Two studies were conducted to determine efficacy of cabergoline for suppressing prolactin (PRL) and the possible effects on vernal transition in mares. In experiment 1, six mares each received either vehicle or cabergoline (5 mg, intramuscularly) every 10 days for 12 treatments beginning February 4, 2013. Blood samples were drawn regularly, and mares were challenged with sulpiride periodically to assess PRL suppression. Weekly hair samples were obtained to determine shedding. Prolactin was suppressed ( P < .05) by cabergoline, but suppression waned in spring. There was no effect ( P > .05) of treatment on day of first ovulation, luteinizing hormone, or follicle stimulating hormone. Hair shedding was generally suppressed ( P = .05). In 2014 (experiment 2), eight of the same 12 mares were used in a similar experiment to determine if the rise in PRL observed in experiment 1 was due to refractoriness to cabergoline or perhaps another factor. Treatment began on April 6, 2014, corresponding to the increase in PRL in treated mares in experiment 1. Mares were treated with cabergoline or vehicle until June 5. Prolactin was suppressed ( P < .05) by cabergoline, and the pattern of apparent escape from suppression was similar to year 1. We conclude that (1) cabergoline at this dose alters hair shedding but does not alter the time of first ovulation in mares and (2) relative to our previous reports of cabergoline treatment in the fall, there is a seasonal effect on the ability of this dose of cabergoline to suppress unstimulated PRL secretion. Ovary is the main functional organ in reproduction and has obvious difference in the biological activity during the estrous cycle. To investigate the genes associated with follicular development, ovulation, or non-season reproduction, gene expression differences in the ovaries of Qira black sheep estrous stage and anestrous stage in May were assessed using RNA-seq technology. This analysis obtained approximately 1.17 Gb and 1.22 Gb clean reads in Qira black sheep anestrous stage and estrous stage libraries, respectively. Six hundred and fifty one differentially expressed genes (DEGs) were identified, in which 400 genes were up-regulated and 251 genes were down-regulated in the anestrous stage samples compared with estrous stage samples. GO enrichment analysis revealed that these DEGs were significantly enriched in 52 functional groups for the 3 GO categories. Using KEGG pathway analysis, 35 significant signaling pathways were enriched, including calcium signaling pathway, steroid biosynthesis, cell adhesion molecules, and steroid hormone biosynthesis. The experiment was conducted to study ovarian follicular population, its diameter, steroid profile (estradiol 17β and progesterone) and corpus luteum development during estrous cycle and seasonal anestrous in Murrah buffaloes. They were categorized into 2 groups based on the estrus signs i.e. cyclic (6) and anestrous (6) and subjected to ultrasound scanning of ovaries on day 0 (day of estrus in cyclic and day of first scanning in anestrous buffaloes), 6, 10 and 16 using B-mode scanner equipped with 6.0 MHz linear array transducer. Blood samples were collected on the above days for the estimation of the progesterone and estradiol-17β. The mean number of small follicles on day 0 and 10 and medium follicles on day 10 were significantly higher within as well as between the cyclic and anestrous buffaloes, while mean number of large follicles on day 0, 10 and 16 were significantly higher in the cyclic as compared to anestrous buffaloes. However, mean number of total follicles were significantly higher in cyclic as compared to anestrous buffaloes on all the days of scanning. The maximum diameter of large follicle was observed in cyclic buffaloes on day 0 (1.40±0.49 cm). The diameter of CL increased with its development and reached to its peak on day 10 (1.39±0.04 cm), thereafter, it reduced on day 16 (0.72±0.30 cm) of the cycle. The mean serum progesterone concentration was significantly higher on day 6, 10 and 16 than day 0 of the estrous cycle indicating development of functional corpus luteum, however, mean serum estradiol-17β concentration was significantly higher on day 0 than the other days of cycle signifying follicular growth and development secreting higher amount of estrogen. Abstract: Thecomas are tumors originating from the ovarian stroma and of rare occurrence in the equine species. Clinical, ultrasonographic, endocrine, and histopathologic descriptions of thecoma are scarce. This article aims to describe a case of ovarian changes in a mare with anomalous growth of the right ovary and anestrus for 1 year. After surgical removal, the ovarian growth was diagnosed as a thecoma. Histopathological findings were consistent with the findings previously described in the literature, characteristic of a thecoma, and decisive for the correct diagnosis. [Copyright &y& Elsevier]

