Abstract

A study of human myometrium obtained from 127 cesarean section deliveries shows that corticotropin-releasing hormone (CRH) may enhance the myometrial response to prostaglandin F2-alpha (PGF2-alpha). Myometrium is a muscle lining of the uterus. CRH concentrations are known to increase throughout pregnancy, with the highest levels found during labor. CRH may play a critical role in facilitating the onset of human labor by its interaction with PGF2-alpha. Myometria that received PGF2-alpha followed by administration of CRH had a significantly increased response to PGF2-alpha. CRH was found to be ineffective if administered along with a second dose of PGF2-alpha. Thus, the interaction of CRH and PGF2-alpha may work in a local loop mechanism to facilitate labor. CRH is produced by the placenta and membranes during pregnancy, and its production is stress-related in response to the presence of glucocorticoids, oxytocin, and prostaglandins.
