Membrane traffic mediated by a soluble N-ethylmaleimide sensitive factor attachment protein receptor (SNARE) complex contributes to plant growth and development. However, the functional significance of SNAREs involved in cell wall deposition and seedling development has not been sufficiently explored. In this study, we explored the roles of R-SNAREs VAMP721 (At1g04750) and VAMP722 (At2g33120) in seedling growth of Arabidopsis thaliana by histochemical staining, fluorescence labeling, and analyzing mutant phenotypes. Our results show a massive intracellular accumulation of cellulose and callose, and an abnormal deposition of callose at the expanding cell plate in vamp721vamp722 root cells compared with the wild type. Particularly, ectopic lignin accumulation was also observed in vamp721vamp722 root cells. The alteration of cell wall components was confirmed using Fourier transform infrared analysis. Plasma membrane integrity and cell viability were disturbed in the vamp721vamp722 seedling. Morphological observation shows that vamp721vamp722 mutations impaired development of roots, hypocotyl, cotyledon, and true leaf, and inhibited lateral root formation. Confocal images reveal that green fluorescent protein-tagged VAMP721 and VAMP722 showed a similar expression pattern and were expressed throughout all cells and tissues examined, including root and shoot apical meristems and cells of hypocotyls, cotyledons, and true leaves. Taken together, our results suggest that membrane traffic mediated by VAMP721 and VAMP722 is involved in seedling growth in A. thaliana.

To study the biological function of activity of bcl complex (ABC1) proteins in Lepidium apetalum Willd., genes encoding ABC1 family proteins were identified from the seed transcriptome. The sequence most closely related to germination at a low temperature was selected and gene expressions in response to low temperature stress further studied. The results show that 21 ABC1 genes were expressed in seeds germinating at the low temperature: 4 genes were upregulated, 6 were downregulated, and 11 were not significantly different from controls. The results of fluorescence quantification of the low temperature stress on the seedlings of 7-d-old L. apetalum showed that seven genes were up-regulated, six genes were down-regulated, and eight genes had no significant difference. Real-time quantitative PCR results show that under the low temperature stress, the expression of the LaAbc1-3 gene increased, but its expression decreased after some time. The expression of this gene increased again after removing the low temperature stress. The expression of LaAbc1-21 gene in L. apetalum seedlings showed a trend of decreasing first and then increasing. The LaAbc1-3 gene was insensitive to salt stress. Expression of the LaAbc1-21 gene was significantly up-regulated during the salt stress. Under osmotic stress, the expression of the LaAbc1-3 gene was down-regulated, and the expression was negatively correlated with polyethylene glycol (PEG-6000) concentration. Under the PEG-6000 treatment, the expression of the LaAbc1-21 gene was significantly up-regulated, and the expression was positively correlated with concentration. These results provide a basis for further analysis of the role of the ABC1 genes in the stress resistance of L. apetalum. [ABSTRACT FROM AUTHOR]

The roles of ethylene (ET) or salicylic acid (SA) in plant response to low temperature (LT, 5 °C) have been implicated. However, the combined effect of ET- and SA-signaling on plant growth and metabolism under LT remains to be evaluated. In this study, we comparatively analyzed the response of Arabidopsis ethylene insensitive (ein) 2-1 (an ET insensitive mutant), nonexprressor of pathogenesis relative (npr)1-1 (an SA insensitive mutant) and double mutant ein2-1/npr1-1 plants to LT. The results show that a LT of 5 °C induced plant growth retardation to a less degree in ein2-1, an intermediate degree in npr1-1, but a much larger in ein2-1/npr1-1 compared to the wild-type (WT) plants. The LT susceptibility of the ein2-1/npr1-1 plants was correlated to a lower net photosynthetic rate and proline content, and a higher content of H2O2 and malondialdehyde and electrolyte leakage relative to the WT plants. Lower activities of superoxide dismutase, peroxidase, and catalase, as well as a lower glutathione content and a ratio of its reduced form to its oxidized form were also observed in the double mutant plants as compared with the WT plants. However, at normal conditions (23 °C), all the tested physiological and biochemical parameters were comparable between the ein2-1/npr1-1 and WT plants, and plant growth was even better in the double mutant than in the WT plants. On the contrary, most of the above-mentioned parameters were advantageous in the ein2-1 and npr1-1 plants over the WT plants under the LT conditions. These data suggest that a parallel function or physiological redundancy of nonexpressor of pathogenesis relative 1 and ethylene insensitive 2 existed in the Arabidopsis plant response to the LT. On the other hand, an interaction between ET- and SA-signaling occurred during this process.

Phenylalanine ammonia lyase (PAL) is the first enzyme in the phenylpropanoid pathway. In this study, we describe the molecular characteristics of four PAL genes (MaPAL1, MaPAL2, MaPAL3, and MaPAL4) cloned from banana (Musa acuminata L. AAA group, cv. Cavendish) using a rapid amplification of cDNA ends and real time quantitative PCR. The predicted molecular masses of corresponding PALs ranged from 70.2 to 77.3 kDa and their isoelectric points were acidic. At the amino acid level, they shared a high sequence similarity with PALs in the banana DH-Pahang (AA group) genome. Phylogenetic analysis shows that the deduced amino acid sequences of MaPALs had also a high similarity with PALs of other plant species. Expression analysis by semi-quantitative reverse transcriptase PCR reveals that these genes were differentially expressed in various tissues. Enzyme activity of PAL and expression of MaPALs in bananas were induced after infection with Fusarium oxysporum f. sp cubense Tropical Race 4. Our findings suggest that MaPALs play important roles in banana resistance to F. oxysporum.
