2. Application of Experimental Design (DoE) to Improve a Very Dilute Workup Procedure

- Abstract
A three-level face-centered central composite experimental design was applied to the reaction parameters (concentration, stoichiometry, temperature) of an acid-catalyzed rearrangement of a morphine derivative, with the aim of reducing the very dilute workup procedure required for isolation of the aporphine product without compromising yield, quality, or other parameters. This enabled the concentration of starting material in the acid catalyst to be increased 2-fold while maintaining other parameters, resulting in nearly 60% overall volume reduction on workup. This formal experimental design study required only 17 tube-scale experiments, yielding a more robust process overall, and additionally provided a greater understanding of the reaction design space for potential future scale-up.

Previous evidence indicates that adjuvant, short-course androgen deprivation therapy (ADT) improves metastasis-free survival when given with primary radiotherapy for intermediate-risk and high-risk localised prostate cancer. However, the value of ADT with postoperative radiotherapy after radical prostatectomy is unclear.

6. Square condenser apertures for square cameras in low-dose transmission electron microscopy

In transmission electron microscopy (TEM), cameras are square or rectangular but beams are round so the circular lobes irradiate adjacent areas, precluding further neighboring acquisition for beam-sensitive samples. We present condenser aperture plates with square and rectangular shapes that improve the efficiency of area usage by 70% and enhance montage imaging for beam-sensitive specimens. We demonstrate the compatibility of these condenser aperture plates with high-resolution cryogenic TEM by reconstructing a 1.8-Å map of equine apo-ferritin.

7. Gold Nanodrum Resonators

- Abstract
Nanodrum resonators have been fabricated using nanometer-thick gold films as the drumheads. The fabrication method is favorable for large-area array manufacture of arbitrary shapes. The drum resonators exhibit fundamental mode vibration frequencies in the MHz regime. We use the stretched-plate model to describe the natural vibrations of the drum. The Qfactor of the fundamental mode increases as the thickness of the drum increases and decreases as the drum diameter goes up. The highest Qfactor of the fundamental mode reaches 290 at room temperature and atmospheric pressure. Based on the deduced material properties we estimate that the resonator has a mass sensitivity of 1.11 × 10–22g/Hz.

The staging and surveillance of testicular cancer is a complex topic, which integrates clinical, biochemical, and imaging components. The use of imaging for staging and surveillance of testicular cancer is individually tailored to each patient by considering tumor histology and prognosis. This document discusses the rationale for use of imaging by imaging modality during the initial staging of testicular seminoma and nonseminoma tumors and during the planned surveillance of stage IA and IB testicular cancer by histological subtype integrating clinical suspicion for disease recurrence in surveillance protocols. The American College of Radiology Appropriateness Criteria are evidence-based guidelines for specific clinical conditions that are reviewed annually by a multidisciplinary expert panel. The guideline development and revision include an extensive analysis of current medical literature from peer reviewed journals and the application of well-established methodologies (RAND/UCLA Appropriateness Method and Grading of Recommendations Assessment, Development, and Evaluation or GRADE) to rate the appropriateness of imaging and treatment procedures for specific clinical scenarios. Digital soil mapping (DSM) has been used from the national to paddock scale in Australia over the past 20years. However, there has been insufficient objective evaluation of the limitations of DSM. The continual evolution of DSM methods over time have led to a lack of operational stability that creates an ongoing risk associated with the method. The inherent modelling paradigm of DSM (a reliance on systematic variation) is a key factor that creates potentially significant constraints to the use of DSM in Australia, particularly in the context of different scales of application. Inherent covariate limitations create a further potential ceiling to what can be achieved with DSM at any point in time. As part of a more critical and objective approach to the use of the method in Australia, there is a need for more effective exploration and discussion of these and other constraints in the DSM approach. This will hopefully ensure that it is used in a fit-for-purpose and effective manner in the future.

