Aberrant activation of the Wnt/β-catenin signaling is associated with tumor development, and blocking β-catenin/BCL9 is a novel strategy for oncogenic Wnt/β-catenin signaling. Herein, we presented two novel β-catenin variations and exposed conformational dynamics in several β-catenin crystal structures at the BCL9 binding site. Furthermore, we identified a class of novel urea-containing compounds targeting β-catenin/BCL9 interaction. Notably, the binding modalities of inhibitors were greatly affected by the conformational dynamics of β-catenin. Among them, 28had a strong affinity for β-catenin (Kd= 82 nM), the most potent inhibitor reported. In addition, 13and 35not only activate T cells but also promote the antigen presentation of cDC1, showing robust antitumor efficacy in the CT26 model. Collectively, our study demonstrated a series of potent small-molecule inhibitors targeting β-catenin/BCL9, which can enhance antigen presentation and activate cDC1 cells, delivering a potential strategy for boosting innate and adaptive immunity to overcome immunotherapy resistance.

Under the background of “carbon neutral”, lithium-ion batteries (LIB) have been widely used in portable electronic devices and large-scale energy storage systems, but the current commercial electrolyte is mainly liquid organic compounds, which have serious safety risks. In this paper, a bilayer heterogeneous composite solid-state electrolyte (PLPE) was constructed with the 3D LiX zeolite nanofiber (LiX-NF) layer and in-situ interfacial layer, which greatly extends the life span of lithium metal batteries (LMB). LiX-NF not only offers a continuous fast path for Li+, but also zeolite’s Lewis acid–base interaction can immobilize large anions, which significantly improves the electrochemical performance of the electrolyte. In addition, the in-situ interfacial layer at the electrode–electrolyte interface can effectively facilitate the uniform deposition of Li+and inhibit the growth of lithium dendrites. As a result, the Li/Li battery assembled with PLPE can be stably cycled for more than 2500 h at 0.1 mA cm–2. Meanwhile, the initial discharge capacity of the LiFePO4/PLPE/Li battery can be 162.43 mAh g–1at 0.5 C, and the capacity retention rate is 82.74% after 500 cycles. These results emphasize that this bilayer heterogeneous composite solid-state electrolyte has distinct properties and shows excellent potential for application in LMB.

Skeletal muscle regeneration is a complex process where various cell types and cytokines are involved. Single-cell RNA-sequencing (scRNA-seq) provides the opportunity to deconvolute heterogeneous tissue into individual cells based on their transcriptomic profiles. Recent scRNA-seq studies on mouse muscle regeneration have provided insights to understand the transcriptional dynamics that underpin muscle regeneration. However, a database to investigate gene expression profiling during skeletal muscle regeneration at the single-cell level is lacking. Here, we collected over 105 000 cells at 7 key regenerative time-points and non-injured muscles and developed a database, the Single-cell Skeletal Muscle Regeneration Database (SCSMRD). SCSMRD allows users to search the dynamic expression profiles of genes of interest across different cell types during the skeletal muscle regeneration process. It also provides a network to show the activity of regulons in different cell types at different time points. Pesudotime analysis showed the state changes trajectory of muscle stem cells (MuSCs) during skeletal muscle regeneration. This database is freely available at https://scsmrd.fengs-lab.com.

Schizophrenia (SCZ) is influenced by a combination of genetic and environmental factors. Although several studies have been conducted to identify the causative loci and genes, few of these loci or genes can be repeated due to the high phenotypic and genetic heterogeneity of disease, and their mechanisms are not fully understood. There may be some “missing heritability” that has not yet been found. In order to investigate the deleterious heritable mutations, whole‐exome sequencing (WES) in pedigrees with SCZ was used in the current work. Two unrelated pedigrees with SCZ were recruited to perform WES. Genetic analysis was next performed to find potential variants in accordance with the prioritized strategy. Followed by genetic analysis to detect candidate variants according to the prioritized strategy. Next, a series of algorithms was used to predict the pathogenicity of variants. Sanger sequencing was finally conducted to verify the co‐segregation. Recessive mutations in six genes (TFEB, SNAI2, TFAP2B, PRKDC, ST18in Pedigree 1 and PKHD1L1in Pedigree 2) that co‐segregated with SCZ in two families were discovered through genetic analysis by WES. Sanger sequencing verified that all of the mutations in the affected siblings were homozygous. These results corroborated the hypothesis that SCZ exhibits strong heterogeneity and complex inheritance patterns. The newly discovered homozygous variations deepen our understanding of the mutation spectrum and offer more proof for the involvement of TFEB, SNAI2, TFAP2B, PRKDC, ST18,and PKHD1L1in the development of SCZ.

