Background and Aims: Plant regeneration by seeds is driven by a set of physiological traits, many of which show functional intraspecific variation along biogeographic gradients. In many species, germination phenology depends on a germination delay imposed by the need for post-dispersal embryo growth (a.k.a. morphological dormancy). Such growth occurs as a function of environmental temperatures and shows base, optimum and ceiling temperatures (i.e. cardinal temperatures or thermal thresholds). However, the biogeographical variation in such thresholds has not been tested., Methods: We used a thermal time approach and field experiments to assess intraspecific variation at the continental scale in the embryo growth thermal thresholds of the geophyte Conopodium majus (Apiaceae) across its distribution from the Iberian Peninsula to Scandinavia., Key Results: Thermal thresholds for embryo growth varied across the latitudinal gradient, with the estimated optimum temperatures between 2.5 and 5.2 ºC, ceiling temperatures between 12 and 20.5 ºC and base temperatures between -6.6 and -2.7 ºC. Germination in the field peaked in the months of January and February. The limiting factor for embryo growth was the ceiling temperature, which was negatively correlated with latitude and the bioclimatic environment of each population. In contrast, the optimal and base temperature were independent of local climate., Conclusions: These results indicate that thermal thresholds for embryo growth are functional ecophysiological traits that drive seed germination phenology and seed responses to soil climatic environment. Background and Aims: Orchid seeds are reputed to be short lived in dry, cold storage conditions, potentially limiting the use of conventional seed banks for long-term ex situ conservation. This work explores whether Cattleya seeds are long lived or not during conventional storage (predried to ~12 % relative humidity, then stored at -18 °C)., Methods: We explored the possible interaction of factors influencing seed lifespan in eight species of the genus Cattleya using physiological (germination and vigour), biochemical (gas chromatography), biophysical (differential scanning calorimetry) and morphometric methods. Seeds were desiccated to ~3 % moisture content and stored at -18 °C for more than a decade, and seed quality was measured via three in vitro germination techniques. Tetrazolium staining was also used to monitor seed viability during storage. The morphometric and germination data were subjected to ANOVA and cluster analysis, and seed lifespan was subjected to probit analysis., Key Results: Seeds of all Cattleya species were found to be desiccation tolerant, with predicted storage lifespans (P50y) of ~30 years for six species and much longer for two species. Cluster analysis showed that the three species with the longest-lived seeds had smaller (9-11 %) airspaces around the embryo. The post-storage germination method impacted the quality assessment; seeds equilibrated at room temperature for 24 h or in 10 % sucrose solution had improved germination, particularly for the seeds with the smallest embryos. Chromatography revealed that the seeds of all eight species were rich in linoleic acid, and differential scanning calorimetry identified a peak that might be auxiliary to selecting long-lived seeds., Conclusions: These findings show that not all orchids produce seeds that are short lived, and our trait analyses might help to strengthen prediction of seed longevity in diverse orchid species., (© The Author(s) 2024. Background and Aims: European ancient woodlands are subject to land use change, and the distribution of herbaceous understorey species may be threatened because of their poor ability to colonize isolated forest patches. The regeneration niche can determine the species assembly of a community, and seed germination traits may be important descriptors of this niche., Methods: We analysed ecological records for 208 herbaceous species regarded as indicators of ancient woodlands in Europe and, where possible, collated data on seed germination traits, reviewed plant regeneration strategies and measured seed internal morphology traits. The relationship between plant regeneration strategies and ecological requirements was explored for 57 species using ordination and classification analysis., Key Results: Three regeneration strategies were identified. Species growing in closed-canopy areas tend to have morphological seed dormancy, often requiring darkness and low temperatures for germination, and their shoots emerge in early spring, thus avoiding the competition for light from canopy species. These species are separated into two groups: autumn and late winter germinators. The third strategy is defined by open-forest plants with a preference for gaps, forest edges and riparian forests. They tend to have physiological seed dormancy and germinate in light and at higher temperatures, so their seedlings emerge in spring or summer., Conclusion: Seed germination traits are fundamental to which species are good or poor colonizers of the temperate forest understorey and could provide a finer explanation than adult plant traits of species distribution patterns. Background and Aims: Environmental temperature regulates plant regeneration via seed in several superimposed ways, and this complex regulation will be disrupted by climate change. The role of diurnally alternating temperatures (ΔT) in terminating dormancy will be a major factor in this disruption, as its effects on seed germination are immediate., Methods: The effect of ΔT on seed germination was modelled using two populations of the wetland sedge Carex diandra, one from a montane site and one from a subalpine site. A cardinal-temperature model was fitted to germination results obtained from a thermal gradient plate, and the model was used to simulate changes in germination under two possible future climate scenarios (RCP2·6 and RCP8·5, for representative concentration pathways) as defined by the Intergovernmental Panel on Climate Change., Key Results: Scenario RCP2·6 projected moderate increases in average temperatures and ΔT, whereas RCP8·5 projected greater warming and higher ΔT. Increasing ΔT decreased the base temperature for seed germination and the thermal time required for germination. The effect of higher ΔT together with the higher temperatures increased germination under both climate scenarios., Conclusions: Carex diandra germination is highly responsive to potential changes in ΔT, and thus this study highlights the role of ΔT in seed responses to climate change. Background and Aims Desert plants possess excellent water-conservation capacities to survive in extreme environments. Cuticular wax plays a pivotal role in reducing water loss through plant aerial surfaces. However, the role of cuticular wax in water retention by desert plants is poorly understood. Methods