Zusammenfassung: Zwei atomabsorptionsspektrometrische Verfahren, eine Direktbestimmung im Graphitrohrofen und eine Naßveraschung mit Salpetersäure, gefolgt von einer Messung mit der Hydridtechnik, werden für die Bestimmung von Selen in menschlichem Blutserum verglichen. Das Direktverfahren mit Graphitrohrofen ist störfrei, wenn mit einer Zeemaneffekt-Untergrundkompensation, Atomisierung von einer L'vov-Plattform, und einer Isoformierungshilfe aus Kupfer und Eisen zur Matrixmodifikation gearbeitet wird. Auch die Bestimmung mit der Hydridtechnik ist störungsfrei, wenn mit einem genügend kleinen Volumen Aufschlußlösung (entsprechend 20 μl Serum) in einem ausreichend großen Volumen Salzsäure gemessen wird. Die mit der Graphitrohrofenbzw. der Hydrid-AAS-Technik gefundenen Mittelwerte für Selen in Serum sind 60 bzw. 68 μg/1. Die niedrigeren Werte, die mit dem Direktverfahren gefunden werden, beruhen möglicherweise auf Verlusten an einer flüchtigen Selenverbindung, die im unbehandelten Serum vorliegt. Aber selbst mit einer Naßveraschung liegen die von uns gefundenen Werte niedriger als die meisten anderen publizierten „Normalwerte“ für Selen in Serum.

Summary: Cadmium and nickel are determined in Coal, Coal Fly Ash and Urban Particulate Matter Reference Materials using solid sampling graphite furnace AAS. The evaluation of integrated absorbance is required to overcome the effect of the matrix on the rate of atomization. The homogeneity of the samples under investigation in the range between 0.3 mg and 1.5 mg sample weight is good enough to make possible relative standard deviations around 10% for cadmium and between 10 and 20% for nickel. Cadmium can be determined against reference solutions, the nickel results are slightly lower than the certified value if calibration is performed against aqueous solutions. Calibration against a solid reference is therefore recommended. Less sensitive resonance lines and an internal gas flow through the tube are required for some of the samples to keep the absorbance in the linear calibration range. The direct determination of chromium in these samples is hampered by the lack of less sensitive resonance lines. Due to high concentrations of chromium in the samples, direct determination without dilution of the sample e.g. by spectrally pure graphite powder is impossible. The peak for the refractory carbide forming element vanadium is strongly suppressed by the Coal and the Urban Particulate matrix. Integration of the signal within a reasonable time and at an atomization temperature of 2650 C is impossible.

Für eine Reihe von Elementen wird die Empfindlichkeit in Rohren für die elektrothermische Atomabsorptionsspektrometrie aus pyrolytisch beschichtetem und unbeschichtetem polykristallinem Elektrographit und aus glasartigem Kohlenstoff verglichen. Die wirksamste Atomisierung wird stets in pyrolytisch beschichteten Graphitrohren erzielt, unabhängig von der Flüchtigkeit des Elements und von dem Atomisierungsmechanismus. Wir nehmen an, daß die ausgeprägten Hafteigenschaften von Metallatomen an aktiven Stellen von polykristallinem Elektrographit und glasartigem Kohlenstoff für dieses Verhalten verantwortlich sind. Diese Theorie wird gestützt von den beobachteten Verschiebungen der Peakmaxima, Peakverbreiterungen und den höheren Atomisierungstemperaturen, die in diesen Rohren verwendet werden müssen. Für Elemente, die stabile Carbide bilden, können diese Effekte durch chemische Reaktionen mit dem Rohrmaterial noch verstärkt werden.

An experimental setup for gas-temperature diagnostics using Coherent Anti-Stokes Raman Scattering (CARS) in very small sample volumes with great spatial and temporal temperature variations is presented. Studies have been started of a newly designed transversely heated graphite-tube atomizer for atomic absorption spectrometry (Perkin-Elmer, 4100 ZL). For the investigations, high spatial resolution and precise time synchronization of the measurements and also automatic control of the time-dependent intensity of the anti-Stokes signal were realized. The graphite-tube atomizer also offers the possibility of high-temperature-gas spectroscopy. A high-temperature CARS spectrum of CO2, recorded in the graphite tube at 2300 K, is shown for the first time. A number of hot-band transitions in the spectral region of the Fermi doublet at a Raman shift from 1230 to 1450cm-1 was observed and assigned, making possible the use of CO2 as “spectroscopic thermometer gas”, similar to N2.

