This study presents the design and implementation of a low-speed intrusion experiment under microgravity, with a strong motivation to support the pre-research of the Chinese asteroid exploration mission. In the experiment apparatus, we reconstructed an effective gravity of 10 - 2 g in a free-falling capsule in the drop tower. The simulant of regolith particles is processed using natural basalt and sorted using layered screens. The irregular shapes of the particles are retained to mimic the realistic regolith. A detailed description of the experimental system components and the full-cycle procedure is first presented, which gives 3.6 s of microgravity time for each trial. Repeatable controlled experiments are performed to characterize the dynamical responses of the simulant particles under centimeter-per second intrusion. The results reveal diverse responses that depend on different variables, and the mechanism of different responses is discussed based on theories of granular mechanics. This experimental study is expected to improve our understanding of the contact mechanics of regolith material, which will facilitate the operation design of future asteroid missions. The research on particle size classification in lunar microgravity has gradually attracted the attention of scholars. Based on the discrete element method (DEM), this work proposes a double-layer auxiliary screening vibrating screen in the form of intermediate feeding, and takes the screening efficiency and screening time as the evaluation indicators. Firstly, the Helmholtz-Maxwell coil is used to simulate the force of particles under lunar microgravity, which verifies the reliability of the numerical simulation of particles under lunar microgravity based on the discrete element method. Then, the influence of the parameters of each screen on the comprehensive performance of screening was analyzed by a single-factor experiment. Finally, based on the BP (Back Propagation) neural network optimized by genetic algorithm, the evaluation model of screen parameters and screen performance is established, and the multi-objective genetic algorithm is used to optimize the screen parameters combination scheme with excellent performance. • A compact electro-adhesive hexapod robot is designed and prototyped. • A novel convertible limb that integrates manipulation and locomotion is proposed. • An optimized foot-trajectory subject to velocity and acceleration is developed. • Extensive simulations and ground experiments are performed. Service space robotics can provide on-orbit maintenance for faulty spacecrafts to extend life and reduce economic losses. In many cases, spacecraft failures are minor and can be easily recovered by completing simple manipulations or by providing observations. In this study, we propose a spacecraft surface adhesion-crawling-manipulation hexapod robot that can provide such services. To avoid increasing the payload, both of the robot's forelimbs were designed to be convertible, integrating manipulation and locomotion capabilities; the manipulation methods for the novel convertible limb are analyzed. A novel foot trajectory generation method is proposed to reduce the contact force generated when the foot of the robot touches the spacecraft surface and increase dynamic stability. Theoretical and experimental analyses of our robot are presented. Kinematic analyses are performed to validate the manipulation methods using convertible forelimbs. The stability of the robot's static adherence and dynamic crawling are analyzed in a microgravity simulation environment. • A ray-tracing algorithm is applied to melting PCM bridges. • Support material for the optical diagnosis of the MarPCM experiment. • The validation of the algorithm is performed via ground experiments. • Three reference bridge shapes are defined to account for volume changes. • A minimum of 60% liquid fraction is needed to accurately determine the front shape. A ray-tracing algorithm is developed to obtain synthetic images for tracking the melting of the phase change material (PCM) n-octadecane in a liquid bridge configuration. The translucent nature of the solid n-octadecane is modelled through a sigmoid function which provides a distance-dependent illumination intensity throughout the solid. The code is validated against two types of experiments: front shapes made of 3D-printed PLA and cast front shapes of solid n-octadecane. Comparison with these experiments allows the numerical constants involved in the model to be tuned to appropriate values. For application to the MarPCM microgravity experiment (Porter et al., 2023), the current study provides the expected images of the solid/liquid front obtained by the optical diagnosis module for three different values of the dimensionless volume (defined with respect to a perfect cylinder), V = 0.9 , 1, and 1.1, and two applied temperature differences, Δ T = 5 and 30 K. The solid/liquid front shapes are obtained numerically from a two-dimensional axisymmetric code as in Varas et al. (2021). The results reveal a strong dependence of image quality on the liquid fraction L. Visual inspection leads to the conclusion that, below L ≈ 60 % , an accurate determination of the front location may be difficult, particularly for the relatively shallow layer of liquid near the thermocapillary interface. This paper attempts to present a comprehensive review of adhesion and locomotion technology for exploring small celestial bodies (SCBs). The long-term landing, adhesion, and locomotion of space robots on the surface of SCBs can maximize the scientific value of exploration missions. They can also provide technical reserves and support for future SCBs resource exploitation and crewed missions. Based on this background, the current progress and planning of SCBs exploration missions are first reviewed. Due to the special microgravity environment and complex surface characteristics of SCBs, adhesion and locomotion on SCBs surfaces are extremely difficult. The surface environmental characteristics of SCBs and their influences and limitations on the adhesion and locomotion of space robots are analyzed. The progress of classic adhesion and locomotion technology on SCBs is classified and studied, including anchoring adhesion, instantaneous adhesion, and locomotion technologies. Based on the research of climbing and soft robots, various bionic adhesion and locomotion techniques are introduced, with emphasis on their application prospects in SCBs exploration missions. Finally, according to the trend of future SCBs exploration missions, the critical issues and research direction of related technologies are presented, and concluding remarks are provided. • A leg-type asteroid probe landing cushioning device is proposed based on electromagnetic damping. • The vertical landing speed and landing tilt angle will affect the cushioning characteristics. • The horizontal movement direction and the landing tilt direction have a coupling effect on the landing dynamics. • Soft landing media will cause the footpad to collide and bounce on the surface, thereby increasing the cushioning time. In-situ scientific exploration is of great significance to asteroid exploration, and resource development and utilization. The asteroid surface landing cushioning is the basis of in-situ scientific exploration of asteroids. Therefore, a leg-type asteroid landing cushioning device is proposed based on the principle of electromagnetic damping. A dynamic simulation model is used to study the cushioning dynamics of the cushioning device in a microgravity environment. For the proposed landing cushioning device, the simulation investigates the influence of vertical landing speed and landing vertical tilt angle on the dynamic characteristics and landing stability. The dynamics simulation method is also adopted to investigate the influence of the landing horizontal tilt angle and the horizontal movement direction on the landing stability of the three-legged asteroid probe. The horizontal movement direction and the landing horizontal tilt angle have a coupling effect on the landing cushioning dynamics. Finally, based on the developed simulation device of asteroid microgravity environment, landing cushioning experiments are conducted to investigate the influence of soft and hard landing media on the cushioning process and verify the feasibility of the landing cushioning device. • Impossible experiments are performed in microgravity environment. • International collaboration/joint ventures are increasing in microgravity research. • Clinostats and RPMs simulate partial/reduced gravity conditions of Mars and Moon. • New microgravity analogs, models and simulators of better benefits can be developed. Gravity is always present on Earth. However, the influence of gravity can be modified or compensated. Generally, the products created under microgravity