1. Trends in cardiovascular health metrics and associations with long-term mortality among US adults with coronary heart disease.

Abstract

Our study examined the trends of cardiovascular health metrics in individuals with coronary heart disease (CHD) and their associations with all-cause and cardiovascular disease mortality in the US. The cohort study was conducted based on the National Health and Nutrition Examination Survey 1999–2018 and their linked mortality files (through 2019). Baseline CHD was defined as a composite of self-reported doctor-diagnosed coronary heart disease, myocardial infarction, and angina pectoris. Cardiovascular health metrics were assessed according to the American Heart Association recommendations. Long-term all-cause and cardiovascular disease mortality were the primary outcomes. Survey-adjusted Cox regression models were used to estimate hazard ratios and corresponding 95% confidence intervals for the associations between cardiovascular health metrics and all-cause and cardiovascular disease mortality. The prevalence of one or fewer ideal cardiovascular health metrics increased from 14.15% to 22.79% (P < 0.001) in CHD, while the prevalence of more than four ideal cardiovascular health metrics decreased from 21.65% to 15.70 % (P < 0.001) from 1999 to 2018, respectively. Compared with CHD participants with one or fewer ideal cardiovascular health metrics, those with four or more ideal cardiovascular health metrics had a 35% lower risk (hazard ratio, 0.65; 95% confidence interval: 0.51, 0.82) and a 44% lower risk (0.56; 0.38, 0.84) in all-cause and cardiovascular disease mortality, respectively. Substantial declines were noted in ideal cardiovascular health metrics in US adults with CHD. A higher number of cardiovascular health metrics was associated with lower all-cause and cardiovascular disease mortality in them. • The prevalence of one or less ideal CVH metrics increased in CHD in the US, while the prevalence of three and more than four ideal CVH metrics decreased. • Ideal CVH metrics were inversely associated with all-cause and CVD mortality in CHD. • Effective measures are needed to improve the CVH for people with CHD. 2. Association of cardiovascular health metrics with all-cause and cardiovascular disease mortality in chronic kidney disease: A cohort study.

Abstract

Since the global burden of chronic kidney disease (CKD) is rising rapidly, the study aimed to assess the association of cardiovascular health (CVH) metrics with all-cause and cardiovascular disease (CVD) mortality among individuals with CKD. The cohort study included 5834 participants with CKD from the National Health and Nutrition Examination Survey 1999–2018. A composite CVH score was calculated based on smoking status, physical activity, body mass index, blood pressure, total cholesterol, diet quality, and glucose control. Primary outcomes were all-cause and CVD mortality as of December 31, 2019. Multivariable-adjusted Cox proportional hazards models were used to estimate the association between CVH metrics and deaths in CKD patients. During a median follow-up of 7.2 years, 2178 all-cause deaths and 779 CVD deaths were documented. Compared to participants with ideal CVH, individuals with intermediate CVH exhibited a 46.0% increase in all-cause mortality (hazard ratio, 1.46; 95% confidence interval: 1.17, 1.83), while those with poor CVH demonstrated a 101.0% increase (2.01; 1.54, 2.62). For CVD mortality, individuals with intermediate CVH experienced a 56.0% increase (1.56; 1.02, 2.39), and those with poor CVH demonstrated a 143.0% increase (2.43; 1.51, 3.91). Linear trends were noted for the associations of CVH with both all-cause mortality (P for trend <0.001) and CVD mortality (P for trend = 0.02). Lower CVH levels were associated with higher all-cause and CVD mortality in individuals with CKD, which highlights the importance of maintaining good CVH in CKD patients. • Lower CVH levels were associated with higher all-cause and CVD mortality in individuals with CKD. • Maintaining a better CVH status is beneficial for reducing the risk of premature death in patients with chronic kidney disease. • The benefits of ideal cardiovascular health were particularly evident in younger participants and those at an early stage of CKD. 3. Surgical outcomes of patients with genetically refractory epilepsy: A systematic review and meta-analysis.