We investigated leaf epidermal morphology and wax composition of five desert shrubs from north-west China and characterized the wax morphology and composition for the typical xerophyte Zygophyllum xanthoxylum under salt, drought and heat treatments. Moreover, we examined leaf water loss and chlorophyll leaching of Z. xanthoxylum and analysed their relationships with wax composition under the above treatments. Key Results The leaf epidermis of Z. xanthoxylum was densely covered by cuticular wax, whereas the other four desert shrubs had trichomes or cuticular folds in addition to cuticular wax. The total amount of cuticular wax on leaves of Z. xanthoxylum and Ammopiptanthus mongolicus was significantly higher than that of the other three shrubs. Strikingly, C31 alkane, the most abundant component, composed >71 % of total alkanes in Z. xanthoxylum , which was higher than for the other four shrubs studied here. Salt, drought and heat treatments resulted in significant increases in the amount of cuticular wax. Of these treatments, the combined drought plus 45 °C treatment led to the largest increase (107 %) in the total amount of cuticular wax, attributable primarily to an increase of 122 % in C31 alkane. Moreover, the proportion of C31 alkane within total alkanes remained >75 % in all the above treatments. Notably, the water loss and chlorophyll leaching were reduced, which was negatively correlated with C31 alkane content. Conclusion Zygophyllum xanthoxylum could serve as a model desert plant for study of the function of cuticular wax in water retention because of its relatively uncomplicated leaf surface and because it accumulates C31 alkane massively to reduce cuticular permeability and resist abiotic stressors. Background and Aims Desiccation-tolerant vascular plants (DT plants) are able to tolerate the desiccation of their vegetative tissues; as a result, two untested paradigms can be found in the literature, despite contradictions to theoretical premises and empirical findings. First, it is widely accepted that DT plants form a convergent group of specialist plants to water deficit conditions. A derived paradigm is that DT plants are placed at the extreme end of stress tolerance. Here, we tested the hypotheses that DT plants (1) are in fact convergent specialists for water deficit conditions and (2) exhibit ecological strategies related to stress tolerance, conservative resource-use and survival. Methods We used biogeographical and functional-traits approaches to address the mentioned paradigms and assess the species' ecological strategies. For this, 27 DT plants were used and compared to 27 phylogenetically related desiccation-sensitive vascular plants (DS plants). Key Results We could not confirm either of the two hypotheses. We found that despite converging in desiccation tolerance, DT plants differ in relation to the conditions in which they occur and the ecological strategies they use to deal with water deficit. We found that some DT plants exhibit advantageous responses for higher growth and resource acquisition, which are suitable responses to cope with more productive conditions or with higher disturbance. We discuss that the ability to tolerate desiccation could compensate for a drought vulnerability promoted by higher investment in growth and bring advantages to deal with quick and pronounced variation of water, rather than to drought solely. Conclusions DT plants are not only selected by drought as an environmental constraint. The alternative functional designs could promote the diversity of ecological strategies, which preclude their convergence to the same resources and conditions. Background Plant seeds have many traits that influence ecological functions, ex situ conservation, restoration success and their sustainable use. Several seed traits are known to vary significantly between tropical and temperate regions. Here we present three additional traits for which existing data indicate differences between geographical zones. We discuss evidence for geographical bias in availability of data for these traits, as well as the negative consequences of this bias. Scope We reviewed the literature on seed desiccation sensitivity studies that compare predictive models to experimental data and show how a lack of data on populations and species from tropical regions could reduce the predictive power of global models. In addition, we compiled existing data on relative embryo size and post-dispersal embryo growth and found that relative embryo size was significantly larger, and embryo growth limited, in tropical species. The available data showed strong biases towards non-tropical species and certain families, indicating that these biases need to be corrected to perform truly global analyses. Furthermore, we argue that the low number of seed germination studies on tropical high-mountain species makes it difficult to compare across geographical regions and predict the effects of climate change in these highly specialized tropical ecosystems. In particular, we show that seed traits of geographically restricted páramo species have been studied less than those of more widely distributed species, with most publications unavailable in English or in the peer-reviewed literature. Conclusions The low availability of functional seed trait data from populations and species in the tropics can have negative consequences for macroecological studies, predictive models and their application to plant conservation. We propose that global analyses of seed traits with evidence for geographical variation prioritize generation of new data from tropical regions as well as multi-lingual searches of both the grey- and peer-reviewed literature in order to fill geographical and taxonomic gaps. Background and Aims Interactions between ecological factors and seed physiological responses during the establishment phase shape the distribution of plants. Yet, our understanding of the functions and evolution of early-life traits has been limited by the scarcity of large-scale datasets. Here, we tested the hypothesis that the germination niche of temperate plants is shaped by their climatic requirements and phylogenetic relatedness, using germination data sourced from a comprehensive seed conservation database of the European flora (ENSCOBASE). Methods We performed a phylogenetically informed Bayesian meta-analysis of primary data, considering 18 762 germination tests of 2418 species from laboratory experiments conducted across all European geographical regions. We tested for the interaction between species' climatic requirements and germination responses to experimental conditions including temperature, alternating temperature, light and dormancy-breaking treatments, while accounting for between-study variation related to seed sources and seed lot physiological status. Key Results Climate was a strong predictor of germination responses. In