Grooming is a common readout in multiple rat models of neuropsychiatric diseases. It is usually associated with distress and negative emotionality, but also with emotional de-arousal after stress. These seemingly conflicting interpretations may result from specific grooming sequences appearing at different arousal levels and during distinct phases of the stress response. To further explore this hypothesis, we analyzed how distinct stressors affect grooming syntaxis and kinetics. To that end, we explored the independent and interacting effects of foot shocks, corticosterone (CORT), and novelty on exploratory activity, grooming, and ultrasonic vocalizations (USVs) in an open-field test (OF). Wistar rats were intraperitoneally injected either with vehicle or CORT, placed in a chamber where half of them were foot-shocked and then assessed in the OF. The next day, animals were re-exposed to the shock chamber and then tested in the OF without receiving any treatment. On day 1, foot shocks and –to a less extent CORT– increased freezing and inhibited rearing in the chamber, but only foot shocks increased distress USVs. In the OF, both treatments suppressed complex grooming, with foot-shocks also inhibiting exploration and CORT marginally reducing rearing. On day 2, foot-shocked rats showed conditioned fear when re-exposed to the chamber. When tested in the OF, foot-shocked and CORT-treated animals still showed low levels of complex grooming, with the former group also showing increased distress USVs. In this study, all different stressors inhibited complex grooming, suggesting an inverse association between these grooming subtypes and negative emotionality. • Foot shocks (FS) and CORT reduced exploratory behaviors in the Open Field (OF). • Both stressors reduced complex grooming but only FS induced 22-kHz calls in the OF. • Foot-shocked rats showed conditioned fear when re-exposed to the FS chamber 24 h later. • Conditioned fear reduced complex grooming and evoked 22-kHz calls in the OF. • All stressors inhibited complex grooming while novelty habituation promoted it. Abstract Spatial orientation is a ubiquitous feature of animal behavior. Environmental and self-movement cues are sources of information used to maintain spatial orientation. The literature has typically focused on differences between mice and rats using environmental cues to guide movement. The current study uses the organization of exploratory behavior under dark conditions to investigate species differences in self-movement cue processing. Mouse and rat exploratory behavior was recorded under dark conditions on a circular table without walls. The resulting movements were segmented in progressions (movement ≥ 3 cm/s) and stops (movement < 3 cm/s). Mice exhibited longer travel distances, faster progression peak speeds, and weaker tendency to scale progression peak speeds to Euclidean distances relative to rats. In contrast, similar levels of performance were observed on measures (progression path circuity, change in heading, stability of stopping behavior) sensitive to vestibular pathology. These results are consistent with species differences in a variety of performance variables; however, self-movement cue based spatial orientation did not differentiate between mice and rats. Highlights • People prefer to find out early about uncertain gambles. • People will even pay for useless information about upcoming rewards. • Reward magnitude does not impact willingness to pay for the information. • Humans are similar to other animals in their willingness to sacrifice rewards for information. Abstract Humans are inherently curious creatures, continuously seeking out information about future outcomes. Such advance information is often valuable, potentially allowing people to select better courses of action. In non-human animals, this drive for information can be so strong that they forego food or water to find out a few seconds earlier whether an uncertain option will provide a reward. Here, we assess whether people will exhibit a similar sub-optimal preference for advance information. Participants played a card-flipping task where they were probabilistically rewarded based on the pattern of 3 cards that were revealed after a 5-s delay. During this delay, participants could instead pay a cost to find out the next card's identity immediately. This choice to find out early did not influence the eventual outcome. Participants preferred to find out early about 80% of the time when the information was free; they were even willing to incur an expense to get advance information about the eventual outcome. The expected magnitude of the outcome, however, had little impact on the likelihood of finding out early. Anxiety is a protective behavior when animals face aversive conditions. The open field test (OFT) is used to assess the spatio-temporal dynamics of exploration, in which both homebase formation and recognition of environmental cues may reflect habituation to unfamiliar conditions. Because emotional- and affective-like states influence exploration patterns and mnemonic