have important properties that usually surpasses the best terrestrial counterparts. Commercially, these products have striking features that aids marketing. Fast responding microgravity platforms can be used for short-term experiments, where the reactions or observations takes less time. This can be provided in drop towers, parabolic flights and sounding rockets. To study long-term effects of microgravity, human tended space laboratories have been used, such as the International Space Station. Scientists have developed various kinds of ground-based facilities and equipment for some of them to achieve the condition of functional weightlessness as a result of limited access to space laboratories. Slow responding microgravity platforms can be used for long-term experiments, where the reactions or observations takes more time. Microgravity analog, model or simulations are slow responding platforms and are able to provide long-term effects of microgravity. This includes long-duration bedrests, water immersions and in magnetic levitation, clinostats, random positioning machines, rotating wall vessels and even centrifuges. This study compares and gives an overview of currently known real, analog, model or simulated microgravity environments and platforms, and demonstrates their individual capacities, benefits and limitations. Three-dimensional and oscillatory thermo-capillary flow in a liquid bridge of 5 cSt silicone oil (Pr = 67) has been numerically investigated by performing dynamic mode decomposition (DMD) analysis applied to the time-resolved thermal data generated by the 'method of snapshots'. For various heat transfer conditions defined on the free surface of the curved liquid bridge, a series of dynamic modes has been extracted to unfold the underlying physical mechanism that governs the considered dynamical system. Unstable and neutral modes primarily accountable for the transition to the supercritical state established in the liquid bridge have been identified by decomposing the acquired temperature disturbances into spatial and temporal coherent structures. Under microgravity conditions, unsteady and three-dimensional computations are carried out in a liquid bridge of different volume ratios (VR = 0.9 and 1.1) using the finite volume method (FVM) to capture the dynamical evolution of temperature disturbances at the onset of oscillatory instability. The present numerical investigation is mainly aimed at providing broader insight into the complex fluid dynamic processes associated with the oscillatory thermal convection by determining the growth/decay rate and frequency of each characteristic mode with the help of DMD spectra. For each oscillation pattern, the dominant behavior of oscillatory thermo-capillary flow is revealed by its coherent structures. Present work describes a numerical investigation concerning the response of oscillatory thermo-capillary convection to free surface heat loss in a cylindrical half floating zone of high Prandtl number liquid (Pr =67) under microgravity conditions. Unsteady and three-dimensional computations have been carried out in the liquid domain using the finite volume method (FVM) to reveal the oscillatory thermo-fluid dynamic regimes at the onset of instability. The influence of different free surface heat loss conditions on the bifurcation from the basic steady axisymmetric state has been discussed in terms of critical azimuthal wavenumber, m. The dynamic evolution of the temperature disturbances at supercritical conditions are characterized by 'pulsating' and 'rotating' modes of spatio-temporal convection. The development of counter-propagating hydrothermal waves in azimuthal direction is found to be followed by an even mode of convection characterised by m = 2. The liquid bridge system exhibiting the well-known transition from periodic (m =2) to quasi-periodic behaviour (m =1) with an increase in Biot number, Bi has been analysed from both spatial and temporal points of view. The existence of both symmetric and asymmetric modes of the supercritical state for different Bi are presented. The dominant fluid dynamic information of 3D oscillatory state are extracted from a specific set of disturbance temperature data by employing a data-driven computational technique 'dynamic mode decomposition' (DMD). Various neutral, stable and unstable dynamic modes responsible for bifurcation to the 3D supercritical state are depicted with the help of DMD spectrum and the frequency spectrum. The CALorimetric Electron Telescope (CALET) space experiment, developed by Japan in collaboration with Italy and the United States, is a high-energy astroparticle physics mission installed on the International Space Station (ISS). The primary goals of the CALET mission include investigating on the possible presence of nearby sources of high-energy electrons, studying the details of galactic particle propagation and searching for dark matter signatures. During a two-year mission, extendable to five years, CALET can measure the flux of cosmic-ray electrons (including positrons) to 20 TeV, gamma-rays to 10 TeV and nuclei with Z = 1 to 40 up to 1,000 TeV. The instrument consists of two layers of segmented plastic scintillators for cosmic-ray charge identification (CHD), a 3 radiation length thick tungsten-scintillating fiber imaging calorimeter (IMC) and a 27 radiation length thick lead-tungstate calorimeter (TASC). CALET has sufficient depth, imaging capabilities and excellent energy resolution to allow for a clear separation between hadrons and electrons and between charged particles and gamma rays. The instrument was launched on August 19, 2015 to the ISS with the H-II Transfer Vehicle 5 (HTV-5) and installed on the Japanese Experiment Module-Exposed Facility (JEM-EF) on August 25. Since the start of operations in mid-October, 2015, a continuous observation has been going on mainly by triggering high energy (>10 GeV) showers without any major interruption. The number of triggered events above 10 GeV is nearly 20 million per month. By using the data obtained during the first two years, we give a summary of CALET observations: (1) Electron + Positron energy spectrum, (2) Proton and Nuclei spectrum, (3) Gamma-ray observation, with results of the performance study on orbit. We also present the results of observations of the electromagnetic counterparts to LIGO-VIRGO gravitational wave events and high-energy counterparts to GRB events measured with the CALET Gamma-ray Burst Monitor (CGBM). Abstract A plant growth system for crop production under microgravity is part of a life supporting system designed for long-duration space missions. A plant growth in soil in space requires the understanding of water movement in soil void spaces under microgravity. Under 1G-force condition, on earth, water movement in porous media is driven by gradients of matric and gravitational potentials. Under microgravity condition, water movement in porous media is supposed to be driven only by a matric potential gradient, but it is still not well understood. We hypothesized that under microgravity water in void spaces of porous media hardly moved comparing in void spaces without obstacles because the concave surfaces of the porous media hindered water movement. The objective of this study was to investigate water movement on the convex surfaces of porous media under microgravity. We conducted parabolic flight experiments that provided 20–25 s of microgravity at the top of a parabolic flight. We observed water movement in void spaces in soil-like porous media made by glass beads and glass spheres (round-bottomed glass flasks) in the different conditions of water injection under microgravity. Without water injection, water did not move much in neither glass beads nor glass spheres. When water was injected during microgravity, water accumulated in contacts between the particles, and the water made thick fluid films on the particles surface. When the water injection was stopped under microgravity, water was held in the contacts between the particles. This study showed that water did not move upward in the void spaces with or without the water injection. In addition, our results suggested that the difficulty of water movement on the convex (i.e. particle surfaces) might result in slower water move in porous media under microgravity than at 1G-force. Highlights • We have identified the necessary modifications to enable CubeSat deep space operations. • ASPECT (Asteroid Spectral Imaging Mission) is a deep space CubeSat technology demonstrator. • ASPECT can deliver valuable technology and scientific data at high performance to mass and cost ratios. Abstract Operation of a small CubeSat in the deep-space microgravity environment brings additional challenging factors including the increased radiation environment, the significant contribution of non-gravitational forces to the