warm and seasonally dry climates the seed germination niche includes a cold-cued germination response and an inhibition determined by alternating temperature regimes and cold stratification, while in climates with high temperature seasonality opposite responses can be observed. Germination responses to scarification and light were related to seed mass but not to climate. We also found a significant phylogenetic signal in the response of seeds to experimental conditions, providing evidence that the germination niche is phylogenetically constrained. Nevertheless, phylogenetically distant lineages exhibited common germination responses under similar climates. Conclusion This is the first quantitative meta-analysis of the germination niche at a continental scale. Our findings showed that the germination niches of European plants exhibit evolutionary convergence mediated by strong pressures at the macroclimatic level. In addition, our methodological approach highlighted how large datasets generated by conservation seed banking can be valuable sources to address questions in plant macroecology and evolution. Background and Aims Seed germination is strongly influenced by environmental temperatures. With global temperatures predicted to rise, the timing of germination for thousands of plant species could change, leading to potential decreases in fitness and ecosystem-wide impacts. The thermogradient plate (TGP) is a powerful but underutilized research tool that tests germination under a broad range of constant and alternating temperatures, giving researchers the ability to predict germination characteristics using current and future climates. Previously, limitations surrounding experimental design and data analysis methods have discouraged its use in seed biology research. Methods Here, we have developed a freely available R script that uses TGP data to analyse seed germination responses to temperature. We illustrate this analysis framework using three example species: Wollemia nobilis , Callitris baileyi and Alectryon subdentatus. The script generates >40 germination indices including germination rates and final germination across each cell of the TGP. These indices are then used to populate generalized additive models and predict germination under current and future monthly maximum and minimum temperatures anywhere on the globe. Key Results In our study species, modelled data were highly correlated with observed data, allowing confident predictions of monthly germination patterns for current and future climates. Wollemia nobilis germinated across a broad range of temperatures and was relatively unaffected by predicted future temperatures. In contrast, C. baileyi and A. subdentatus showed strong seasonal temperature responses, and the timing for peak germination was predicted to shift seasonally under future temperatures. Conclusions Our experimental workflow is a leap forward in the analysis of TGP experiments, increasing its many potential benefits, thereby improving research predictions and providing substantial information to inform management and conservation of plant species globally. Background and Aims European mesic meadows are semi-natural open habitats of high biodiversity and an essential part of European landscapes. These species-rich communities can be a source of seed mixes for ecological restoration, urban greening and rewilding. However, limited knowledge of species germination traits is a bottleneck to the development of a competitive native seed industry. Here, we synthesize the seed ecology of mesic meadows. Methods We combined our own experimental data with data obtained from databases to create a combined dataset containing 2005 germination records of 90 plant species from 31 European countries. We performed a Bayesian meta-analysis of this dataset to test the seed germination response to environmental cues including scarification, stratification, temperature, alternating temperature and light. We also used multivariate ordination to check the relationship between seed traits (germination and morphology) and species ecological preferences, and to compare the seed ecology of mesic meadows with that of other herbaceous plant communities from the same geographic area. Key Results The seed ecology of mesic meadows is characterized by (1) high seed germinability when compared with other herbaceous plant communities; (2) low correspondence between seed traits and species ecological preferences; and (3) a deep phylogenetic separation between the two major families, Poaceae and Fabaceae. Poaceae produce many light seeds that respond to gap-detecting germination cues (alternating temperatures and light); Fabaceae produce fewer heavy seeds, which need scarification to break their physical dormancy. Conclusions High germinability of meadow seeds will reduce their capacity to form persistent seed banks, resulting in dispersal limitations to passive regeneration. For centuries, human activities have shaped the regeneration of meadows, leading to a loss of seed dormancy and decoupling seeds from seasonal cycles, as has been found in many domesticated species. The same anthropic processes that have shaped semi-natural mesic meadows have left them dependent on continued human intervention for their regeneration, highlighting the importance of active restoration via seed supply. Background and Aims The long-term conservation of seeds of plant genetic resources is of key importance for food security and preservation of agrobiodiversity. Nevertheless, there is scarce information available about seed longevity of many crops under germplasm bank conditions. Methods Through germination experiments as well as the analysis of historical monitoring data, we studied the decline in viability manifested by 1000 maize (Zea mays subsp. mays) seed accessions conserved for an average of 48 years at the CIMMYT germplasm bank, the largest maize seedbank in the world, under two cold storage conditions: an active (–3 °C; intended for seed distribution) and a base conservation chamber (–15 °C; for long-term conservation). Key Results Seed lots stored in the active chamber had a significantly lower and more variable seed germination, averaging 81.4 %, as compared with the seed lots conserved in the base chamber, averaging 92.1 %. The average seed viability detected in this study was higher in comparison with that found in other seed longevity studies on maize conserved under similar conditions. A significant difference was detected in seed germination and longevity estimates (e.g. p85 and p50) among accessions. Correlating seed longevity with seed traits and passport data, grain type showed the strongest correlation, with flint varieties being longer lived than floury and dent types. Conclusions The more rapid loss of seed viability detected in the active chamber suggests that the seed conservation approach, based on the storage