Abstract

• Meticulous presurgical evaluation and the selection of appropriate surgical procedure can effectively control seizure to a certain extent. • Pooled outcomes of resective surgery showed that 74 % of patients achieved Engel I at the last follow-up. • Pooled outcomes of neurostimulation showed that 9 % patients achieved seizure free and 61 % of patientsachieved 50 % reduction in seizure frequency at the last follow-up. • Univariate analysis indicated that being female, having somatic mutations, and presenting with focal seizure symptoms were associated with a better prognosis. To summarize the surgical outcomes of genetically refractory epilepsy and identify prognostic factors for these outcomes. A literature search of the PubMed, Web of Science, and Embase databases for relevant studies, published between January 1, 2002 and December 31, 2023, was performed using specific search terms. All studies addressing surgical outcomes and follow-up of genetically refractory epilepsy were included. All statistical analyses were performed using STATA software (StataCorp LLC, College Station, TX, USA). This review was performed in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses, 2020 (i.e., "PRISMA") reporting guidelines. Of the 3833 studies retrieved, 55 fulfilled the inclusion criteria. Eight studies were eligible for meta-analysis at the study level. Pooled outcomes revealed that 74 % of patients who underwent resective surgery (95 % confidence interval [CI] 0.55–0.89; z = 9.47, p < 0.05) achieved Engel I status at the last follow-up. In the study level analysis, pooled outcomes revealed that 9 % of patients who underwent vagus nerve stimulation achieved seizure-free status (95 % CI 0.00–0.31; z = 1.74, p < 0.05), and 61 % (95 % CI 0.55–0.89; z = 11.96, p < 0.05) achieved a 50 % reduction in seizure frequency at the last follow-up. Fifty-three studies comprising 249 patients were included in an individual-level analysis. Among patients who underwent lesion resection or lobectomy/multilobar resection, 65 % (100/153) achieved Engel I status at the last follow-up. Univariate analysis indicated that female sex, somatic mutations, and presenting with focal seizure symptoms were associated with better prognosis (p < 0.05). Additionally, 75 % (21/28) of patients who underwent hemispherectomy/hemispherotomy achieved Engel I status at the last follow-up. In the individual-level analysis, among patients treated with vagus nerve stimulation, 21 % (10/47) were seizure-free and 64 % (30/47) experienced >50 % reduction in seizure frequency compared with baseline. Meticulous presurgical evaluation and selection of appropriate surgical procedures can, to a certain extent, effectively control seizures. 4. Prenylated Flavonoids Isolated from the Root of Sophora flavescensas Potent Antifungal Agents against Botrytis cinerea

Abstract

Sophora flavescens, a traditional Chinese herb, produces a wide range of secondary metabolites with a broad spectrum of biological activities. In this study, we isolated six isopentenyl flavonoids (1–6)from the roots of S. flavescensand evaluated their activities against phytopathogenic fungi. In vitro activities showed that kurarinone and sophoraflavanone G displayed broad spectrum and superior activities, among which sophoraflavanone G displayed excellent activity against tested fungi, with EC50values ranging from 4.76 to 13.94 μg/mL. Notably, kurarinone was easily purified and showed potential activity against Rhizoctonia solani, Botrytis cinerea, and Fusarium graminearumwith EC50values of 16.12, 16.55, and 16.99 μg/mL, respectively. Consequently, we initially investigated the mechanism of kurarinone against B. cinerea. It was found that kurarinone disrupted cell wall components, impaired cell membrane integrity, increased cell membrane permeability, and affected cellular energy metabolism, thereby exerting its effect against B. cinerea. Therefore, kurarinone is expected to be a potential candidate for the development of plant fungicides.

8. Histopathological staging and differential diagnosis of marginal zone lymphoma of gastric mucosa-associated lymphoid tissue

Abstract

The purpose of this study was to explore the histopathological staging and differential diagnosis of marginal zone lymphoma in gastric mucosa-associated lymphoid tissue (MALT lymphoma). We performed detailed histomorphology and immunohistochemistry investigations as well as genetic testing on endoscopic biopsy and endoscopic mucosal resection specimens from 18 patients with gastric MALT lymphoma. We found that gastric MALT lymphoma typically begins as a small, isolated area outside the lymphoid follicular mantle zone or proliferates in a multifocal, patchy manner, gradually spreads to the interfollicular zone, forming diffuse proliferation, invades the gastric mucosal glands, and infiltrates or proliferates into the center of peripheral reactive lymphoid follicles. Abnormally proliferating lymphocytes invade the surrounding lymphoid follicles, resulting in damage, atrophy, and disappearance of their normal follicles as well as of the gastric mucosa glands, forming diffuse proliferation. Redifferentiation and proliferation lead to the transformation of lymphocytes; that is, MALT transitions into highly invasive lymphoma. Based on our findings in this study, we propose the following five stages in the process of development and progression of gastric MALT lymphoma: the stage of cell proliferation outside the lymphoid follicular mantle zone; the stage of heterogeneous proliferative lymphoepithelial lesion; the stage of reactive lymphoid follicular implantation; the stage of lymphoid follicular clonal proliferation; and the stage of MALT transforming into highly invasive lymphoma. We examined the differential diagnosis of histopathological features at each stage. The clinicopathological staging of gastric MALT lymphoma can help clinicians provide accurate treatment and track malignant cell transformation, thus playing a significant role in controlling its development and progression.

9. Design of novel DABO derivatives as HIV-1 RT inhibitors using molecular docking, molecular dynamics simulations and ADMET properties

Abstract

AbstractHIV-1 reverse transcriptase is an important target for developing effective anti-HIV-1 inhibitors. Different types of small molecules have been designed based on this target, showing different levels of inhibitory activity against various types of HIV-1 strains. The relationship between structure and activity of DABO derivatives was investigated by means of 3D-QSAR molecular model, molecular docking, molecular dynamics and ADMET properties. The statistical results of molecular models show that the CoMFA and CoMSIA models have good internal stability (CoMFA: q2= 0.623, r2= 0.946; CoMSIA: q2= 0.668, r2= 0.983) and external prediction ability (CoMFA: rpred2= 0.961; CoMSIA: rpred2= 0.961). In addition, molecular docking has explored the mechanism of action between small molecules and receptor proteins, and the results show that hydrogen bonding between amino acid Lys101 and small molecules can improve the affinity of ligands to receptor binding. A total of 12 novel molecules were designed and their activities were predicted based on the 3D-QSAR model and molecular docking results. The results showed that the designed molecules had higher predictive activity. Subsequently, 100 ns MD simulation and binding free energy verified the stability of molecular docking results. Finally, the pharmacokinetic properties of the novel designed molecule were verified by using ADMET to predict its properties. These results can provide reference for the design and development of novel and effective HIV-1 RT inhibitors, and provide new ideas for the design of subsequent drugs.Communicated by Ramaswamy H. 12. MiR-548t-5p regulates pancreatic ductal adenocarcinoma metastasis through an IL-33-dependent crosstalk between cancer cells and M2 macrophages