The construction joint is the weak part of the structure, and the P-δ effect is mostly ignored when considering the second-order effect, so it is necessary to study the influence of second-order effect on frame columns. Based on the above considerations, under different axial compression ratios, the mechanical properties of the construction joints of the frame columns with construction joints and the cast-in-situ frame columns were studied by low cycle repeated load testsand analyzed the influence of the second-order effect on the shear capacity of frame columns with joints. The test results indicate that the existence of construction joints reduces the shear-bearing capacity of the specimens, and the second-order effect has a greater impact on the columns with joints under the same axial compression ratio, and the shear capacity decreases more. With the increase of the axial compression ratio, the second-order effect will be weakened on the frame column with seam, but when the axial compression ratio exceeds a certain limit, the second-order effect will be increased.

This article through to the two common cast-in-situ frame structure (casting of a whole, with a construction joint) and 6 root not construction joints under axial pressure ratio through concrete columns of different processing low reversed cyclic loading experiment, study their experimental phenomena, hysteresis curves, displacement ductility, stiffness degradation, thus draw the conclusion: 1, the existence of the construction joint can lead to the decline of ductility of frame structure. 2. When the test axial pressure is relatively low, the presence or absence of construction joints and different treatment methods will obviously affect the ductility of the cast-in-place frame structure. 3. When the test axial pressure is relatively high, the presence or absence of construction joints and different treatment methods will not have a significant impact on the ductility of the cast-in-place frame structure.

The interaction between Pt and its various supports can regulate the intrinsic electronic structure of Pt particles and their catalytic performance. Herein, Pt/CeO2and Pt/SiC catalysts were successfully prepared via a facile Pt colloidal particle deposition method, and their catalytic performance in CO oxidation was investigated. XRD, TEM, XPS and H2-TPR were used to identify the states of Pt particles on the support surface, as well as their effect on the performance of the catalysts. Formation of the Pt–O–Ce interaction is one of the factors controlling catalyst activity. Under the oxidative treatment at low temperature, the Pt–O–Ce interaction plays an important role in improving the catalytic activity. After calcining at high temperature, enhanced Pt–O–Ce interaction results in the absence of metallic Pt0on the support surface, as evidenced by the appearance of Pt2+species. It is consistent with the XPS data of Pt/CeO2, and is the main reason behind the deactivation of the catalyst. By contrast, either no interaction is formed between Pt and SiC or Pt nanoparticles remain in the metallic Pt0state on the SiC surface even after aging at 800 °C in an oxidizing atmosphere. Thus, the Pt/SiC shows better thermal stability than Pt/CeO2. The interaction between Pt and the active support may be concluded to be essential for CO oxidation at low temperature, but strong interactions may induce serious deactivation of catalytic activity.

Due to the high safety, flexibility, and excellent compatibility with lithium metals, composite solid-state electrolytes (CSEs) are the best candidates for next-generation lithium metal batteries, and the construction of fast and uniform Li+transport is a critical part of the development of CSEs. In this paper, a stable three-dimensional metal–organic framework (MOF) network was obtained using polydopamine as a medium, and a high-performance CSE reinforced by the three-dimensional MOF network was constructed, which not only provides a continuous channel for Li+transport but also restricts large anions and releases more mobile Li+through a Lewis acid–base interaction. This strategy endows our CSEs with an ionic conductivity (7.1 × 10–4S cm–1at 60 °C), a wide electrochemical window (5.0 V), and a higher Li+transfer number (0.54). At the same time, the lithium symmetric batteries can be stably cycled for 2000 h at 0.1 mA cm–2, exhibiting excellent electrochemical stability. The LiFePO4/Li cells have a high initial discharge specific capacity of 153.9 mAh g–1at 1C, with a capacity retention of 80% after 915 cycles. This paper proposes an approach for constructing three-dimensional MOF network-enhanced CSEs, which provides insights into the design and development of MOFs for the positive effects of high-performance CSEs.

