Nanoparticles (NPs) concentration directly impacts the dose delivered to target tissues by nanocarriers. The evaluation of this parameter is required during NPs developmental and quality control stages, for setting dose-response correlations and for evaluating the reproducibility of the manufacturing process. Still, faster and simpler procedures, dismissing skilled operators and post-analysis conversions are needed to quantify NPs for research and quality control operations, and to support result validation. Herein, a miniaturized automated ensemble method to measure NPs concentration was established under the lab-on-valve (LOV) mesofluidic platform. Automatic NPs sampling and delivery to the LOV detection unit were set by flow programming. NPs concentration measurements were based on the decrease in the light transmitted to the detector due to the light scattered by NPs when passing through the optical path. Each analysis was accomplished in 2 min, rendering a determination throughput of 30 h -1 (6 samples h -1 for n = 5) and only requiring 30 μL (≈0.03 g) of NPs suspension. Measurements were performed on polymeric NPs, as these represent one of the major classes of NPs under development for drug-delivery aims. Determinations for polystyrene NPs (of 100, 200, and 500 nm) and for NPs made of PEGylated poly-d,l-lactide- co -glycolide (PEG-PLGA, a biocompatible FDA-approved polymer) were accomplished within 10 8 -10 12 particles mL -1 range, depending on the NPs size and composition. NPs size and concentration were maintained during analysis, as verified for NPs eluted from the LOV by particle tracking analysis (PTA). Moreover, concentration measurements for PEG-PLGA NPs loaded with an anti-inflammatory drug, methotrexate (MTX), after their incubation in simulated gastric and intestinal fluids were successfully achieved (recovery values of 102-115%, as confirmed by PTA), showing the suitability of the proposed method to support the development of polymeric NPs targeting intestinal delivery.

In recent years, there has been an increasing interest in nitro fatty acids (NO2-FA) as signaling molecules formed under nitroxidative stress. NO2-FA were detected in vivo in a free form, although it is assumed that they may also be esterified to phospholipids (PL). Nevertheless, insufficient discussion about the nature, origin, or role of nitro phospholipids (NO2-PL) was reported up to now. The aim of this study was to develop a mass spectrometry (MS) based approach which allows identifying nitroalkenes derivatives of three major PL classes found in living systems: phosphatidylcholines (PCs), phosphatidylethanolamine (PEs), and phosphatidylserines (PSs). NO2-PLs were generated by NO2BF4 in hydrophobic environment, mimicking biological systems. The NO2-PLs were then detected by electrospray ionization (ESI-MS) and ESI-MS coupled to hydrophilic interaction liquid chromatography (HILIC). Identified NO2-PLs were further analyzed by tandem MS in positive (as [M + H](+) ions for all PL classes) and negative-ion mode (as [M - H](-) ions for PEs and PSs and [M + OAc](-) ions for PCs). Typical MS/MS fragmentation pattern of all NO2-PL included a neutral loss of HNO2, product ions arising from the combined loss of polar headgroup and HNO2, [NO2-FA + H](+) and [NO2-FA - H](-) product ions, and cleavages on the fatty acid backbone near the nitro group, allowing its localization within the FA akyl chain. Developed MS method was used to identify NO2-PL in cardiac mitochondria from a well-characterized animal model of type 1 diabetes mellitus. We identified nine NO2-PCs and one NO2-PE species. The physiological relevance of these findings is still unknown.