Abstract

ABSTRACTIL-33 has been associated with pro- and anticancer functions in cancer. However, its role in pancreatic cancer metastasis remains unknown. This study aimed to explore the role of miR-548t-5p/IL-33 axis in the metastasis of pancreatic cancer. Luciferase activity assay, qRT-PCR, Western blot and ELISA were performed to prove whether IL-33 is the target of miR-548t-5p. In vivo metastasis assay and cellular transwell assay were performed to explore the role of miR-548t-5p/IL-33 axis in the invasion and metastasis of pancreatic cancer. Co-culture experiments and immunohistochemistry were performed to observe whether IL-33 affects cell invasion and metastasis dependent on the involvement of M2 macrophages. THP-1 cell induction experiment and flow cytometry were performed to explore the effect of IL-33 on macrophage polarization. CCK-8, colony formation, cell apoptosis, cell cycle, cell wound healing and transwell assay were performed to investigate the effect of IL-33 induced M2 macrophages on cell malignant biological behavior by coculturing pancreatic cancer cells with the conditioned medium (CM) from macrophages. We found that miR-548t-5p regulated the expression and secretion of IL-33 in pancreatic cancer cells by directly targeting IL-33 mRNA. IL-33 secreted by cancer cells promoted the recruitment and activation of macrophages to a M2-like phenotype. In turn, IL-33 induced M2 macrophages promoted the migration and invasion of cancer cells. Moreover, IL-33 affected pancreatic cancer cell invasion dependent on the involvement of M2 macrophages in the co-culture system. Thus, our study suggested that manipulation of this IL-33-dependent crosstalk has a therapeutic potential for the treatment of pancreatic cancer metastasis.

Abstract

ObjectiveThe remodelling of gut mycobiome (ie, fungi) during pregnancy and its potential influence on host metabolism and pregnancy health remains largely unexplored. Here, we aim to examine the characteristics of gut fungi in pregnant women, and reveal the associations between gut mycobiome, host metabolome and pregnancy health.DesignBased on a prospective birth cohort in central China (2017 to 2020): Tongji-Huaxi-Shuangliu Birth Cohort, we included 4800 participants who had available ITS2 sequencing data, dietary information and clinical records during their pregnancy. Additionally, we established a subcohort of 1059 participants, which included 514 women who gave birth to preterm, low birthweight or macrosomia infants, as well as 545 randomly selected controls. In this subcohort, a total of 750, 748 and 709 participants had ITS2 sequencing data, 16S sequencing data and serum metabolome data available, respectively, across all trimesters.ResultsThe composition of gut fungi changes dramatically from early to late pregnancy, exhibiting a greater degree of variability and individuality compared with changes observed in gut bacteria. The multiomics data provide a landscape of the networks among gut mycobiome, biological functionality, serum metabolites and pregnancy health, pinpointing the link between Mucorand adverse pregnancy outcomes. The prepregnancy overweight status is a key factor influencing both gut mycobiome compositional alteration and the pattern of metabolic remodelling during pregnancy.ConclusionThis study provides a landscape of gut mycobiome dynamics during pregnancy and its relationship with host metabolism and pregnancy health, which lays the foundation of the future gut mycobiome investigation for healthy pregnancy.

14. Effect of tannase on sediment formation in green tea infusion

Abstract

Reducing sediment is important for the utilization and production of green tea infusion and concentrate. The effect of tannase on sediment formation in green tea infusion and concentrate was investigated. The results showed that the amount of sediment of green tea infusion significantly decreased after tannase treatment, which was due to the decrease in the concentration of gallated catechins. Significant positive correlations between the sediment amount and the gallated catechin (r= 0.980, p< 0.01) concentration were found. The formation of sediment in green tea infusion was significantly influenced by the tannase concentration and hydrolysis time. 0.5 mg/mL tannase concentration, 120 min hydrolysis time, and 35 °C hydrolysis temperature were found to be the optimized hydrolysis parameters for reducing the sediment. Tannase treatment not only lowered the amount of sediment of green tea concentrate (74.63%), but it also improved the taste quality of green tea concentrate after storage. These results could be used to improve the quality and utilization of green tea infusion and concentrate.

