Background: Statins represent a group of drugs that are currently indicated in the primary and secondary prevention of cardiovascular events. Their administration can be associated with side effects and the insufficient reduction of triacylglyceride (TAG) levels. This study aimed to assess the effect of the triple combination of statins with omega-3 fatty acids and coenzyme Q10 (CoQ10) on parameters associated with atherogenesis and statin side effects. Methods: In this pilot randomized double-blind trial, 105 subjects who met the criteria of combined dislipidemia and elevated TAG levels were randomly divided into three groups. In the control group, unaltered statin therapy was indicated. In the second and third groups, omega-3 PUFA 2.52 g/day (Zennix fa Pleuran) and omega-3 PUFA 2.52 g + CoQ10 200 mg/day (Pharma Nord ApS) were added, res//. At the end of the 3-month period (±1 week), all patients were evaluated. Results: Significant reduction of hepatic enzymes activity, systolic blood preasure, inflammatory markers and TAG levels were detected in both groups in comparison to the control group. Activity of SOD and GPx increased significantly after additive therapy. Coenzyme Q10 addition significantly reduced most of the abovementioned parameters (systolic blood preasure, total cholesterol, LDL, hsCRP, IL-6, SOD) in comparison with the statin + omega-3 PUFA group. The intensity of statin adverse effects were significantly reduced in the group with the addition of CoQ10. Conclusions: The results of this pilot study suggest the possible beneficial effects of triple combination on the lipid and non-lipid parameters related to atherogenesis and side effects of statin treatment. - Abstract
Many studies have highlighted the important role of PCSK9 in the development of cardiometabolic changes and its possible function as a biomarker of myocardial infarction or ischemic heart disease. This study aimed to determine the relationship between circulating PCSK9 levels and subclinical vascular changes in the group of low risk patients without manifest cardiovascular diseases. In this study, 120 healthy patients, free of manifest cardiovascular diseases, diabetes mellitus, and without lipid-lowering therapy, were divided into three groups based on BMI: normal weight (N= 50), overweight (N= 30), and obese (N= 40). Biochemical parameters, including basic lipid and non-lipid ones, were analyzed. PCSK9 levels were measured by ELISA, vascular changes were quantified by carotid ultrasound (carotid artery intima-media thickness, cIMT), and arterial stiffness parameters (pulse wave velocity, PWV; augmentation index, AI; stiffness parameter, β) were measured by an echo-tracking method. Plasma levels of PCSK9 significantly increased in obese (172.78 ± 51.67 ng/mL) in comparison with overweight (120.14 ± 37.64, p< 0.001) and normal weight groups (114.92 ± 35.87, p< 0.001). Differences between the overweight and normal weight groups were not significant (p= 0.85). The level of PCSK9 significantly correlated with values of BMI (p< 0.001, r= 0.38). In addition to increase in laboratory parameters associated with moderate metabolic changes, significant increase in cIMT and parameters of vascular changes (β, AI, PWV) were detected in groups with elevated BMI. Significant positive linear correlation of PCSK9 concentrations and cIMT (p< 0.001, r= 0.39), PWV (p< 0.001, r= 0.31), and β(p< 0.001, r= 0.3) were found. In multivariable regression analysis after adjusting for gender, age, BMI, and LDL, the impact of PCSK9 on cIMT, β, and PWV remained significant (p= 0.006, 0.03, and 0.002, respectively). PCSK9 plasma levels significantly correlated with subclinical vascular changes and their values were significantly elevated in obese subjects. We assume that PCSK9 could be used as a predictor of early vascular involvement, prior to the existence of manifest atherosclerosis. These results also highlight the role of anti-PCSK9 treatment in primary prevention.

14. Impact of alanyl-glutamine dipeptide on proliferative and inflammatory changes in jejunal mucosa after acute mesenteric ischemia.

- Abstract
Purpose The aim of our study was to determinate the impact of dipeptide (alanyl-glutamine) administration on inflammatory and proliferative changes in jejunal mucosa after acute mesenteric ischemia. Methods Male Wistar rats (n = 30) were divided into three groups: ischemia/reperfusion (IR) group which undergoes 60 min of mesenteric ischemia and 1 or 24 h of reperfusion (IR1, IR24, n = 12). Groups with dipeptide administration (D + IR1, D + IR24, Dipeptiven con inf., i.v., 0.75 g/kg) prior to IR injury were followed by 1 and 24 h of reperfusion. At the end of reperfusion period jejunal bioptic samples were obtained for histological (H&E), histochemical (Alcian blue) and immunohistochemical (anti-PCNA, anti-MPO) evaluations. Results Our results pointed out a significant (p < 0.001) increase of histopathological injury score in IR1 group compared to D + IR1 group. Immunohistochemical evaluation showed that MPO-positivity was significantly increased in IR groups after 1 (p < 0.001) as well as 24 h of reperfusion (p < 0.01) compared to dipeptide pretreated groups. Proliferative/reparatory rate was assessed using anti-PCNA antibody and showed a significant increase (p < 0.01) in PCNA cell positivity in lamina propria in dipeptide treated group compared to IR group. Conclusion In conclusion we may suggest that administration of alanyl-glutamine dipeptide prior to IR injury may help to protect small intestine and its mucous membrane integrity against insult such as intestinal ischemic/reperfusion injury presents. 19. Morphological changes in basement membrane associated with jejunal graft injury

