Multidrug-resistant Edwardsiella tardathreatens both sustainable aquaculture and human health, but the control measure is still lacking. In this study, we adopted functional proteomics to investigate the molecular mechanism underlying norfloxacin (NOR) resistance in E. tarda. We found that E. tardahad a global proteomic shift upon acquisition of NOR resistance, featured with increased expression of siderophore biosynthesis and Fe3+-hydroxamate transport. Thus, either inhibition of siderophore biosynthesis with salicyl-AMS or treatment with another antibiotic, kitasamycin (Kit), which was uptake through Fe3+-hydroxamate transport, enhanced NOR killing of NOR-resistant E. tardaboth in vivoand in vitro. Moreover, the combination of NOR, salicyl-AMS, and Kit had the highest efficacy in promoting the killing effects of NOR than any drug alone. Such synergistic effect not only confirmed in vitroand in vivobacterial killing assays but also applicable to other clinic E. tardaisolates. Thus, our data suggest a proteomic-based approach to identify potential targets to enhance antibiotic killing and propose an alternative way to control infection of multidrug-resistant E. tarda.

The relationship between antibiotic resistance and bacterial virulence has not yet been fully explored. Here, we use Edwardsiella tardaas the research model to investigate the proteomic change upon oxytetracycline resistance (LTB4-ROTC). Compared to oxytetracycline-sensitive E. tarda(LTB4-S), LTB4-ROTChas 234 differentially expressed proteins, of which the abundance of 84 proteins is downregulated and 15 proteins are enriched to the Type III secretion system, Type VI secretion system, and flagellum pathways. Functional analysis confirms virulent phenotypes, including autoaggregation, biofilm formation, hemolysis, swimming, and swarming, are impaired in LTB4-ROTC. Furthermore, the in vivobacterial challenge in both tilapia and zebrafish infection models suggests that the virulence of LTB4-ROTCis attenuated. Analysis of immune gene expression shows that LTB4-ROTCinduces a stronger immune response in the spleen but a weaker response in the head kidney than that induced by LTB4-S, suggesting it's a potential vaccine candidate. Zebrafish and tilapia were challenged with a sublethal dose of LTB4-ROTCas a live vaccine followed by LTB4-S challenge. The relative percentage of survival of zebrafish is 60% and that of tilapia is 75% after vaccination. Thus, our study suggests that bacteria that acquire antibiotic resistance may attenuate virulence, which can be explored as a potential live vaccine to tackle bacterial infection in aquaculture.

Most researches of the threshold pressure gradient in tight gas reservoirs are experimental and mainly focus on the transient pressure response, without paying much attention to the transient rate decline. This paper establishes a dual-porosity rate transient decline model for the horizontal well with consideration of the threshold pressure gradient, which represents the non-Darcy flow in a fracture system. The solution is obtained by employing the Laplace transform and the orthogonal transform. The bi-logarithmic type curves of the dimensionless production rate and derivative are plotted by the Stehfest numerical inversion method. Seven different flow regimes are identified and the effects of the influence factors such as the threshold pressure gradient, the elastic storativity ratio, and the cross flow coefficient are discussed. The presented research could interpret the production behavior more accurately and effectively for tight gas reservoirs.

Applying 3D digital image correlation (3D-DIC) method to observe and investigate on crack forming process of C45 cylinder concrete under axial compression. The three-dimensional displacement and strain of the exterior surface of C45 concrete during the failure process are obtained. Furthermore, the location and shape of the crack are predicted with the generation, variation and aggregation of the maximum strain equipotential line on the concrete surface. The results show that the applying of 3D-DIC method can accurately obtain the concrete strain distribution data, predict the crack development trend and provide an important basis for further study on the mechanism of damage and fracture characteristics of concrete structure.

To determine how fracture–vuggy structure, pressure, and injection–production mode influence displacement efficiency in the water flooding process in fracture–vuggy reservoirs, two types of models were built based on the characteristic of such reservoir: two fracture models with different fracture apertures and two fracture–vuggy models with different sizes of vugs. A series of two-phase fluid flow experiments was conducted to study oil and water distribution as well as flow pattern in the water flooding process. Results show that displacement efficiency is significantly influenced by injection–production mode and injection pressure. By contrast, its relationship with the size of vugs is insignificant. Under the same injection–production mode, high-to-low recovery that corresponds to model type includes the small fracture, large fracture, small vug, and large vug models. Under the same pore volume multiple, high-to-low recovery that corresponds to model includes the small fracture, small vug, large fracture, and large vug models.

In this work, the influence of a rigid cylindrical inclusion on the elastic field in a transparent soft material is studied by an embedded-grating technique. The same experimental procedure was implemented on two specimens, a homogeneous bulk soft material and the same material containing a rigid cylindrical inhomogeneity. Under the indentation of a rigid wedge, two kinds of deformation sectors underneath the contacting boundary are observed from experimental results. Furthermore, the shape of the deformation sectors is horizontally expanded due to the inclusion. A local fluctuation of contact stresses between the soft matrix and rigid inclusion are discussed.

