Abstract

A recent study conducted at China Medical University in Taichung, Taiwan, has found that Coenzyme Q0 (CoQ0), a derivative from Antrodia camphorata, has anti-melanogenic, anti-photoaging, and anti-inflammatory effects on keratinocyte cells. The researchers discovered that CoQ0 activates Nrf2-mediated antioxidant pathways, downregulates alpha-MSH expression, and inhibits melanogenesis and melanin generation. Additionally, CoQ0 enhances Nrf2 nuclear translocation, resulting in antioxidant expression and the inhibition of ROS-mediated inflammation. Researchers at the National University of Cordoba in Argentina have conducted a study on the effects of a high-fat diet (HFD) on cognitive function and neuroinflammation. The study found that HFD consumption, combined with an immune challenge, led to cognitive deficits and oxidative stress in the hippocampus, a region of the brain important for learning and memory. However, the researchers also discovered that the neuropeptide alpha-melanocyte stimulating hormone (alpha-MSH) had a protective effect, reversing the negative effects of the HFD on memory, oxidative stress, and astrocyte proliferation. Abstract

A staphylococcal growth inhibition experiment was performed using chitosan-cholesterol alpha-MSH and individual controls. alpha-MSH and chitosancholesterol both show bacterial growth inhibition by a magnitude of 50 and 79%, respectively. According to the news editors, the research concluded: "In a nutshell, our analysis demonstrated novel antimicrobial activity of nanoparticle-conjugated alpha-MSH, which could be used as future therapeutics against multidrug-resistant Staphylococcus aureus and other types of bacterial cells." Phenotypic alternation was observed in the presence of alpha-MSH causing changes in the cell structure and shape, indicating stress on Staphylococcus aureus. [Extracted from the article]

Abstract

Abstract: Long-term regulation of energy balance involves two major trends: first age-related obesity develops in the middle-aged, later it is followed by anorexia of aging (sarcopenia and/or cachexia). A dynamic balance between orexigenic and anorexigenic neuropeptides is essential for the regulation of energy homeostasis. Special imbalances of neuropeptide effects may be assumed corresponding to different age-periods. Anorexia induced by acute alpha-MSH (alpha-melanocyte stimulating hormone; endogenous melanocortin agonist) injections was analyzed in male Wistar rats aged 6–9 weeks (juvenile), 3–4 months (young adult), 6 or 12 months (two middle-aged groups), 18 months (aging) and 24–26 months (old). Alpha-MSH injected through a preimplanted intracerebroventricular (ICV) cannula (compared with saline injection) dose-dependently suppressed spontaneous food intake and also re-feeding following 24-h fasting, but the rate of suppression varied between age-groups. An ICV injection of 5μg alpha-MSH attenuated the 2-h re-feeding by 21.9±3.2% in juvenile rats, strongly (68.7±2.5%) suppressed it in young adults, the suppression became progressively weaker in the two middle-aged groups (55.7±4.9%, vs. 26.4±4.9%, respectively), but it turned extreme in aging (94.7±4.2%) and old (74.3±4.5%) rats. Body composition also changed with age: unlike the tibialis anterior muscle, the epididymal and retroperitoneal fat pads increased until middle-age and remained large even in old animals, while the measured indicator of muscle mass decreased in the oldest group. The food intake suppressing and body weight decreasing effects of a 7-day-long ICV infusion of 1μg/h alpha-MSH were weakest in the 12-month-old and most pronounced in the 24month-old rats. In conclusion, responsiveness to the anorexic effect of alpha-MSH varies with age, with a nadir of the curve in the middle-aged, and a peak in the aging and old animals. This age-related nadir of melanocortin-responsiveness may promote obesity in middle-aged rats, while the tendency for anorexia and incipient sarcopenia of old (still obese) rats may result from age-related melanocortin-hypersensitivity rather than from adiposity. Abstract

Hypothalamic melanocortin 4 receptors (MC4R) are a key regulator of energy homeostasis. Brain-penetrant MC4R agonists have failed, as concentrations required to suppress food intake also increase blood pressure. However, peripherally located MC4R may also mediate metabolic benefits of MC4R activation. Mc4r transcript is enriched in mouse enteroendocrine L cells and peripheral administration of the endogenous MC4R agonist, α-melanocyte stimulating hormone (α-MSH), triggers the release of the anorectic hormones Glucagon-like peptide-1 (GLP-1) and peptide tyrosine tyrosine (PYY) in mice. This study aimed to determine whether pathways linking MC4R and L-cell secretion exist in humans. GLP-1 and PYY levels were assessed in body mass index–matched individuals with or without loss-of-function MC4R mutations following an oral glucose tolerance test. Immunohistochemistry was performed on human intestinal sections to characterize the mucosal MC4R system. Static incubations with MC4R agonists were carried out on human intestinal epithelia, GLP-1 and PYY contents of secretion supernatants were assayed. Fasting PYY levels and oral glucose-induced GLP-1 secretion were reduced in humans carrying a total loss-of-function MC4R mutation. MC4R was localized to L cells and regulates GLP-1 and PYY secretion from ex vivo human intestine. α-MSH immunoreactivity in the human intestinal epithelia was predominantly localized to L cells. Glucose-sensitive mucosal pro-opiomelanocortin cells provide a local source of α-MSH that is essential for glucose-induced GLP-1 secretion in small intestine. Our findings describe a previously unidentified signaling nexus in the human gastrointestinal tract involving α-MSH release and MC4R activation on L cells in an autocrine and paracrine fashion.
