Heparan sulfate (HS) is a major component of cell surface glycocalyx with extensive negative charges and plays a protective role by preventing toxins, including small molecule drugs and anticancer cationic lytic peptides (ACLPs), from cells. However, this effect may compromise the treatment efficiency of anticancer drugs. To overcome the impedance of cancer cell glycocalyx, an HS-targeting ACLP PTP-7z was designed by fusion of an ACLP and a Zn 2+ -binding HS-targeting peptide. Upon Zn 2+ ion binding, PTP-7z could self-assemble into uniform nanoparticles and show improved serum stability and reduced hemolysis, which enable it to self-deliver to tumor sites. The peptide PTP-7z showed a pH- and Zn 2+ ion-dependent HS-binding ability, which triggers the HS-induced in situ self-assembling on the cancer cell surface in the acidic tumor microenvironment (TME). The self-assembled PTP-7z can overcome the impedance of cell glycocalyx by either disrupting cell membranes or translocating into cells through endocytosis and inducing cell apoptosis. Moreover, PTP-7z can also inhibit cancer cell migration. These results proved that HS-responsive in situ self-assembling is a practical strategy to overcome the cancer cell glycocalyx barrier for ACLPs and could be extended to the design of other peptide drugs to promote their in vivo application.

Typical methods for stable immobilization of proteins often involve time-consuming surface modification of silicon-based materials to enable specific binding, while the nonspecific adsorption method is faster but usually unstable. Herein, we fused a silica-binding protein, Si-tag, to target proteins so that the target proteins could attach directly to silica substrates in a single step, markedly streamlining the immobilization process. The adhesion force between the Si-tag and glass substrates was determined to be approximately 400-600 pN at the single-molecule level by atomic force microscopy, which is greater than the unfolding force of most proteins. The adhesion force of the Si-tag exhibits a slight increase when pulled from the C-terminus compared to that from the N-terminus. Furthermore, the Si-tag's adhesion force on a glass surface is marginally higher than that on a silicon nitride probe. The binding properties of the Si-tag are not obviously affected by environmental factors, including pH, salt concentration, and temperature. In addition, the macroscopic adhesion force between the Si-tag-coated hydrogel and glass substrates was ∼40 times higher than that of unmodified hydrogels. Therefore, the Si-tag, with its strong silica substrate binding ability, provides a useful tool as an excellent fusion tag for the rapid and mechanically robust immobilization of proteins on silica and for the surface coating of silica-binding materials.

Thorium separation has recently become a hot topic because of the potential application of thorium as a future nuclear fuel, while metal-organic framework (MOF) materials have received much attention in the separation field due to their unique properties. Herein, a highly porous and stable MOF, UiO-66, and its carboxyl derivatives (UiO-66-COOH and UiO-66-(COOH) 2 ) were synthesized and explored for the first time for Th(IV) capture from a weak acidic solution. Although the introduction of carboxyl groups into UiO-66 leads to an obvious decrease in the surface area and pore volume, the adsorbability toward Th(IV) is greatly enhanced. At pH = 3.0, the saturated sorption capacity for Th(IV) into UiO-66-(COOH) 2 reached 350 mg/g, representing one of the largest values for Th(IV) capture by solid extraction. Moreover, the functionalized MOFs show fast sorption kinetics and desirable selectivity toward Th(IV) over a range of competing metal ions. A possible mechanism for the selective recognition of Th(IV) by these MOFs was explored on the basis of extended X-ray absorption fine structure and Fourier transform infrared analysis. It is concluded that UiO-66-COOH and UiO-66-(COOH) 2 sorb Th(IV) through the coordination of carboxyl anions in the pores of the MOFs, whereas in the case of UiO-66, both the precipitation and the exchange with the organic solvent contribute to the Th(IV) uptake. This study contributes to the assessment of the feasibility of MOFs applied in actinides separation and better understanding of actinides sorption behavior in this kind of hybrid porous solid materials.
