Aroclor 1254, a technical PCB mixture (polychlorinated biphenyls) and TBT (tributyltinchloride) are environmental pollutants that cause a broad spectrum of acute toxic and chronic effects in aquatic animals. In this paper, the sensitivity of Daphnia magna to chronic exposure to mixed xenobiotics was evaluated under laboratory conditions. The results show that xenobiotic mixtures (50 % each of the single compounds) were more toxic than individual xenobiotics alone. By measuring behavioral parameters of animals, it becomes evident that exposure to single xenobiotics significantly affects daphnids: exposure led finally to a rapid decrease in mean swimming activity and also caused changes in preferred swimming depth, with daphnids preferring the upper layers of aquaria. The mixture altered the swimming behavior even more strongly compared to the group stressed by single chemicals. Finally, all daphnids sank to the bottom of the aquaria, still alive, but inactive at the end of the exposure period. In addition, we investigated the reproductive capacity (number of newborn per female and day). PCB did not affect the number of newborn significantly, TBT-stress led to an evidently decreased number of young daphnids and the xenobiotic mixture decreased reproduction even more. In conclusion, we found significant effects of the single compounds as well as approximately additive (swimming behavior) and synergistic (reproduction) effects of the chemical mixture on daphnids indicating the possibility of dramatic ecological consequences of the occurrence of mixed xenobiotic substances in the aquatic environment. Einfluss einer xenobiotischen Mischung (PCB und TBT) auf Schwimmverhalten oder Reproduktion von Daphnia magna Aroclor 1254, eine technische PCB-Mischung (polychlorierte Biphenyle), und TBT (Tributylzinn) sind Umweltbelastungen, die ein breites Spektrum akut toxischer und chronischer Wirkungen in aquatischen Tieren verursachen. Untersucht wird die Sensitivitat von Daphniamagna bei chronischer Exposition gegenuber gemischten Xenobiotika unter Laborbedingungen. Die Ergebnisse belegen, dass xenobiotische Mischungen (50 % jeder Substanz) starker toxisch wirken als jede Substanz fur sich allein. Durch Messung von Verhaltens-Parametern der Tiere wird deutlich, dass die Exposition gegen einzelne Xenobiotika Daphnien signifikant beeinflusst: Die Wirkung fuhrt letztlich zu einer rapiden Abnahme der Schwimmaktivitat und verursacht bei den Daphnien Veranderungen in der Aufenthaltstiefe gegenuber den sonst bevorzugten oberen Schichten eines Aquariums. Die Stoffmischung verandert das Schwimmverhalten starker als die Einzelstoffe: Zu Versuchsende sinken alle Daphnien lebend, aber inaktiv zu Boden. Die zusatzlich untersuchte Reproduktionskapazitat (Anzahl der Neonaten je Weibchen) zeigt: PCB beeinflusst die Zahl der Neonaten nicht signifikant, TBT-Stress fuhrt zu einer deutlich verringerten Zahl junger Daphnien und die Mischung beider Stoffe senkt die Reproduktion noch starker. Zusammenfassend wurden signifikante Wirkungen der Einzelstoffe beobachtet sowie additive (Schwimmverhalten) als auch synergistische (Reproduktion) Wirkungen der Stoffmischung. Dies indiziert die Moglichkeit dramatischer okologischer Konsequenzen aus dem Vorkommen gemischter xenobiotischer Stoffe in der aquatischen Umwelt.

