The effect of noise in the input data for learning potential energy surfaces (PESs) based on neural networks for chemical applications is assessed. Noise in energies and forces can result from aleatoric and epistemic errors in the quantum chemical reference calculations. Statistical (aleatoric) noise arises for example due to the need to set convergence thresholds in the self consistent field (SCF) iterations whereas systematic (epistemic) noise is due to, {\it inter alia}, particular choices of basis sets in the calculations. The two molecules considered here as proxies are H$_{2}$CO and HONO which are examples for single- and multi-reference problems, respectively, for geometries around the minimum energy structure. For H$_2$CO it is found that adding noise to energies with magnitudes representative of single-point calculations does not deteriorate the quality of the final PESs whereas increasing the noise level commensurate with electronic structure calculations for more complicated, e.g. metal-containing, systems is expected to have a more notable effect. However, the effect of noise on the forces is more noticeable. On the other hand, for HONO which requires a multi-reference treatment, a clear correlation between model quality and the degree of multi-reference character as measured by the $T_1$ amplitude is found. It is concluded that for chemically "simple" cases the effect of aleatoric and epistemic noise is manageable without evident deterioration of the trained model - although the quality of the forces is important. However, considerably more care needs to be exercised for situations in which multi-reference effects are present.

The value of uncertainty quantification on predictions for trained neural networks (NNs) on quantum chemical reference data is quantitatively explored. For this, the architecture of the PhysNet NN was suitably modified and the resulting model was evaluated with different metrics to quantify calibration, quality of predictions, and whether prediction error and the predicted uncertainty can be correlated. The results from training on the QM9 database and evaluating data from the test set within and outside the distribution indicate that error and uncertainty are not linearly related. The results clarify that noise and redundancy complicate property prediction for molecules even in cases for which changes - e.g. double bond migration in two otherwise identical molecules - are small. The model was then applied to a real database of tautomerization reactions. Analysis of the distance between members in feature space combined with other parameters shows that redundant information in the training dataset can lead to large variances and small errors whereas the presence of similar but unspecific information returns large errors but small variances. This was, e.g., observed for nitro-containing aliphatic chains for which predictions were difficult although the training set contained several examples for nitro groups bound to aromatic molecules. This underlines the importance of the composition of the training data and provides chemical insight into how this affects the prediction capabilities of a ML model. Finally, the approach put forward can be used for information-based improvement of chemical databases for target applications through active learning optimization.

All-atom simulations can provide molecular-level insights into the dynamics of gas-phase, condensed-phase and surface processes. One important requirement is a sufficiently realistic and detailed description of the underlying intermolecular interactions. The present perspective provides an overview of the present status of quantitative atomistic simulations from colleagues' and our own efforts for gas- and solution-phase processes and for the dynamics on surfaces. Particular attention is paid to direct comparison with experiment. An outlook discusses present challenges and future extensions to bring such dynamics simulations even closer to reality.

Atomistic simulations using accurate energy functions can provide molecular-level insight into functional motions of molecules in the gas- and in the condensed phase. Together with recently developed and currently pursued efforts in integrating and combining this with machine learning techniques provides a unique opportunity to bring such dynamics simulations closer to reality. This perspective delineates the present status of the field from efforts of others in the field and some of your own work and discusses open questions and future prospects.

Machine learning techniques applied to chemical reactions has a long history. The present contribution discusses applications ranging from small molecule reaction dynamics to platforms for reaction planning. ML-based techniques can be of particular interest for problems which involve both, computation and experiments. For one, Bayesian inference is a powerful approach to include knowledge from experiment in improving computational models. ML-based methods can also be used to handle problems that are formally intractable using conventional approaches, such as exhaustive characterization of state-to-state information in reactive collisions. Finally, the explicit simulation of reactive networks as they occur in combustion has become possible using machine-learned neural network potentials. This review provides an overview of the questions that can and have been addressed using machine learning techniques and an outlook discusses challenges in this diverse and stimulating field. It is concluded that ML applied to chemistry problems as practiced and conceived today has the potential to transform the way with which the field approaches problems involving chemical reactions, both, in research and academic teaching.

Extensions and improvements of empirical force fields are discussed in view of applications to computational vibrational spectroscopy and reactive molecular dynamics simulations. Particular focus is on quantitative studies which make contact with experiments and provide complementary information for a molecular-level understanding of processes in the gas phase and in solution. Methods range from including multipolar charge distributions to reproducing kernel Hilbert space approaches and machine learned energy functions based on neural networks.