satellite orbit, or the limited communication opportunities. These factors need to be taken into account in the form of modifications to the classic CubeSat architecture. Increased radiation resistance, the semi-autonomous satellite operation, navigation, and the active orbit correction are required. Such a modified CubeSat platform can potentially deliver a high performance to mass and cost ratios. The Asteroid Spectral Imaging Mission (ASPECT) is a three unit (3U) CubeSat mission built on these principles. It is part of the AIDA (Asteroid Impact & Deflection Assessment) project to the binary asteroid Didymos. ASPECT is equipped with a visible to near-infrared hyperspectral imager and will deliver both technological knowledge as well as scientific data about the origin and evolution of Solar System small bodies. Through the striving of humanity into space, new production processes and technologies for the use under microgravity will be essential in the future. Production of objects in space demands for new processes, like additive manufacturing. This paper presents the concept and the realization for a new machine to investigate microgravity production processes on earth. The machine is based on linear long stator drives and a vacuum chamber carrying up to 1000 kg. For the first time high repetition rate and associated low experimental costs can provide basic research. The GeoFlow experiment on the ISS is designed to study convective flows in a spherical gap under microgravity conditions. The main challenge, however, is the visualization of the fluid flow especially under the safety requirements of the Columbus module. The Wollaston shearing interferometry unit of the Fluid Science Laboratory works by optical means alone and is therefore utilized as measurement device for temperature fluctuations. The resulting interferograms in terms of fringe patterns are the base for the presented advanced post-processing techniques. They are used to identify convective patterns, to track these structures and to reconstruct the inaccessible three-dimensional temperature field. A comparison between experimentally gained results and numerically calculated interferograms is given, too. We show that convective patterns are automatically recognized and tracked accurately in experimental images by means of the generalized structure tensor. Furthermore, generic numerical simulations are used to deduce the internal temperature distribution by comparison with interferograms from the experiment. [ABSTRACT FROM AUTHOR]

Freely suspended films of smectic liquid crystals are unique examples of quasi two-dimensional fluids. Mechanically stable and with quantized thickness of the order of only a few molecular layers, smectic films are ideal systems for studying fundamental fluid physics, such as collective molecular ordering, defect and fluctuation phenomena, hydrodynamics, and nonequilibrium behavior in two dimensions (2D), including serving as models of complex biological membranes. Smectic films can be drawn across openings in planar supports resulting in thin, meniscus-bounded membranes, and can also be prepared as bubbles, either supported on an inflation tube or floating freely. The quantized layering renders smectic films uniquely useful in 2D fluid physics. The OASIS team has pursued a variety of ground-based and microgravity applications of thin liquid crystal films to fluid structure and hydrodynamic problems in 2D and quasi-2D systems. Parabolic flights and sounding rocket experiments were carried out in order to explore the shape evolution of free floating smectic bubbles, and to probe Marangoni effects in flat films. The dynamics of emulsions of smectic islands (thicker regions on thin background films) and of microdroplet inclusions in spherical films, as well as thermocapillary effects, were studied over extended periods within the OASIS (Observation and Analysis of Smectic Islands in Space) project on the International Space Station. The paper presents a new physical model describing the processes in materials and electronic devices under the influence of cosmic rays in microgravity. The model identifies specific features of formation of the area of radiation defects (ARD) in the electronic materials in microgravity. The mechanism of interaction between the ARD and memory modules in microgravity causing malfunction and failure of onboard electronics is considered. Experiments with granular gases do not only provide statistical information on multi-particle systems undergoing random dissipative interactions, they may also help to test predictions of numerical simulations and to gain understanding of the self-organization of dilute granular matter to clusters and stable assemblies. We shortly review an experiment under zero gravity conditions on different platforms. Implementations on a sounding rocket flight, parabolic flights and drop tower shots are analyzed. The effect of gravity on plant growth is an interesting topic in its own right, but it is also important because it impacts the possibility of long-term space travel. Plants may be grown in microgravity simulated by diamagnetic levitation within superconducting magnet, but this approach is limited by the size and other objective conditions of the superconducting magnet. Tremendous difficulties exist in evaluating the effects of simulated microgravity on plant seedling growth under lighting conditions. Therefore, we developed a lighting system and culturing system that can meet the demands of growing plant seedlings in a superconducting magnet. This system mainly consists of an illumination system, suitable containers and a method to cultivate Arabidopsis thaliana seedlings. In order to prove the suitability of this light-growing system, A . thaliana was cultured in a superconducting magnet for four days. The status of seedlings was recorded and total RNA was extracted for gene expression analysis. Our results showed that Arabidopsis seedlings could germinate and grow successfully in this light-growing system. In addition, it was observed that under diamagnetic levitation conditions, the seedling bended and gene expression of PGM and MOR1 decreased significantly compared to a control group. Nonetheless, there were no substantial differences between the diamagnetic levitation group and RPM group. Our results suggest that this light-growing system is expedient and beneficial for plants grown in a superconducting magnet. Our experiment also provides a way to utilize diamagnetic levitation in a superconducting magnet that simulates the conditions necessary to study plant physiology and biochemical responses in a microgravity environment. Abstract: The microgravity environment of spaceflight expeditions has been associated with altered microbial responses. This study explores the characterization of Serratia marcescensis grown in a spaceflight environment at the phenotypic, transcriptomic and proteomic levels. From November 1, 2011 to November 17, 2011, a strain of S. marcescensis was sent into space for 398h on the Shenzhou VIII spacecraft, and ground simulation was performed as a control (LCT-SM213). After the flight, two mutant strains (LCT-SM166 and LCT-SM262) were selected for further analysis. Although no changes in the morphology, post-culture growth kinetics, hemolysis or antibiotic sensitivity were observed, the two mutant strains exhibited significant changes in their metabolic profiles after exposure to spaceflight. Enrichment analysis of the transcriptome showed that the differentially expressed genes of the two spaceflight strains and the ground control strain mainly included those involved in metabolism and degradation. The proteome revealed that changes at the protein level were also associated with metabolic functions, such as glycolysis/gluconeogenesis, pyruvate metabolism, arginine and proline metabolism and the degradation of valine, leucine and isoleucine. In summary S. marcescens showed alterations primarily in genes and proteins that were associated with metabolism under spaceflight conditions, which gave us valuable clues for future research. Abstract: An experiment in microgravity conditions aboard the TEXUS-23 mission was performed to obtain the deformation up to breakage of a cylindrical liquid column in isorotation around an eccentric axis. In previous work, breakage rotation speed was predicted by a numerical method. This method was validated by comparison with analytical and experimental results. The non-symmetric breakage of the liquid column observed in the experiment, however, was not explained by the combined effect of rotation and eccentricity. In this work we use an extension of that method that includes an axial gravitational field. Thus the combined effect of rotation, eccentricity and axial gravity in the stability threshold and in the shape of the liquid bridge is analyzed. Abstract: Vegetable cultivation plays a crucial role in dietary supplements and psychosocial benefits of the crew during manned space flight. Here we developed a ground-based prototype of horn-type sequential vegetable production facility, named Horn-type Producer (HTP), which was capable of simulating the microgravity effect and the continuous cultivation of leaf–vegetables on root modules. The growth chamber of the facility had a volume of 0.12m3, characterized by a three-stage space expansion with plant growth. The planting surface of 0.154m2 was comprised of six ring-shaped root modules with a fibrous ion-exchange resin substrate. Root modules were fastened to a central porous tube supplying water, and moved forward with plant growth. The total illuminated crop area of 0.567m2 was provided by a combination of red and white light emitting diodes on the internal surfaces. In tests with a 24-h photoperiod, the productivity of the HTP at 0.3kW for lettuce achieved 254.3g eatable biomass per week. Long-term operation of the HTP did not alter vegetable nutrition composition to any great extent. Furthermore, the efficiency of the HTP, based on the Q-criterion, was 7×10−4 g2 m−3 J−1. These results show that the HTP exhibited high productivity, stable quality, and good efficiency in the process of planting lettuce, indicative of an interesting design for space vegetable production. Abstract: Numerous studies have indicated that the microgravity environment of space has harmful effects on several tissues throughout the body. Although this phenomenon is well documented, research in this area is still in its relative infancy. This study investigates the effects of space flight on mucin production of the uterine tubes of mice. This study examined the epithelium of the uterine tubes from female mice that were flown on the space shuttle Endeavour for 13days in August, 2007 and their concomitant controls. The tissue was qualitatively analyzed for the type of mucin produced, i.e., acidic, neutral, acidic/neutral mixture. Further, the tissue was quantitatively analyzed for the amounts of mucins produced by measuring the thickness of the mucin layer for each region of the uterine tube: isthmus, ampulla, and infundibulum. One way ANOVA tests were used to compare mucin thickness between all three sets of animals. Results indicate similar but not identical results between the three regions of the uterine tube. The Baseline tissue had the thickest mucin layer regardless of treatment group. In the ampulla the mucin layer was the thinnest in the Flight tissue, followed by the Ground Control, with the Baseline being the thickest. Analysis of the mucin layer of the infundibulum of the three treatment groups indicated no difference in its thickness between the three regions of the uterine tube. These results indicate a trend toward thinning of the mucin layer of the uterine tube in space flight, but also indicate an influence by the housing environment. [Copyright &y& Elsevier]

Jatropha (Jatropha curcas) is a tropical perennial species identified as a potential biofuel crop. The oil is of excellent quality and it has been successfully tested as biodiesel and in jet fuel mixes. However, studies on breeding and genetic improvement of jatropha are limited. Space offers a unique environment for experiments aiming at the assessment of mutations and differential gene expression of crops and in vitro cultures of plants are convenient for studies of genetic variation as affected by microgravity. However, before microgravity studies can be successfully performed, pre-flight experiments are necessary to characterize plant material and validate flight hardware environmental conditions. Such preliminary studies set the ground for subsequent spaceflight experiments. The objectives of this study were to compare the in vitro growth of cultures from three explant sources (cotyledon, leaf, and stem sections) of three jatropha accessions (Brazil, India, and Tanzania) outside and inside the petriGAP, a modified group activation pack (GAP) flight hardware to fit petri dishes. In vitro jatropha cell cultures were established in petri dishes containing a modified MS medium and maintained in a plant growth chamber at 25±2°C in the dark. Parameters evaluated were surface area of the explant tissue (A), fresh weight (FW), and dry weight (DW) for a period of 12weeks. Growth was observed for cultures from all accessions at week 12, including subsequent plantlet regeneration. For all accessions differences in A, FW and DW were observed for inside vs. outside the PetriGAPs. Growth parameters were affected by accession (genotype), explant type, and environment. The type of explant influenced the type of cell growth and subsequent plantlet regeneration capacity. However, overall cell growth showed no abnormalities. The present study demonstrated that jatropha in vitro cell cultures are suitable for growth inside PetriGAPs for a period of 12weeks. The parameters evaluated in this study provide the basic ground work and pre-flight assessment needed to justify a model for microgravity studies with jatropha in vitro cell cultures. Abstract: The bone loss induced by microgravity is partly due to the decrease of mature osteoblasts. In the present study, we employed the random positioning machine (RPM) to simulate microgravity and investigated the acute effects of simulated microgravity on the differentiation of 2T3 preosteoblasts. Following 7days’ culture under normal (1 g) condition, cells were exposed to simulated microgravity for 24h. The results showed that 24h treatment of simulated microgravity significantly decreased alkaline phosphatase (ALP) activity without changing the cell morphology. In addition, the mRNA expressions of osteogenic genes, including runt-related gene 2 (Runx2), osterix, osteocalcin (OC), type I collagen (Col I) and bone morphogenetic protein (BMP), were dramatically downregulated. Moreover, western blot analysis of total extracellular signal-regulated kinase (Erk) and phosphorylated Erk (p-Erk) indicated that p-Erk level, which represents the Erk activation status, was increased. Taken together, our results suggested that acute exposure to simulated microgravity inhibited osteoblast differentiation through modulating the expression of osteogenic genes and the Erk activity. Abstract: Arabidopsis gene expression microarray (44K) was used to detect genes highly induced under simulated microgravity stress (SMS). Ten SMS-inducible genes were selected from the microarray data and these 10 genes were found to be abundantly expressed in 3-week-old plants. Nine out of the 10 SMS-inducible genes were also expressed in response to the three abiotic stresses of drought, touch, and wounding in 3-week-old Arabidopsis plants respectively. However, WRKY46 was elevated only in response to SMS. Six other WRKY genes did not respond to SMS. To clarify the characteristics of the genes expressed at high levels in response to SMS, 20 cis-elements in the promoters of the 40 selected genes including the 10 SMS-inducible genes, the 6 WRKY genes, and abiotic stress-inducible genes were analyzed and their spatial positions on each promoter were determined. Four cis-elements (M/T-G-T-P from MYB1AT or TATABOX5, GT1CONSENSUS, TATABOX5, and POLASIG1) showed a unique spatial arrangement in most SMS-inducible genes including WRKY46. Therefore the M/T-G-T-P cis-element patterns identified in the promoter of WRKY46 may play important roles in regulating gene expression in response to SMS. The presences of the cis-element patterns suggest that the order or spatial positioning of certain groups of cis-elements is more important than the existence or numbers of specific cis-elements. Taken together, our data indicate that WRKY46 is a novel SMS inducible transcription factor and the unique spatial arrangement of cis-elements shown in WRKY46 promoter may play an important role for its response to SMS. Abstract: This study investigated intracellular oxidative stress and its underlying mechanisms in a rotary cell culture system used to achieve a simulated microgravity (SMG) environment. Experiments were conducted with human breast cancer cell lines MCF-7 (an estrogen receptor (ER) α positive cell line) and MDA-MB-231 (an ERα negative cell line) encapsulated in alginate/collagen carriers. After 48h, SMG led to oxidative stress and DNA damage in the MDA-MB-231 cells but a significant increase in mitochondrial activity and minimal DNA damage in the MCF-7 cells. The activity of superoxide dismutase (SOD) significantly increased in the MCF-7 cells and decreased in MDA-MB-231 cells in the SMG environment compared with a standard gravity control. Moreover, SMG promoted expression of ERα and protein kinase C (PKC) epsilon in MCF-7 cells treated with PKC inhibitor Gö6983. Overall, exposure to SMG increased mitochondrial activity in ERα