A DNA sequence encoding for the human proinsulin was designed according to the codon bias of Escherichia coli and then chemically synthesized. The synthesized DNA fragment was subcloned into pGEX-3X for expression in E. coli BL21 (DE3) and E. coli BL21 Star (DE3), respectively. Conditions for the highest expression of the GST-proinsulin fusion proteins were optimized. These conditions are that cells of E. coli BL21 star (DE3) are incubated in 100mL of the LB medium with 2 mmol/L IPTG and 60μ?g/mL ampicillin at 26oCfor 4h. After disrupted E. coli cells with ultrasonication, inclusion bodies were precipitated from cell lysis and washed. Fusion proteins from the inclusion bodies were redissolved in 8mmol/L of urea. After dialysed in purified water, fusion proteins were analysed by SDS-PAGE. The purity of the fusion protein is about 80.5% in total. The fusion protein from SDS-PAGE was further identified by mass/mass spectrum. GST in the dyad protein is confirmed by the 9 matched sequences. However, the left part is proved a polypeptide of which is completely different from the human proinsulin.

Coal blending is widely utilized in coal chemical industry and power plants, and ash slagging propensity, a non-additive property of the pure coals, is difficult to predict and related to its AFTs (ash fusion temperatures). A combustion experimental approach was undertaken to investigate the influence of coal blending ratio on the AFTs of three coal samples Sh-coal, Qi-coal, Que-coal from Ningdong mining areas. The results showed that a nonlinear decrease of the AFTs was obtained with the blending ratios (Sh-coal/Qi-coal or Sh-coal/Que-coal) increasing, the AFTs dropped by 30 °C from 0.5 to 0.8 ratio; the content of Al2O3 can be obviously lowered AFTs. Therefore, coal blending could lower the AFTs evidently. The obtained results in this research can be used in coal gasification process and further theoretical studies.

To investigate biomass classification technology, such as rotary classifier, and to select appropriate biomass classifier. Through the change of different feeding rates and rotation speeds of the classifier, the classification efficiency and Newton classification efficiency, specific particle size and uniformity index, the recoveries of coarse and fine powder and energy consumption were measured. The results indicate that feeding rate and rotation speed have an important effect on the classification. When the rotation rate of classifier is 300rpm, the classification efficiency decrease basically, but in 78.96kg/h, it is the maximum. And Newton classification efficiency would decrease at first and then increase, in 88.96kg/h, it is the lowest. When the feed rate are 78.96kg/h and 88.96kg/h, as the rotation speed increases from 50r/min to 150r/min, there is a moderate increase in uniformity coefficient, that means the biomass size distribution becomes narrower. The energy consumption means the pulverizer capacity is larger, the less is the wasted energy. It is a common sense that the larger the mill, the higher the efficiency.

TiO2 nano film with different nano structure were prepared on the surfaces of titanium TA1 and titanium alloy TC4 via electrochemical anodization. The surface morphology was observed with SEM, and it is found that TiO2 films prepared on TA1 surface have porous structures, while the TiO2 films synthesized on TC4 surface look like nano rod. The contact angle(CA) test results show that the TiO2 nano film prepared on TA1 is hydrophilicity and the CA is about 19°,the film on the TC4 is super hydrophilicity and its CA is smaller than 2°. After combined with fluorinated silane the self-assembled film synthesized on the surfaces and they are superhy- drophobic and the Contact angle is 150° and 158° respectively. It is concluded that both nano- structures and fluoroalkysilanes with low surface energy have play an important role in the wettability with the contact angle is more than 150°. The surface only have nano roughness without the low energy does not show superhydrophobicity.

Vibrating lamella thickener (VLT) developed by Kunming University of Science and Technology is a new generation of inclined plate settling device, and in this device such long-standing problems in traditional lamella thickener (LT) as material bonding and blockage between channels in inclined plates, material deposits and blockage in cone bucket, as well as unsteady discharging of high solids underflow, etc. are effectively resolved, through periodic vibration of inclined plate modules, the use of built-in device for removal of deposited material on cone wall and of forced discharging device for high solids underflow. Paste discharging is now a new way for the effective treatment of tailings in recent years, due to its superiority for mining safety and environmental protection. The industrial tests for paste thickening of iron tailings were carried out in a mineral processing plant of Mining Co., Ltd. of Panzhihua Iron and Steel Group, using a ZXNQP-32 VLT. In the test the size distribution and settling characteristics of the tailings were analyzed, and the main operating parameters in the VLT were tested under the natural settling and flocculated settling conditions, respectively. The results of test indicate that the solids concentration of underflow from the VLT is capable of reaching higher than 70%; meanwhile, the solids content in overflow from the VLT is as low as less than 300 mg/l while flocculant is added in the ore pulp for pretreatment and such an overflow can be directly discharged into environment or be recycled in the plant. It is thus concluded that VLT is effective for paste thickening of iron tailings.