aspects, we aimed to verify whether the exploratory behaviors of two zebrafish populations showing distinct baselines of anxiety differ in two OFT sessions. Firstly, we assessed the baseline anxiety-like responses of short fin (SF) and leopard (LEO) populations using the novel tank test (NTT) and light-dark test (LDT) in 6-min trials. Fish were later tested in two consecutive days in the OFT, where the spatial occupancy and exploratory profile were analyzed for 30 min. In general, LEO showed pronounced diving behavior and scototaxis in the NTT and LDT, respectively, in which an "anxiety index" corroborated their exacerbated anxiety-like behavior. In the OFT, the SF population spent less time to establish the homebase in the 1st trial, while only LEO showed a markedly reduction in the latency to homebase formation in the 2nd trial. Both locomotion and homebase-related activities were decreased in the 2nd trial, in which animals also revealed increased occupancy in the center area of the apparatus. Moreover, we verified a significant percentage of homebase conservation for both populations, while only SF showed reduced the number of trips and increased the average length of trips. Principal component analyses revealed that distinct factors accounted for total variances between trials for each population tested. While homebase exploration was reduced in the 2nd trial for SF, an increased occupancy in the center area and hypolocomotion were the main factors that contribute to the effects observed in LEO during re-exposure to the OFT. Behavioral responses vary between individuals and may be repeated in different contexts over time. When a behavioral response set is linked and present regardless of the context, it characterizes a behavioral syndrome. By evaluating how bold and shy (profiles related to risk-taking) individuals perform about exploration and anxiety, we can predict relationships of behavioral syndromes and better understand how different axis of personality is formed. Here we classified the profiles by risk-taking and evaluated their exploration behavior in the open field test. In this context, the two groups showed significant differences in thigmotaxis behavior: bold individuals habituate faster and show decreased thigmotaxis (less anxiety), while shy ones are less prone to leave the security of the side areas of the open tank and present higher anxiety. Individuals within animal groups may differ in personality and degree of familiarity raising the question of how this influences their social interactions. In Iberian magpies Cyanopica cooki , a portion of first-year males engage in cooperative behaviours and dispersal, allowing addressing this question. In this study, we first investigate the relationship between colony familiarity (native versus foreign) and reproductive status (breeding versus helping) of males during 21 years. Secondly, we measure the exploratory behaviour and monitor reproductive status of a sample of individuals with different colony familiarity during 2 years. Long-term monitoring revealed that foreign individuals were more likely breeders. The analysis on the subset of individuals in which exploratory behaviour was measured revealed a mediatory effect of exploratory behaviour in the association between colony familiarity and helping behaviour. Specifically, among foreign individuals, higher explorative males were more frequently involved in helping behaviour than lower explorative ones. Conversely, among native males, breeders were more explorative than helpers. It has been shown that rearranging the spatial properties of a familiar environment consistently elicits a positive response in rats directed toward the source of novelty. Previous studies have been conducted under red light or darkness. The purpose of this study was to test the effect of rearranging the spatial properties of a familiar environment in conjunction with a change in lighting conditions. The results have shown specific effects of the light presence and its intensity on different behavioral measures. We propose that this study provides a basis for hypothesizing a two-way mechanism of the behavioral response to light regulation in rats. The first is based on ON/OFF states. This level may be related to fundamental, evolutionarily early, emergent components of behavioral antipredator adaptations. Another level of behavioral regulation involves mechanisms sensitive to light intensity. These appear to be involved in the regulation of more advanced behavioral acts, such as exploratory responses. Nicotine, an important component of cigarette smoke, is a neurotransmitter that contributes to stress, depression and anxiety in smokers. In rodents, it increases anxiety and reduces exploratory behaviours. However, so far, the measurements of exploratory behaviour in rodents have only been semi-quantitative and lacking in sufficient detail to characterise the temporal effect of smoking cessation. As rodents, such as mice and rats, primarily use whiskers to explore their environment, we studied the effect of 3 months smoking with 