A combination of new
40 Ar/39 Ar dating results, major- and trace-element data, plus Sr-Nd-Pb-Hf isotope data, are used to investigate the petrogenesis of Triassic high-Si adakite (HSA), Cretaceous low-Si adakite-like (LSA) lavas, and Cretaceous high-K and shoshonitic trachyandesite lavas, from eastern and south-central Mongolia. All samples are light rare-earth element and large-ion lithophile element enriched but depleted in some high-field strength elements (notably Nb, Ta and Ti). Two alternative models are proposed to explain the petrogenesis of the HSA samples. (1) A southward-subducting Mongol-Okhotsk slab underwent partial melting in the Triassic during the closure of the Mongol-Okhotsk Ocean, with the resultant melts assimilating mantle and crustal material. Alternatively (2), a basaltic underplate of thickened (>50 km; >1.5 GPa), eclogitic lower crust foundered into the underlying mantle, and underwent partial melting with minor contamination from mantle material and some shallow-level crustal contamination. The LSA samples are interpreted as melts derived from a lithospheric mantle wedge that was previously metasomatised by slab melts. Similarly, the trachyandesite lavas are interpreted as melts deriving from a subduction-enriched subcontinental lithospheric mantle. The spatial distribution of these samples implies that metasomatism likely occurred due to a southward-subducting Mongol-Okhotsk slab associated with the closure of the Mongol-Okhotsk Ocean. When this interpretation is combined with previous evidence for a northward-subducting Mongol-Okhotsk slab it advocates that the Mongol-Okhotsk Ocean closed with double-sided subduction. 15. On the accuracy of soil survey in Queensland, Australia

- Abstract
The accuracy of soil survey is not well described in a consistent manner for either conventional or digital soil mapping in Queensland or more generally in Australia. Concepts of accuracy are often poorly understood and the rise of digital soil mapping has led to further terminology confusion for clients. Despite long-standing recommendations for derivation of accuracy statistics of soil surveys via statistically-based external validation, accuracy assessment by this method has been limited. Concepts for accuracy description (overall, producers and users accuracy) from the remote sensing discipline are applicable to soil survey and their use should be encouraged. An analysis of 12 published 1:50000 and 1:100000 soil surveys in Queensland revealed a 73% to 97% match between mapped polygonal and site data. This, in conjunction with accuracy standards for similar mapping disciplines and published soil survey accuracy assessments, leads us to recommend that a benchmark of 80% accuracy is realistic for all types of soil surveys. The adoption of a benchmark is however dependent upon further development and evaluation of accuracy assessment methods and standards, particularly in relation to minimum sample size and acceptance criteria. These outcomes will only be achieved if all surveys include accuracy assessment within the survey design.

16. Ketone Reductase Biocatalysis in the Synthesis of Chiral Intermediates Toward Generic Active Pharmaceutical Ingredients

A range of generic active pharmaceutical ingredients were examined for potential chiral alcohol motifs and derivatives within their structures that could be employed as key synthetic intermediates. For seven generic active pharmaceutical ingredients (APIs), eight precursor ketones were acquired and then subjected to reduction by >400 commercially available ketone reductases from different suppliers. Positive screening results were achieved for five ketones screened, with multiple ketone reductases available for each successful ketone. Selectivity was typically >99.5% ee in most cases, including for the opposite enantiomer. The three best examples were then optimized and quickly scaled up to 1 L scale in high conversion and isolated yield while retaining selectivity of >99.5% ee for the desired chiral alcohol enantiomer. This work illustrates that where a wide range of enzymes are available, productive enzymes to give either alcohol enantiomer can be readily identified for many ketones and rapidly scaled up to produce chiral alcohols. This approach is particularly applicable to generating chiral API intermediates.