positive cells but induced cellular oxidative damage in ERα negative cells. Abstract: The effects of real and simulated microgravity on the eye tissue regeneration of newts were investigated. For the first time changes in Müller glial cells in the retina of eyes regenerating after retinal detachment were detected in newts exposed to clinorotation. The cells divided, were hypertrophied, and their processes were thickened. Such changes suggested reactive gliosis and were more significant in animals exposed to rotation when compared with desk-top controls. Later experiments onboard the Russian biosatellite Bion-11 showed similar changes in the retinas that were regenerating in a two-week spaceflight. In the Bion-11 animals, GFAP, the major structural protein of retinal macroglial cells, was found to be upregulated. In a more recent experiment onboard Foton-M3 (2007), GFAP expression in retinas of space-flown, ground control (kept at 1 g), and basal control (sacrificed on launch day) newts was quantified, using microscopy, immunohistochemistry, and digital image analysis. A low level of immunoreactivity was observed in basal controls. In contrast, retinas of space-flown animals showed greater GFAP immunoreactivity associated with both an increased cell number and a higher thickness of intermediate filaments. This, in turn, was accompanied by up-regulation of stress protein (HSP90) and growth factor (FGF2) expressions. It can be postulated that such a response of Müller cells was to mitigate the retinal stress in newts exposed to microgravity. Taken together, the data suggest that the retinal population of macroglial cells could be sensitive to gravity changes and that in space it can react by enhancing its neuroprotective function. Abstract: All life on earth is accustomed to the presence of gravity. When gravity is altered, biological processes can go awry. It is of great importance to ensure safety during a spaceflight. Long term exposure to microgravity can trigger detrimental physiological responses in the human body. Fluid redistribution coupled with fluid loss is one of the effects. In particular, in microgravity blood volume is shifted towards the thorax and head. Sympathetic nervous system-induced vasoconstriction is needed to maintain arterial pressure, while venoconstriction limits venous pooling of blood prevents further reductions in venous return of blood to the heart. In this paper, we modify an existing one dimensional blood flow model with the inclusion of the hydrostatic pressure gradient that further depends on the gravitational field modified by the oblateness and rotation of the Earth. We find that the velocity of the blood flow V B is inversely proportional to the blood specific volume d, also proportional to the oblateness harmonic coefficient J 2, the angular velocity of the Earth ω E , and finally proportional to an arbitrary constant c. For c =−0.39073 and ξ H =−0.5mmHg, all orbits result to less blood flow velocities than that calculated on the surface of the Earth. From all considered orbits, elliptical polar orbit of eccentricity e =0.2 exhibit the largest flow velocity V B =1.031m/s, followed by the orbits of inclination i =45°and 0°. Abstract: Simulated microgravity (SMG) can inhibit proliferation and enhance microcystin production of Microcystis aeruginosa. We investigated the role of nitric oxide (NO) in regulating the SMG induced changes of proliferation, photochemical system II photochemical activity, pigment, soluble protein and microcystin production in M. aeruginosa. M. aeruginosa was exposed to 0.1mM sodium nitroprusside (SNP, NO donor) or 0.02mM 2-(4-carboxyphenyl)-4, 4, 5, 5-tetramethylimidazoline-1-oxyl-3-oxide (c-PTIO, NO scavenger) alone or in combination with SMG for 48h. SMG and SNP inhibited the growth of M. aeruginosa while c-PTIO had no effect on cell number. As to yield, the negative effect of SMG was augmented by SNP and suppressed by c-PTIO. The intracellular concentrations of chlorophyll a, carotenoid, phycocyanin, soluble protein and microcystin were increased by SMG after 48h. The effects of SMG on these metabolic processes could be enhanced by SNP and be partly eliminated by c-PTIO. Moreover, SNP and c-PTIO only functioned in these biochemical processes under SMG, unlike in the regulation of cell proliferation and yield. These results showed that the effects of SMG could be enhanced by adding exogenous NO and be mitigated by scavenging endogenous NO, revealing the involvement of NO in the changes in biochemistry processes induced by SMG in M. aeruginosa. Abstract: Space flight has been shown to have many adverse effects on various systems throughout the body. Because the opportunity to place research animals on board a Space Shuttle or the International Space Station is infrequent, various techniques have been designed to simulate the effects of microgravity in Earth based laboratories. A commonly used technique is known as antiorthostatic suspension, also often referred to as hind limb suspension. In this technique the hind portion of the animal is raised so that its hind limbs are non-weight bearing. This places the animal in roughly a 30° head down tilt position. This results in cephalic fluid shifts similar to those seen in actual space flight. This technique has also been shown to mimic other physiological parameters that are affected during space flight. This study examined testicular tissue from rats subjected to a 7 day antiorthostatic suspension. This tissue was acquired through a tissue sharing program and some of the experimental animals were injected with Interleukin 1 receptor antagonist (IL-1ra) which was hoped to ameliorate some of the effects of antiorthostatic suspension. The injection of IL-1ra was not expected to have any effect on testicular tissue, however this tissue was included in the morphological and statistical analysis to conduct a more complete study. All tissues were embedded in paraffin, sectioned, and stained using standard H&E staining. The tissue was then qualitatively ranked according to the “health” of the seminiferous tubules. Our findings indicate that 7 days of antiorthostatic suspension had adverse effects on the tissue that comprises the walls of the seminiferous tubules. It has long been known that antiorthostatic suspension has deleterious effects on testicular tissue, however this research indicates that these effects occur much faster than indicated by previous researchers. This is a significant finding because it indicates that meaningful earth based studies in this area can be carried out in a shorter time span. This could result in more studies per year as well as saving money by avoiding longer than necessary animal suspensions. This is especially important as we enter an era when, without Space Shuttle, flight opportunities will become scarce. These antiorthostatic suspension studies indicate that space flight, even short duration spaceflight, may have harmful effects on the seminiferous tubules and blood-testis barrier of astronauts. Abstract: In view of the concern for the health of astronauts that may one day journey to Mars or the Moon, we investigated the effect that space radiation and microgravity might have on DNA damage and repair. We sent frozen human lymphoblastoid TK6 cells to the International Space Station where they were maintained under frozen conditions during a 134-day mission (14 November 2008 to 28 March 2009) except for an incubation period of 8 days under 1G or μG conditions in a CO2 incubator. The incubation period started after 100 days during which the cells had been exposed to 54mSv of space radiation. The incubated cells were then refrozen, returned to Earth, and compared to ground control samples for the determination of the influence of microgravity on cell survival and mutation induction. The results for both varied from experiment to experiment, yielding a large SD, but the μG sample results differed significantly from the 1G sample results for each of 2 experiments, with the mean ratio of μG to 1G being 0.55 for the concentration of viable cells and 0.59 for the fraction of thymidine kinase deficient (TK−) mutants. Among the mutants, non-loss of zygosity events (point mutations) were less frequent (31%) after μG incubation than after 1G incubation, which might be explained by the influence of μG on cellular metabolic or physiological function. Abstract: The calculation of two-phase frictional pressure drop (TPFPD) is required by two-phase systems operating under microgravity and reduced gravity. There are a large number of correlations for the TPFPD in tubes under normal gravity. However, it is hard to find out a TPFPD correlation obtained from microgravity and/or reduced gravity conditions, and thus people have to use TPFPD correlations for normal gravity to calculate TPFPD under microgravity and reduced gravity. It is necessary