of the same seed accessions in two chambers with different temperatures, might be counterproductive for overall long-term conservation and that base conditions should be applied in both. The significant differences detected in seed longevity among accessions underscores that different viability monitoring and regeneration intervals should be applied to groups of accessions showing different longevity profiles. Background and Aims The structure of plant communities, which is based on species abundance ratios, is closely linked to ecosystem functionality. Seed germination niche plays a major role in shaping plant communities, although it has often been neglected when explaining species coexistence. The aim of this work is to link the seed germination niche to community ecology, investigating how functional seed traits contribute to species coexistence. Methods Species selection was based on a database of 504 vegetation surveys from the Veneto coast (Italy). Through cluster analysis we identified the foredune community and selected all of its 19 plant species. By using the 'Phi coefficient' and frequency values, species were pooled in different categories (foundation species, accidental species of the semi-fixed dune and aliens), then the 19 species were grouped according to their germination responses to temperature and photoperiod through cluster analyses. For each germination cluster, we investigated germination trends against temperature and photoperiod by using generalized linear mixed models. Key Results We identified four germination strategies: (1) high germination under all tested conditions ('high-germinating'); (2) high germination at warm temperatures in the dark ('dark warm-cued'); (3) high germination at warm temperatures in the light ('light warm-cued'); and (4) low germination, regardless of conditions ('low-germinating'). Foredune foundation species showed a narrow germination niche, being 'low-germinating' or 'dark warm-cued'. Annual species of semi-fixed dunes were 'high-germinating', while alien species were the only members of the 'light warm-cued' cluster. Conclusions Our research suggests that different categories of species have dissimilar seed germination niches, which contributes to explaining their coexistence. Climatic events, such as rising temperature, could alter germination patterns, favouring seed regeneration of certain categories (i.e. alien and semi-fixed dune species) at the expense of others (i.e. foundation species, pivotal to ecosystem functioning), and hence potentially altering the plant community structure. Background and Aims Branching is an important mechanism of plant shape establishment and the direct consequence of axillary bud outgrowth. Recently, hydrogen peroxide (H2O2) metabolism, known to be involved in plant growth and development, has been proposed to contribute to axillary bud outgrowth. However, the involvement of H2O2 in this process remains unclear. Methods We analysed the content of H2O2 during bud outgrowth and characterized its catabolism, both at the transcriptional level and in terms of its enzymatic activities, using RT–qPCR and spectrophotometric methods, respectively. In addition, we used in vitro culture to characterize the effects of H2O2 application and the reduced glutathione (GSH) synthesis inhibitor l -buthionine sulfoximine (BSO) on bud outgrowth in relation to known molecular markers involved in this process. Key Results Quiescent buds displayed a high content of H2O2 that declined when bud outgrowth was initiated, as the consequence of an increase in the scavenging activity that is associated with glutathione pathways (ascorbate–glutathione cycle and glutathione biosynthesis); catalase did not appear to be implicated. Modification of bud redox state after the application of H2O2 or BSO prevented axillary bud outgrowth by repressing organogenesis and newly formed axis elongation. Hydrogen peroxide also repressed bud outgrowth-associated marker gene expression. Conclusions These results show that high levels of H2O2 in buds that are in a quiescent state prevents bud outgrowth. Induction of ascorbate–glutathione pathway scavenging activities results in a strong decrease in H2O2 content in buds, which finally allows bud outgrowth. [ABSTRACT FROM AUTHOR]

Background and Aims Understorey species in temperate deciduous woodlands such as wild daffodil (Narcissus pseudonarcissus) and common snowdrop (Galanthus nivalis) have complex dormancy: seeds that are shed in late spring require warm summer temperatures for embryo elongation and dormancy alleviation, but then cooler temperatures for germination in autumn. As seasons warm and tree canopies alter, how will different seasonal temperature sequences affect these complex dormancy responses? Methods The effect of different sequences of warmer (+5 °C), current or cooler (–5 °C) seasons (summer to spring) on seed germination patterns over seven successive seasons were investigated, with all sequences combined factorially to determine the consequences of differential seasonal temperature change for the temporal pattern of germination (and so seedling recruitment). Key Results Little (<1 %, G. nivalis) or no (N. pseudonarcissus) seed germination occurred during the first summer in any treatment. Germination of N. pseudonarcissus in the first autumn was considerable and greatest at the average (15 °C) temperature, irrespective of the preceding summer temperature; germination was also substantial in winter after a warmer autumn. Germination in G. nivalis was greatest in the warmest first autumn and influenced by preceding summer temperature (average > warmer > cooler); the majority of seeds that germinated over the whole study did so during the two autumns but also in year 2's cooler summer after a warm spring. Conclusions Warmer autumns and winters delay first autumn germination of N. pseudonarcissus to winter but advance it in G. nivalis ; overall, warming will deplete the soil seed bank of these species, making annual seed influx increasingly important for recruitment and persistence. This study provides a comprehensive account of the effects of temperature changes in different seasons on seed germination in these early spring-flowering geophytes and consequently informs how these and other temperate woodland species with complex seed dormancy may respond to future climate change. Background and Aims Oaks are the foundation and dominant tree species of most Mediterranean forests. As climate models predict dramatic changes in the Mediterranean basin, a better understanding of the ecophysiology of seed persistence and germination in oaks could help define their regeneration niches. Tunisian oaks occupy distinct geographical areas, which differ in their rainfall and temperature regimes, and are thus a valuable model to investigate relationships between seed traits and species ecological requirements. Methods