The 1997 European Conference on Spectroscopy of Biological Molecules (ECSBM) is the seventh in a biennial series of conferences devoted to the applications of molecular spectroscopy to biological molecules and related systems. The interest of these conferences rests mainly on the relationship between the structure and physiological activity of biological molecules and related systems of which these molecular species form part. This volume ofECSBM contains articles prepared by the invited lecturers and those making poster presentations at the seventh ECSBM. The reader will find mainly applications of vibrational spectroscopy to protein structure and dynamics, biomembranes, molecular recognition, nucleic acids and other biomolecules and biological systems containing specific chromophors. In the first 20 years that followed the purinergic signalling hypothesis in 1972, most scientists were sceptical about its validity, largely because ATP was so well established as an intracellular molecule involved in cell biochemistry and it seemed unlikely that such a ubiquitous molecule would act as an extracellular signalling molecule. However, after the receptors for ATP and adenosine were cloned and characterized in the early 1990s and ATP was established as a synaptic transmitter in the brain and sympathetic ganglia, the tide turned. More recently it has become clear that ATP is involved in long-term (trophic) signalling in cell proliferation, differentiation and death, in development and regeneration, as well as in short-term signalling in neurotransmission and secretion. Also, important papers have been published showing the molecular structure of P2X receptors in primitive animals like Amoeba and Schistosoma, as well as green algae. This has led to the recognition of the widespread nature of the purinergic signalling system in most cell types and to a rapid expansion of the field, including studies of the pathophysiology as well as physiology and exploration of the therapeutic potential of purinergic agents.In two books, Geoffrey Burnstock and Alexej Verkhratsky have aimed at drawing together the massive and diverse body of literature on purinergic signalling. The topic of this first book is purinergic signalling in the peripheral and central nervous systems and in the individual senses. Post-Traumatic Stress Disorder (PTSD) is a common and severe psychiatric disorder precipitated by exposure to a psychologically distressing event. PTSD is associated with significant morbidity and mortality and is characterized by the presence of three distinct, but co-occurring, symptom clusters. Research evidence suggests that PTSD has a neurobiological basis. Current research on the neurobiology of PTSD include the utilization of functional brain imaging; molecular genetic research; the incorporation of cross-system research including neuroendocrine, neurochemical, and neuroimmunological systems. Post-traumatic stress disorder is a psychiatric illness that can occur in anyone who has experienced a life-threatening or violent event. The trauma can be due to war, terrorism, torture, natural disasters, violence, or rape. In PTSD the brain areas that are likely to be affected are the hippocampus (memory), amygdala (fear association), the prefrontal cortex (cognitive processing), and the ascending reticular activating system (arousal). The chemical of interest is norepinephrine, which is released during a stressful event and is part of the fight-or-flight response meant to mobilize the body to action.The objective of this title is to outline the neurobiology of post-traumatic stress disorder and provide treatment strategies for clinicians. Innate immunity is one the most evolutionally conserved systems, designed to protect the organism from viruses and bacterial infections, stress and many other types of attacks from the outside world. During the past decade, the capacity of molecular biology and information technology to produce and analyse data have grown exponentially, rapidly reforming many aspects of immunology research in the post-genomics era. As a result, scientific understanding of signalling networks governing the innate immunity response in human tissues and other organisms has evolved beyond recognition, compared to even just a decade ago. Many strategies have been designed over the years to identify novel proteins, which have a crucial role in innate immunity responses by regulating particular signalling pathways. These projects had many advantages, including the definition of novel drug targets, as exemplified by the recent success of anti-TNF therapy, as well as leading to a better, system-wide understanding of the molecular control of innate immunity. In the past few years, a new concept, Immunomics, has been adopted to define an emerging, multidisciplinary field of research (Schonbach, 2003). Although rapid progress has been made to identify the proteins playing pivotal roles in the innate immunity–related signalling pathways (for example, TIR signalling pathways), the catalogue of proteins with a key regulatory function identified and studied is far from completed.