15. Fast System Matrix Generation Based on Single Angle Calibration in Open-Sided Field Free Line Magnetic Particle Imaging

Abstract

Objective: Open-sided field-free line magnetic particle imaging (OS FFL MPI) is a novel medical imaging system configuration that has received significant attention in recent years. However, the measurement-based system matrix (SM) image reconstruction for OS FFL MPI typically requires multiple angle calibration (MAC), which is time-consuming in practice. Methods: To address this issue, we propose a fast 2D SM generation method that requires only a single angle calibration (SAC). The SAC method exploits the rotational invariance of the system function. Based on the measured single angle system function, the system function is rotated to generate system functions at other angles, and then the SM for image reconstruction is constructed. Then, we conducted various simulation experiments and built an OS FFL MPI scanner to evaluate the proposed SAC method. Results: The experiments demonstrating the effectiveness of SAC in reducing calibration workload, requiring fewer scanning numbers while maintaining a similar image reconstruction quality compared to MAC method. Furthermore, the SM generated by SAC produces consistent imaging results with the SM generated by MAC, regardless of the interpolation algorithms, the number of rotation angles, or the signal-to-noise ratios employed in phantom imaging experiments. Conclusion: SAC has been experimentally verified to reduce acquisition time while maintaining accurate and robust reconstruction performance. Significance: The significance of SAC lies in its contribution to improving calibration efficiency in OS FFL MPI, potentially facilitating the implementation of MPI in a wider range of applications.

16. Sequential Scan-Based High-Resolution 3-D Open-Sided Magnetic Particle Imaging System

Abstract

Magnetic particle imaging (MPI) is an emerging molecular imaging technique that relies on the nonlinear magnetization response of superparamagnetic iron oxide nanoparticles. Open-sided MPI (OS-MPI) has attracted considerable interest for its potential to enhance interventional procedures. However, improving the spatial resolution of OS-MPI poses a significant challenge, mainly due to the inefficiencies of selection coils in generating, rotating, and shifting a high-gradient field-free line (FFL). To address this issue, we propose a novel approach called the permanent magnet-based sequential scan to develop a high-resolution 3-D OS-MPI system with ultralow power consumption. By utilizing permanent magnets, we can create high-gradient selection fields without any power consumption. The sequential scan technique involves rotating the FFL angle by angle and shifting the FFL layer by layer. To maintain thermal stability during sequential scans, we used drive coils wound with hollow copper wires and implemented forced oil cooling. Additionally, measures such as copper shielding, feed-through compensation, and filtering were employed to minimize interference from the permanent magnets, drive coils, and motor drivers. Experimental results demonstrated that the developed system successfully achieved a spatial resolution of 1 mm while operating with an overall low power consumption of 282.16 W.

19. Consumption of ultra-processed foods and all-cause and cause-specific mortality in the Southern Community Cohort Study.

Abstract

Higher intake of ultra-processed foods (UPF) has been linked with higher risks of cancer, cardiovascular disease, and diabetes, as well as all-cause mortality. However, studies on UPF and cause-specific mortality remain limited, especially among disadvantaged populations. We aimed to examine associations of UPF intake with all-cause and cause-specific mortality among low-income Americans. In the Southern Community Cohort Study (SCCS), a prospective cohort of mostly low-income Black and White Americans, we included 77,060 participants who completed a food frequency questionnaire (FFQ) at baseline (2002–2009) and had at least 1 year follow-up. All 89 items in the FFQ were categorized using the Nova classification. UPF intake was calculated as % of daily foods intake by weight (grams). Cox regression was used to estimate HR (95% CI) for the association of UPF intake (quartile or per 10% increase) with total and cause-specific mortality (cancer, coronary heart disease [CHD], stroke, and diabetes) after adjusting for sociodemographics, lifestyles, and disease history. Of 77,060 participants, 46,175 (59.9%) were women, 49,857 were Black (64.7%), and mean age was 52.4 (SD: 8.8) years at baseline. The mean intake of UPF was 41.0% (SD: 15.7%). UPF intake was inversely associated with Healthy Eating Index and intakes of fiber, minerals, and vitamins but positively associated with intakes of sugars and fats (all P FDR <0.0001). During an average follow-up of 12.2 years, we documented 17,895 total deaths, including 4267 from cancer, 2208 from CHD, 867 from stroke, and 997 from diabetes. In the fully adjusted model, higher UPF intake was not associated with all-cause, cancer, CHD, or stroke mortality but showed a significant association with increased diabetes mortality (HR [95% CI] = 1.32 [1.07, 1.62] for the highest versus lowest quartiles [>51.1% vs. <29.3%] and 1.09 [1.04, 1.15] per 10% increase). The adverse UPF-diabetes mortality association was noted regardless of sex, race, income, neighborhood deprivation, lifestyles, and cardiometabolic disease history, while particularly evident in participants with no more than high school education or a history of hypercholesterolemia (HR [95% CI] per 10% increase = 1.12 [1.05, 1.18] and 1.14 [1.07, 1.22], respectively; both P interaction <0.05). Among predominantly low-income Black and White American adults, UPF intake was associated with increased diabetes mortality, especially for individuals with limited education or hypercholesterolemia. Our findings suggest the potential impact of increasing access and intake of un/minimally processed food to replace UPF on reducing diabetes-related mortality among populations facing socioeconomic and health disparities. 20. Exposure to Trihalomethanes and Bone Mineral Density in US Adolescents: A Cross-Sectional Study (NHANES)