Abstract: Acupuncture is part of an Eastern system of medicine used for diagnosing, treating, and preventing disease. It has been used for the treatment of reproductive disorders in both human beings and domestic species. We hypothesized that acupuncture could be used to induce cyclicity in anestrous mares. In experiment 1, six anestrous mares were treated with acupuncture at Bai Hui, governing vessel (GV)-2, bladder (BL)-22, BL-23, BL-31, and BL-33 twice a week for 3 weeks, then once a week for an additional 8 weeks, whereas six anestrous mares served as controls. In experiment 2, six anestrous mares received acupuncture stimulation at Bai Hui, GV-1, BL-22, BL-23, and BL-28, with electroacupuncture (10 minutes at 50 Hz) at BL-22 and BL-28 bilaterally. In addition, vitamin B12 (3 mL, 1,000 mcg/mL) was injected in the vaginal fornix at the 3, 6, and 9 o’clock positions around the cervix. Six anestrous mares served as controls. Examinations and treatments were carried out twice a week until ovulation. In both experiments, ovarian activity was examined in all mares by transrectal ultrasonography and blood sample was obtained twice a week to determine progesterone concentrations. The time to first ovulation for experiment 1 was 14.0 ± 0.7 and 13.5 ± 1.0 weeks (mean ± SD) for the control and acupuncture groups, respectively (P = .60). In experiment 2, the time to ovulation was 9.0 ± 0.6 and 10 ± 0.3 weeks (mean ± SD) for the control and acupuncture groups, respectively (P = .19). The aim of this study was to evaluate the effect of eCG treatment on follicular response, diameter of CL and P4 level on buffaloes synchronized with P4 device. Forty buffalo were assigned into two Groups (GnoeCG and GeCG) and received a P4 device (DIB®) plus 2.0mg of estradiol benzoate (D0). On D9, the DIB® was removed and a PGF2a was administered. On this day, buffalo in GeCG received 400IU of eCG. After two days (D11), each buffalo received 10μg of GnRH. Buffaloes were examined by ultrasonography on D -12, D0, D9, D11 to D14, D16, D20 and D24. All animals were bled in order to measure P4 level from D16 to D24. There were no significant differences between GnoeCG and GeCG on dominant follicle diameter, device removal/ovulation interval, ovulation rate, CL diameter and volume on D20 and D24. Notwithstanding, there were differences between GnoeCG and GeCG on CL diameter and volume on D16 and P4 level from D16 to D24. These results indicate that treatment with eCG can increase the CL diameter and volume and the P4 level on anestrous buffaloes synchronized with P4 device. The beneficial effect of eCG needs to be further investigated on pregnancy rate of anestrous buffaloes submitted to FTAI. [ABSTRACT FROM AUTHOR]

The article discusses a study that assessed the effect of intra-vaginal Fluorogesterone-Acetate (FGA) sponges on the fertility of ewes during the anestrous season. Grouped into 4 were 96 ewes and treated with intra-vaginal Fluorogestone-Acetate (FGA) and equine chorionic gonadotropin (eCG) for 6 and 12 days and examined for estrus and bred by fertile rams. Results showed that using intra-vaginal FGA sponges with eCG regimen in 12 days can help improve the reproductive performance of ewes in the anestrous season.

The objective of this study was to quantify serum progesterone levels, uterine features, and pregnancy rates in acyclic, embryo recipient mares using a bovine progesterone-releasing intravaginal device in a commercial embryo transfer (ET) program. The study included 73 recipient mares of unknown breed, aged 3–10 years, weighing 350–500 kg, and kept under an intensive management system on Tifton 85 (Cynodon spp.) pastures with water and mineral salt ad libitum. The horses were divided into two groups: a group with a progesterone-releasing intravaginal device (1 g progesterone, G-IVP4, n = 24) and a control group (G-iP4, n = 49) receiving an injection of 1,500 mg long-acting progesterone. Jugular blood was collected for the G-IVP4 group for subsequent progesterone measurement by radioimmunoassay on three occasions: Day 0 (D0), intravaginal device was placed; Day 5 (D5), day of the ET; and Day 9 (D9), day of pregnancy diagnosis. There was an increase (P <.0001) in serum progesterone levels on D5 and D9 compared with D0 (4.09 ± 0.81 and 6.45 ± 1.03 ng/mL vs. 0.71 ± 0.14 ng/mL). There were no differences among groups in the pregnancy rate (P >.05), with rates of 83.33% and 73.46% for G-IVP4 and G-iP4, respectively. In conclusion, the intravaginal route for absorption of 1 g of progesterone device increased the serum level of progesterone sufficiently to prepare the uterus of acyclic recipient mares for ET, and the conception rate was similar to the standard protocol using long-acting injectable progesterone. • Bovine progesterone-releasing intravaginal device increases progesterone levels in mare. • Pregnancy rates are similar to the standard progesterone injection protocol. • Time to embryo retransfer to recipient mares is reduced. • Easy administration and excellent pregnancy rates in acyclic recipients. • Recipient availability after a short delay.