Commercial Li-ion batteries use LiPF6-based carbonate electrolytes extensively, but there are many challenges associated with them, like dendritic Li growth and electrolyte decomposition, while supporting the aggressive chemical and electrochemical reactivity of lithium metal batteries (LMBs). This work proposes 1,1,1,3,3,3-hexafluoroisopropyl methacrylate (HFM) as a multifunctional electrolyte additive, constructing protective solid-/cathode-electrolyte interphases (SEI/CEI) on the surfaces for both lithium metal anode (LMA) and Ni-rich cathode to solve these challenges simultaneously. The highly fluorinated group (−CF3) of the HFM molecule contributes to the construction of SEI/CEI films rich in LiF that offer excellent electronic insulation, high mechanical strength, and surface energy. Accordingly, the HFM-derived LiF-rich interphases can minimize the electrolyte–electrode parasitic reactions and promote uniform Li deposition. Also, the problems of LiNi0.8Co0.1Mn0.1O2particles’ inner microcrack evolution and the growth of dendritic Li are adequately addressed. Consequently, the HFM additive enables a Li/LiNi0.8Co0.1Mn0.1O2cell with higher capacity retention after 200 cycles at 1 C than the cell with no additive (74.7 vs 52.8%), as well as a better rate performance, especially at 9 C. Furthermore, at 0.5/0.5 mAh cm–2, the Li/Li symmetrical battery displays supersteadfast cyclic performance beyond 500 h when HFM is present. For high-performance LMBs, the HFM additive offers a straightforward, cost-effective route.

Li-metal batteries (LMBs), especially in combination with high-energy-density Ni-rich materials, exhibit great potential for next-generation rechargeable Li batteries. Nevertheless, poor cathode–/anode–electrolyte interfaces (CEI/SEI) and hydrofluoric acid (HF) attack pose a threat to the electrochemical and safety performances of LMBs due to aggressive chemical and electrochemical reactivities of high-Ni materials, metallic Li, and carbonate-based electrolytes with the LiPF6salt. Herein, the carbonate electrolyte based on LiPF6is formulated by a multifunctional electrolyte additive pentafluorophenyl trifluoroacetate (PFTF) to adapt the Li/LiNi0.8Co0.1Mn0.1O2(NCM811) battery. It is theoretically illustrated and experimentally revealed that HF elimination and the LiF-rich CEI/SEI films are successfully achieved via the chemical and electrochemical reactions of the PFTF additive. Significantly, the LiF-rich SEI film with high electrochemical kinetics facilitates Li homogeneous deposition and prevents dendritic Li from forming and growing. Benefiting from the collaborative protection of PFTF on the interfacial modification and HF capture, the capacity ratio of the Li/NCM811 battery is boosted by 22.4%, and the cycling stability of the symmetrical Li cell is expanded over 500 h. This provided strategy is conducive to the achievement of high-performance LMBs with Ni-rich materials by optimizing the electrolyte formula.

A new evaluation index system, which includes five dimensions is put forward to evaluate the competitiveness of construction subcontracting enterprise properly. Based on GA optimized BP neural network model,construction subcontracting enterprises’ competitiveness can be quantitative analysis systematically. Use of Matlab simulation analysis,research has shown that this system can well solve the problem of construction subcontracting enterprise competitiveness evaluation.

By way of the soaking and drying cycle test on the different mix proportions vertical section of 10 pieces of RC beams suffered artificial sea water (ASW) corrosion under0,35,70,105,140 times of dry-wet cycles, the compared result of exerting pressure test of these beams under simply supporting were investigated. The law about the changes of the mechanical performance for RC beams with different mix proportions under different time periods for suffering corrosion of dry-wet cycles was as follows: The resistibility to ASW corrosion for the concrete specimens with various water cement ratio (various initial strength) are different;The characters of normal section failure for RC beams attacked by sea water are about the same as that for ordinary RC beam; Along with the extension of the time for seawater attack, the bearing capacity for normal section of RC beams varies wave upon wave. In this paper, in order to simulate the corrosion of seawater, the drying-wetting cycling test was used. The mechanical properties of carbon fiber-reinforced concrete beam under salt damage were studied and the reinforcement effect of the CFRP was analyzed. At the same time, the bearing capacity of cross section of CFRP strengthened PC beam under corroded was analyzed.