Abstract

Animal and human studies have suggested that trihalomethane (THM) has toxicity to bone. In this study, we included adolescents from the National Health and Nutrition Examination Survey who had quantified blood and tap water THM concentrations [chloroform (TCM), bromodichloromethane (BDCM), dibromochloromethane (DBCM), and bromoform (TBM)] and lumbar spine or total body less head (TBLH) bone mineral density (BMD). A 2.7-fold increase in concentrations of blood TCM, DBCM, chlorinated THMs (the sum of TCM, BDCM, and DBCM), and total THMs (the sum of 4 THMs) was associated with lower lumbar spine BMD z-scores by −0.06 [95% confidence interval (CI): −0.12, −0.01], −0.06 (95% CI: −0.11, −0.003), −0.08 (95% CI: −0.14, −0.02), and −0.07 (95% CI: −0.13, −0.003), respectively, in adjusted models. Similarly, a 2.7-fold increase in blood BDCM, DBCM, and chlorinated THM concentrations was associated with lower TBLH BMD z-scores by −0.10 (95% CI: −0.17, −0.02), −0.10 (95% CI: −0.17, −0.03), and −0.11 (95% CI: −0.20, −0.01), respectively. Low-to-moderate predictive power was attained when tap water THM concentrations were used to predict blood THM measurements. Notably, the inverse associations for blood THMs persisted exclusively between water concentrations of DBCM and Br-THMs and the TBLH BMD z-scores. Our findings suggest that exposure to THMs may adversely affect the adolescent BMD.

21. Anomaly Detection in Dynamic Graphs via Transformer

Abstract

Detecting anomalies for dynamic graphs has drawn increasing attention due to their wide applications in social networks, e-commerce, and cybersecurity. Recent deep learning-based approaches have shown promising results over shallow methods. However, they fail to address two core challenges of anomaly detection in dynamic graphs: the lack of informative encoding for unattributed nodes and the difficulty of learning discriminate knowledge from coupled spatial-temporal dynamic graphs. To overcome these challenges, in this paper, we present a novel Transformer-based Anomaly Detection framework for DYnamic graphs (TADDY). Our framework constructs a comprehensive node encoding strategy to better represent each node’s structural and temporal roles in an evolving graphs stream. Meanwhile, TADDY captures informative representation from dynamic graphs with coupled spatial-temporal patterns via a dynamic graph transformer model. The extensive experimental results demonstrate that our proposed TADDY framework outperforms the state-of-the-art methods by a large margin on six real-world datasets.

23. Optimization and Tradeoff of DC Bus Voltage Ripple and DAB Current Stress in Two-Stage Single-Phase Converter by Double-Line-Frequency Power Control

Abstract

With consideration of double-line-frequency instantaneous power in two-stage single-phase system, a double-line-frequency power control method is proposed in this article to achieve optimization and tradeoff control between the current stress of dual-active-bridge (DAB) dc/dc converter and the dc bus voltage ripples in both sides. Different double-line-frequency powers in DAB will lead to different values of DAB current stress and dc bus voltage ripples in both sides. To find out the best double-line-frequency power in DAB, the optimal control trajectory (OCT) is proposed in this article so that the current stress of DAB can be minimized under any certain voltage ripple. An approximate analytical calculation method is derived to solve the optimal double-line-frequency power in OCT. The comparative analysis indicates that the approximate OCT has similar optimization effects as OCT. Based on the OCT, two tradeoff control methods for the DAB current stress and the dc bus voltage ripples are presented so that the current stress and the voltage ripples can be flexibly adjusted to adapt various load conditions. The initial phase of the single-phase ac load is also optimized to maximize the adjustable range of voltage ripple. The experimental results verify the feasibility and effectiveness of the proposed OCT and control methods.

24. Toward economic function of elite social networks

Abstract

Quantifying historical developments is crucial for understanding a large variety of complex processes from economy to political conflict, population dynamics and urban evolution. Yet the driving force of emergent processes in history is a product of complex interactions among massive individuals, in which the elite class plays a vital role. With the social relations of elites in ancient China, the dynamical elite social networks are compiled from the year 618 to 1840. We systematically investigate the dynamical structure of elite social networks, which significantly evolves with major historical events. We detect that the macroeconomic performance affects the structure of elite social networks comprehensively, while only the assortative coefficient and average shortest path length have a predictive ability on the economy. Then the evolutionary game theory is introduced to simulate how the structure of elite social network influences the economy. Our work provides a framework to study the economic function of real-world social networks from the perspective of the elite class.