The model can be solved using the Laplace conversion method. The dimensionless bottom hole pressure can be obtained using the Stehfest numerical inversion method. The bi-logarithmic type curves for the tree-shaped fractal reservoirs are thus obtained. The pressure transient responses under different fractal factors are discussed. The factors with a primary effect on the inter-porosity flow regime include the initial branch number N, the length ratio α, and the branch angle θ. The diameter ratio β has a significant effect on the fracture radial flow, the inter-porosity and the total system radial flow regimes. The total branch level Mof the network mainly influences the total system radial flow regime. The model presented in this paper provides a new methodology for analyzing and predicting the pressure dynamic characteristics of naturally fractured reservoirs.

Bulk Nd60-xCo15+xAl25 (where x =0, 2, 5, 8, 11) sheet alloys were prepared by argon arc melting and suction casting a copper mold. Glassing forming ability (GFA) of these alloys was investigated by X-ray diffraction (XRD) and differential scanning calorimetry (DSC). Due to the dissimilarity reflected from DSC curves the thermodynamic calculation was applied. The values of Gibbs free energy (∆Gl-x (Tg)) for the amorphous alloys were gave out and some GFA criterions were adopted to make a comparison among the samples.

Anisotropic BaBi4Ti4O15 powder was synthesized by a molten salt synthesis (MSS) method in K2SO4-Na2SO4 flux and the effects of different process parameters such as calcining temperature, and ratio of salt to reactant (R) on the phase formation and morphology of anisotropic BaBi4Ti4O15 particles were also investigated. The as-synthesized powder calcined at 850-950°C exhibits a single tetragonal BaBi4Ti4O15 phase. The morphology of BaBi4Ti4O15 powder could be adjusted by changing the synthesis conditions. The average particle size (APS) of BaBi4Ti4O15 powder increased with R changing from 0.8 to 1.0, while it decreased with further increasing of R to 1.2. In addition, the APS increased with increasing calcining temperature and it showed an Arrhenius dependence on the temperature. The corresponding apparent activation energy for particle growth is 31.9kJ/mol for calcining temperature of 850-1000ºC.

With the increasing investigation on reliability and stability of engineering structures, it becomes important for reseach on mechanical properties of materials under fracutre conditions. Concerning to fracture problems of large deformation materials, deformation characteristics of the crack tip field is necessarily needed. This paper presents an experimental analysis on a double-cracked rubber disk under the I-II mixed-mode fracture condition. Deformation properties around the crack tip are described through analyzing displacement information acquired by digital moiré method of circular and radial gratings. Also the existence of sector division mode under mixed loading condition is verified, and it is also found experimentally that the shear component makes the sectors shift.

The glass-forming ability, thermal stability and magnetic properties of the Nd60-xDyxFe30Al10 (x=0, 2, 5) bulk amorphous alloys were investigated by x-ray diffraction (XRD), differential scanning calorimetry (DSC), scanning electron microscope (SEM) and the vibrating sample magnetometer (VSM). The results show that the glass forming ability of the Nd60-xDyxFe30Al10 (x=0, 2, 5) alloys decrease with increasing Dy content. The as-cast Nd60-xDyxFe30Al10 (x=0, 2, 5) alloys show hard magnetic behavior at room temperature. With increasing Dy content, the intrinsic coercivity of the alloys increase significantly while the saturation magnetization and remanence of the alloys decrease monotonously. With increasing annealed temperature, the intrinsic coercivity of the Nd55Fe30Al10Dy5 alloy decreased significantly, while the saturation magnetization and remanence decrease monotonously. The Nd55Fe30Al10Dy5 alloy shows soft magnetic behavior after annealed at 773K for 30 min.

On the base of some kinetics model analysis and kinetic observation of hydrate formation process, a new prediction model of gas hydrate formation is proposed. The analysis of the present model shows that the formation of gas hydrate not only relevant with gas composition and free water content but also relevant with temperature and pressure. Through contrast experiment, the predicted result of the new prediction method of gas hydrate crystallization kinetics is close to measured result, it means that the prediction method can reflect the hydrate crystallization accurately.

Horizontal well (HW) was divided into several elements after acidizing. In each element, there would exist a composite zone that was made up of damage zone (DZ) and acidizing zone (AZ). Local skin factor after acidizing was used to describe the resistance in each composite zone. The models for local skin factor and productivity of HW were created using the method of equivalent filtration resistance and displacement between two similar flow modes. According to the solution of the models, type curves of skin factor and production-increasing ratio after acidizing were illustrated, and the effect of different parameters in DZ and AZ on distribution of skin factor and production-increasing ratio after acidizing were discussed. The present model has a significant guide on the practice of acidizing technology.

After multistage fracturing, the flowback of fracturing fluid will cause two-phase flow through hydraulic fractures in shale gas reservoirs. With the consideration of two-phase flow and desorbed gas transient diffusion in shale gas reservoirs, a two-phase transient flow model of multistage fractured horizontal well in shale gas reservoirs was created. Accurate solution to this flow model is obtained by the use of source function theory, Laplace transform, three-dimensional eigenvalue method, and orthogonal transformation. According to the model’s solution, the bilogarithmic type curves of the two-phase model are illustrated, and the production decline performance under the effects of hydraulic fractures and shale gas reservoir properties are discussed. The result obtained in this paper has important significance to understand pressure response characteristics and production decline law of two-phase flow in shale gas reservoirs. Moreover, it provides the theoretical basis for exploiting this reservoir efficiently.
