Subepithelial blood vessels of the rat lingual papillae and their spatial relations to the connective tissue papillae and surface structures were demonstrated by light and scanning electron microscopy. In the rat, four types of papillae were distinguished on the dorsal surface of the tongue, i.e. the filiform, fungiform, foliate and circumvallate papillae. Vascular beds of various appearance were found in all four types of lingual papillae: a simple or twisted capillary loop in the filiform papilla; a basket- or petal-like network in the fungiform papilla; a ring-like network in the foliate papilla, and a conglomerated network surrounded by double heart-shaped capillary networks in the circumvallate papilla. These characteristic vascular beds corresponded to the shape of the connective tissue papillae and surface structures. The vascular bed beneath the gustatory epithelium in the fungiform, foliate and circumvallate papilla consisted of fine capillary networks next to the taste buds.

In the Indian buffalo (Bubalus bubalis), the gross anatomical and neurohistological aspects of the tongue, its tunics and various lingual papillae have been studied. The observations indicate that the widest part of the tongue is also the thickest. The papillae encountered therein include filiform (four sub-types), fungiform and circumvallate papillae. Simple branched tubulo-acinar serous glands are located in the lamina propria of the circumvallate papillae. The mucous glands are limited to the root of the tongue. The nerve endings such as the muscle spindle, ovoid or elliptical and motor end-plates are found to be associated with the skeletal muscle. In the lingual epithelium, only simple free nerve endings are seen. In the taste buds, the intragemmal fibres end in close proximity of the gustatory cells.

Human deep posterior lingual glands (von Ebner's glands) are located beneath the circumvallate papillae. They are formed by tubuloalveolar adenomeres, intercalated ducts and excretory ducts coming together in the main excretory duct. The tubuloalveolar cells, pyramid-shaped, show large and dense secretory granules (clear cored) throughout the cytoplasm, rare basal folds and packed cisternae of rough endoplasmic reticulum (RER) at the basal pole. The columnar cells of the intercalated ducts are arranged in a monolayer. They are characterized by dense, clear-core secretory granules (mostly in the apical cytoplasm), a basal nucleus, well-developed RER and Golgi apparatus, and thin filaments distributed in supra- and perinuclear cytoplasm. Striated ducts are absent. Excretory ducts, coming together in the main duct, are lined by a bistratified epithelium. The inner layer consists of columnar cells showing bundles of tonofilaments with scarce secretory activity. The outer layer is composed of basal cells lying on the basal lamina. The main excretory duct, which opens at the bottom of the vallum, shows a stratified epithelium. The outer side is composed of 2-3 layers of malpighian cells lying on the basal lamina. The inner side consists of a single layer of cuboidal-columnar cells with dense apical granules and well-developed organelles synthesizing and condensing secretions. These cells interpolate with goblet cells, rare mitochondria-rich cells, ciliated cells and numerous small globous cells showing a clear matrix and lacking secretory granules. The cilia show a 9 + 2 microtubular structure with basal bodies provided with striated rootlets. Myoepithelial cells surround with their processes the basal portions of the secretory cells and the intercalated ducts. The conclusions concern some comparative aspects and some hypothesis on the functional role of goblet cells, ciliated cells and epithelial cells lining the different ducts, also in relation to the final secretory product.

Some type III cells in taste buds of rabbit foliate papillae have greatly increased numbers of dense-cored vesicles. Such cells also contain unusual lamellar bodies that resemble those in alveolar type II cells; they consist of alternating dense and light layers with a periodicity of about 4.4 nm. The precise relationship between the vesicles and the lamellar bodies is unknown.

The anatomy and neurohistology of the tongue of the Indian goat, Capra aegagrus, have been described. The apex linguae is notched in the centre. The foramen caecum is found to be absent. The sublingua could not be traced. The filiform papillae are the most common and divided into three types: the simple, giant, and true filiform papillae. The true filiform papillae are the most developed of the three types. The foliate papillae are absent. There are 13--14 circumvallate papillae arranged in two rows in a V-shaped pattern. The fungiform papillae are large and could easily be seen with the naked eye. They are scattered over the entire dorsum, being in abundance at the tip. The tongue of the goat is richly innervated. On the dorsum, the lamina propria is innervated by thick nerve fibres. In the fungiform papillae quite a large number of nerve fibres could be seen. The circumvallate papillae are also abundantly provided with nerves. A few ganglion cells are found below the circumvallate papillae. Thick nerve fibres are seen across the numerous glands and their ducts. Muscle fibres and connective tissue are also richly innervated.

The three-dimensional and surface structures of the simple conical papillae of the rat tongue have been demonstrated with scanning electron microscopy. The papillary projection was organized into the anterior, posterior and central core cell populations, whereas the basal region of the papilla which consisted of circularly arranged cells showed no differentiation into three autonomic cell populations. It is considered that the anterior and posterior cell populations around the central core tend to be mutually attached at the bilateral sides, and that the posterior and core cell contacts are rather close than the anterior one. The anterior papillary cells showed relatively smooth surface with little micropits and without microridges. The reticular microridges on the basal cell surface of the posterior papillary cells appear to later develop the micropits and linear microridges on the tip cell surface. These suggest that the anterior cell surface is more highly keratinized than the posterior one. The microridges or micropits on the outer cell surface and the microprojections on the inner cell surface organizing filiform papilla are considered to be the structures for the purpose of cell adhesion.

The tongue, pinna and dorsal skin of adult male C-1 mice were removed at 03.00, 06.00, 09.00, 12.00, 15.00, 18.00, 21.00 and 24.00 h, fixed in basic lead acetate and stained with Alcian blue-safranin or 0.5% toluidine blue. The mast cell numbers of these regions were counted and analyzed statistically by analysis of variance. It was found that there were circadian variations in the mast cell number in the tongue, pinna and dorsal skin. The difference between the minimum and maximum of circadian variation in mast cell number in all three regions was highly significant (p less than 0.01). Furthermore, the time points of the maximum and minimum of mast cell number varied between the different regions. The time point of the minimum in the tongue and pinna was at 06.00 h, whereas it was at 09.00 h in the dorsal skin. The time point of the maximum in the tongue and dorsal skin was at 21.00 h, but in the pinna it was at 18.00 h.

A study of secondary palate formation in man employing histochemical techniques indicates that (1) tongue muscles are sufficiently differentiated at the time of palatal shelf elevation to produce active tongue movements; (2) an embryo with asymmetrically arranged shelves and tongue suggests that the tongue is indeed active at this time; (3) few blood vessels are present beneath the epithelium at fusion sites both before and after fusion has occurred suggesting that this epithelium is not metabolically active; (4) only slight lysosomal enzyme activity is seen in the disintegrating epithelium at the junction of palatal shelves and nasal septum, and (5) macrophages are infrequent near to the disintegrating epithelium and their scarcity is consistent with the prolonged survival of epithelial rests in the midline of the human secondary palate.
