Abstract

The activity of recombinant murine G9a toward lysine 9 of histone H3 was investigated. It was suggested that the sequence context of the modified residue affects G9a activity and the modification in the proximal amino acids influences methylation.

Abstract

A recombinant purified SUV39H1 is used for studying the substrate specificity and steady-state kinetic analysis with peptides representing the un- or dimethylated lysine 9 histone H3 tail or full length human recombinant H3 (rH3). The results have shown that SUV39H1 utilizes both the chromodomain and the SET domain for catalysis.