Seed morphological traits, desiccation sensitivity level, lethal freezing temperature, embryonic axis and cotyledon sugar and lipid composition, and seed and acorn germination rates at various constant temperatures were measured in Quercus canariensis , Q. coccifera , Q. ilex and Q. suber , using seeds sampled in 22 Tunisian woodlands. Key Results Only faint differences were observed for desiccation sensitivity in the oak species studied. By contrast, the species differed significantly in sensitivity to freezing, germination rates at low temperature and base temperature. Quercus ilex and Q. canariensis , which occur at high elevations where frost events are frequent, showed the lowest freezing sensitivity. A significant correlation was found between hexose contents in the embryonic axis and freezing tolerance. Significant interspecific differences in the time for seeds to germinate and the time for the radicle to pierce the pericarp were observed. The ratio of pericarp mass to acorn mass differed significantly among the species and was negatively correlated with the acorn germination rate. Quercus coccifera , which is frequent in warm and arid environments, showed the highest acorn germination rate and synchrony. Conclusions Seed lethal temperature, seed germination time at low temperatures, the ratio of pericarp mass to acorn mass and the embryonic axis hexose content appeared to be key functional traits that may influence the geographical ranges and ecological requirements of Mediterranean oaks in Tunisia. Background and Aims Hydroperiod drives plant community composition in wetlands, resulting in distinct zonation patterns. Here, we explored the role of seed germination traits in shaping wetland community assembly along a hydroperiod gradient. Specifically, we tested the hypothesis that seeds of reed, mudflat, swamp, shallow- and deep-water communities only germinate under a specific set of environmental factors characterized by the community-specific optimal conditions for seedling survival and growth. Methods In a three-factorial experiment, we tested the seed germination response of 50 species typical for temperate wetlands of Europe to temperature fluctuations (constant vs. fluctuating temperature), illumination (light vs. darkness) and oxygen availability (aerobic vs. hypoxia). Phylogenetic principal component analysis, cluster analysis and phylogenetic linear regressions were used to confirm the community-specific seed germination niches. Key Results Our study revealed the presence of five distinct, community-specific seed germination niches that reflect adaptations made by the study communities to decreasing light intensity, temperature fluctuations and oxygen availability along the hydroperiod gradient. Light as a germination trigger was found to be important in mudflats, swamps and shallow water, whereas the seeds of reed and deep-water species were able to germinate in darkness. A fluctuating temperature is only required for seed germination in mudflat species. Germination of species in the communities at the higher end of the hydroperiod gradient (reed and mudflat) demonstrated a strict requirement for oxygen, whereas swamp, shallow- and deep-water species also germinated under hypoxia. Conclusions Our study supports the recent argument that the inclusion of seed germination traits in community ecology adds significant insights to community response to the abiotic and biotic environment. Furthermore, the close relationship between seed germination adaptations and community assembly could help reach a better understanding of the existing patterns of wetland plant distribution at local scales and wetland vegetation dynamics, as well as facilitate nature conservation measures and aquatic habitat restoration. Background and Aims Understanding variation in seed longevity, especially within closely related germplasm, will lead to better understanding of the molecular basis of this trait, which is particularly important for seed genebanks, but is also relevant to anyone handling seeds. We therefore set out to determine the relative seed longevity of diverse Indica rice accessions through storage experiments. Since antioxidants are purported to play a role in seed storability, the antioxidant activity and phenolic content of caryopses were determined. Methods Seeds of 299 Indica rice accessions harvested at 31, 38 and 45 d after heading (DAH) between March and May 2015 and differing in harvest moisture content (MC) were subsequently stored at 10.9 % MC and 45 °C. Samples were taken at regular intervals and sown for germination. Germination data were subjected to probit analysis and the resulting parameters that describe the loss of viability during storage were used for genome-wide association (GWA) analysis. Key Results The seed longevity parameters, K i [initial viability in normal equivalent deviates (NED)], − σ −1 (σ is the time for viability to fall by 1 NED in experimental storage) and p 50 [time for viability to fall to 50 % (0 NED)], varied considerably across the 299 Indica accessions. Seed longevity tended to increase as harvest MC decreased and to decrease as harvest MC increased. Eight major loci associated with seed longevity parameters were identified through GWA analysis. The favourable haplotypes on chromosomes 1, 3, 4, 9 and 11 enhanced p 50 by ratios of 0.22–1.86. Conclusions This is the first study to describe the extent of variation in σ within a species' variety group. A priori candidate genes selected based on rice genome annotation and gene network ontology databases suggested that the mechanisms conferring high seed longevity might be related to DNA repair and transcription, sugar metabolism, reactive oxygen species scavenging and embryonic/root development. Background and Aims Determining seed longevity by identifying chemical changes that precede, and may be linked to, seed mortality, is an important but difficult task. The standard assessment, germination proportion, reveals seed longevity by showing that germination proportion declines, but cannot be used to predict when germination will be significantly compromised. Assessment of molecular integrity, such as RNA integrity, may be more informative about changes in seed health that precede viability loss, and has been shown to be useful in soybean. Methods A collection of seeds stored at 5 °C and 35–50 % relative humidity for 1–30 years was used to test how germination proportion and RNA integrity are affected by storage time. Similarly, a collection of seeds stored at temperatures from −12 to +32 °C for 59 years was used to manipulate ageing rate. RNA integrity was calculated using total RNA extracted from one to five seeds per sample, analysed on an Agilent Bioanalyzer. Results Decreased RNA integrity was usually observed before viability loss. Correlation of RNA integrity with storage time or storage temperature was