Social insects display extreme phenotypic differences between sexes and castes even though the underlying genome can be almost identical. Epigenetic processes have been proposed as a possible mechanism for mediating these phenotypic differences. Using whole genome bisulfite sequencing of queens, males, and reproductive female workers we have characterised the sex- and caste-specific methylome of the bumblebee Bombus terrestris. We have identified a potential role for DNA methylation in histone modification processes which may influence sex and caste phenotypic differences. We also find differentially methylated genes generally show low levels of DNA methylation which may suggest a separate function for lowly methylated genes in mediating transcriptional plasticity, unlike highly methylated genes which are usually involved in housekeeping functions. We also examined the relationship between the underlying genome and the methylome using whole genome re-sequencing of the same queens and males. We find DNA methylation is enriched at zero-fold degenerate sites. We suggest DNA methylation may be acting as a targeted mutagen at these sites, providing substrate for selection via non-synonymous changes in the underlying genome. However, we did not see any relationship between DNA methylation and rates of positive selection in our samples. In order to fully assess a possible role for DNA methylation in adaptive processes a specifically designed study using natural population data is needed.

Abstract.??In insect societies, eggs laid by workers are frequently killed by other workers ? a behaviour known as ?worker policing?. The traditional explanation of worker policing is that it is a mechanism to resolve intracolony conflict, and maintain the reproductive monopoly of the queen. Recently, Pirk et al. (2004) proposed that worker policing instead is aimed at removing unviable worker-laid eggs and is ultimately just another example of hygienic behaviour. Here we test this hypothesis for the common waspVespula vulgaris, a species with highly effective worker policing. We show that worker-laid eggs from queenless colonies have a lower hatch rate (68%) than queen-laid eggs (82%). Analysis of egg laying rates of queens and workers, however, shows that the difference is not big enough to explain the apparent absence of adult worker-derived males in this species.

Abstract: In ca. 150 species of queenless ants, a specialized queen caste is rare or absent, and mated workers take over the role of the queen in some or all of the colonies. Previously, it has been shown that reproduction in queenless ants is regulated by a combination of dominance behavior and chemical fertility signaling. It is unknown, however, whether chemical signals alone can sufficiently regulate reproduction. To investigate this possibility, we studied reproductive regulation in the facultatively queenless ant Gnamptogenys striatula, a species where dominance behavior is rare or absent. Active egg layers and infertile workers showed qualitative and quantitative differences in their cuticular hydrocarbon profile. Five long-chain methyl alkanes, 3,13- and 3,15-dimethyl pentriacontane, 3,13- and 3,15-dimethyl heptentriacontane, and 3,11,15-trimethyl heptentriacontane occurred only on the cuticles of virgin and mated egg layers. Pronounced quantitative differences were found in a further 27 alkenes; alkanes; and mono-, di-, and trimethyl alkanes. Workers that had recently stopped laying eggs had profiles similar to infertile workers, and mating status did not affect this chemical pattern. We conclude that the cuticular hydrocarbon profiles of G. striatula workers provide reliable information about their current fertility. In the interest of colony productivity, this allows reproduction to be regulated without the use of aggression.

Wolbachiais a maternally inherited bacterium that may manipulate the reproduction of its arthropod hosts. In insects, it is known to lead to inviable matings, cause asexual reproduction or kill male offspring, all to its own benefit, but to the detriment of its host. In social Hymenoptera, Wolbachiaoccurs widely, but little is known about its fitness effects. We report on a Wolbachiainfection in the wood ant Formica truncorum, and evaluate whether it influences reproductive patterns. All 33 colonies of the study population were infected, suggesting that Wolbachiainfection is at, or close to, fixation. Interestingly, in colonies with fewer infected workers, significantly more sexuals are produced, indicating that Wolbachiahas deleterious effects in this species. In addition, adult workers are shown to have significantly lower infection rates (45) than worker pupae (87) or virgin queens (94), suggesting that workers lose their infection over life. Clearance of Wolbachiainfection has, to our knowledge, never been shown in any other natural system, but we argue that it may, in this case, represent an adaptive strategy to reduce colony load. The cause of fixation requires further study, but our data strongly suggest that Wolbachiahas no influence on the sex ratio in this species.

Here we report on territorial behavior in the desert ant Cataglyphis niger. We show that, similar to the “bourgeois strategy,” contests are settled quickly through nonescalating fights, with the owner of the territory usually being placed in the winning role. The chemical cue that advertises ownership is shown to come from pheromones secreted by the cloacal gland. Differences in size did not influence the outcome of the experimental contests, but resource value did. The observed results fit in well with the biology of Cataglyphis, where, due to the harsh semidesert environment, fighting costs (C) are expected to be high relative to the value of the contested resources (V). A situation where V < C is exactly that in which bourgeois behavior is expected to be seen.

In hymenopteran societies, workers are not always sterile, and may produce parthenogenetically either males (arrhenotoky) or females (thelytoky). Thelytoky however is exceptional, and has been recorded in only four ant species. Here we provide evidence for worker thelytoky in an additional species, the harvesting ant Messor capitatus (Latreille) (Hymenoptera Formicidae Myrmicinae). Two orphaned colonies produced a large amount of worker offspring during 10 months in the laboratory. Dissections showed that reproduction was performed by workers and not by mated worker-like individuals (ergatoids). In some parasitoid wasps, parthenogenetic reproduction is caused by the maternally transmitted bacterium Wolbachia. Using a PCR-based assay we showed that Wolbachia can not be involved in parthenogenesis induction in this species. Finally, we point out reasons for the low Wolbachia susceptibility of parthenogenetic ant species.

For more than 20 years, sex allocation in hymenopteran societies has been a major topic in insect sociobiology. A recurring idea was that relatedness asymmetries arising from their haplodiploid sex determination system would lead to various parent–offspring conflicts over optimal reproduction. A possible weakness of existing theory is that only interests of nuclear genes are properly accounted for. Yet, a diversity of maternally transmitted elements manipulate the reproduction of their host in many solitary arthropod groups. The bacterium Wolbachiais a striking example of such a selfish cytoplasmic element, with effects ranging from reproductive incompatibility between host strains, induction of parthenogenesis and feminization of males. This paper reports on a first PCR–based Wolbachiascreening in ants. Out of 50 Indo–Australian species, 50% screened positive for an A–group strain. One of these species also harboured a B–group strain in a double infection. Various factors that might explain the unusually high incidence of Wolbachiain ants are discussed. In general, Wolbachiamay represent a widespread and previously unrecognized party active in the conflicts of interest within social insect colonies.