The detailed mechanism of divorced pearlite in bearing steels began to be actively discussed more recently. The survey of divorced pearlite in higher Mn bearing steel was carried out through scanning electronic microscope following isothermal transformation heat treatment. The results show that while divorced transformation taking place in the higher Mn bearing steel just slightly below A1, the cementite could be produced in such way as growing on the base of pre-existing cementite particles in austenite, or emerge either in globular particles or in short rod directly from the austenite. Under isothermal transformation, the carbon atoms in austenite prefer to precipitate on the original boundary of the austenite. The enough small size of the cementite particles remained in the austenite is helpful for getting spheroidal cementite particles structure through divorced eutectoid transformation.

We used the iron tailings with different concentrations of raw slurry and the classifier overflow after desliming for natural sedimentation and adding flocculants flocculating sedimentation test research. The results show that: the settling velocity is slower during the natural sedimentation, The effect that concentration on the settling velocity of material is larger, with the increase of concentration, particle settling velocity drop, When the concentration reached 20%, material group is in a state of suspension, particle is essentially not settlement; Adding polyacrylamide flocculant for the pulp material pretreatment, the settling velocity is obviously improved than natural sedimentation, Supernatant turbidity also decreased significantly, but the pulp concentration still has a great influence on the sedimentation rate, the higher the concentration of the settlement, the slower settling velocity. The test result is according to the theoretical formula of settlement.

this review summarizes an important factor of clogging in subsurface flow constructed wetlands ¬(SSF)—field distribution. The efficiency of filter media and the spatial distribution of biological growth are determined by the flow field distribution directly. If the flow field distribution is un-uniform, it would accelerate the clogging of substrate. The influencing factors for flow field distribution, including hydraulic loads, properties of filter media, filling structures and root distribution were investigated, and the clogging reasons were discussed additionally. At last, this study showed some corresponding research methods, and helping in the process of research and development of subsurface flow constructed wetlands in solving clogging problem.

The paper analyzed the sulfur in steel for civil engineering in 150t converter in TangSteel. Based on the analysis of desulfurization process and the thermodynamic conditions in converter, the operation of desulfurization was optimized. The industrial tests for desulfurization were carried out, and the results were analyzed. The results were shown that the distribution coefficient would be the highest value, when the alkalinity of slag was 3.5 and the FeO content was 21% in making SPHD steel. The distribution coefficient would reduce, when the temperature was increased, especially more than 1700°C, so end point temperature should be best less than 1700°C.

Calcium-activated chloride channels (CaCCs) play pivotal roles in many physiological Activities, including transepithelial fluid secretion, smooth muscle contraction and sensory transduction. TMEM16A is a bona fide calcium-activated chloride channel, which was discovered by three independent labs in 2008 after Calcium-activated chloride channel current was recorded about thirty years ago. In this study, DNA fragments encoding mouse TMEM16A with green fluorescence protein (GFP) fusion protein were subcloned into pcDNA3.1/Zeo. Transient transfection condition was optimized and Fischer Thyroid epithelial cells (FRT) expressing TMEM16A were got by stable transfection. The classical calcium-activated chloride channels current was recorded in FRT cells stably expressing TMEM16A by whole cell patch clamp technique. These results were beneficial for the delving into the effects of other bivalent cations on TMEM16A-CaCCs and the role of TMEM16A-CaCCs in cell proliferation and migration.

In this paper, the electromagnetic shielding fabric with Ni-Cu-Ni three layers and colorized appearance was prepared by the electroless plating and colorized electroplating. The current density was distributed unevenly in the course of colorized electroplating, which resulted in the nonuniform color of the electroplated fabric. In this paper, this problem was solved by improving the electroplating fixture. The adhesion strength and the conductivity of the coating were measured respectively. The results showed that the Ni-Cu- Ni multilayered coating possessed stronger adhesion strength to the fabric substrate than single layered coating, and the conductivity of coating was better than that of pure metal.

A frosting test bench was designed, and unsteady frosting on vertical plate surface was dynamically described with liquid nitrogen as working medium under natural convection conditions. The variation of frost thickness was analyzed, and the flow state of moist air and temperature distribution near the vertical plate were described. The experimental results show the unsteady frosting of vertical plate surface under natural convection and cryogenic condition actually includes several steps: formation of frost crystals, growth of frost crystals and growth of frost layer. The initial frosting rate is large, but the rate decreases as time goes on until the frost layer thickness is unchanged. Moist air conducts a top-down flow near the vertical plate, and the rest of moist air flows into the main flow. The temperature of moist air decreased systematically along the tangential direction of vertical plate from top to bottom and along the normal direction of vertical plate from far to vertical plate surface.