to evaluate the feasibility of such practice. This paper offers a comprehensive review of the TPFPD correlations for normal gravity and an up-to-data survey of the TPFPD experimental study under microgravity and reduced gravity. There are 23 TPFPD correlations for normal gravity reviewed and 135 experimental data under microgravity obtained from the literature. These experimental data are used to evaluate the reviewed TPFPD correlations. It is found that the smallest mean absolute relative deviation (MARD) of the correlations is greater than 34%. Using TPFPD correlations for normal gravity to reduced gravity and microgravity may be acceptable for the first approximation, but correlations intended for microgravity and reduced gravity are needed and more experiments are desired to obtain more data with high accuracy. Abstract: In microgravity, one of the major challenge encountered in biological life support systems (BLSS) is the gas–liquid transfer with, for instance, the necessity to provide CO2 (carbon source, pH control) and to recover the evolved O2 in photobioreactors used as atmosphere bioregenerative systems. This paper describes first the development of a system enabling the accurate characterization of the mass transfer limiting step for a PTFE membrane module used as a possible efficient solution to the microgravity gas–liquid transfer. This original technical apparatus, together with a technical assessment of membrane permeability to different gases, is associated with a balance model, determining thus completely the CO2 mass transfer problem between phases. First results are given and discussed for the CO2 mass transfer coefficient obtained in case of absorption experiments at pH 8 using the hollow fiber membrane module. The consistency of the proposed method, based on a gas and liquid phase balances verifying carbon conservation enables a very accurate determination of the value as a main limiting step of the whole process. Nevertheless, further experiments are still needed to demonstrate that the proposed method could serve in the future as reference method for mass transfer coefficient determination if using membrane modules for BLSS in reduced or microgravity conditions. Abstract: Animal models are frequently used to assist in the determination of the long- and short-term effects of space flight. The space environment, including microgravity, can impact many physiological and immunological system parameters. It has been found that ground based models of microgravity produce changes in white blood cell counts, which negatively affects immunologic function. As part of the Center of Acute Radiation Research (CARR), we compared the acute effects on white blood cell parameters induced by the more traditionally used animal model of hindlimb unloading (HU) with a recently developed reduced weightbearing analog known as partial weight suspension (PWS). Female ICR mice were either hindlimb unloaded or placed in the PWS system at 16% quadrupedal weightbearing for 4h, 1, 2, 7 or 10days, at which point complete blood counts were obtained. Control animals (jacketed and non-jacketed) were exposed to identical conditions without reduced weightbearing. Results indicate that significant changes in total white blood cell (WBC), neutrophil, lymphocyte, monocyte and eosinophil counts were observed within the first 2days of exposure to each system. These differences in blood cell counts normalized by day 7 in both systems. The results of these studies indicate that there are some statistically significant changes observed in the blood cell counts for animals exposed to both the PWS and HU simulated microgravity systems. Abstract: Single cell was capable of sensing and responding to alterations of gravity. Osteocytes, as the most abundant cells of the bone tissue playing an important role in the bone mechanotransduction, are very sensitive to mechanical stimuli. However, the effect of altered gravity on osteocytes so far is less known according to the public papers. Further study on this issue will help to verify and develop the theory of how cells perceive and respond to gravity. It also brings new ideas to the study of space bone loss. In our study, Osteocyte-like MLO-Y4 cells were exposed to 30 parabolic flights three times on ZERO-G airbus A300 to investigate the comprehensive effect on osteocytes stimulated by hyper- and hypo-gravity forces. It showed that the cell morphology, as well as cell area and height, was not changed significantly by hyper-gravity and hypo-gravity. However, the cytoskeleton was reorganized. In flight cells, F-actin polymerization was enhanced at the cell periphery and microtubule organizing center disappeared, but no apoptotic feathers were detected. The results of western blot showed that connexin 43 (Cx43) expression was down-regulated, indicating an decrease of gap-junction. In conclusion, hyper- and hypo-gravity stimulation altered the cytoskeleton architecture and suppressed gap-junction of osteocyte-like MLO-Y4 cells. [Copyright &y& Elsevier]

Abstract: The Gravity Recovery and Climate Experiment (GRACE) satellite mission has been estimating temporal changes in the Earth’s gravitational field since its launch in 2002. While it is not yet fully resolved what the limiting source of error is for GRACE, studies on future missions have shown that temporal aliasing errors due to undersampling signals of interest (such as hydrological variations) and errors in atmospheric, ocean, and tide models will be a limiting source of error for missions taking advantage of improved technologies (flying drag-free with a laser interferometer). This paper explores the option of reducing the effects of temporal aliasing errors by directly estimating low degree and order gravity fields at short time intervals, ultimately resulting in data products with improved spatial resolution. Three potential architectures are considered: a single pair of polar orbiting satellites, two pairs of polar orbiting satellites, and a polar orbiting pair of satellites coupled with a lower inclined pair of satellites. Results show that improvements in spatial resolution are obtained when one estimates a low resolution gravity field every two days for the case of a single pair of satellites, and every day for the case of two polar pairs of satellites. However, the spatial resolution for these cases is still lower than that provided by simply destriping and smoothing the solutions via standard GRACE post-processing techniques. Alternately, estimating daily gravity fields for the case of a polar pair of satellites coupled with a lower inclined pair results in solutions with superior spatial resolution than that offered by simply destriping and smoothing the solutions. Abstract: Primordial germ cells (PGCs), precursors of germline cells, display a variety of antigens during their migration to target gonads. Here, we used silk chicken offspring (Gallus gallus domesticus) embryos subjected to space microgravity to investigate the influence of microgravity on PGCs. The ShenZhou-3 unmanned spaceship carried nine fertilized silk chicken eggs, named the flight group, returned to Earth after 7 days space flight. And the control group has the same clan with the flight group. PGCs from flight and control group silk chicken offspring embryos were examined during migration by using two antibodies (2C9 and anti-SSEA-1), in combination with the horseradish peroxidase detection system, and using periodic acid-Schiff’s solution (PAS) reaction. After incubation for about 30h, SSEA-1 and 2C9 positive cells were detected in the germinal crescent of flight and control group silk chicken offspring embryos. After incubation of eggs for 2–2.5 days, SSEA-1 and 2C9 positive cells were detected in embryonic blood vessels of flight and control group silk chicken offspring embryos. After incubation of eggs for 5.5 days, PGCs in the dorsal mesentery and gonad could also be identified in flight and control group silk chicken offspring embryos by using SSEA-1 and 2C9 antibodies. Based on location and PAS staining, these cells were identified as PGCs. Meanwhile, at the stage of PGCs migration and then becoming established in the germinal ridges, no difference in SSEA-1 or 2C9 staining was detected between female and male PGCs in flight and control group silk chicken offspring embryos. Although there were differences in the profiles of PGC concentration between male and female embryos during the special circulating stage, changing profile of PGCs concentration was similar in same sex between flight and control group offspring embryos. The configuration consisted of a liquid bridge between two solid supporting discs. An expected breakage occurred during the experiment. The recorded images are analysed and the measured behaviour compared with the results of a three dimensional model of the liquid dynamics, obtaining a much better fit than with linear models. Abstract: Hematopoietic progenitor cell proliferation can be altered in either spaceflight or under simulated microgravity experiments on the ground, however, the underlying mechanism remains unknown. Our previous study showed that exposure of the human erythropoietin (EPO)-dependent leukemia cell line UT-7/EPO to conditions of simulated microgravity significantly inhibited the cellular proliferation rate and induced cell apoptosis. We postulated that the downregulation of the erythropoietin receptor (EPOR) expression in UT-7/EPO cells under simulated microgravity may be a possible reason for microgravity triggered apoptosis. In this paper, a human EPOR gene was transferred into UT-7/EPO cells and the resulting expression of EPOR on the surface of UT-7/EPO cells increased approximately 61% (p <0.05) as selected by the antibiotic G418. It was also shown through cytometry assays and morphological observations that microgravity-induced apoptosis markedly decreased in these UT-7/EPO–EPOR cells. Thus, we concluded that upregulation of EPOR in UT-7/EPO cells could inhibit the simulated microgravity-induced cell apoptosis in this EPO dependent cell line. [Copyright &y& Elsevier]

