A total of 46 pregnant women with premature delivery and preserved amniotic sac were examined. The effect of tocolysis applied was recorded at different control periods of time with the aid of obstetrical monitor. The basic frequency of uterine contractions was more than 3--84.78%. In the 120th min after tocolysis with Partusisten 50% of the cases did not show any labour activity: after 120 min the inhibition was very successful--82.61%. This success was mainly recorded in the cases with uterine contractions over 4 for 10 min (31.61%) followed by those with 3-4 contractions (34.78%) and those with up to 2 uterine contractions (15.22%). On the basis of the results obtained it is concluded that the quicker and earlier the tocolytic was applied, the quicker and lasting was the effect. The statistical analysis revealed that all uterine contractions in the second and particularly in the third group depending on the time of tocolysis gradually passed into the first and second group.

Clinical and experimental studies have revealed that salbupart, a beta-mimetic agent, administered in a dose of 0.14 mg/kg (the maximal therapeutic dose) markedly inhibited the spontaneous and oxytocin-induced uterine contractility, caused no embryotoxic or teratogenic effects, and did not influence fetal growth and development. The efficacy of salbupart tocolytic therapy was estimated as 75%. Blood plasma cAMP levels normalized in the pregnant women over the course of therapy. Clinical and experimental results recommend salbupart for pregnancy protecting therapy in cases with threatened spontaneous abortions.

It is established that the mean velocity of blood flow (Vmean) of the fetal aorta in pregnant women with pre-term deliveries during Doppler examination is 40.70-3.26 cm/s, but in women with normal pregnancy (a control group) is 32.46-2.13 cm/s. The difference is statistically significant (p less than 0.001). It is indicated that the tocolytic Partusisten passes through the placental barrier, penetrates in the fetal circulation and accelerates the velocity of the blood flow. This shows that it improves utero-placental circulation and fetal oxygenation and with this the general state of the fetus as well. It is pointed out that the results from this study support the rightness for usage of Partusisten in treatment of chronic and acute placental insufficiency for improvement of fetal hemodynamics as well as the general state of the fetus.

Experiments on pregnant rats revealed that a combination of low doses of partusisten and clophelin (1.25 mg/kg and 0.15 ml of 0.001% solution, respectively) potentiated a tocolytic effect. A combination of prostenon and oxytocin given in half doses (0.15 mg/kg and 0.25 IU in 0.1 ml, respectively) summarized their stimulant effect on the myometrium. Premedication with partusisten (2.5 micrograms/kg) failed to change the stimulant effect produced by a combination of prostenon and oxytocin given in half doses.

Partusisten was examined for effects on uterine contractility in 28-36- or 38-42-week pregnant women and parturients. The agent was shown to result in inhibited uterine contractility in the pregnant and parturient females, the contractility was decreased before and particularly during labor, which was indicative of lower beta-adrenoreceptor inhibitory mechanism force.