1 and 2 weeks smoking cessation on whisker movements in mice, using high-speed video camera footage and image analysis. Both protraction and retraction whisker velocities were increased in smoking mice (p < 0.001) and returned to normal following just one week of smoking cessation. In addition, locomotion speeds were decreased in smoking mice, and returned to normal following smoking cessation. Lung function was also impacted by smoking and remained impaired even following smoking cessation. We suggest that the increased whisker velocities in the smoking mice reflect reduced exploration and impeded tactile performance. The increase in whisker velocity with smoking, and its reduction following smoking cessation, also lends support to acetylcholine being involved in awareness, attention and alertness pathways. A major challenge in behavior and evolutionary ecology is to understand the evolution and maintenance of animal personality. Theory suggests that females can benefit by choosing a high-quality mate, but largely ignores the potential interaction between male and female personality during mate choice. Here, we examined the influence of exploration on mate choice by captive female Java sparrows (Lonchura oryzivora). Females preferred high exploratory males as mates rather than choosing mates according to their own exploration, and thus showed no assortative mating. To be able to reproduce, animals need to survive and interact with an ever changing environment. Therefore, they create a cognitive representation of that environment, from which they derive expectancies regarding current and future events. These expected events are compared continuously with information gathered through exploration, to guide behaviour and update the existing representation. When a moderate discrepancy between perceived and expected events is detected, exploration is employed to update the internal representation so as to alter the expectancy and make it match the perceived event. When the discrepancy is relatively large, exploration is inhibited, and animals will try to alter the perceived event utilizing aggression or fear. The largest discrepancies are associated with a tendency to flee. When an exploratory, fear, or aggressive behaviour pattern proofs to be the optimal solution for a particular discrepancy, the response will become conditioned to events that previously preceded the occurrence of that discrepancy. When primary needs are relatively low, animals will actively look for or create moderately violated expectancies in order to learn about objects, behaviour patterns, and the environment. In those situations, exploratory tendencies will summate with ongoing behaviour and, when all primary needs are satiated, may even be performed exclusively. Stress encompasses reactions to stimuli that promote negative and positive effects on cognitive functions, such as learning and memory processes. Herein, we investigate the effect of restraint stress on learning, memory, anxiety levels and locomotor activity of male and female mice. We used the plus-maze discriminative avoidance task (PMDAT), a behavioral task based on the innate exploratory response of rodents to new environments. Moreover, this task is used to simultaneously evaluate learning, memory, anxiety-like behavior and locomotor activity. Male and female mice were tested after repeated daily restraint stress (4 h/day for 3 days). The results showed stress-induced deficits on aversive memory retrieval only in female mice, suggesting a sexual dimorphism on memory acquisition. Furthermore, stressed females exhibited increased anxiety-like behavior and decreased exploratory behavior. Plasma corticosterone levels were similarly increased by restraint stress in both sexes, suggesting that the behavioral outcome was not related to hormonal secretion. Our findings corroborate previous studies, showing a sexually dimorphic effect of restraint stress on cognition. The existence of animal personality is well-established across a wide range of species, with the majority of evidence for this being obtained from individuals held in captivity. However, there has been little work assessing the influence of commonly-measured personality traits on fitness, which is pertinent when the genetic basis of personality is considered. We measured whether the reproductive behaviour and success of zebra finches in a captive mixed-sex aviary environment was influenced by an aspect of their personality, their exploratory behaviour in a single-sex social aviary. We found that more exploratory males made a greater number of breeding attempts and raised more nestlings than less exploratory males. These results were not confounded by extra-pair paternity, which was not related to personality, or by the individuals that did not initiate any reproductive attempts at all. Our work provides evidence that attributes of personality may influence the degree to which individuals cope with, and thrive in a captive environment and this should be accounted for in both experimental design and the interpretation of