Based on experiment of a two-bay two-story reinforced concrete frame with construction joint under low-reversed cyclic loading, hysteretic performance, displacement ductility, bearing capacity degeneration, rigidity degeneration, energy dissipation and displacement restoring capacity are studied systematically. The studies show that the frame with construction joint has not better seismic performance than the monolithic cast frame.

According to the quasi static test of reinforced concrete frame with construction joints in, the fracture crack sequence, fracture morphology and plastic hinge are studied in detail. Research shows that two kinds processing method of construction joint will not affect the destruction form of a frame structure, but destruction order .

In this paper,in order to study the load-bearing capacity of frame columns with construction joint under low-cyclic reversed loading, nine specimens were made, divided into three groups.Every group had three specimens, the first were made without construction joint, the second had fine processing construction joint, the third had simple processing construction joint. The three groups were loaded in axial compression ratio of 0.23,0.34,0.46 respectively.The results show that the influence of different processing with construction joint to crack position ultimate bearing capacity,skeleton curves.

Abstract. In order to generate mesh for arch dam abutment slip block automatically, virtual grid method is proposed based on grid-based automatic generation algorithm. The proposed algorithm avoids generating initial grid in one time, but intersects the model with a series of plane sections firstly. Then form initial grid by using vector intersection method for adjacent sections. By modifying initial grid, a layer of mesh is formed. The algorithm proposed can better adapt to complex topography and generates hexahedral meshes for the block of arch dam abutment automatically. The method can present data for limit equilibrium analysis and numerical calculation method such as finite element method. Based on original topography and structural plane data, the automatic identification, generation and meshing of arch dam abutment block is accomplished.

The finite element models of biaxial shear capacity of RC frame columns are investigated.The calculation results and experimental results are compared using the different models.It is shown that different finite element models are effectively used to simulate the RC frame column. However, the simplified model may save computer time and be more convenient for the numerical simulation. Finally, the bilateral shear correlation of RC frame column is investigated using the simplified model. The results indicate that the bilateral shear correlation of the RC frame columns with different quantity of stirrups in two directions accords with the ellipse law and the fitting formula is presented.

Elaborates the phenomenon of enhancement of the concrete compressive strength and the bearing capacity and cracking load of test beams, and makes an indepth analysis on the mechanism of this phenomenon, at the same time the concept of the additional expansion prestress is introduced; a mathematical model of the additional expansion prestress is establish and the computational model of the cracking load is derived.

In this paper, a comprehensive mathematical model of load flow for Flexible AC Transmission Systems (FACTS) with Unified Power Flow Controller (UPFC) is presented basing on the analysis of basic principle and operation states for UPFC. The model can use fast P-Q decoupled load flow method. Examples of test system show that the proposed model and method have good convergence. An effective tool is provided by the load flow computer program to analyze the load flow and calculate initial value of dynamic state for FACTS with UPFC.

A highly sensitive and selective method for the determination of low concentrations of nitrite in aqueous solutions using surface-enhanced resonance Raman spectroscopy (SERRS) is presented. The SERRS-active substrate was silver hydrosol. In order to induce the SERRS effect, nitrite was transformed into a colored product (an azo dye) by a standard chemical reaction. The detection limit for nitrite is 0.02 nM in distilled water, and analytical calibration curves yield straight lines in the concentration range below 30 nM. The sensitivity of this method is better than that of colorimetry and other techniques such as chemiluminescence. This improvement indicates that SERRS shows great promise as a useful analytical tool for trace analysis of nitrite and dyes.

The electrochemical characteristics of boron-doped polycrystalline diamond thin film electrodes with 4×4 mm2 in size have been studied using cyclic voltammetry and a potentiostatic method. The diamond electrode exhibits adequate electrochemical activity and the response current changes linearly with the K3Fe(CN)6 concentrations. The response current is proportional to the square root of the scan rate, reflecting the mass transport controlled by planar diffusion. Stable mass transport was quickly established within 4 s. Good stability and reproducibility are attractive properties of the diamond electrode for sensor applications.
