Com o foco voltado a essas limitações, buscamos produzir uma matriz integralmente descelularizada e fiel ao complexo tridimensional e de alta conectividade. O scaffold naturalmente vascularizado e morfologicamente integro apresentou todos os quesitos físicos e mecânicos necessários para seguir as próximas etapas da validação biológica, conforme o mensurado pelas diversas caracterizações realizadas. Esse scaffold apresenta um grande potencial como plataforma de sustenc¸a~o para a formação de tecidos funcionais capazes de gerenciar o recrutamento de células-tronco progenitoras e suprir todas as necessidades sistêmicas inerentes a` proliferação e diferenciação celulares. Com esse novo biomaterial será´ possível desenvolver novas abordagens capazes de oferecer a regeneração das funções endócrinas do pâncreas no combate ao diabetes.

Over the last years, a large range of different omics have led to the development of valuable biomarkers, based on genomics, proteomics, transcriptomics, metabolomics etc. Only few attention has been paid to the study of glycomics, an attractive though not well known technology that can lead to the development of biomarkers in a variety of diseases. Chapter Four provides information that amoeboid protists are a taxonomically eclectic group encompassing ‘amoebae'and ‘amoeboflagellates.'They are single-celled organisms that move by a crawling-like motion, while extending pseudopodia (slender or finger-shaped protrusions) from the periphery of the cell. Their shape is highly variable; and the number and organization of the pseudopodia, in addition to cell shape, are important features used to categorize and taxonomically identify the amoebae. Chapter Five subject dealt with in this chapter may be useful for improving strategies to treat certain diseases as well as for the design of materials for tissue engineering, among other applications. Chapter Six indicates that abnormalities in physical properties of the cell membranes may be strongly linked to hypertension and other circulatory disorders. X-ray imaging is a corner stone of breast cancer diagnosis. By exploiting the phase shift of X-rays rather than their attenuation, phase-contrast tomography has the potential to dramatically increase the visibility of small and low contrast features, thus leading to better diagnosis. This thesis presents research on the first synchrotron-based project developing a clinical phase-contrast breast computed tomography (CT) setup at Elettra, the Italian Syncrotron Radiation Facility. This book includes a comprehensive theoretical background on propagation-based phase-contrast imaging, exploring and extending the most recent image formation models. Along with theory, many practical implementation and optimization issues, ranging from detector-specific processing to setup geometry, are tackled on the basis of a large number of experimental evidences. Most of the modelling results and data analysis have general validity, being a valuable framework for optimization of phase-contrast setups. Results obtained at synchrotron are also compared with'real world'laboratory sources: both a first-of-its-kind comparison with one of the few hospital breast CT systems and a state-of-the-art implementation of monochromatic phase-contrast micro-tomography with a conventional rotating anode source are presented. On a more general level, this work sheds a light on the importance of synchrotron-based clinical programs, which are key to trigger the long-anticipated transition of phase-contrast imaging from synchrotrons to hospitals.
