Despite the high diversity of histamine H3receptor (H3R) antagonist/inverse agonist structures, partial or full H3R agonists have typically been imidazole derivatives. An in-house screening campaign intriguingly afforded the non-imidazole 4-(3-azetidin-1-yl)pyrimidin-2-amine 11bas a partial H3R agonist. Here, the design, synthesis, and structure–activity relationships of 11banalogues are described. This series yields several non-imidazole full agonists with potencies varying with the alkyl substitution pattern on the basic amine following the in vitro evaluation of H3R agonism using a cyclic adenosine monophosphate response element-luciferase reporter gene assay. The key compound VUF16839 (14d) combines nanomolar on-target activity (pKi= 8.5, pEC50= 9.5) with weak activity on cytochrome P450 enzymes and good metabolic stability. The proposed H3R binding mode of 14dindicates key interactions similar to those attained by histamine. In vivo evaluation of 14din a social recognition test in mice revealed an amnesic effect at 5 mg/kg intraperitoneally. The excellent in vitro and in vivo pharmacological profiles and the non-imidazole structure of 14dmake it a promising tool compound in H3R research.

The different weed species and its density influenced the nutrient content of pea in a different way. In our experiment the measured numerical values of phosphorus and potassium were similar to previous results of Debreczeniné (1994) and Nádasyné (1999). The growth rate of weed species was not uniform, S. arvensisgrew the quickest, at the end of the experiment it covered the most of the pot’s surface. E. crus-galligrew well too. We observed slender growth by A. retroflexusand C. albumplants, it follows from this that nutrients uptake and competition ability was rather moderate. Weeds through intensive nutrient uptake mostly decreased the nutrient content of green pea. S. arvensishindered strongest the intensive nutrient uptake of crop plants. The phosphorus and potassium content of wild mustard were not influenced by its density. The nitrogen, phosphorus and potassium content of A. retroflexusreduced with increased density, so it has an intraspecific competitive effect.

The metabolite‐modulated association of a fraction of hexokinase to mitochondria in brain is well documented, however, the involvement of other non‐mitochondrial components in the binding of the hexokinase is controversial. Now we present evidence that the hexokinase binds both tubulin and microtubules in brain in vitro systems. The interaction of tubulin with purified bovine brain hexokinase was characterized by displacement enzyme‐linked immunosorbent assay using specific anti‐brain hexokinase serum (IC50=4.0±1.4 μM). This value virtually was not affected by specific ligands such as ATP or glucose 6‐phosphate. Microtubule‐bound hexokinase obtained in reconstituted systems using microtubule and purified hexokinase or brain extract was visualized by transmission and immunoelectron microscopy on the surface of tubules. The association of purified bovine brain hexokinase with either tubulin or microtubules caused about 30% increase in the activity of the enzyme. This activation was also observed in brain, but not in muscle cell‐free extract. The possible physiological relevance of the multiple heteroassociation of brain hexokinase is discussed.

In a Hungarian family with severe decrease in triosephosphate isomerase (TPI) activity, 2 germ line–identical but phenotypically differing compound heterozygote brothers inherited 2 independent (Phe240Leu and Glu145stop codon) mutations. The kinetic, thermodynamic, and associative properties of the recombinant human wild-type and Phe240Leu mutant enzymes were compared with those of TPIs in normal and deficient erythrocyte hemolysates. The specific activity of the recombinant mutant enzyme relative to the wild type was much higher (30%) than expected from the activity (3%) measured in hemolysates. Enhanced attachment of mutant TPI to erythrocyte inside-out vesicles and to microtubules of brain cells was found when the binding was measured with TPIs in hemolysate. In contrast, there was no difference between the binding of the recombinant wild-type and Phe240Leu mutant enzymes. These findings suggest that the missense mutation by itself is not enough to explain the low catalytic activity and “stickiness” of mutant TPI observed in hemolysate. The activity of the mutant TPI is further reduced by its attachment to inside-out vesicles or microtubules. Comparative studies of the hemolysate from a British patient with Glu104Asp homozygosity and with the platelet lysates from the Hungarian family suggest that the microcompartmentation of TPI is not unique for the hemolysates from the Hungarian TPI-deficient brothers. The possible role of cellular components, other than the mutant enzymes, in the distinct behavior of TPI in isolated form versus in hemolysates from the compound heterozygotes and the simple heterozygote family members is discussed.

Brain cell-free extract greatly stimulates the polymerization rate of purified tubulin with a reduction of the nucleation period and without a significant alteration of the final assembly state. This effect is mimicked by neuroblastoma extract at 10-fold lower extract concentration, but not by excess muscle extract. Copper inhibits microtubule assembly in vitrobut in the presence of brain extract the copper effect is suspended. Electron microscopic images showed that intact microtubules are formed and decorated by cytosolic proteins in the absence and presence of copper, while the copper alone induces the formation of S-shaped sheets and oligomeric threads. The flux of triosephosphate formation from glucose is enhanced by microtubules in brain extract, but not in muscle extract. Copper inhibits the glycolytic flux; however, the presence of microtubules not only suspends the inhibition by copper but the activation of glycolysis by microtubules is also preserved. We conclude that the organization of neuronal proteins modifies both the rates of microtubule assembly and glycolysis, and reduces their sensitivities against the inhibition caused by copper.