Abstract: The budding yeast Saccharomyces cerevisiae has been proposed as an ideal model organism for clarifying the biological effects caused by spaceflight conditions. The postmitotic S. cerevisiae cells onboard Practice eight recoverable satellite were subjected to spaceflight for 15days. After recovery, the viability, the glycogen content, the activities of carbohydrate metabolism enzymes, the DNA content and the lipid peroxidation level in yeast cells were analyzed. The viability of the postmitotic yeast cells after spaceflight showed a three-fold increase as compared with that of the ground control cells. Compared to the ground control cells, the lipid peroxidation level in the spaceflight yeast cells markedly decreased. The spaceflight yeast cells also showed an increase in G2/M cell population and a decrease in Sub-G1 cell population. The glycogen content and the activities of hexokinase and succinate dehydrogenase significantly decreased in the yeast cells after spaceflight. In contrast, the activity of malate dehydrogenase showed an obvious increase after spaceflight. These results suggested that microgravity or spaceflight could promote the survival of postmitotic S. cerevisiae cells through regulating carbohydrate metabolism, ROS level and cell cycle progression. Abstract: The effects of gravity on bone metabolism are unclear, and little has been reported about the effects of hypergravity on the mature skeleton. Since low gravity has been shown to decrease bone volume, we hypothesized that hypergravity increases bone volume. To clarify this hypothesis, adult female rats were ovariectomized and exposed to hypergravity (2.9G) using a centrifugation system. The rats were killed 28days after the start of loading, and the distal femoral metaphysis of the rats was studied. Bone architecture was assessed by micro-computed tomography (micro-CT) and bone mineral density was measured using peripheral quantitative CT (pQCT). Hypergravity increased the trabecular bone volume of ovariectomized rats. Histomorphometric analyses revealed that hypergravity suppressed both bone formation and resorption and increased bone volume in ovariectomized rats. Further, the cell morphology, activity, proliferation, and differentiation of osteoclasts and osteoblasts exposed to hypergravity were evaluated in vitro. Hypergravity inhibited actin ring formation in mature osteoclasts, which suggested that the osteoclast activity was suppressed. However, hypergravity had no effect on osteoblasts. These results suggest that hypergravity can stimulate an increase in bone volume by suppressing bone resorption in ovariectomized rats. [Copyright &y& Elsevier]

Abstract: HORACE (HOrloge à Refroidissement d’Atomes en Cellule=clock based on atoms cooled from vapour cell) is a compact cold caesium atom clock developed in SYRTE at Paris Observatory. This clock can operate both on ground and in microgravity environment. Design of HORACE is based on isotropic light cooling, allowing performing the whole clock sequence (cooling, atomic preparation, Ramsey interrogation and detection) at the same place. Compared to more conventional cold atom clocks such as atomic fountains, the use of isotropic light cooling simplifies both the optical part of the setup and the detection sequence, and leads to a drastic size reduction of the physics package. Very good short-term performances have been demonstrated at SYRTE since relative frequency instability of 2.2×10−13 τ −1/2 has been obtained. Optimization of the long term stability is still under progress and current results show relative frequency instability around 3×10−15 in 104 s of integration. Abstract: With the advent of space flights questions concerning the effects of microgravity (0×G) on human reproductive physiology have received great attention. The aim of this study was to evaluate the influence of 0×G on Sertoli cells. A Sertoli cell line from mouse testis (42GPA9) was analyzed for cytoskeletal and Sex Hormone Binding Globilin (SHBG) changes by immunohistochemistry, for antioxidant content by RT-PCR and for culture medium lactate concentrations by protein chemistry. Cells were cultured for 6, 24 and 48h on a three-dimensional Random Positioning Machine (3D-RPM); static controls (1×G) were positioned on the supporting frame. At the end of each experiment, cultured cells were either fixed in paraformaldehyde or lysed and RNA-extracted or used for culture medium lactate measurements as needed. At 0×G, Sertoli cytoskeleton became disorganized, microtubules fragmented and SHBG undetectable already after 24h, with alterations worsening by 48h. It was evident that various antioxidant systems appreciably increased during the first 24h but significantly decreased at 48h. No changes occurred in the 1×G samples. Initially, 0×G seemed to disturb antioxidant protection strategies allowing the testes to support sperm production, thus generating an aging-like state of oxidative stress. Lactate production at 0×G slightly decreased after 24h. Further experiments are needed in space to investigate upon steroidogenesis and germ cell differentiation within the testis, to rule out male infertility as a possible consequence, which could be a problem, as life expectancy increases. Abstract: In order to effectively study phototropism, the directed growth in response to light, we performed a series of experiments in microgravity to better understand light response without the “complications” of a 1-g stimulus. These experiments were named TROPI (for tropisms) and were performed on the European Modular Cultivation System (EMCS), a laboratory facility on the International Space Station (ISS). TROPI-1 was performed in 2006, and while it was a successful experiment, there were a number of technical difficulties. We had the opportunity to perform TROPI-2 in 2010 and were able to optimize experimental conditions as well as to extend the studies of phototropism to fractional gravity created by the EMCS centrifuge. This paper focuses on how the technical improvements in TROPI-2 allowed for a better experiment with increased scientific return. Major modifications in TROPI-2 compared to TROPI-1 included the use of spaceflight hardware that was off-gassed for a longer period and reduced seed storage (less than 2months) in hardware. These changes resulted in increased seed germination and more vigorous growth of seedlings. While phototropism in response to red illumination was observed in hypocotyls of seedlings grown in microgravity during TROPI-1, there was a greater magnitude of red-light-based phototropic curvature in TROPI-2. Direct downlinking of digital images from the ISS in TROPI-2, rather than the use of analog tapes in TROPI-1, resulted in better quality images and simplified data analyses. In TROPI-2, improved cryo-procedures and the use of the GLACIER freezer during transport of samples back to Earth maintained the low temperature necessary to obtain good-quality RNA required for use in gene profiling studies. Abstract: Gene expression analysis using microarrays has proved to be an important method in life science. The opportunity to grow higher plants on the International Space Station (ISS) opens up the possibility for gene expression profiling of plants grown in microgravity. The work presented focuses on how to meet the scientific requirements of plant growth and the sample preservation, given the technical and operational constraints associated with space research. The growth chamber (Multigen-2 Science Testing Unit) and a protocol suggested to be used in the European Modular Cultivation System (EMCS) Multigen-2 experiment on the