results. Abstract: This study aimed to develop and characterize a novel (standard) open field test adapted for larval zebrafish. We also developed and characterized a variant of the same assay consisting of a colour-enriched open field; this was used to assess the impact of environmental complexity on patterns of exploratory behaviours as well to determine natural colour preference/avoidance. We report the following main findings: (1) zebrafish larvae display characteristic patterns of exploratory behaviours in the standard open field, such as thigmotaxis/centre avoidance; (2) environmental complexity (i.e. presence of colours) differentially affects patterns of exploratory behaviours and greatly attenuates natural zone preference; (3) larvae displayed the ability to discriminate colours. As reported previously in adult zebrafish, larvae showed avoidance towards blue and black; however, in contrast to the reported adult behaviour, larvae displayed avoidance towards red. Avoidance towards yellow and preference for green and orange are shown for the first time, (4) compared to standard open field tests, exposure to the colour-enriched open field resulted in an enhanced expression of anxiety-like behaviours. To conclude, we not only developed and adapted a traditional rodent behavioural assay that serves as a gold standard in preclinical drug screening, but we also provide a version of the same test that affords the possibility to investigate the impact of environmental stress on behaviour in larval zebrafish while representing the first test for assessment of natural colour preference/avoidance in larval zebrafish. Abstract: Impacts of individual personality on group distribution were investigated using sheep (Ovis aries) as a model. In an indoor exploration test, individuals who visited <4 (out of 6) objects in a novel environment were classified as ‘shy’ (n =10), and those who visited 5 or 6 objects were classified as ‘bold’ (n =10). Nine weeks later, using a series of groups (n =40) of either 5 shy or 5 bold sheep, we measured distribution at pasture and responses to disturbance and the approach of a human handler. When grazing undisturbed, the mean nearest neighbour distance and spread (minimum convex hull area) of shy groups were less than those of bold groups, with shy individuals moving towards one another more often. Shy groups explored a smaller area than bold groups. When disturbed, shy sheep were more likely to stop grazing and move closer together. Shy sheep kept further away from the handler and moved faster when driven. The results demonstrate a link between personality and group distribution, suggesting that our ‘shy’ and ‘bold’ individuals may occupy different positions on the shy–bold continuum documented for other species. Abstract: Individual variation in exploratory behaviour has been demonstrated in a diverse array of animal species. Understanding the evolutionary antecedents and ecological consequences of this variation is an active research area within animal behaviour. Here we investigate whether different exploration styles exhibited by black-capped chickadees (Poecile atricapillus) in a novel environment are related to how quickly these birds learn an acoustic discrimination task. We found that birds that readily enter a novel environment learn an acoustic discrimination task faster than birds that do not readily enter a novel environment. This result contrasts with previous work suggesting no correlation between exploration style and learning a spatial or associative task in great tits (Parus major), a close relative of the black-capped chickadee. Abstract: The present study examined the acute behavioral responses of pigeons to separation from conspecifics and exposure to an unfamiliar environment (UE). The effects of (1) repeated exposure to the UE; (2) visual isolation from surroundings, or saline injections; and (3) diazepam treatment (i.p., 0.25, 0.75, 2.5 or 7.5mg/kg) before the trial were also examined. UE exposure evoked intense ballistic head movements (peeping), gradually replaced with angular head movements (AHM), both associated with immobility of the trunk and legs. These behaviors failed to habituate after three trials (7-day intertrial intervals). Visual isolation from the surroundings and saline injection prior to exposure to the UE increased the AHM and reduced peeping. Doses of diazepam (0.25 and 0.75mg/kg) that have demonstrated anti-conflict effects in other tests did not affect the behavioral responses to the UE. Diazepam at 2.5 and 7.5mg/kg doses consistently increased time spent in immobility. These data suggest that peeping, although expressed in potentially threatening or harmful situations appears not to be a fear-motivated behavior or, alternatively, this specific behavioral response is not diazepam sensitive. Abstract: Berlyne [Berlyne, D.E., 1950. Novelty and curiosity as determinants of exploratory behaviour. Brit. J. Psychol. 