Ingestion of plant and fungal glucosylceramides is known to reduce colon carcinogenesis and skin barrier damage in mice and humans. However, such effects in animal experiments have not been revealed for plant and fungal ceramides because the content of ceramides contained in plants and fungi is so low that the large amount required for animal experiments is difficult to obtain. Noting that the fungus shiitake mushroom (Lentinula edodes) is rich in a glucosylceramide, (4E,8E)-N-d-2 -hydroxypalmitoyl-1-O-β-d-glucopyranosyl-9-methyl-4,8-sphingadienine [Glc-d19:2(4E,8E,9Me)-h16:0], we developed a new method to purify this fungal glucosylceramide using ethanol precipitation and high-performance liquid chromatography. We also developed a new method to produce large amounts of a ceramide [d19:2(4E,8E,9Me)-h16:0] from this purified glucosylceramide using human glycoside hydrolase family 30 glucocerebrosidase (imiglucerase). Glucosylceramides and ceramides with 8 E and 8 Z isomers of the long chain base are found in plants. These isomers have been difficult to quantify separately using liquid chromatography–tandem mass spectrometry (LC–MS/MS) because the isomers have the same retention time, their precursor and product ions have the same m / z values, and plant ceramide standards are not commercially available. Here we tested trial separations using various ODS columns and prepared plant ceramide standards generated by human glucocerebrosidase (imiglucerase) using commercially available plant glucosylceramide standards as the substrates. Consequently, we were able to quantify the isomers based on differences in retention times in a TSKgel ODS-120A column (Tosoh, Tokyo Japan) using LC–electrospray ionization–MS/MS (LC–ESI–MS/MS). The purpose of this study was to evaluate the effects of intragastrical administration of Glucerabacter canisensis NATH-2371T on glucosylceramide (GluCer) digestion in mice. Although G. canisensis was unable to utilize starch and cellulose, coculture of G. canisensis with mouse fecal bacteria greatly increased GluCer hydrolysis in polysaccharide medium, indicating that G. canisensis grew in competition with other intestinal bacteria. Although most of the administered G. canisensis cells were detected in feces, some cells were present in the colorectum contents, which had GluCer-hydrolyzing activity. These results indicate that G. canisensis can viably transit through the mouse gut. Administration of G. canisensis to mice fed diets supplemented with GluCer or GluCer-containing foods significantly enhanced GluCer hydrolysis. Since G. canisensis did not show acute toxicity, it may be useful as a probiotic to augment GluCer hydrolysis in the large intestine. Sphingolipids, including ceramides, glycosphingolipids, sphingomyelin, and sphingosine-1-phosphate, have been recognized as important molecules that regulate critical cellular functions. Although originally studied in the context of lysosomal storage diseases, the roles of these compounds in more common disorders involving metabolism, vascular disease, and aberrant growth has been the focus of recent studies, including in disorders that affect the kidneys. These efforts have led to new insights into Fabry disease, a classic disorder of lysosomal function that results in renal failure as well as in more common renal diseases including diabetic nephropathy and polycystic kidney disease. Pathways for glycosphingolipid synthesis can be targeted with orally available small-molecule inhibitors, creating new opportunities for the treatment of both rare and common kidney diseases. Gaucher disease is caused by inherited deficiency of lysosomal glucocerebrosidase. Proteome analysis of laser-dissected splenic Gaucher cells revealed increased amounts of glycoprotein nonmetastatic melanoma protein B (gpNMB). Plasma gpNMB was also elevated, correlating with chitotriosidase and CCL18, which are established markers for human Gaucher cells. In Gaucher mice, gpNMB is also produced by Gaucher cells. Correction of glucocerebrosidase deficiency in mice by gene transfer or pharmacological substrate reduction reverses gpNMB abnormalities. In conclusion, gpNMB acts as a marker for glucosylceramide-laden macrophages in man and mouse and gpNMB should be considered as candidate biomarker for Gaucher disease in treatment monitoring. Background: It has been shown that exogenic administration of glycosphingolipids (GSLs) induces outgrowth of neurites from cultured nerve cells. Furthermore, the activator of glucosylceramide synthase, L-threo-1-phenyl-2-decanoylamino-3-morpholino-1-propanol (L-PDMP), is thought to exhibit stimulatory effects on both the biosynthesis and neurotrophic actions of GSL in the same culture system. To investigate the effect of GSLs on focal cerebral ischemia in vivo, L-PDMP was injected into the intraperitoneal space of rats during the chronic phase following permanent occlusion of the left middle cerebral artery (MCA) and thereafter, the levels of GSLs and their effects on behavioral changes were examined Methods: The levels of cerebrosides, sphingomyelin (SM) and ceramide in the ischemic cortex were measured by gas–liquid chromatography (GLC) after separation by high-performance thin-layer chromatography, using the internal standards N-heptadecanoyl-D-cerebroside, N-heptadecanoyl-D-sphingomyelin and N-heptadecanoyl-D-sphingosine, respectively. To determine the sugar components of the cerebrosides, the trimethylsilylated derivatives of their methylglycosides after methanolysis were analysed directly by GLC. Results: The L-PDMP treatment induced a 2.4-fold increase in glucosylceramide, the precursor of gangliosides, but no changes were evident in the levels of SM and ceramide in the ischemic cerebral cortex. The ischemic rats treated with L-PDMP showed improved re-acquisition of memory and learning in the Morris water maze task. Conclusion: These results suggest that the pharmacological effects of L-PDMP include significant facilitation of glucosylceramide biosynthesis and improvement of neural function. This enhancement occurred when the dietary glucosylceramide increased the expression of claudin - I in an ultraviolet B (UVB) radiation in epidermis of a mouse. This caused increase in electrical resistance of Sphingosine and phytosphingosine. This showed that increase in skin barrier resistance is due to enhancement of tight junction.