Abstract The western spruce budworm (Choristoneura occidentalis Freeman = C. freemani (Razowski), Lepidoptera: Tortricidae) (WSB) is a recurrent defoliator of Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) and other conifers in western North America. Repeated defoliation causes timber depletions, which often require control using pest management programs involving combinations of aerial sprays with biological controllers (Bacillus thuringiensis) and other tactics. Anticipating the recurrence cycle of this periodic defoliator is important for forest management planning, which needs to consider budworm-caused depletions in timber supply analyses, and for risk management and control. We studied the historical periodicity of outbreaks at the western edge of the insect's distribution, on southern Vancouver Island, British Columbia, using the methods of dendrochronology applied to a living Douglas-fir chronology spanning 734 years (1281–2014 CE) and a floating chronology dating from 1863 to 1528 before the Common Era (BCE). To separate the growth fluctuations due to budworm defoliation from climatic fluctuations in the living chronology, we compared the temporal growth patterns of Douglas-fir with those of the non-budworm host western red cedar (Thuja plicata Donn ex D. Don), which is sympatric with Douglas-fir in this area. We concluded that before the 1930s, outbreaks were frequent, severe, and often spatially synchronized. Following the 1930s, periods of growth reductions indicative of outbreaks were present at the same frequency, but were less severe and not as synchronized as before. We provide evidence to suggest that periodic budworm disturbances on southern Vancouver Island have persisted through the centuries but that this regime has gradually shifted outside its historic range at about 1930. We concluded that although WSB is still present in this setting, population levels have not reached detectable outbreak levels for the last 90 years. We hypothesized that the absence of budworm outbreaks in the last 90 years is a response to the drastic ecosystem changes in this region caused by exploitation and fragmentation of the virgin forests, to possible changes in climate and to the accidental introduction of novel budworm predators into the ecosystem. 22. Normal saline versus Normosol™-R in sepsis resuscitation: A retrospective cohort study

Objective To determine the effect of Normosol™-R as compared to normal saline on the outcomes of acute kidney injury and the need for renal replacement therapy in the resuscitation phase of sepsis.Design Our study is a retrospective before-and-after cohort study.Setting The study occurred at a 700-bed tertiary academic level 1-trauma center.Patients A total of 1218 patients were enrolled through emergency department admissions. The normal saline (before) cohort was defined as the dates between 1 March and 30 September 2014 and the Normosol™-R (after) cohort was assessed from 1 March to 30 September 2015.Interventions None.Measurements and main results Intravenous fluid volumes received during the first 24 h, 72 h, and total hospital stays were compared. Sodium, chloride, potassium, and bicarbonate levels at 72 h were also compared. The medical coded diagnosis of acute kidney failure, need for renal replacement therapy, hospital LOS, ICU admission, ICU LOS, in-hospital mortality, and need for mechanical ventilation were all compared. There was no significant difference in intravenous fluid volumes between groups. Regression modelling controlling for baseline characteristics and 24-h fluid intake volume found no differences between groups for the primary outcomes of acute kidney injury (P= 0.99) and renal replacement therapy (P= 0.88). Patients in the Normosol™-R cohort were found to have a lower rate of hyperchloremia at 72 h post-admission (28% vs. 13%, P< 0.0001). There was a trend toward a decrease in the hospital and ICU LOS in the Normosol™-R cohort; however, the data were not statistically significant.Conclusions This study was unable to detect any difference in outcomes between sepsis patients who received intravenous fluid resuscitation with either a balanced crystalloid (Normosol™-R) or normal saline, except for a decreased rate of hyperchloremia.

Environmentally friendly, ultrafast display pixels of micrometer sizes are fabricated with nanometer-thick gold films and Si/SiO2wafers. The color displayed is due to both the plasmon response of the gold film and the optical interference from the Fabry–Peerot cavity formed by the underlying silicon substrate, the semitransparent gold film and the air gap between them. When an electric potential is applied to the gold film, the electrostatic force induces an attraction between the gold film and the silicon wafer. Due to the flexibility of the film, the size of the air gap changes, resulting in a changing color. By applying different driving signals, we have achieved cyan, magenta, and yellow reflected colors. The maximum switching rate of the pixel is primarily determined by the thickness dependence of the metal drum and its Young’s modulus and is typically in the MHz regime.