41, 68–80] first illustrated that rats prefer to explore novel objects over ones with which they have had previous experience. Recently, variants on this novel object recognition (NOR) task have become widely popular and have been employed in numerous neuroscience and behavioral pharmacological studies investigating memory processes. Given this popularity, a thorough understanding of the various behavioral processes involved in novelty reaction and preference is essential. The current study compared the effects of spaced and massed initial stimulus exposures upon later object exploration and novel stimulus preference in Long-Evans rats. Results illustrated that a distributed initial stimulus familiarization procedure promoted greater novel object preference than did a massed procedure, and suggest that the novel object recognition task is sensitive to spacing effects in a similar fashion to more traditional learning paradigms. Abstract: The exploratory behaviour of laboratory rodents is of interest within a number of areas of behavioural pharmacology. However, how best to measure exploratory behaviour in rodents remains a contentious issue. Many unconditioned tests, such as the open field, potentially confound general locomotor activity with exploration. The hole-board apparatus appears to avoid this confound, as head-dipping into holes in the floor is assumed to be a valid measure of the subject''s attraction towards novelty (neophilia). This study aimed to investigate whether head-dipping should be considered a valid measure of neophilia by comparing performance of adult male and female Lister hooded rats on the hole-board task (a) over repeated sessions and (b) when novel objects were absent or present underneath the holes. The results show that head-dipping initially decreased across repeated exposures, while time spent in the aversive central area increased. No change in head-dipping was seen in response to objects being placed underneath the holes. Rather than being a measure of neophilia, these results support the hypothesis that head-dipping represents an escape response, which declines as the subject becomes less fearful. Abstract: The present work investigated whether the number of rats housed in a cage affects exploration of an elevated plus-maze. Male Wistar-derived rats were kept 1, 2, 3, 4, 6, 8, 12, 16, or 24 to same size cages either for 1 or 14 days and tested in the elevated plus-maze. Rats kept 6 to a cage were arbitrarily considered controls because this is the housing condition adopted in many laboratories, ours included. In comparison to controls, 1-day housed rats kept 1, 2, 16, and 24 to a cage decreased the percentage of entries into the open arms. Similar decreases were also found in the time spent in the open arms, the only exception being the group with rats kept 16 to a cage which failed to show significant differences from the control group. Fourteen-day housed rats kept 1, 2, 16, or 24 to a cage decreased the percentage of entries and time spent in the open arms. We found plus-maze exploration to be similar in groups in which rats were kept from 4 to 12 to a cage. The present data indicate that anxiogenic effects resulting from under- and overcrowding should be taken into consideration in behavioral studies. [Copyright &y& Elsevier]

Abstract: The effect of isolation on exploratory behavior has been shown to differ depending on the developmental stages of male rats. However, there has been little systematic comparison of the frequencies and the patterns of exploratory behavior across the developmental stages. The present study assessed the frequencies of exploration using the emergence test and exploratory patterns in the open-field test in three developmental stages of male rats: juvenile, post-puberty, and adult. A lower propensity for exploration was observed in rats isolated during the juvenile stage, as assessed by increased latency and decreased duration of exploratory behaviors compared to pair-reared rats, and this tendency was maintained in adulthood. Altered patterns of exploratory behavior were demonstrated both in rats isolated in adulthood, who showed an increased active pattern, and those pair-reared following puberty, who shifted to a more passive pattern. However, rats isolated during the juvenile stage did not change their exploratory patterns following puberty. These results suggest that the changes in the exploratory pattern, which can be observed in adulthood, are associated with the emergence of adult-like dominance relationships. Juvenile-isolated rats did not show these changes following puberty, suggesting the importance of social interaction as juveniles for the ontogenetic emergence of behavioral flexibility implicated in the regulation of exploratory patterns. The purpose of the present study was to investigate the response to novelty in Roman high- and low-avoidance rats under non-stressful conditions. To reduce fear, a procedure of repetitive placing in the experimental chamber consisting of start, screen, and tunnel zones was applied. Each animal was placed in the experimental chamber daily for a 6 min period. The first 11 sessions were the habituation sessions. In the 12th session, the novelty was