Sediment humic substances from an acidification chronology (210Pb- and 137Cs-dating) of Groser Arbersee, a small cirque lake in the Bavarian Forest, Germany, have been studied by means of 1H NMR spectroscopy. The acidification of the lake and, particularly, its catchment caused a more or less continuous decrease in diatom-inferred pH from 5.4 to 4.75. Combined with diagenetic processes in the sediment, acidification reduced the bulk concentration of humic material in the water column and altered the composition of humic material, too: The share of humic acids (IHSS protocol) increased from 12 to 17% of the total humic carbon. Statistically, the reduced pH values account for 60%. While the constituents changed in the fulvic acid fraction, they remained more or less unchanged in the humic acid fraction. Within the fulvic acid fraction, percentages of carbohydrates increase, whereas those of aliphatic as well as aromatic compounds decrease. These changes may be explained (a) by reductions in enzymatic activity, particularly of cellulases and proteases, in the raw humic material in the catchment, (b) protonation of acidic functional groups (carboxylic acids, phenolic compounds) which leads to an immobilization and retention of these compounds in the catchment, or/and (c) by increased chemical oxidation or precipitation of the aromatic portions, leaving the more transparent aliphatic chains. As a consequence of reduced chromophoric DOC content, light penetration, particularly of UV irradiation, has increased and the biogeochemical detoxification mechanism will be reduced. The residual fauna and flora in acidified Groser Arbersee will be exposed to another severe physical and chemical stress, because acidification is generally accompanied by a variety of organic micro-contaminants. Versauerungsbedingte Veranderungen der Huminstoff-Bestandteile im Grosen Arbersee Die Huminstoffe des Sedimentes aus dem Grosen Arbersee, einem kleinen Karstsee im Bayerischen Wald, wurden mittels 1H-NMR-Spektroskopie auf ihre Bestandteile hin untersucht. Uber subfossile Diatomeen und 210Pb- sowie 137Cs-Datierung war die Versauerungschronologie rekonstruiert worden. Die Versauerung des Sees und insbesondere seines Einzugsgebietes fuhrte zu einer mehr oder weniger kontinuierlichen Abnahme des pH-Wertes im See von 5.4 auf 4.75. Neben nicht weiter studierten diagenischen Veranderungen im Sediment war die Versauerung ein masgeblicher Prozess, der die Gesamtkonzentration an Huminstoffen in der Wassersaule und ebenfalls die Zusammensetzung der Huminstoffe veranderte: Der Anteil der Huminsauren (IHSS-Verfahren) stieg von 12% auf 17% der Huminstoffe. Statistisch bestimmt die Versauerung diese Zunahme zu 60%. In den Fulvosauren veranderte sich die Zusammensetzung deutlich, wahrend sie in den Huminsauren weitgehend unverandert blieb. In den Fulvosauren nehmen die Anteile der Kohlenhydrate zu, wahrend aliphatische als auch aromatische Komponenten abnehmen. Diese Veranderungen konnen erklart werden (a) uber eine Reduktion der enzymatischen Aktivitat im Grobhumus, insbesondere solcher, die Makromolekule wie Cellulose oder Proteine spalten, (b) uber Protonierungen von sauren funktionellen Gruppen (Carboxyl- und Phenolgruppen), die zu einer Immobilisierung und Retention dieser Verbindungen im Einzugsgebiet fuhrten, und/oder (c) durch eine erhohte chemische Oxidation oder Prazipitation von aromatischen Anteilen im See, wobei die transparenteren aliphatischen Ketten ubrigbleiben. Da die Gehalte des chromophoren DOC stark abgenommen haben, hat als Konsequenz die Lichteindringung (insbesondere des UV-B) deutlich zugenommen. Ferner wird der geochemische Detoxierungsmechanismus geschwacht, so dass insgesamt die verbliebene Biota einem zusatzlichen Stress ausgesetzt ist.

Direct effects of pure humic substance (BS1 FA) on three different aquatic organisms (Ceratophyllum demersum, Dreissena polymorpha, and Chaetogammarus ischnus) were demonstrated in this study. Exposure to environmentally relevant concentrations (0.5 mg/L) of this humic substance led to the activation of the microsomal and soluble glutathione S-transferase (GST) in Ceratophyllum demersum. Exposure to 3-chlorobiphenyl showed also an elevation of GST activity, which is due to the proposed detoxication metabolism of these chlorinated biphenyl. Coexposure to a mixture of 3-chlorobiphenyl and BS1 FA showed a reduction of enzyme elevation, but still significant over an untreated control. The direct impact of humic substances seems not to be restricted to a specific class of organisms.

Several papers report on obviously contradictory biological and ecological effects of humic substances (and NOM). For instances, growth promotion as well as growth inhibition of algae and macrophytes, promotion as well as inhibition of net-heterotrophy in non-eutrophic lakes, promotion as well as inhibition of reproduction of the nematode, Caenorhabditis elegans, are reported. If one takes direct effects of humic substances (and NOM) on aquatic organisms into account that are supported by increasing empirical evidence, these obvious contradictions can be solved as trade-offs between specific and non-specific effects. For instance, net-heterotrophy in non-eutrophic lakes can be considered as (weak) inhibition of photosynthesis of algae and macrophytes (specific effect) and simultaneous promotion of heterotrophic growth (non-specific effect). If net-autotrophy predominates, a positive feed-back effect of bacteria on algae, based on growth promoting substances can be observed. These assumptions are based on recent experimental laboratory findings that include even quantitative structure activity relationships. Two more contradictory effects of humic substances (and NOM) from reports are discussed with respect to trade-offs between specific and non-specific effects: reduction/increase of adverse mineral acid effects on invertebrates as well as effects of lipophilic chemicals upon invertebrates and fishes. We present a simple concept model that is based on specific and non-specific effects of humic substances and NOM towards aquatic organisms. Applying this model to hormone-like effects of humic substances and NOM upon the nematode C. elegans, we show that both effects inhibitory and promoting can qualitatively be predicted.