28. Radiomics Based on DCE-MRI for Predicting Response to Neoadjuvant Therapy in Breast Cancer.

Abstract

To compare the value of radiomics and diameter% based on pre- and early-treatment dynamic enhanced MR (DCE-MRI) of the breast in predicting response to neoadjuvant therapy (NAT) in breast cancer and to construct a tool for early noninvasive prediction of NAT outcomes. Retrospective analysis of clinical and imaging data of 142 patients with primary invasive breast cancer who underwent DCE-MRI before and after two cycles of NAT at our institution. Enroled patients were randomly assigned in a 7:3 ratio to the training group and the test group. Patients were divided into pathological complete response (pCR) and non-pathological complete response groups based on surgical pathology findings after NAT. The maximum diameter relative regression values (Diameter%) before and after treatment were calculated and the conventional imaging Diameter% model was constructed. Based on pre- and early-NAT DCE-MRI, the optimal features of pre-NAT, early-NAT, and delta radiomics were screened using redundancy analysis, least absolute shrinkage, and selection operator methods to construct the corresponding radiomics model and calculate the Radscores. Indicators that were statistically significant in the univariate analysis of clinical data were further screened by stepwise regression and combined with Radscores to construct the fusion model. All models were evaluated and compared. In the test set, the area under the curve (AUC) of the delta radiomics model (0.87) was higher than that of the pre-NAT, early-NAT radiomics models (0.57, 0.78) and the Diameter% model (0.83). The fusion model had the best efficacy in predicting pCR after NAT, with AUCs of 0.91 in the training and test sets. And its nomogram plot showed that Radscore of early-NAT radiomics had the greatest weight. In the test set, the fusion model and Delta radiomics model improved the efficacy of predicting pCR by 35.56% and 14.19%, respectively, compared to the Diameter% model (P = 0 and.039). Clinical decision curves showed the highest overall clinical benefit for the fusion model. Radiomics, especially delta and early-NAT radiomics, may be potential biomarkers for early noninvasive prediction of NAT outcomes. 29. Excessive weight gain among preschool children during the COVID-19 lockdown in China: a retrospective observational study.

Abstract

We aimed to analyse weight change and risk factors associated with excessive weight gain in preschool children during the COVID-19-related lockdown in western China. A retrospective observational study of eight randomly selected kindergartens was conducted. Data was collected via online electronic questionnaires during the lockdown. During the COVID-19 lockdown period, the incidence of overweight and obesity among preschool children involved in the study was 7.6 and 14.7 %, respectively. In addition, the incidence of obesity among children aged 3–4 years was 26.3 %, which was higher than that of other age groups. Children with excessive weight gain (weight gain ≥1.0 kg) spent more time on TV and video viewing during lockdown than children with normal gain. Among children with excessive weight gain, weight before lockdown and fathers' BMI were higher than those of children with normal weight gain. Heavier weight before lockdown (OR 1.044, p<0.05), higher father's BMI (OR 1.022, p<0.05), fresh fruit consumption during lockdown (frequently, ≥5 times/week) (OR 5.946, p<0.05), and long touch-screen device time during lockdown (OR 1.259, p<0.05) were found to be risk factors significantly associated with excessive weight gain. Living space (80–100 m2, OR=0.499, p<0.05; 100–150 m2, OR=0.467, 95 % CI 0.26, 0.83) and good mental behavior during the lockdown (OR=0.056, p<0.05) were found to be significant protective factors against excessive weight gain. There was a clear trend in excessive weight gain among preschool children during the COVID-19 lockdown, especially those who had already had heavier weight before the pandemic. The obesity-promoting environment resulting from the COVID-19 pandemic may lead to a further serious exacerbation of the childhood obesity pandemic. 31. Effects of Sample Dilution on Nuclear Magnetic Resonance-Derived Metabolic Profiles of Human Urine

Abstract

Traditionally, a relatively big urine volume (e.g., 500 μL) is used in nuclear magnetic resonance (NMR)-based human metabolomics, which is not feasible for studies with limited/precious samples. Although urine may be diluted before conventional high-throughput metabolomics analysis, the comprehensive effect of urine dilution on metabolic profiles is unknown. Here, for the first time, we systematically investigated the effect of urine dilution on 1H NMR metabolic profiles, by evaluating signal detectability, integration, signal-to-noise ratio (SNR), chemical shift (δ) and its variation, and signal overlapping of 47 metabolites in 10 volunteers. We observed significant linear changes along with increased dilution, including decreased integration and SNR, altered δ, decreased intersample variation of δ, and increased separation between overlapped signals, e.g., lactate and threonine, β-d-glucose and an unassigned signal, and histidine and 3-methylhistidine. We further tested the 40% dilution level (i.e., employing 300 μL urine) in an epidemiological study containing 1018 pregnant women from the Tongji-Shuangliu Birth Cohort, showing acceptable detectability and chemical shift variability for most of the 47 metabolites profiled. It indicated that mild (e.g., 40%) dilution of human urine can largely preserve the high-abundance metabolites profiled, reduce intersample chemical shift variations, and increase separations of overlapped signals, which is an improvement of routine sample preparation methods in NMR-based metabolomics and is applicable for studies with limited urine volumes, including large-scale epidemiological studies.

32. Task Scheduling in Three-Dimensional Spatial Crowdsourcing: A Social Welfare Perspective

Abstract

Until recently, a novel spatial crowdsourcing paradigm, namely Three-Dimensional (3D) spatial crowdsourcing, has emerged, in which the task requestors and the workers need travel to their designated third-party workplaces, e.g., shared offices, to deliver certain services, such as DiDi station ride-sharing service, Quyundong sport training service in the Online-To-Offline (O2O) applications. In 3D spatial crowdsourcing applications, a core issue is to develop an efficient global tasklist plan, based on the tripartite matching among the three parties, i.e., task requestors, workers and workplaces, which is different from the conventional spatial crowdsourcing. In this context, one key challenge is how to suitably schedule the available workers with the consideration of the interests of all the parties, under the constraint of worker resource. To answer the questions, in this paper, we propose and study a new problem, namely Social-Welfare-driven Task Scheduling (SWTS) problem, which strives to schedule the workers’ continuous routines, i.e., successively implementing tasks for different requestors at different workplaces, to promote the social welfare for all the involved parties. We prove our studied problem is NP-hard, and devise two heuristic optimization algorithms to solve it. Finally, we conduct extensive experiments which verify the efficiency and effectiveness of the proposed algorithms on both real and synthetic data sets.