introduced into the screen and tunnel zones. The subsequent two sessions were conducted under novelty conditions. Behavioral activities such as walking, object contacts, time spent in given zones, and entering the tunnels were measured. All the comparisons were made for two 3-min intervals, using a three-factor MANOVA, involving 2sex×2subline×83-mininterval. All subjects increased time spent in the tunnel zone, but RHA/Verh rats responded to a greater extent, especially the males. All subjects spent shorter times in the screen zone, but the RHA/Verh rats responded to a much greater extent. RHA/Verh rats, especially males responded with a substantial increase of time spent inside the tunnels. All subjects responded with an increased amount of object contacts. In general the RHA/Verh subjects showed a more pronounced response to novelty, as evidenced by a significant shift toward the tunnel zone. They spent more time in this zone than their RLA/Verh counterparts. Among the RLA/Verh rats, males tended to behave similarly to RHA/Verh rats, especially during the second 3 min interval of session “12.” The differences between the rat lines obtained in this study may be attributed to mechanisms specific to exploration, making them promising subjects to study the relationships between reactivity, novelty detection, adaptation, and environmental information processing. The elevated plus-maze test is usually run with a short edge surrounding the open arms in order to prevent the rats from falling. The present experiment investigated the role of transparent edges differing in heights: 1 (used as control), 5, 10, 20 and 40 cm, the latter the same height as the closed arm walls. Additionally, this 40-cm high transparent edge was also studied covered by white translucent or black opaque paper. The data show that the time spent in the open arms was significantly greater when the edge height was 5, 10 or 40 cm covered by the white or black paper. However, there were no differences from the 1-cm control edge when the height was 40 cm transparent. A similar effect was observed when entries in the open arms and total entries were analyzed. The facts that there were no differences when the open arms were surrounded by 1- or 40-cm transparent edges (which allow thigmotaxis) and that the same 40-cm edge caused increases in exploratory behavior when covered by papers indicate that vision triggers aversion to open spaces. Sexually dimorphic performance has been observed across humans and rodents in many spatial tasks. In general, these spatial tasks do not dissociate the use of environmental and self-movement cues. Previous work has demonstrated a role for self-movement cue processing in organizing open field behavior; however, these studies have not directly compared female and male movement characteristics. The current study examined the organization of open field behavior under dark conditions in female and male rats. Significant differences between female and male rats were observed in the location of stopping behavior relative to a cue and the topography exhibited during lateral movements. In contrast, no sex differences were observed on measures used to detect self-movement cue processing deficits. These results provide evidence that female and male rats are similar in their use of self-movement cues to organize open field behavior; however, other factors may be contributing to differences in performance. • We analyzed boldness and novelty response in chimangos from different urbanization levels. • Flight initiation distance was higher in rural than in more urbanized raptors. • Urban raptors were less neophobic and faster explorers than rural birds. • Both rural and urban raptors showed similar exploration times. • Only in rural raptors there was a correlation between neophobia and exploration. The study of traits that enable species to thrive in urban habitats is critical to a better understanding the evolution of urban ecosystems. Here, we examined variation in boldness, neophobia, and exploratory behavior in a generalist raptor, the Chimango Caracara, Milvago chimango , occurring in areas with different urbanization level. We also focused on the relationship between object exploration and neophobia in rural and urban population of this species. The results showed that birds from more urbanized habitats showed more tolerance to humans than rural raptors, without any effect of age and sex in this trait. Rural birds were also more neophobic and were slower to explore than urban raptors, though they dedicated a similar amount of time to exploring novel objects, indicating a non-correspondence between the speed to approach and contact these objects and the amount of exploration performed. Finally, we found a correlation between exploration speed and neophobia in rural birds, whereas for urban raptors this correlation was not observed. Our results show that urbanization not only influences the expression of risk-taking behaviors and novelty responses in the chimango, but can also modify the relationship between exploration and neophobia.
