultivation for over 5 generations, whose high selectivity in lignin degradation was observed within 16 days’ cultivation (SV = 2.78). Meanwhile, a remarkable reduction in the fiber crystallinity of tree trimmings (10.35%) resulted from the bio-degradation of DM-1, displayed a greater exposure of cellulose by selective removal of lignin. The diversity analysis of DM-1 was investigated by PCR amplification and 16S rDNA sequencing, indicated that mesorhizobium, cellulosimicrobium, pandoraea, achromobacter and stenotrophomones were the predominant genera. The performance, microbial community and functional genes of a sequencing batch biofilm reactor (SBBR) were investigated in treating mariculture wastewater under different aerobic/anoxic alternation number. The removal efficiency of chemical oxygen demand (COD) and NH 4 + -N kept at 95.66 ± 1.83 % and 90.28 ± 2.42 % under aerobic/anoxic alternation number between 1 and 4. The total nitrogen (TN) removal efficiency gradually decreased from 94.45 ± 1.12 % to 83.06 ± 1.25 % with the increase of aerobic/anoxic alternative number from 1 to 4. The nitrification rates and their corresponding enzymatic activities increased slightly with the increase of aerobic/anoxic alternation number, whereas the denitrifying process had the contrary results. The variation of aerobic/anoxic alternation number obviously affected the microbial diversity and abundance. The microbial network structure and keystone taxa were different under different aerobic/anoxic alternation number. Microbial biomass and lipid production with mixed-culture of Rhodotorula glutinis and Chlorella vulgaris using acetate as sole carbon source was investigated. Synergistic effect of mixed-culture using 20 g/L acetate significantly promoted cell growth and acetate utilization efficiency. Increasing the proportion of algae in co-culture was beneficial for biomass and lipid accumulation and the optimal ratio of yeast/algae was 1:2. Light exposure further enhanced biomass and lipid titer with 6.9 g/L biomass and 2.6 g/L lipid (38.3 % lipid content) obtained in a 5L bioreactor. The results of lipid classes and fatty acid profiles moreover indicated that more neutral lipids and linolenic acid were synthesized in mixed-culture under light exposure condition, suggesting the great potential in applications of biofuels production. Bioaugmentation using microbial consortia is helpful in some anaerobic digestion (AD) systems, but accelerated acidification to produce methane has not been performed effectively with corn stalks and cow dung. In this study, the thermophilic microbial consortia MC1 was inoculated into a complex open environment (unsterilized and sterilized systems) to evaluate the feasibility of bioaugmentation to improve acidification efficiency. The results indicated that MC1 itself degraded lignocellulose efficiently, and accumulated more organic acids within 3 days. Similar trends were also observed in the unsterilized system, where the hemicellulose degradation rate and organic acid concentrations increased significantly by two-fold and 20.1% (P < 0.05), respectively, and clearly reduced the loss of product. Microbial composition did not change obviously after inoculating MC1, but the abundance of members of MC1, such as Bacillus and Clostridium , increased clearly on day 3. In this study, the production of lactic acid from food waste in industrially relevant conditions was investigated. Laboratory assays were first performed in batch conditions to determine the suitable operational parameters for an efficient lactic acid production. The use of compost as inoculum, the regulation of temperature at 35 °C and pH at 5 enhanced the development of Lactobacillus sp. resulting in the production of 70 g/L of lactic acid with a selectivity of 89% over the other carboxylic acids. Those parameters were then applied at pilot scale in successive fed-batch fermentations. The subsequent high concentration (68 g/L), yield (0.38 g/gTS) and selectivity (77%) in lactic acid demonstrated the applicability of the process. To integrate the process into a complete value chain, fermentation residues were then converted into biogas through anaerobic digestion. Synthetic microbial consortia provide a versatile and efficient platform for biochemicals production through the labor division. Especially, microbial communities composed of phototrophs and heterotrophs offer a promising alternative, as they can directly convert carbon dioxide (CO 2 ) into chemicals. Within this system, photoautotrophic microbes can convert CO 2 into organic carbon for microbial growth and metabolites synthesis by the heterotrophic partners. In return, heterotrophs can provide additional CO 2 to support the growth of photoautotrophic microbes. However, the unmatched growing conditions, low stability and production efficiency of synthetic microbial consortia hinder their further applications. Thus, design and construction of mutualistic and stable synthetic light-driven microbial consortia are urgently needed. In this review, the progress of synthetic light-driven microbial consortia for chemicals production was comprehensively summarized. In addition, space-efficient synthetic light-driven microbial consortia in hydrogel system were reviewed. Nanobubbles have the potential to curtail the loss of oxygen during activated sludge aeration due to their extensive surface areas and lack of buoyance in solution. In this study, nanobubble aeration was explored as a novel approach to enhance aerobic activated sludge treatment and benchmarked against coarse bubble aeration at the lab scale. Nanobubble aerated activated sludge reactors achieved greater dissolved oxygen levels at faster rates. Higher soluble chemical oxygen demand removal by 10% was observed when compared to coarse bubble aeration with the same amount of air. The activated sludge produced compact sludge yielding easier waste sludge for subsequent sludge handling. The samples showed fewer filamentous bacteria with a lower relative abundance of floc forming Corynebacterium, Pseudomonas, and Zoogloea in the sludge. Abstract: Microbes bound to a wheat bran/red wood powder/diatomaceous earth carrier were used as inoculants for a biotrickling filter (BTF) for treating gases contaminated with a mixture of benzene, toluene, and o-xylene (BTo-X). An overall removal efficiency of more than 87.9% was achieved after a start-up period of as low as 4days. At BTo-X loading rates (LRs) below 60.0g/m3 h, the BTF’s performance was similar for EBRTs of 90, 60, 45 and 30s with an elimination capacity (EC) almost approaching the LR; stable REs above 91.3% for benzene and toluene and above 82.8% for o-xylene were achieved. A maximum EC of 97.7g/m3 h was obtained at inlet load of 146.4g/m3 h. The mass ratio of carbon dioxide produced to the BTo-X removed was approximately 2.62, which confirmed complete degradation of BTo-X. The results demonstrate that microbes bound to a solid carrier can be an alternative to traditional liquid inoculums applied in BTFs and highlight their potential applicability to BTF technologies. Abstract: This paper presents the co-production of hydrogen and methane from cornstalks by a two- or three-stage anaerobic fermentation process augmented with effective artificial microbial community. Two-stage fermentation by using the anaerobic sludge and DGGE analysis showed that effective and stable strains should be introduced into the system. We introduced Enterobacter aerogens or Clostridium paraputrificum into the hydrogen stage, and C. paraputrificum was proven to be more effective. In the three-stage process consisting of the improved hydrolysis, hydrogen and methane production stages, the highest soluble sugars (0.482kg/kg cornstalks) were obtained after the introduction of Clostridium thermocellum in the hydrolysis stage, under the thermophilic (55°C) and acidic (pH 5.0) conditions. Hydrolysates from 1kg of cornstalks could produce 2.61mol (63.7l) hydrogen by augmentation with C. paraputrificum and 4.69mol (114.6l) methane by anaerobic granular sludge, corresponding to 54.1% energy recovery. The carboxylate platform employs a diverse microbial consortium of anaerobes in which the methanogens are inhibited. Nearly all biomass components are digested to a mixture of C1–C8 monocarboxylic acids and their corresponding salts. The methane-arrested anaerobic digestion proceeds readily without needing to sterilize biomass or equipment. It accepts a wide range of feedstocks (e.g., agricultural residues, municipal solid waste, sewage sludge, animal manure, food waste, algae, and energy crops), and produces high product yields. This review highlights several important aspects of the platform, including its thermodynamic underpinnings, influences of inoculum source and operating conditions on product formation, and downstream chemical processes that convert the carboxylates to hydrocarbon fuels and oxygenated chemicals.
