International audience; Endogenous electric fields drive many essential functions relating to cell proliferation, motion, differentiation and tissue development. They are usually mimicked in vitro by using electrochemical systems to apply direct current or voltage stimuli to cell cultures. The many studies devoted to this topic have given rise to a wide variety of experimental systems, whose results are often difficult to compare. Here, these systems are analysed from an electrochemical standpoint to help harmonize protocols and facilitate optimal understanding of the data produced. The theoretical analysis of single-electrode systems shows the necessity of measuring the Nernst potential of the electrode and of discussing the results on this basis rather than using the value of the potential gradient. The paper then emphasizes the great complexity that can arise when high cell voltage is applied to a single electrode, because of the possible occurrence of anode and cathode sites. An analysis of two-electrode systems leads to the advice to change experimental practices by applying current instead of voltage. It also suggests that the values of electric fields reported so far may have been considerably overestimated in macro-sized devices. It would consequently be wise to revisit this area by testing considerably lower electric field values.

Recently, bacteriophage particles have started to be applied as a new biomaterial for developing sensing platforms. They can be used as both a recognition element or/and as building blocks, template/scaffold. In this paper, we studied a bacteriophage selected through phage-display technology. The chosen bacteriophage acted as a building block for creating a carbon nanofiber-based electrode and as a new receptor/binding element that recognizes C-reactive protein (CRP) - one of the markers of inflammatory processes in the human body. The binding efficiency of the selected phage towards CRP is two orders of magnitude higher than in the wild type. We demonstrate that the phage-based sensor is selective against other proteins. Finally, we show that layer-by-layer methods are suitable for deposition of negatively charged phages (wild or CRP-binding) with positively charged carbon nanofibers for electrode surface modification. A three-layered electrode was successfully used for molecular recognition of CRP, and the molecular interactions were studied using electrochemical, biological, and optical methods, including microscopic and spectroscopic analyses.

In this paper we studied the performance of bioanodes under different experimental conditions using polarization curves and impedance spectroscopy. We have identified that the large capacitances of up to 1 mF·cm− 2 for graphite anodes have their origin in the nature of the carbonaceous electrode, rather than the microbial culture. In some cases, the separate contributions of charge transfer and diffusion resistance were clearly visible, while in other cases their contribution was masked by the high capacitance of 1 mF·cm− 2. The impedance data were analyzed using the basic Randles model to analyze ohmic, charge transfer and diffusion resistances. Increasing buffer concentration from 0 to 50 mM and increasing pH from 6 to 8 resulted in decreased charge transfer and diffusion resistances; lowest values being 144 Ω·cm2 and 34 Ω·cm2, respectively. At acetate concentrations below 1 mM, current generation was limited by acetate. We show a linear relationship between inverse charge transfer resistance at potentials close to open circuit and saturation (maximum) current, associated to the Butler–Volmer relationship that needs further exploration. The extension of the photon correlation spectroscopy (PCS) in multiple scattering regime, so-called diffusing wave spectroscopy (DWS) was employed to the study of blood samples. Multiple scattered light from a helium-neon (He-Ne) laser beam incident on the blood samples was detected by a photomultiplier, and both the temporal autocorrelation intensity functions g 2(tau) and power spectra S(omega) were measured by a spectrum analyzer. The potentials of using DWS for the qualitative and quantitative determination of the structural characteristics of the blood elements were studied experimentally. The experimental studies made, permits the use of DWS for blood cells monitoring in a multiple scattering regime. This paper describes our initial attempts at applying DWS to the study of the discrete blood samples of both healthy donors and patients with the cardiac ischemia. The subsequent experiments provide a verification of DWS of blood cells shape monitoring under multiple scattering.