This study examines spatially variable stand structure and fire-climate relationships at a low elevation forest-grassland ecotone in west central British Columbia, Canada. Fire history reconstructions were based on samples from 92 fire-scarred trees and stand demography from 27 plots collected over an area of about 7 km
2 . We documented historical chronologies of widespread fires and localized grassland fires between AD 1600 and 1900. Relationships between fire events, reconstructed values of the Palmer Drought Severity Index, and annual precipitation were examined using superposed epoch and bivariate event analyses. Widespread fires occurred during warm, dry years and were preceded by multiple anomalously dry, warm years. Localized fires that affected only grassland-proximal forests were more frequent than widespread fires. These localized fires showed a lagged, positive relationship with wetter conditions. The landscape pattern of forest structure provided further evidence of complex fire activity with multiple plots shown to have experienced low-, mixed-, and/or high-severity fires over the last four centuries. We concluded that this forest-grassland ecotone was characterized by fires of mixed severity, dominated by frequent, low-severity fires punctuated by widespread fires of moderate to high severity. This landscape-level variability in fire-climate relationships and patterns in forest structure has important implications for fire and grassland management in west central British Columbia and similar environments elsewhere. Forest restoration techniques such as prescribed fire and thinning are oftentimes applied at the forest-grassland ecotone on the basis that historically high frequency, low-severity fires defined the character of past fire activity. This study provides forest managers and policy makers with important information on mixed-severity fire activity at a low elevation forest-grassland ecotone, a crucial prerequisite for the effective management of these complex ecosystems. - Abstract
Current first-line treatments for Hemophilia A patients are clotting factor replacement or bi-specific antibodies. These treatments require continuous, lifelong infusions, yet many patients have breakthrough bleeds. There remains a high unmet need for safe, effective, and durable therapies. We have developed a liver-directed non-viral in vivogene insertion approach using the piggyBac® DNA insertion system. Unlike conventional AAV-based gene therapy, our platform enables delivery of large transgenes, the ability to stably and efficiently integrate the therapeutic transgene into the genome, and the potential for re-dosing to titrate to target FVIII activity levels. A key challenge for all gene insertion systems relates to the need to safely and efficiently deliver the transgene DNA. In the current study we evaluated a novel hepatocyte-targeted non-viral platform able to co-deliver both DNA and mRNA with superior safety and specificity relative to the traditional liver-directed LNP concept.

32. Interannual climate variability drives regional fires in west central British Columbia, Canada

- Abstract
We investigated the influence of climate variability on forest fire occurrence at eight sites in west central British Columbia, Canada. Forty‐six local fire years affecting a single site and 16 moderate fire years affecting two or more sites were identified (1600–1900 A.D.). Existing fire history data were incorporated to identify 17 regionally synchronous fire years (fires that affected ≥3 sites). Interannual and multidecadal relationships between fire occurrence and the Palmer Drought Severity Index (PDSI), El Niño–Southern Oscillation (ENSO), Pacific Decadal Oscillation (PDO), and the Pacific North American (PNA) pattern were examined, in addition to the effects of additive positive phases of ENSO and PDO. We examined multiple reconstructions of ENSO, PDO, and PNA and utilized three methodological approaches to characterize climate‐fire relationships. We found that the influence of interannual climate, expressed as PDSI, increasingly synchronized the occurrence of fires when examined from local to regional scales. An association between local fires and positive antecedent moisture conditions suggests moisture‐driven fine fuel development and the proximity of some sites to grasslands likely function as key determinants of local‐scale fire activity. The relationships between regional fires and ENSO, PDO, and PNA suggest that large‐scale patterns of climate variability exert a weak and/or inconsistent influence over fire activity in west central British Columbia between 1700 and 1900 A.D. Although inconsistent among reconstructions of climate patterns, we identified a significant relationship between regional fires and large‐scale climate patterns when ENSO and PDO were both in positive phases. Ongoing and future climate changes are expected to result in fundamental shifts in forest productivity and wood quality over wide regions of interior British Columbia. This study was conducted to investigate the relationships between climate and wood property attributes within Douglas-fir and spruce forests, and to use those relationships to develop a non-invasive approach wood quality attribute prediction. Historical climate station data was correlated to tree-ring samples collected at five sites and climate–tree growth relationships were established to measure and predict wood density, cell-wall thickness, and microfibril angle attributes. Time series models were developed to reconstruct the measured wood properties, and a strong correspondence between the predicted and measured wood attributes was verified. The results confirm that climate parameters provide a useful index for assigning wood quality attributes to forest stands at many sites in BC’s interior. The findings of this research provide insight into the impact future climates may have on wood quality characteristics and could be applied elsewhere to investigate the impacts of climate change on forest fibre supplies. Examining the Government of Manitoba decision in 2014 to eliminate the Council on Post-Secondary Education, this article argues that government sought to exercise greater control over the public post-secondary system and its institutions in the province for the purpose of exacting greater control over system integration. While the elimination of the agency was consistent with the elimination of similar agencies in other provinces, the article finds that the direction of the new post-secondary governance model is less collegial and less consultative with more emphasis on regulation, and ministerial influence than was the case with the previous intermediary model, continuing trends in Manitoba towards greater government control. Service oriented chatbot systems are designed to help users access information from a website more easily. The system uses natural language responses to deliver the relevant information, acting like a customer service representative. In order to understand what users expect from such a system and how they interact with it we carried out two experiments which highlighted different aspects of interaction. We observed the communication between humans and the chatbots, and then between humans, applying the same methods in both cases. These findings have enabled us to focus on aspects of the system which directly affect the user, meaning that we can further develop a realistic and helpful chatbot. [ABSTRACT FROM AUTHOR]

