Herein, a functional class of microenvironment-associated nanomaterials is reported for improving the second near-infrared (NIR-II) imaging and photothermal therapeutic effect on intracranial tumors via a spontaneous membrane-adsorption approach. Specific peptides, photothermal agents, and biological alkylating agents were designed to endow the nanogels with high targeting specificity, photothermal properties, and pharmacological effects. Importantly, the frozen scanning electron microscopy technology (cryo-SEM) was utilized to observe the self-association of nanomaterials on tumor cells. Interestingly, the spontaneous membrane-adsorption behavior of nanomaterials was captured through direct imaging evidence. Histological analysis showed that the cross-linking adhesion in intracranial tumor and monodispersity in normal tissues of the nanogels not only enhanced the retention time but also ensured excellent biocompatibility. Impressively, in vivo data confirmed that the microenvironment-associated nanogels could significantly enhance brain tumor clearance rate within a short treatment timeframe (only two weeks). In short, utilizing the spontaneous membrane-adsorption strategy can significantly improve NIR-II diagnosis and phototherapy in brain diseases while avoiding high-risk complications.

Grain boundaries (GBs) are a kind of lattice imperfection widely existing in two-dimensional materials, playing a critical role in materials' properties and device performance. Related key issues in this area have drawn much attention and are still under intense investigation. These issues include the characterization of GBs at different length scales, the dynamic formation of GBs during the synthesis, the manipulation of the configuration and density of GBs for specific material functionality, and the understanding of structure-property relationships and device applications. This review will provide a general introduction of progress in this field. Several techniques for characterizing GBs, such as direct imaging by high-resolution transmission electron microscopy, visualization techniques of GBs by optical microscopy, plasmon propagation, or second harmonic generation, are presented. To understand the dynamic formation process of GBs during the growth, a general geometric approach and theoretical consideration are reviewed. Moreover, strategies controlling the density of GBs for GB-free materials or materials with tunable GB patterns are summarized, and the effects of GBs on materials' properties are discussed. Finally, challenges and outlook are provided.

Graphene oxide (GO) and reduced GO (rGO) are the only variants of graphene that can be manufactured at the kilogram scale, and yet the widely accepted model for their structure has largely relied on indirect evidence. Notably, existing high-resolution transmission electron microscopy (HRTEM) studies of graphene oxide report long-range order of sp(2) lattice with isolated defect clusters. Here, we present HRTEM evidence of a different structural form of GO, where nanocrystalline regions of sp(2) lattice are surrounded by regions of disorder. The presence of contaminants that adsorb to the surface of the material at room temperature normally prevents direct observation of the intrinsic atomic structure of this defective GO. To overcome this, we use an in situ heating holder within an aberration-corrected TEM (AC-TEM) to study the atomic structure of this nanocrystalline graphene oxide from room temperature to 700 °C. As the temperature increases to above 500 °C, the adsorbates detach from the GO and the underlying atomic structure is imaged to be small 2-4 nm crystalline domains within a polycrystalline GO film. By combining spectroscopic evidence with the AC-TEM data, we support the dynamic interpretation of the structural evolution of graphene oxide.

Direct imaging of liquids by electron microscopy has been extremely difficult due to their high volatility. Ionic liquids are a unique liquid material with almost zero vapor pressure, which allows us to characterize them under high-vacuum conditions. Here we report the first direct observation of the microstructure and phase behavior of an imidazolium-based room-temperature ionic liquid by high-resolution transmission electron microscopy with the aid of a special carbon nanotube network, which is able to support a freestanding ultrathin ionic liquid film on its nanosized holes. It was found that the existence of cluster structures is one of the intrinsic properties of the ionic liquid in its whole liquid phase range. Furthermore, the size and mobility of the clusters play an important role during phase transition of the ionic liquid. We show that the direct HRTEM imaging on freestanding liquid film is a powerful technique to obtain insight into the structure of ionic liquids and their phase behavior. The present study can provide an important starting point for more sophisticated structural studies on the microstructure of liquidus materials.

The function of bioenergetic membranes is strongly influenced by the spatial arrangement of their constituent membrane proteins. Atomic force microscopy (AFM) can be used to probe protein organization at high resolution, allowing individual proteins to be identified. However, previous AFM studies of biological membranes have typically required that curved membranes are ruptured and flattened during sample preparation, with the possibility of disruption of the native protein arrangement or loss of proteins. Imaging native, curved membranes requires minimal tip–sample interaction in both lateral and vertical directions. Here, long-range tip–sample interactions are reduced by optimizing the imaging buffer. Tapping mode AFM with high-resonance-frequency small and soft cantilevers, in combination with a high-speed AFM, reduces the forces due to feedback error and enables application of an average imaging force of tens of piconewtons.