Abstract

Transarterial chemoembolization (TACE) is an image-guided locoregional therapy used for the treatment of patients with primary hepatocellular carcinoma (HCC). However, conventional TACE formulations such as epirubicin-lipiodol emulsion are rapidly dissociated due to the instability of the emulsion, resulting in insufficient local drug concentrations in the target tumor. To overcome these limitations, we used biodegradable Idarubicin loaded microspheres (BILMs), which were prepared from gelatin and carrageenan and could be loaded with Idarubicin (IDA-MS). The morphology and the ability to load and release IDA of BILMs were characterized in vitro. We evaluated tumor changes and side effects after TACE treatment with IDA-MS in VX2 rabbit and C57BL/6 mice HCC models. In addition, the effect of IDA-MS on the tumor immune microenvironment of HCC tumors was elucidated via mass spectrometry and immunohistochemistry. Result showed that IDA-MS was developed as a new TACE formulation to overcome the poor delivery of drugs due to rapid elimination of the anticancer drug into the systemic circulation. We demonstrated in rabbits and mice HCC models that TACE with IDA-MS resulted in significant tumor shrinkage and no more severe adverse events than those observed in the IDA group. TACE with IDA-MS could also significantly enhance the sensitivity of anti-PD1 immunotherapy, improve the expression of CD8+ T cells, and activate the tumor immune microenvironment in HCC. This study provides a new approach for TACE therapy and immunotherapy and illuminates the future of HCC treatment. 38. Preliminary comparison of total laparoscopic and open radical resection for hepatic hilar cholangiocarcinoma a single-center cohort study.

Abstract

The aim was to compare perioperative outcomes of hilar cholangiocarcinoma (HCCA) radical resection performed through the laparoscopic route or by open surgery in single-center cohort study. A retrospective study included all patients undergoing HCCA radical resection in our center from January 2018 to November 2019. Short-term outcomes of patients with laparoscopic and open surgery were compared demographic characteristics, intraoperative and postoperative parameters and extent of resection. Among 64 patients included, 34 were performed with laparoscopic route and 30 were with open surgery. Laparoscopic versus open group was associated with a longer operation duration [475.50(219.00–630.00) vs. 375.00(220.00–557.00) min, P = 0.031], similar intraoperative bleeding volume [300.00(50.00–3500.00) vs. 350.00(100.00–1500.00) ml, P = 0.426]. No significant differences in tumor diameter [2.00(1.50–3.00) vs. 2.00(1.50–3.00) cm, P = 0.965], the number of lymph nodes retrieved [9.50(6.00–15.00) vs. 8.50(5.00–12.00), P = 0.706], and resection margins (94.1% vs. 86.7%, P = 0.407) between the two approaches were found. The LOS [20.00(10.00–44.00) vs. 21.00(6.00–27.00) d, P = 0.622], bile leakage (23.5% vs. 26.7%, P = 0.781), and liver failure (8.80% vs. 6.70%, P >0.99) were similar in the two groups. There was one patient in open surgery group developing liver failure and finally expired. The safety and radicality of laparoscopic resection for HCCA are similar to open surgery and can be technically improved by rational trocar distribution, total caudate lobectomy and appropriate cholangioenteric anastomosis strategy. Abstract

Recognizing the risk factors for dyslipidemia during pregnancy is crucial for safeguarding the health of both the mothers and the offspring. Growing evidence emerged and suggested links between environmental factors, including metals, and alteration in lipid levels or dyslipidemia in general populations. However, knowledge of the associations during pregnancy remains extremely lacking. Herein, we aimed to explore whether elevated metal exposure constitutes a risk factor for dyslipidemia in pregnant women. Based on the Tongji-Shuangliu Birth Cohort (TSBC), a total of 663 pregnant women were recruited and their urinary levels of 17 metals and blood lipid biomarkers in early pregnancy were measured, namely triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C). The multivariable linear regression models revealed that exposure to selected metals during early pregnancy was significantly associated with some important biomarkers. In particular, after natural log-transformed for the levels of lipid biomarkers and metals, copper (Cu) exposure was positively associated with HDL-C (β = 0.024, 95% CI: 0.001, 0.046), while zinc (Zn) was associated with TG (β = 0.062, 95% CI: 0.013, 0.110) and selenium with TC (β = 0.028, 95% CI: 0.004, 0.054). Exposure to rubidium (Rb) was positively associated with multiple lipid biomarkers, including HDL-C (β = 0.020, 95% CI: 0.002, 0.037) and LDL-C (β = 0.022, 95% CI: 0.001, 0.042). Mixture exposure analysis further identified significant associations between Cu and HDL-C, Zn and TG, Rb and HDL-C, when multiple metal exposures were considered in the Bayesian kernel machine regression model simultaneously. Our findings showed that exposure to several metals during early pregnancy was associated with an increased prevalence of blood lipid abnormalities in pregnant women. These findings underscore the potential impact of metal combinations on lipid metabolism and increase our understanding of the risk factors associated with abnormal lipid metabolism during pregnancy. 44. Quantitative imaging for predicting hematoma expansion in intracerebral hemorrhage: A multimodel comparison.

