Advanced analytical imaging techniques, including matrix-assisted laser desorption/ionization high-resolution mass spectrometry (MALDI-HRMS) imaging, can be used to visualize the distribution, localization, and dynamics of target compounds and their precursors with limited sample preparation. Herein we report an application of MALDI-HRMS imaging to map, in high spatial resolution, the accumulation of the medicinally important naphthodianthrone hypericin, its structural analogues and proposed precursors, and other crucial phytochemical constituents in the leaves of two hypericin-containing species, Hypericum perforatum and Hypericum olympicum. We also investigated Hypericum patulum, which does not contain hypericin or its protoforms. We focused on both the secretory (dark glands, translucent glands, secretory canals, laminar glands, and ventral glands) and the surrounding non-secretory tissues to clarify the site of biosynthesis and localization of hypericin, its possible precursors, and patterns of localization of other related compounds concomitant to the presence or absence of hypericin. Hypericin, pseudohypericin, and protohypericin accumulate in the dark glands. However, the precursor emodin not only accumulates in the dark glands but is also present outside the glands in both hypericin-containing species. In hypericin-lacking H. patulum, however, emodin typically accumulates only in the glands, thereby providing evidence that hypericin is possibly biosynthesized outside the dark glands and thereafter stored in them. The distribution and localization of related compounds were also evaluated and are discussed concomitant to the occurrence of hypericin. Four geometric isomers of trifloxystrobin (TFS)—namely EE, EZ, ZE, and ZZ—were hydrolyzed by 0.05 M NaOH, resulting in four corresponding acid metabolites. These compounds—namely EE-, EZ-, ZE-, and ZZ-acids—were purified by preparative HPLC and authentically characterized by a combination of infrared, Raman, GC–MS, LC–MS/MS, and NMR spectroscopies. The spectra were found to be very characteristic of the individual isomers, and so they could be used to distinguish the isomers from each other. The detailed spectral features of the individual isomers are presented and compared. EE-acid was identified as being the major metabolite of TFS in soil, which indicates that hydrolysis is the principal route of degradation of TFS. This finding further justifies the importance of the present study in relation to assessing the risk associated with the release of TFS into the environment. [ABSTRACT FROM AUTHOR]

The behaviour of sulfadiazine (SDZ) and its metabolites was investigated by administering the 14C-labelled veterinary drug to fattening pigs. The excretion kinetics were determined after daily collection of manure. Two known metabolites, N-acetylsulfadiazine and 4-hydroxysulfadiazine, and two hitherto unidentified minor metabolites were recovered. Various mass spectrometric techniques such as parent, product ion scans and accurate mass measurement were used. The new compounds were identified as N-formylsulfadiazine (For-SDZ) and N-acetyl-4-hydroxysulfadiazine (Ac-4-OH-SDZ). The identification of SDZ, Ac-SDZ and For-SDZ was confirmed by comparison of the spectroscopic and chromatographic data of the synthesized authentic references. The identification of the hydroxylated compounds 4-OH-SDZ and Ac-4-OH-SDZ was performed by MSn, and accurate mass measurements. Only 4% of the administered radioactivity remained in the pig after ten days and SDZ accounted for 44% of the 96% radioactivity excreted. More than 93% of the labelled compounds were detected and identified in the manure. The key analytical problem, namely a high concentration of matrix in sample extracts, was overcome by advanced measurement techniques and with the use of a suitable internal standard. The mean recoveries for all compounds were ≥96%. Linearity was established over a concentration range of 0.5 to 10,000 μg kg−1 manure with a correlation coefficient ≥0.99. The photoisomerization kinetics of trifloxystrobin (TFS) in acetone under artificial sunlight is reported. HPLC analysis showed the TFS, a strobilurine fungicide of EE conformation, was converted into an equilibrium mixture of four isomers after illumination for 7 h. The isomers were identified as EZ, EE, ZZ, and ZE and were separated in the crystalline form by preparative HPLC and characterized by use of a variety of spectroscopic techniques. The quantum yield and reaction constants for the isomerization reactions were determined. The detailed spectral features of the individual isomers measured by UV, IR, Raman, NMR and mass spectroscopy are presented and compared. A method for the analysis of several macrolide and ionophore antibiotics as well as tiamulin in liquid manure was developed. Reversed-phase liquid chromatography and atmospheric pressure chemical ionisation (APCI) tandem mass spectrometry was used for detection. High-performance liquid chromatographic (HPLC) separation of the antibiotics was achieved in 35 min. The analytes were extracted with ethyl acetate and the extracts were cleaned up by solid-phase extraction on a diol SPE cartridge. Recovery experiments with spiked liquid manure concentrations varying from 6 to 2,000 µg kg-1 gave constant recovery rates. The recovery rates for the macrolides erythromycin, roxithromycin and oleandomycin were 75–94%, that for the ionophore salinomycin was 119%, while that for the pleuromutilin tiamulin was 123%, when using a macrolide internal standard. The relative standard deviation was found to be 15–36% and the limits of detection were 0.4–11.0 µg kg-1. The maximum concentrations found in manure samples were 43 µg kg-1 for tiamulin and 11 µg kg-1 for salinomycin. [ABSTRACT FROM AUTHOR]