Jejunal mucosa injury and morphometric continuity of the intestinal epithelial basement membrane (BM) in experimental groups after 1 h and 6 h of reperfusion following intestinal heterotopic allotransplation in male Wistar rats were observed. Intestinal graft tissue was compared to the jejunum of recipients. Significant damage to the jejunal mucosa was detected and followed by complete intestinal villi destruction, damage to epithelial intestinal crypts and the surrounding lamina propria mucosae. There was a local interruption of BM continuity, as well as its absence in particular areas. The percentage of continuous, clearly visible and undamaged BM detected using the periodic acid and Schiff reagent (PAS) method was 22.4% in the graft samples taken 1 h after transplantation. In the same samples, 43.1% undamaged BM was visualised by silver impregnation. Disappearance of the BM was significant in comparison with recipient samples using both staining methods (both p < 0.001) Biopsies of the intestinal mucosa graft 6 h after transplantation showed significant reduction of the damage to the jejunal mucosa (p < 0.05 and p < 0.01, respectively). The percentage of continuous, clearly visible and undamaged PAS-positive BM was 54%, and argyrophilic BM rose to approximately 83%. The difference between BM continuity detected by PAS method was significant when comparing graft and recipient samples (p < 0.001), but impregnation method did not revealed significant distinction. Based on our results, the silver impregnation method seems to be more sensitive to damage assessment of the jejunal graft.

22. Dynamic of apoptosis of cells in duodenal villi infected with Eimeria acervulina in broiler chickens

- Abstract
The aim of our study was to evaluate the parasite — host interactions at apoptosis level. We studied histopathological changes and time course of apoptosis in the duodenum during Eimeria acervulina infection. One-day-old broiler chicks were randomly allocated into two equal groups. At the age of two weeks the first group was experimentally infected with a pure suspension of sporulated E. acervulina oocysts. The second group served as a negative control. Tissue samples from the upper part of duodenum were obtained at 0.5, 1, 2, 3, 4, 5 and 6 days post infection. Biopsies of duodenum were studied immunohistochemically using DeadEnd™ Colometric TUNEL System for apoptosis detection in duodenal mucosa. Number of parasites in duodenal epithelium was also investigated. Our experimental results demonstrate: (i) macroscopic and histopathological changes in epithelium detected mainly in proximal segment of duodenum in infected groups; (ii) the number of developmental stages of E. acervulina (DSEA) during our trial increased, reaching the maximum 5 days post infection (dpi) (332.2 ± 16.12) (mean ± SEM), whereas the amount of DSEA declined significantly as late as 6 dpi (124.6 ± 3.91); (iii) the highest apoptosis level was recorded in initiatory 0.5 dpi (13.2 ± 1.02) and on the end of parasite development cycle after 5 dpi (12.6 ± 1.36). Finally, results showed that there was a period of inhibition of apoptosis during infection by E. acervulina.

23. Detection of early stages of apoptosis in experimental intestinal ischemia-reperfusion injury

- Abstract
Apoptosis is a form of programmed cell death that plays an important role in small intestine ischemia-reperfusion (IR) injury. The aim of this study was to determine the total proportion of apoptotic cell death (apoptotic index) following injury induced by ischemia and during various subsequent reperfusion periods, total histopathological status and the intestine regeneration dynamics after the IR injury. Experimental animals, Wistar rats (n = 45) were divided into three experimental and one control groups. In the experimental groups 1 h ischemia was followed by 1, 4 and 24 h reperfusion. Intestinal ischemia was induced by superior mesenteric artery (SMA) occlusion. Segments of jejunum were stained with hematoxylin and eosin and studied immunohistochemically using M30 CytoDEATH and in situ TUNEL methods for apoptosis detection. Our experimental data showed that: (i) apoptosis is an important form of cell death in the small intestine after IR injury induced by SMA occlusion; (ii) maximum levels of histopathological damage and apoptotic index of mucosa occurred after 1 h ischemia and 1 h of reperfusion; and (iii) mucosa possesses great regeneration ability. The lowest levels of histopathological damage and apoptotic index were observed in the group with 1 h ischemia and 24 h reperfusion where, however, the highest mitotic index was present.

- Abstract
We present a novel method of assessing damage to the liver using fluorescence analysis of hepatic mitochondria following ischaemia/reperfusion of the small intestine in a rat model. This work is of substantial importance in understanding the syndrome of multiorgan failure after ischemia/transplantation of the small intestine. Mitochondria were isolated from six sample groups that had undergone three different experimental treatments: a control group; a treatment with ischaemia followed by reperfusion of the small intestine (IRx); and, a one hour ischaemia followed by reperfusion after transplant of the small intestine (TRx). The IR treatment was further subdivided into three groups: 1, 24 h and 30 days reperfusion – IR1, IR24, IR720, respectively. Concomitantly, the TR treatment was further subdivided: one group underwent a 1 h reperfusion and another group a 6 h reperfusion following ischaemia and transplant – groups TR1 and TR6, respectively. Once treatment had been undergone, mitochondria were isolated and all five experimental groups – IR1, IR24, IR720, Tr1, Tr6 – and their emission matrices were analysed compared with that of the control group (C). Comparing fluorescence values in zone A of all experimental groups with those of the control group indicated a reduction in aromatic amino acids in the mitochondria of all experimental groups. Comparison of fluorescent zone B of experimental groups with the control group identified a lack of oxygen in samples IR1, IR24, which was indicated though an increase in the fluorescence of the reduced pyridine nucleotide NADH+H+.