Matching schemas at an element level or structural level is generally categorized as either hybrid, which uses one algorithm, or composite, which combines evidence from several different matching algorithms for the final similarity measure. We present an approach for combining element-level evidence of similarity for matching XML schemas with a composite approach. By combining high recall algorithms in a composite system we reduce the number of real matches missed. By performing experiments on a number of machine learning models for combination of evidence in a composite approach and choosing the SMO for the high precision and recall, we increase the reliability of the final matching results. The precision is therefore enhanced (e.g., with data sets used by Cupid and suggested by the author of LSD, our precision is respectively 13.05% and 31.55% higher than COMA and Cupid on average). Abstract: The growth of mountain hemlock trees in Pacific North America demonstrates a complex relationship to two or more seasonal environmental variables. In order to examine the radial growth response of mountain hemlock to subseasonal climate variables, ring-width and X-ray densitometric analyses were used to construct intra-annual dendroclimatic records. The intent was to highlight the difference between the dendroclimatic outcomes of standard ring-width analyses to those derived from density chronologies collected at high elevation locations in the British Columbia Coast Mountains. This study highlights the importance of using multiple tree-ring parameters to better define the complex growth behaviour in mountain hemlock trees for the construction of more robust proxy climate records. Tree-ring chronologies from three sites were used to describe the inherent climate-growth trends. Maximum tree-ring density values provided a robust data series for constructing site-specific proxy records of late-summer temperature. Annual ring-width measurements provided independent proxies of spring snowpack trends. Significant decreases in temperature and an increase in snowpack depth during the early 1700s and early 1800s coincides with documented PDO phases and Little Ice Age glacier advances. Identification of early and late growing season climate signals within mountain hemlock trees demonstrates the value of documenting the characteristics of multiple tree ring parameters in future dendroclimatic studies. Abstract: A white spruce ring-width chronology was used to reconstruct June–July mean temperatures in the northern Canadian Rocky Mountains back to 1772 A.D. Samples were collected in an old growth subalpine forest in the remote Kwadacha Wilderness Provincial Park. Two chronologies were created, one using standard dendroclimatological methods and one through the use of principal components analysis. The ring-width chronologies both showed a strong positive relationship with minimum, maximum, and mean temperatures during the current growing season. The principal component based chronology was deemed superior for use as a proxy record due to its greater ability to explain the variance in the instrumental temperature record and stronger performance during reconstruction verification. Comparison of this reconstruction with other dendroclimatological reconstructions from western Canada revealed a coherent pattern of low-frequency variability, whereas comparisons at annual times-scales showed considerable temporal and spatial variability in the level of agreement between reconstructions. The northern Canadian Rocky Mountains reconstruction showed no evidence of the reduction in sensitivity to climatic variability that has been found in many other northern spruce chronologies during the late 20th century.