Spatial non-uniformities in analyte, atomizer gas phase temperature and radiant intensity distributions characteristic for graphite furnace atomic absorption spectrometry are briefly summarized and their effect on the analyte detection is analysed. It is shown that conventional detection of analyte based on the use of photomultiplier tubes provides excellent temporal resolution, sufficient wavelength isolation but totally ignores the spatial aspects of the interaction of the probing radiation beam with the analyte in the atomizer. A new, spatially resolved, method of analyte detection based on the use of a solid state detector located along the monochromator slit is presented. The approach is illustrated by the temporally and spatially resolved detection of Cd atomization and NaCl vaporization. It is shown that severe non-uniformities in atomic and/or background absorbance may be a potential source of analytical error. Advantages of spatially resolved detection as compared with conventional detection are discussed.

The results of an investigation of the three-dimensional distribution of radiant intensity in the probing radiation beam produced in a conventional atomic absorption spectrometer by hollow cathode lamps and electrodeless discharge lamps are presented. The investigation is based on the use of a photodiode array-based digital imaging system. The results obtained revealed that the probing radiation beam is highly non-uniform, both longitudinally and radially. The character of the non-uniformities is greatly dependent on the type of the primary source. An interpretation of the results is given and the possible consequences of the radiation beam non-uniformities for the application of Beer-Lambert's law are discussed.

Es wird ein Überblick über morphologische Studien an Oberflächen der elektrothermischen AAS mit Hilfe der Rasterelektronenmikroskopie gegeben. Prinzipielle Möglichkeiten werden diskutiert wie Störungen, die mit dem Material oder der Oberfläche der Atomisierungseinrichtung zusammenhängen, verringert oder beseitigt werden können. An zahlreichen Materialien, die entweder schon verbreitet Anwendung finden, oder sich noch in der Entwicklungsphase befinden, wurden Oberflächen von neuen und korrodierten Teilen studiert. Zu den untersuchten Materialien gehörenRohre aus polykristallinem Elektrographit mit Pyrographitbeschichtung oder Metallsalzimprägnierung,Plattformen aus massivem Pyrographit,Rohre aus massivem Pyrographit,Rohre aus glasartigem Kohlenstoff,Rohre aus Wolfram,Rohre und Plattformen mit Refraktärmetallcarbidbeschichtung durch physikalische Abscheidung aus der Dampfphase.

Ein neuartiges Gerät zur automatischen Probendosierung bei der flammenlosen AAS mit Graphitrohren wurde untersucht. Mit Lösungen von Cd, Pb, Cu und Ni wurde die erreichbare Reproduzierbarkeit, der Verschleppungsfaktor durch zusätzliche Experimente mit 89Sr, die Lebensdauer kommerziell erhältlicher Graphitrohre, der Einfluß einiger ausgewählter Matrices sowie einer pyrolytischen Beschichtung der Oberfläche der Graphitrohre auf die Meßwerte ermittelt. Die Ergebnisse der Studien, die hieraus ersichtlichen Vorteile des Systems sowie zukünftige Einsatzmöglichkeiten werden diskutiert.

A graphite tube furnace for flameless Atomic Absorption Spectroscopy and its application in the analysis of biological materials is described. The values obtained for copper in 12 sera are in good agreement with colorimetric results and Flame-AAS. The sample volume meeded for one determination is only 2 µl of serum. Several further application possibilities for the determination of trace elements and for smallest sample volumes are discussed.

Al, Cr, Cu, Fe, Hg, Mn, and Pb are determined in Lake Constance and the major rivers to the lake, and compared with the contents of ammonium, chloride and phosphate. Correlations between elevated contents of trace elements and the general degree of contamination are checked. As a direct method, flameless Atomic Absorption offers the advantage of high accuracy and precision in the µg/l-range.

Al, Cr, Cu, Fe, Hg, Mn und Pb werden im Bodensee und seinen wichtigsten Zuflüssen bestimmt und mit den Werten für Ammonium, Chlorid und Phosphat verglichen. Zusammenhänge zwischen erhöhten Spurenelementgehalten und dem allgemeinen Verschmutzungsgrad werden geprüft. Die flammenlose Atom-Absorption bietet als Direktverfahren den Vorteil hoher Präzision und Richtigkeit im μg/l-Bereich.

A method has been developed for the direct determination of copper in petroleum condensate using detergentless micro-emulsions three-component solutions and graphite furnace atomic absorption spectrometry. The optimization was performed using a three-level full factorial design, resulting in pyrolysis and atomization temperatures of 920 °C and 2600 °C, respectively. Using the secondary copper line at 327.4 nm, which is less affected by the magnetic field of the Zeeman-effect background correction system, a characteristic mass of 39 pg, and detection limit of 0.4 μg l−1was obtained using 20 μl of micro-emulsion, which corresponds to 1.2 μg l−1in petroleum condensate. The precision, expressed as relative standard deviation, was 2.0 or 2.8 for copper concentrations of 30 μg l−1and 5.0 μg l−1, respectively. The spike recovery of 105 for copper added to a petroleum condensate sample proved that this procedure could be applied satisfactorily to a determination in this matrix.