Abstract

Several studies report that radiomics provides additional information for predicting hematoma expansion in intracerebral hemorrhage (ICH). However, the comparison of diagnostic performance of radiomics for predicting revised hematoma expansion (RHE) remains unclear. The cohort comprised 312 consecutive patients with ICH. A total of 1106 radiomics features from seven categories were extracted using Python software. Support vector machines achieved the best performance in both the training and validation datasets. Clinical factors models were constructed to predict RHE. Receiver operating characteristic curve analysis was used to assess the abilities of non-contrast computed tomography (NCCT) signs, radiomics features, and combined models to predict RHE. We finally selected the top 21 features for predicting RHE. After univariate analysis, 4 clinical factors and 5 NCCT signs were selected for inclusion in the prediction models. In the training and validation dataset, radiomics features had a higher predictive value for RHE (AUC = 0.83) than a single NCCT sign and expansion-prone hematoma. The combined prediction model including radiomics features, clinical factors, and NCCT signs achieved higher predictive performances for RHE (AUC = 0.88) than other combined models. NCCT radiomics features have a good degree of discrimination for predicting RHE in ICH patients. Combined prediction models that include quantitative imaging significantly improve the prediction of RHE, which may assist in the risk stratification of ICH patients for anti-expansion treatments. 45. Structural Simplification of Cryptolepine to Obtain Novel Antifungal Quinoline Derivatives against Phytopathogenic Fungi

Abstract

A series of quinoline derivatives were designed and synthesized by the structural simplification of cryptolepine and evaluated for their fungicidal activity against six phytopathogenic fungi. Most of these compounds exhibited remarkable activities against Botrytis cinereain vitro. Among them, compounds A18and L01showed superior antifungal activity. Significantly, compared to cryptolepine, compound A18exhibited broad-spectrum inhibitory activities against B. cinerea, Sclerotinia sclerotiorum, Rhizoctonia solani, Phytophthora capsica, Magnaporthe oryzae, and Fusarium graminearumwith the respective EC50values of 0.249, 1.569, 3.915, 0.505, 0.246, and 4.999 μg/mL. Compound L01displayed the best antifungal activity against B. cinereawith an EC50value of 0.156 μg/mL. Preliminary mechanistic studies showed that compound A18could inhibit spore germination, affect the permeability of the cell membrane, increase the content of reactive oxygen species, and affect the morphology of hyphae and cells. Moreover, compound A18showed excellent in vivoprotective effect against B. cinerea, which was more potent than pyrimethanil and equitant to cryptolepine. These results evidenced that compound A18displayed superior fungicidal activities and could be a potential fungicidal candidate against plant fungal diseases.

Abstract

Research Object: Taking the acquisition mechanism of oscillatory flapping-wing wave energy as the research object, the scheme design and dynamic analysis of the acquisition mechanism are studied. Research Methods: According to the working principle of the acquisition mechanism of oscillatory flapping-wing wave energy, the rotation dynamics model of the acquisition mechanism and the corresponding relationship between the model parameters and the design parameters of the mechanism are established. Based on the momentum moment theorem, the differential equation of the dynamics of the acquisition mechanism and the transfer function of the system are established. The dynamic response of the acquisition mechanism is analyzed, the system parameters are optimized, and the scheme design is carried out. The conversion efficiency of the acquisition mechanism and the energy collection efficiency under different sea conditions are calculated. The motion response of the acquisition mechanism is simulated and analyzed by MATLAB and AQWA software. Research Results: The results show that the response of the crank and swing slider acquisition mechanism is fast and stable, and the average conversion efficiency is 34%. The research process provides an effective reference for the development of the acquisition mechanism of oscillatory flapping-wing wave energy.

Abstract

The inhibitory effect of tavaborole on the invasion of Botrytis cinereain grapes and tomatoes, as well as the potential mechanism involved, was discovered in this study. Our findings showed that tavaborole inhibited Botrytis cinereaspore germination and mycelial expansion in vitro and that the control efficiency in vivo on fruit decay was dose-dependent, which was effective in reducing disease severity and maintaining the organoleptic quality of the fruit, such as reducing weight loss and retaining fruit hardness and titratable acid contents during storage. Furthermore, the precise mechanism of action was investigated further. Propidium iodide staining revealed that Botrytis cinereatreated with tavaborole lost membrane integrity. For further validation, cytoplasmic malondialdehyde accumulation and leakage of cytoplasmic constituents were determined. Notably, the inhibitory effect was also dependent on inhibiting the activities of aminoacyl-tRNA synthetases involved in the aminoacyl-tRNA biosynthesis pathway in Botrytis cinerea. The above findings concluded that tavaborole was effective against Botrytis cinereainfection in postharvest fruit, and a related mechanism was also discussed, which may provide references for the drug repurposing of tavaborole as a postharvest fungicide.