negative and significant for most species tested. Exceptions were watermelon, for which germination proportion and storage time were poorly correlated, and tomato, which showed electropherogram anomalies that affected RNA integrity number calculation. Temperature dependencies of ageing reactions were not significantly different across species or mode of detection. The overall correlation between germination proportion and RNA integrity, across all experiments, was positive and significant. Conclusions Changes in RNA integrity when ageing is asymptomatic can be used to predict onset of viability decline. RNA integrity appears to be a metric of seed ageing that is broadly applicable across species. Background and Aims Tropical understorey plant communities are highly diverse and characterized by variable resource availability, especially light. Plants in these competitive environments must carefully partition resources to ensure ecological and evolutionary success. One mechanism of effective resource partitioning is the optimization of functional traits to enhance competition in highly heterogeneous habitats. Here, we surveyed the ecophysiology of two early lineage vascular plant groups from a tropical forest understorey: Selaginella (a diverse lineage of lycophytes) and ferns. Methods In a lowland rain forest in Costa Rica, we measured a suite of functional traits from seven species of Selaginella and six fern species. We evaluated species microclimate and habitat; several photosynthetic parameters; carbon, nitrogen and phosphorus content; chlorophyll concentration; leaf mass per area (LMA); and stomatal size and density. We then compare these two plant lineages and search for relationships between key functional parameters that already exist on a global scale for angiosperms. Key Results Convergence of trait function filtered Selaginella species into different habitats, with species in heavily shaded environments having higher chlorophyll concentrations and lower light compensation points compared with open habitats. Alternatively, lower foliar nitrogen and higher stomatal densities were detected in species occupying these open habitats. Selaginella species had denser and smaller stomata, lower LMA and lower foliar nutrient content than ferns, revealing how these plant groups optimize ecophysiological function differently in tropical forest floors. Conclusions Our findings add key pieces of missing evidence to global explorations of trait patterns that define vascular plant form and function, which largely focus on seed plants. Broadly predictable functional trait relationships were detected across both Selaginella and ferns, similar to those of seed plants. However, evolutionary canalization of microphyll leaf development appears to have driven contrasting, yet successful, ecophysiological strategies for two coexisting lineages of extant homosporous vascular plants. Background and Aims The relationship between fruiting phenology and seed dispersal syndrome is widely recognized; however, the interaction of dormancy classes and plant life-history traits in relation to fruiting phenology and seed dispersal is understudied. Here we examined the relationship between fruiting season and seed dormancy and how this relationship is modulated by dormancy classes, dispersal syndromes, seed mass and seed moisture content in a Brazilian savanna (cerrado). Methods Dormancy classes (non-dormancy and physical, morphological, morphophysiological, physiological and physiophysical dormancy) of 34 cerrado species were experimentally determined. Their seed dispersal syndrome (autochory, anemochory, zoochory), dispersal season (rainy, dry, rainy-to-dry and dry-to-rainy transitions), seed mass and moisture contents, and the estimated germination date were also determined. Log-linear models were used to evaluate how dormancy and dormancy classes are related to dispersal season and syndrome. Key Results The proportions of dormant and non-dormant species were similar in cerrado. The communityestimated germination date was seasonal, occurring at the onset of rainy season. Overall, anemochorous nondormant species released seeds during the dry-to-rainy transition; autochorous physically dormant species dispersed seeds during the dry season and rainy-to-dry transition; zoochorous species dispersed non-dormant seeds during the dry and rainy seasons, while species with morphological, morphophysiological or physiological dormancy dispersed seeds in the transitional seasons. Seed mass differed among dispersal seasons and dormancy classes, but seed moisture content did not vary with dispersal syndrome, season or dormancy class. Conclusions The beginning of the rainy season was the most favourable period for seed germination in cerrado, and the germination phenology was controlled by both the timing of seed dispersal and seed dormancy. Dormancy class was influenced by dispersal syndrome and season. Moreover, dormancy avoided seed germination during the rainy-to-dry transition, independently of dispersal syndrome. The variability of dormancy classes with dispersal syndrome allowed animal-dispersed species to fruit all year round, but seeds germinated only during the rainy season. • Background and Aims Seed desiccation response plays an important role in plant regeneration ecology, and has significant implications for species conservation. The majority of seed plants produce desiccation-tolerant (orthodox) seeds, whilst comparatively few produce desiccation-sensitive (recalcitrant) seeds that are unable to survive dehydration, and which cannot be conserved in traditional seed banks. This study develops a set of models to predict seed desiccation response in unstudied species. • Methods Taxonomy, trait, location and climate data were compiled to form a global data set of 17 539 species. Three boosted regression trees models were then developed to predict species' seed desiccation responses based on habitat and trait information for the species, and the seed desiccation responses of close relatives (either members of the same genus, family or order, depending on the model). Ten-fold cross-validation was used to test model predictive success. The utility of the models was then demonstrated by predicting seed desiccation response for two floras: Ecuador, and Britain and Ireland. • Key Results The three models had varying success rates for identifying the desiccation-sensitive species: 89 % for the genus-level model, 79 % for the family-level model and 60 % for the order-level model. The most important predictor variables were the seed desiccation responses of a species' relatives, seed mass and annual precipitation. It is predicted that 10 % of seed plants from Ecuador and 1.2 % of those from Britain and Ireland produce desiccation-sensitive seeds. Due to data availability, prediction accuracy is likely to be higher for the British and Irish flora, where