This study evaluated the compostability of rice straw as the main feedstock (75% in dry weight), supplemented with three different nitrogen-rich wastes, namely food waste (FW), dairy manure (DM), and sewage sludge (SS). Organic matter (OM) degradation, maturity and fertility of the end-product, and bacterial community structure during the composting processes were compared. All composting processes generated mature end-product within 51 days. Notably, FW addition was more effective to accelerate rice straw OM degradation and significantly improved end-product fertility with a high yield of Chinese cabbage. The succession of the bacterial community was accelerated with FW supplementation. Genera Geobacillus, Chryseolinea, and Blastocatella were significantly enriched during the composting of rice straw with FW supplementation. Finally, temperature, total nitrogen, moisture, pH, and total carbon were the key factors affecting microorganisms. This study provides a promising alternative method to enhance the disposal of larger amounts of rice straw in a shorter time.

This study evaluated the feasibility, system stability, and microbial community succession of thermophilic semi-continuous composting of kitchen waste (KW). The results revealed that treatment performance was stable at a 10 % feeding ratio, with an organic matter (OM) degradation efficiency of 81.5 % and seed germination index (GI) of 50.0 %. Moreover, the OM degradation efficiency and GI were improved to 83.4 % and 70.0 %, respectively, by maintaining an optimal compost moisture content (50-60 %). However, feeding ratios of ≥ 20 % caused deterioration of the composter system owing to OM overloading. Microbial community analysis revealed that Firmicutes, Actinobacteria, Chloroflexi, Proteobacteria, and Gemmatimonadetes were dominant. Additionally, moisture regulation significantly increased the Proteobacteria abundance by 57.1 % and reduced the Actinobacteria abundance by 57.8 %. Moreover, network analysis indicated that the bacterial community stability and positive interactions between genera were enhanced by moisture regulation. This information provides a useful reference for practical KW composting treatment in the semi-continuous mode.

Quorum sensing (QS) and quorum quenching (QQ) are two antagonistic processes coexisting in various bacterial communities in bioreactors, e.g., activated sludge for biological wastewater treatment. Although QS signal molecules are detected in activated sludge reactors and known to affect sludge properties and reactor performance, there has been no direct evidence to prove the endogenous existence of QQ effects in activated sludge. In this study, for the first time, acyl homoserine lactones-degrading enzymatic activity, a typical QQ effect, was discovered in activated sludge and found to considerably affect the QS detection results. The coexistence of QS and QQ bacteria in activated sludge was further confirmed by bacterial screening and denaturing gradient gel electrophoresis analysis. The method developed in this study could also be used to evaluate QQ activities in bioreactors, and a possible way is provided to tune bioreactor performance through balancing the QS and QQ processes.

The angiotensin-I-converting enzyme (ACE) inhibitory oligopeptide-enriched hydrolysates from Acetes chinensis by treatment with the protease from Bacillus sp. SM98011 were produced at pilot scale (100 L) and plant scale (1000 L). The pilot and plant scaled hydrolysate products almost had the same properties as that at laboratory scale. Spray-drying had little effect on the peptide composition and bioactivity of the hydrolysates. The plant scaled hydrolysates were used to study its blood pressure-depressing effect in vivo. It caused reduce of 18.3-38.6 mmHg of the blood pressure of spontaneously hypertensive rats in dose-dependent manner in the range of 100-1,200 mg/kg/day. Histopathologic study showed that the pathologic changes of heart and brain in SHRs got obvious alleviation after treatment of the hydrolysates.

Marine organisms are potentially an untapped source of drugs and value-added food production. Currently, Acetes chinensis is an underutilized shrimp species with low commercial value from the Bo Hai Gulf of China. In this paper, the shrimp was digested by a crude protease from Bacillus sp. SM98011 and filtered through a 3 kDa ultrafiltration membrane. Biological functions of the hydrolysate and ultrafiltrate were then assayed. The analyses showed that 41% of the oligopeptides in the ultrafiltrate had a molecular mass lower than 3 kDa. The antioxidant activities of the hydrolysate and ultrafiltrate were determined through the scavenger activity of the hydroxyl radical, with inhibitions of 42.38% and 67.95%, respectively. The hydrolysate and ultrafiltrate also had good Angiotensin-I-converting enzyme (ACE) inhibitory activity, with IC50 values of 0.98 mg/ml and 0.22 mg/ml, respectively. In addition, Chitin and chitosan were recovered from the hydrolytic sediment using a much smaller volume of strong acids and bases than is normally needed. With this method, we have shown that A. chinensis can be utilized to generate a high value-added product, and have revealed its hidden potential in the production of functional foods and ACE inhibitory peptides.

The optimal conditions for extraction of astragalosides III and IV (AGs III and IV) in Radix Astragali by negative pressure cavitation-accelerated enzyme pretreatment were studied on the basis of a Box-Behnken design and response surface methodology. Experimental results showed that negative pressure, amount of enzyme and incubation temperature were the main factors governing the enzyme pretreatment of Radix Astragali. The optimum parameters were obtained as follows: negative pressure -0.08 Mpa, amount of enzyme 1.48% (w/w of materials) and incubation temperature 45 degrees C. Under the optimal conditions, the maximal extraction yields of AGs III and IV were 0.103 and 0.325 mg/g, which were 41.67% and 65.31% increased as compared to those without enzyme pretreatment, respectively. The effect of negative pressure cavitation and enzyme pretreatment on the structural changes of plant cells was observed by scanning electron microscopy. In conclusion, negative pressure cavitation-accelerated enzyme pretreatment was proved to be environment-friendly and economical, and could be used in secondary metabolites production.

Trichoderma sp. K9301 secreting endoxylanases with an activity of 2836 U/g (dry weight) was screened for XOs production. Two acidic beta-endoxylanases EX1 (30.1 kDa) and EX2 (20.1 kDa) were purified from crude extract of the strain K9301 in solid fermentation. Action modes of EX1 and EX2 towards XOs showed similar hydrolysis characters to endoxylanases belonging to glycosyl hydrolase family 10 and 11, respectively. EX1 exhibited better affinity but lower hydrolytic efficiency than EX2 to xylans from beechwood, birchwood, and oat-spelt. They had synergistic action on xylan hydrolysis. The optimum condition to prepare XOs from corncobs was obtained as 10 mg/ml corncob xylan incubated with 10 U/mg crude enzymes at 50 degrees C for 3 h. The yield of XOs reached 43.3%, and only a little amount of xylose (3.1%) was simultaneously produced, suggesting the good potential of strain K9301 in XOs production.

Twelve kinds of marine protein materials, including fish, shrimp, seashell, algae and seafood wastes were selected for the hydrolysis using four different proteases. The IC 50 values for angiotensin-converting enzyme (ACE) inhibitory activity of 48 hydrolysates were rapidly determined by capillary electrophoresis (CE). The values ranged from 0.17 to 501.7 mg/ml, and were affected by both the marine protein resources and the selected proteases. Hydrolysates of the lowest IC 50 values were from shrimp ( Acetes chinensis ), shark meat, mackerel bone, Polysiphonia urceolata and Spirulina platensis , indicating these five kinds of marine food proteins contained beneficial materials for the production of ACE inhibitory peptides by proteolysis. The hydrolysates obtained using proteases Protamex and SM98011 had lower IC 50 values, showing these two proteases were superior to others. The CE method achieved the same sensitivity as the high performance liquid chromatography (HPLC) method. However, the CE method was faster and, as a result, more economical. Therefore, CE had potential for rapid screening of marine protein hydrolysates enriched in ACE inhibitory peptides.