ISS to grow and later preserve Arabidopsis seedlings, were tested on ground. The results showed that most of the plants developed normally. In order to avoid high population stress the number of seedlings per growth area should be reduced. The RNAlater preservation method to be used in the space experiment was compared with a quick freeze in Liquid Nitrogen (LN2). The RNA from samples preserved in RNAlater at room temperature for 24h was slightly more degraded than the RNA from the LN2 preserved samples (RNA integrity number, RIN: 7.7 and 8.6, respectively). However, the RNA quality and quantity was satisfactory for microarray analysis. Of the genes analysed, 74 genes (0.28%) were significantly differentially expressed, most of them showing moderate to low regulation. Among the genes induced in the RNAlater preserved samples, three salt inducible transcription factors (ZAT10, SZF1 and SZF2) were identified, suggesting that the high salt concentration in RNAlater causes salt stress before the transcription stopped. In conclusion, the Multigen-2 preservation protocol tested here will allow for the genes regulated by microgravity in the space experiment to be revealed. The results do indicate that not all the biological processes are stopped instantly by the RNAlater. The limited diffusion indirectly caused by the microgravity may potentially result in a different degree of salt stress in the test compared to the 1×g control during the space experiment. This has to be accounted for during the evaluation of the results. Since slightly degraded RNA was observed, further optimalisation of the preservation protocol will be performed. Abstract: The latest advances in bioregenerative strategies for long-term life support in extraterrestrial outposts such as on Mars have indicated soil-based cropping as an effective approach for waste decomposition, carbon sequestration, oxygen production, and water biofiltration as compared to hydroponics and aeroponics cropping. However, it is still unknown if cropping using soil systems could be sustainable in a Martian greenhouse under a gravity of 0.38g. The most challenging aspects are linked to the gravity-induced soil water flow; because water is crucial in driving nutrient and oxygen transport in both liquid and gaseous phases, a gravitational acceleration lower than g =9.806ms−2 could lead to suffocation of microorganisms and roots, with concomitant emissions of toxic gases. The effect of Martian gravity on soil processes was investigated using a highly mechanistic model previously tested for terrestrial crops that couples soil hydraulics and nutrient biogeochemistry. Net leaching of solute, gaseous fluxes of NH3, CO2, N2O, NO and N2, depth concentrations of O2, CO2 and dissolved organic carbon (DOC), and pH in the root zone were calculated for a bioregenerative cropping unit under gravitational acceleration of Earth and for its homologous on Mars, but under 0.38g. The two cropping units were treated with the same fertilizer type and rate, and with the same irrigation regime, but under different initial soil moisture content. Martian gravity reduced water and solute leaching by about 90% compared to Earth. This higher water holding capacity in soil under Martian gravity led to moisture content and nutrient concentrations that favoured the metabolism of various microbial functional groups, whose density increased by 5–10% on Mars as compared to Earth. Denitrification rates became substantially more important than on Earth and ultimately resulted in 60%, 200% and 1200% higher emissions of NO, N2O and N2 gases, respectively. Similarly, O2 and DOC were consumed more rapidly in the Martian soil and resulted in about 10% increase in CO2 emissions. More generally, Martian cropping would require 90% less water for irrigation than on Earth, being therefore favourable for water recycling treatment; in addition, a substantially lower nutrient supply from external sources such as fertilizers would not compromise nutrient delivery to soil microorganisms, but would reduce the large N gas emissions observed in this study. Abstract: Hematopoietic progenitor cell proliferation can be alternated on either spaceflight or under simulated microgravity experiments on the ground; however, the underlying mechanism remains largely unknown. In the present study, we have demonstrated that exposure of human erythropoietin (EPO)-dependent leukemia cell line UT-7/EPO cells to conditions of simulated microgravity with a rotary culture instrument significantly inhibited the cellular proliferation rate. Adding higher concentrations of EPO to the culture medium failed to improve the inhibitory status. Cell apoptosis was detected by fluorescence staining of cell nuclei and a flow cytometry assay using Annexin V/PI double staining. This microgravity-induced apoptosis in UT-7/EPO cells could be blocked by a pancaspase inhibitor Z-VAD-FMK. Immunoblotting demonstrated that rotary culture resulted in a reduction of the expression of Bcl-xL, an anti-apoptotic protein, and the cleavage of caspase-3. Furthermore, rotary culture reduced surface localization and protein content, as well as the mRNA expression of erythropoietin receptor (EPOR) of UT-7/EPO. Take together, the findings indicated that simulated microgravity may induce mitochondrial related apoptosis of UT-7/EPO cell through depressing the EPO–EPOR pathway. [ABSTRACT FROM AUTHOR]

Abstract: Although it has been suggested that microgravity might affect drug absorption in vivo, drug permeability across epithelial barriers has not yet been investigated in vitro during modelled microgravity. Therefore, a cell culture/diffusion chamber was designed specifically to accommodate epithelial cell layers in a 3D-clinostat and allow epithelial permeability to be measured under microgravity conditions in vitro with minimum alteration to established cell culture techniques. Human respiratory epithelial Calu-3 cell layers were used to model the airway epithelium. Cells grown at an air interface in the diffusion chamber from day 1 or day 5 after seeding on 24-well polyester Transwell cell culture inserts developed a similar transepithelial electrical resistance (TER) to cells cultured in conventional cell culture plates. Confluent Calu-3 layers exposed to modelled microgravity in the 3D-clinostat for up to 48h maintained their high TER. The permeability of the paracellular marker 14C-mannitol was unaffected after a 24h rotation of the cell layers in the 3D-clinostat, but was increased 2-fold after 48h of modelled microgravity. It was demonstrated that the culture/diffusion chamber developed is suitable for culturing epithelial cell layers and, when subjected to rotation in the 3D-clinostat, will be a valuable in vitro system in which to study the influence of microgravity on epithelial permeability and drug transport. Abstract: This work compares cell wall regeneration from protoplasts of the fungus Penicillium decumbens under rotary culture (simulated microgravity) and stationary cultures. Using an optimized lytic enzyme mixture, protoplasts were successfully released with a yield of 5.3×105 cells/mL. Under simulated microgravity conditions, the protoplast regeneration efficiency was 33.8%, lower than 44.9% under stationary conditions. Laser scanning confocal microscopy gave direct evidence for reduced formation of polysaccharides under simulated conditions. Scanning electron microscopy showed the delayed process of cell wall regeneration by simulated microgravity. The delayed regeneration of P. decumbens cell wall under simulated microgravity was likely caused by the inhibition of polysaccharide synthesis. Abstract: This study investigated the time-course of stress and recovery states and their relations to social support and personality traits in healthy women during a long-term head-down tilt bed rest. Personality, social support and affective states were assessed in 16 women exposed to simulated microgravity for a 60-day duration involving three stages: a 20-day baseline control period (BDC), a 60-day head-down tilt bed rest (HDT) and a 20-day post-HDT ambulatory recovery period (R+). Participants were divided into two groups: an exercise (Exe, n =8) and a control group (Ctl, n =8). All the participants experienced significantly more stress during the HDT period. But exercise did not improve the impaired effects of simulated microgravity. The Exe group perceived more stress and less recovery than the Ctl group during the HDT period. Among the five major personality factors, only Neuroticism was related to both social and affective variables. Neuroticism was positively associated with stress and negatively associated with recovery and social support (S-SSQ). Practical implications in psychological countermeasures for better dealing with the key human factor in spaceflights are discussed. [Copyright &y& Elsevier]
