In this study, the effects of repeated cycles of drying and rehydration on some physiological traits were assessed in long shelf-life tomatoes cultivated in a typical semi-arid area of Southern Italy. Three Sicilian landraces (‘Custonaci’, ‘Salina’, and ‘Vulcano’) from the germplasm collection at CNR-IBE (Catania, Italy) and a commercial tomato mini-plum (‘Faino Hy., control) were investigated under three water regimes: DRY (no irrigation), IRR (long-season full irrigation) and REW (post-drought rewaterings). Net photosynthetic assimilation rate (Pn), leaf transpiration (E), stomatal conductance (gs), instantaneous water use efficiency (WUEi), leaf intercellular CO2 (Ci, ppm), and leaf temperature (°C), were measured during the growing season. At harvest (late July), fruit production per plant was measured and ripened fruits were analysed for total solids (TS), soluble solids (SS), reducing sugars (RS), vitamin C (AscA), and total phenols (TP). Pn promptly responded to rewatering (REW), quickly increasing immediately after irrigation, and declined with soil drying up. All genotypes had similar physiological pathways in DRY, but in IRR, ‘Faino’ had higher Pn (up to 31 μmol CO2 m−2s−1) and E (up to 18 mmol H2O m−2s−1). Stomatal conductance (gs) after rewatering steeply increased and quickly declined after that. All local landraces had the same gs in IRR and REW. Variations in RWC were less pronounced than those in other physiological parameters. WUEi in REW and DRY proceeded similarly (up to 3 μmol CO2 mmol H2O). Irrigation in REW significantly promoted plant productivity over the DRY control (up to +150% in ‘Vulcano’). TS and SS in REW were lower than those in DRY, but higher (+19 and +7%, respectively) than in IRR. Vitamin C was greater in DRY and REW (26 and 18% higher than in IRR, respectively). TP in all local tomatoes were significantly higher (up to +29% in ‘Vulcano’) than those in the commercial control. Water regime had a minor effect on TP in ‘Custonaci’ and ‘Salina’. Principal Component Analysis (PCA) provided information on the changes in physiological and fruit quality traits in tomatoes in relation to cultivars and water regimes. The results of this study also revealed that a water-saving irrigation strategy where few irrigations are applied after prolonged periods of drought might be profitable in terms of fruit production enhancement in long shelf-life tomatoes and that limited rewaterings in most cases, help retaining high levels of fruit quality traits.

Germination of castor seeds from five dwarf hybrid genotypes, compared to a ‘Local’ genotype selected by the University of Catania from a Tunisian population well adapted to the Mediterranean environment, was studied at five different salt levels (0, −0.3, −0.6, −0.9, and −1.2 MPa) in order to assess seed germination performance under stress conditions. The results confirmed that optimum moisture (0 MPa) ensured 100% of germination; on the contrary, salt concentration negatively influenced the final germination percentage (FGP) and radicle elongation, causing severe consequences for plant establishment. At a level of −1.2 MPa, no germination occurred, while a level of −0.3 MPa slightly affected the seed germination of the dwarf genotypes, which achieved 77.3% of germination, contrary to the ‘Local’ genotype, in which germination was kept stable. Higher levels of salt (−0.6 and −0.9 MPa) caused a linear decrease in FGP and radicle elongation. Overall, the dwarf hybrid ‘C1019’ performed better at higher salt impositions, as did ‘C857’, confirming these genotypes were the most tolerant among the dwarf hybrids. Conversely, ‘C1013’ turned out to be the most susceptible genotype, followed by ‘C1008’. On the other hand, the ‘Local’ castor genotype was the best-performing genotype at −0.3 MPa and the most tolerant genotype in terms of salt concentration and germination time, which were accurately predicted by the hydrotime model, validating it as a valid method of assessing the germination response of castor seeds to Ψ.

To evaluate the performance of dwarf castor hybrids (‘C1012’, ‘C857’, ‘C856’), compared to a local selected genotype, in four subsequent sowing dates (SW1, SW2, SW3, SW4), a trial was conducted at the experimental farm of the University of Catania (Sicily, Italy). The length of the growing season decreased with the increase of the sowing date in the average genotypes from 160 to 94 days, respectively, for the first and the last sowing date. According to the RED—Renewable Energy Directive, the genotype ‘C856’ was the earliest (112 days), resulting in suitability as a catch crop for biomass production. The results showed that early spring sowings negatively impact dwarf hybrid production (1.2 and 1.5 Mg ha−1 in SW1 and SW2, in the average of the three hybrids), which reached the highest yield in the third sowing date (2.0 Mg ha−1), preferring warmer temperatures for the germination of seeds. On the contrary, the ‘Local’ genotype reached the highest yield (1.6 Mg ha−1) in the first sowing date and linearly decreased in the subsequent ones. Nonetheless, the third sowing date positively influenced the oil content and the oil yield in all dwarf genotypes except the ‘Local’ genotype, which showed the highest oil yield in the first sowing date.

Alternaria alternata is a fungus that causes a serious disease in susceptible genotypes of citrus, particularly in mandarins. The Alternaria citri toxin (ACT) produced by the pathogen induces necrotic lesions on young leaves and fruits, defoliation and fruit drop. Here, we describe two methods of marker-assisted selection (MAS) that could be used for the early identification of Alternaria brown spot (ABS)-resistant mandarin hybrids. The first method is based on a nested PCR coupled to high resolution melting (HRM) analysis at the SNP08 locus, which is located at 0.4 cM from the ABS resistance locus, and was previously indicated as the most suitable for the selection of ABS-resistant hybrids. The method was validated on 41 mandarin hybrids of the CREA germplasm collection, and on 862 progenies generated from five crosses involving different susceptible parents. Four out of five populations showed Mendelian segregation at the analyzed locus, while a population involving Murcott tangor as male parent showed distorted segregation toward the susceptible hybrids. The second method is based on a cleaved amplified polymorphic sequences (CAPS) marker that was developed using the same primers as the nested PCR at the SNP08 locus, coupled with BccI restriction enzyme digestion. To verify the reliability of the two genotyping methods, in vitro leaf phenotyping was carried out by inoculating A. alternata spores onto young leaves of 101 hybrids, randomly chosen among the susceptible and resistant progenies. The phenotyping confirmed the SNP08 genotyping results, so the proposed method of selection based on HRM or CAPS genotyping could be routinely used as an alternative to KBioscience competitive allele specific polymerase chain reaction (KASPar) single nucleotide polymorphism (SNP) genotyping system to improve citrus breeding programs. While the study confirmed that the SNP08 marker is a reliable tool for MAS of new citrus hybrids with different genetic backgrounds, it also identified a small group of genotypes where the resistance mechanism requires further investigation.