it is estimated that a desiccation-sensitive species had a 96.7 % chance of being correctly identified, compared with 80.8 % in the Ecuador flora. • Conclusions These models can utilize existing data to predict species' likely seed desiccation responses, providing a gap-filling tool for global studies of plant traits, as well as critical decision-making support for plant conservation activities. Background and Aims: Climate warming has major impacts on seed germination of several alpine species, hence on their regeneration capacity. Most studies have investigated the effects of warming after seed dispersal, and little is known about the effects a warmer parental environment may have on germination and dormancy of the seed progeny. Nevertheless, temperatures during seed development and maturation could alter the state of dormancy, affecting the timing of emergence and seedling survival. Here, the interplay between pre- and post-dispersal temperatures driving seed dormancy release and germination requirements of alpine plants were investigated. Methods: Three plant species inhabiting alpine snowbeds were exposed to an artificial warming treatment (i.e. +1·5 K) and to natural conditions in the field. Seeds produced were exposed to six different periods of cold stratification (0, 2, 4, 8, 12 and 20 weeks at 0 °C), followed by four incubation temperatures (5, 10, 15 and 20 °C) for germination testing. Key Results: A warmer parental environment produced either no or a significant increase in germination, depending on the duration of cold stratification, incubation temperatures and their interaction. In contrast, the speed of germination was less sensitive to changes in the parental environment. Moreover, the effects of warming appeared to be linked to the level of (physiological) seed dormancy, with deeper dormant species showing major changes in response to incubation temperatures and less dormant species in response to cold stratification periods. Conclusions: Plants developed under warmer climates will produce seeds with changed germination responses to temperature and/or cold stratification, but the extent of these changes across species could be driven by seed dormancy traits. Transgenerational plastic adjustments of seed germination and dormancy shown here may result from increased seed viability, reduced primary and secondary dormancy state, or both, and may play a crucial role in future plant adaptation to climate change. Background and Aims Adaptations allowing plants to cope with drying are particularly relevant in the light of predicted climate change. Dehydration tolerance (DhT, also dehydration-tolerant) is one such adaptation enabling tissue to survive substantial drying. A great deal of work has been conducted on highly DhT species. However, bryophytes showing less intense and variable DhT are understudied, despite the potential for these species to provide an informative link between highly tolerant and sensitive species. In this study, we tested the degree to which DhT varies across populations and the sexes of a species expected to exhibit a moderate DhT phenotype. Methods To test predicted patterns of tolerance we assessed DhT in males and females of Marchantia inflexa from two distinct habitat types that differ in water availability. Both common garden and field-collected tissue was subjected to drying assays at multiple intensities and recovery was monitored by chlorophyll florescence. Verification studies were conducted to confirm the level of dehydration, the rate of drying and the associated changes in photosynthetic physiology. Key Results We confirmed our expectation that M. inflexa is able to tolerate moderate dehydration. We also found that females exhibited higher DhT than males, but populations did not differ in DhT when cultured in a common garden. However, field-collected samples exhibited differences in DhT corresponding to environmental dryness, suggesting plasticity in DhT. Conclusions By studying a less extreme DhT phenotype we gained insight into how more sensitive (yet still tolerant) organisms cope with dehydration. Additionally, the identified sex-specific variation in DhT may explain ecological patterns such as female-biased sex ratios. • Background and Aims Despite their significant capacity to propagate vegetatively, most orchids reproduce via seeds. Sexual reproduction via seed is commonly reported, in contrast to apomixis, whereby seeds are clones of the mother. Although insect pollination and autonomous self-pollination exist in mycoheterotrophic plants, the reproductive embryology of these plants remains under-studied. This paper provides evidence for the co-occurrence of both sexual and apomictic reproduction in a population of mycoheterotrophic plants - Epipogium aphyllum. We investigated seed formation via open pollination, induced autogamy, autogamy sensu stricto and autonomous apomixis. • Methods The study was performed on a population of E. aphyllum located in northern Poland. The research included studies of the micromorphology, histochemistry and embryology of four types of reproductive systems. Scanning, fluorescence and light microscopy accompanied by graphical and statistical analyses were employed. • Key Results We observed gametophyte development, from the one-nucleate stage to maturity, in unpollinated flower buds. The lack of zygotes in flower buds indicated that fertilization did not occur at this stage. Manual selfpollination led to a zygote, followed by embryo formation. Fertilization and embryo development derived from embryogenesis via open pollination is delayed compared with hand pollination. Isolation from external pollination resulted only in structures resembling zygotes that may originate either sexually or independent of fertilization. Parthenogenetic structures that resembled zygotes were observed in flowers that were emasculated and isolated from pollination. Zygotes formed at significantly higher frequencies via open pollination and induced autogamy in comparison to the parthenogenetic structures formed in other treatments. • Conclusions We showed the absence of pre-zygotic barriers for autogamy in E. aphyllum. Self-pollination and self-fertilization are possible; however, natural self-pollination is unlikely or rare due to the position of the pollinia. Incidental parthenogenesis in E. aphyllum is very likely, given the biology of ovule development of this mycoheterotrophic orchid. This species therefore has the potential to produce seeds via both sexual and asexual means, although the contribution of apomixis to this process appears largely negligible. [ABSTRACT FROM AUTHOR]