Handling of carbon nanoparticles as sorptive materials in a flow-through packed-bed mode has been to date hampered by undue pressure drop and deteriorated retention efficiency because of nanoparticle bundling and entanglement. To surmount this limitation, a dedicated stirred-flow sorptive microchamber integrated in a fully automated sequential injection (SI) assembly is herein proposed for expedient handling and reuse of carbon nanoparticles in microsolid-phase extraction (μSPE) procedures. The assembled setup features automatic uptake, preconcentration, and retrieval of target organic species using dispersed nanoparticles as a front-end to liquid chromatographic (LC) assays. Chlorotriazine residues (atrazine, simazine, and propazine) and dealkylated metabolites thereof (deisopropyltriazine (DIA) and deethylatrazine (DEA)) were selected as model compounds because of their electron-poor aromatic structure and potentially strong π-π interactions with electron-rich sorptive materials. The effect of several parameters on the analytical performance including the type and amount of nanoparticles (carbon nanofibers (CNFs), multiwalled carbon nanotubes (MWCNTs) and oxidized carbon nanotubes (MWCNT-COOH), the sample volume (breakthrough volume), the nature and volume of eluent, and the interface between the sample processing module and LC was explored in detail. Using dispersed CNFs at-line coupled to LC, absolute recovery percentages for 10 mL sample percolation were >94% for the overall herbicides with enrichment factors of ca. 20, limits of detection (S/N = 3) of 0.004-0.03 ng mL(-1), limits of quantification (S/N = 10) of 0.01-0.09 ng mL(-1) and repeatability within the range 0.5-1.8%. The SI-CNF-LC hyphenated system was harnessed to the analysis of not merely untreated environmental waters at concentration levels below those endorsed by the current EU Water Framework Directives but to crude soil extracts for which CNF reuse with no loss of retention efficiency was proven feasible by resorting to appropriate automatic regeneration procedures and internal standardization.

Molecular imprinted polymers (MIP) have recently drawn much attention as highly selective solid-phase materials for handling and isolation of organic pollutants in complex matrices. Because of the impaired retention capacity for target species as compared with reversed-phase materials and irreversible sorption of interfering compounds by nonspecific interactions, the implementation of MIP-based solid-phase reactors as permanent components in automatic flow-systems has not received widespread acceptance as of yet. To tackle this limitation, a dynamic microscale solid phase extraction (microSPE) method capitalizing on the principle of programmable flow and bead injection analysis is herein proposed as a front end to liquid chromatography for multiresidue assays. It involves in-line renewable tandem-SPE microcolumns composed of molecularly imprinted polymers and copolymeric N-vinylpyrrolidone/divinylbenzene beads integrated within the flow network for multimodal extraction. Chlorotriazine herbicides (namely, atrazine, simazine, propazine) and principal degradation products thereof (namely, deisopropylatrazine and deethylatrazine) were selected as model analytes. The effect of several parameters, including the dimensions and chemical composition of the sorptive microcolumns, the sample loading flow rate, the type and volume of eluent, the interface with liquid chromatography (LC), and the disposable nature of the column on the analytical performance were investigated in detail. The assembled flow setup features appropriate removal of interfering organic species via solvent switch with toluene, the circumvention of analyte band-broadening in LC by in-line merging of the eluate with a water stream, and the transfer of the overall analyte-containing eluate into the LC. For 10-mL sample percolation, limits of detection (S/N = 3) of 0.02-0.04 ng mL(-1), limits of quantification (S/N = 10) of 0.07-0.12 ng mL(-1), absolute recovery percentages >79%, precision within 1.4-5.5%, and enrichment factors of 46-49 were obtained for the suite of assayed herbicides. The multimodal microSPE method with renewable beads was applied to the multiresidue determination of the target herbicides in crude soil extracts and untreated environmental waters at concentration levels below those endorsed by the current EU Water Framework Directives following appropriate sample preconcentration and/or cleanup.

In the present work, a chemiluminometric automatic flow methodology for the in vitro determination of hypochlorous acid scavenging capacity, under pH and concentration conditions similar to those found in vivo, is proposed. As the pH found in physiological conditions (7.4) and the pH required for the chemiluminescence detection reaction (>10) are different, the multisyringe flow injection analysis features were exploited to perform the in-line reaction of HOCl and the scavenger molecule at physiological pH prior to reaction of the remaining HOCl with luminol at alkaline conditions. These two reactions were carried out in about 3 s, allowing the determination of fast reacting antioxidants, in a time frame closer to in vivo generation of HOCl when compared to previously described methods. The developed method was applied to nonsteroidal antiinflammatory drugs of different chemical families, and positive controls (cysteine, gallic acid, lipoic acid). The HOCl scavenging capacity was evaluated at pH 7.4 and 10.0; different results were found for oxicam derivatives, providing evidence that the pH of in vitro methods should be carefully selected to allow assumptions about putative in vivo effects.