Background and Aims Dominant tree species in northern temperate forests, for example oak and beech, produce desiccation-sensitive seeds. Despite the potentially major influence of this functional trait on the regeneration and distribution of species under climate change, little is currently known about the ecological determinants of the persistence of desiccation-sensitive seeds in transient soil seed banks. Knowing which key climatic and microsite factors favour seed survival will help define the regeneration niche for species whose seeds display extreme sensitivity to environmental stress Methods Using the Mediterranean Holm oak (Quercus ilex) forest as a model system, an in situ time-course monitoring of seed water status and viability was performed during the unfavourable winter season in two years with contrasting rainfall, at an instrumented site with detailed climate records. In parallel, the characteristics of the microhabitat and their influence on the post-winter water status and viability of seeds were investigated in a regional survey of 33 woodlands representative of the French distribution of the species. Key Results Time-course monitoring of seed water status in natural conditions confirmed that in situ desiccation is the main abiotic cause of mortality in winter. Critical water contents could be reached in a few days during drought spells. Seed dehydration rates were satisfactorily estimated using integrative climate proxies including vapour pressure deficit and potential evapotranspiration. Seed water status was therefore determined by the balance between water uptake after a rainfall event and water loss during dry periods. Structural equation modelling of microhabitat factors highlighted the major influence of canopy openness and resulting incident radiation on the ground. Conclusions This study provides part of the knowledge required to implement species distribution models which incorporate their regeneration niche. Background: Anthropogenic climate change (ACC) will influence all aspects of plant biology over coming decades. Many changes in wild species have already been well-documented as a result of increased atmospheric CO2 concentrations, warming climate and changing precipitation regimes. A wealth of available data has allowed the use of meta-analyses to examine plant-climate interactions on more sophisticated levels than before. These analyses have revealed major differences in plant response among groups, e.g. with respect to functional traits, taxonomy, life-history and provenance. Interestingly, these meta-analyses have also exposed unexpected mismatches between theory, experimental, and observational studies. Scope: We reviewed the literature on species' responses to ACC, finding ~42 % of 4000 species studied globally are plants (primarily terrestrial). We review impacts on phenology, distributions, ecophysiology, regeneration biology, plant-plant and plant-herbivore interactions, and the roles of plasticity and evolution. We focused on apparent deviations from expectation, and highlighted cases where more sophisticated analyses revealed that unexpected changes were, in fact, responses to ACC. Conclusions: We found that conventionally expected responses are generally well-understood, and that it is the aberrant responses that are now yielding greater insight into current and possible future impacts of ACC. We argue that inconclusive, unexpected, or counter-intuitive results should be embraced in order to understand apparent disconnects between theory, prediction, and observation. We highlight prime examples from the collection of papers in this Special Issue, as well as general literature. We found use of plant functional groupings/traits had mixed success, but that some underutilized approaches, such as Grime's C/S/R strategies, when incorporated, have improved understanding of observed responses. Despite inherent difficulties, we highlight the need for ecologists to conduct community-level experiments in systems that replicate multiple aspects of ACC. * Background and Aims Loss of seed viability has been associated with deteriorative processes that are partly caused by oxidative damage. The breaking of dormancy, a seed trait that prevents germination in unfavourable seasons, has also been associated with oxidative processes. It is neither clear how much overlap exists between these mechanisms nor is the specific roles played by oxygen and reactive oxygen species. * Methods Antioxidant profiles were studied in fresh (dormant) or after-ripened (non-dormant) sunflower (Helianthus annuus) embryos subjected to controlled deterioration at 40 °C and 75% relative humidity under ambient (21%) or high O2 (75%). Changes in seed vigour and viability, dormancy, protein carbonylation and fatty acid composition were also studied. * Key Results After-ripening of embryonic axes was accompanied by a shift in the thiol-based cellular redox environment towards more oxidizing conditions. Controlled deterioration under high O2 led to a faster loss of seed dormancy and significant decreases in glutathione reductase and glutathione peroxidase activities, but viability was lost at the same rate as under ambient O2. Irrespective of O2 concentration, the overall thiol-based cellular redox state increased significantly over 21 d of controlled deterioration to strongly oxidizing conditions and then plateaued, while viability continued to decrease. Viability loss was accompanied by a rapid decrease in glucose-6-phosphatede-hydrogenase, which provides NADPH for reductive processes such as required by glutathione reductase. Protein carbonylation, a marker of protein oxidation, increased strongly in deteriorating seeds. The lipid-soluble tocochromanols, dominated by α-tocopherol, and fatty acid profiles remained stable. * Conclusions After-ripening, dormancy-breaking during ageing and viability loss appeared to be associated with oxidative changes of the cytosolic environment and proteins in the embryonic axis rather than the lipid environment. * Background Reactive oxygen species (ROS) are considered to be detrimental to seed viability. However, recent studies have demonstrated that ROS have key roles in seed germination particularly in the release of seed dormancy and embryogenesis, as well as in protection from pathogens. * Scope This review considers the functions of ROS in seed physiology. ROS are present in all cells and at all phases of the seed life cycle. ROS accumulation is important in breaking seed dormancy, and stimulating seed germination and protection from pathogens. However, excessive ROS accumulation can be detrimental. Therefore, knowledge of the mechanisms by which ROS influence seed physiology will provide insights that may not only allow the development of seed quality markers but also help us understand how dormancy can be broken in several recalcitrant species. * Conclusions Reactive oxygen species have a dual role in seed physiology. Understanding the relative importance of beneficial and detrimental effects of ROS provides great scope for the improvement and maintenance of seed vigour and quality, factors that may ultimately increase crop yields.
