Smoothing of laser beam non-uniformities using gas jets has been studied. The experiment has been performed with the PALS laser working at 0.44 μm with an intensity of about 1015W/cm2. The laser beam has been split in two by a prism thus creating an artificial large-scale non-uniformity (≈90 μm). We recorded time resolved and static images of laser-gas jet interaction with and without an Al target. Multi 1D and 2D simulations show that such interaction acts redistributing the over-intensities over larger surface. This effect has to be attributed to ionization processes with consequent laser beam refraction. Results show that Argon gas jet produces a strong refraction of the laser beam thus strongly reducing the initial two spots separation.

Industrial lasers are an expanding technology of welding and other materials processing. Lasers with optical scanning heads are often used, as these provide more versatility, accuracy, and speed. The output part of the scanning head is covered by a protective glass, which might get contaminated by various particles from the laser processing. This decreases the transmissivity of the glass, and it can affect the production quality. The contamination needs to be checked regularly, but a visual inspection might not always be effective. This paper proposes two alternative methods of inspecting the protective glass: flash-pulse active thermography, and laser active thermography. They are based on the thermal excitation of the glass and measuring the response with an infrared camera. The experimental setup and practical results are described and the advantages and disadvantages are discussed. The presented methods are proven to be effective in detecting the contamination of the glass.

Abstract The efficiency of crater creation for different types of Al targets, namely, single massive targets and double targets consisting of a foil or a disk placed before the massive target at a chosen distance (300 and 500 µm), is studied. Targets were irradiated by the PALS facility laser beam with EL = 100 – 400 J at the first harmonic ? = 1315 nm, a focal spot radius of 125 µm, and pulse duration of 400 ps. Velocities of the accelerated foil’s fragments or disks and electron density distributions of the plasma streams are determined by means of three-frame interferometry. Shapes and volumes of craters are obtained using the crater replica technology and microscopy measurements. It is shown that direct laser action is the most efficient way of energy transfer to the massive target and the most efficient method of crater creation. Somewhat lower efficiencies of shock wave loading and crater creation in comparison with direct laser action are found in the case of double targets where the energy is transferred to the massive target by colliding laser-driven foils or disks. The efficiencies of such a colliding energy transfer are close to 60% for foils and 40% for disks. The experimental results are in a good agreement with two-dimensional hydrodynamic models of shock wave generation under direct laser action and laser-driven macroparticle impact.

The results of investigations are presented which are concerned with laser radiation absorption in a target, the plasma state of its ablated material, the energy transfer to the solid target material, the characteristics of the shock wave and craters on the target surface. The investigation involved irradiation of a planar target by a subnanosecond plasma-producing laser pulse. The experiments were carried out with massive aluminium targets using the PALS iodine laser, whose pulse duration (0.4 ns) was much shorter than the shock wave attenuation and on-target crater formation times (50—200 ns). The investigations were conducted for a laser radiation energy of 100 J at two wavelengths of 0.438 and 1.315 mm. For a given pulse energy, the irradiation intensity was varied in a broad range (1013—1016 W cm-2) by varying the radius of the laser beam. The efficiency of laser radiation-to-shock energy transfer was determined as a function of the intensity and wavelength of laser radiation; also determined were the characteristics of the plasma plume and the shock wave propagating in the solid target, including the experimental conditions under which two-dimensional effects are highly significant.

Soft X-ray contact microscopy (SXCM) experiments have been performed using the Prague Asterix Iodine Laser System (PALS). Laser wavelength and pulse duration were λ = 1.314 μm and τ (FWHM) = 450 ps, respectively. Pulsed X rays were generated using teflon, gold, and molybdenum targets with laser intensities I ≥ 1014 W/cm2. Experiments have been performed on the nematodes Caenorhabditis elegans. Images were recorded on PMMA photo resists and analyzed using an atomic force microscope operating in contact mode. Our preliminary results indicate the suitability of the SXCM for multicellular specimens.

Shock pressure generated in aluminum targets due to the interaction of 0.44 μm (3 ω of iodine laser) laser radiation has been studied. The laser intensity profile was smoothed using phase zone plates. Aluminum step targets were irradiated at an intensity I ≈ 1014 W/cm2. Shock velocity in the aluminum target was estimated by detecting the shock luminosity from the target rear using a streak camera to infer the shock pressure. Experimental results show a good agreement with the theoretical model based on the delocalized laser absorption approximation. In the present report, we explicitly discuss the importance of target thickness on the shock pressure scaling.

AbstractWe present an experimental study of the dynamics of shocks generated by the interaction of a double-spot laser in different kinds of targets: simple aluminum foils and foam–aluminum layered targets. The experiment was performed using the Prague PALS iodine laser working at 0.44 μm wavelength and irradiance of a few 1015W/cm2. Shock breakouts for pure Al and for foam-Al targets have been recorded using time-resolved self-emission diagnostics. Experimental results have been compared with numerical simulations. The shocks originating from two spots move forward and expand radially in the targets, finally colliding in the intermediate region and producing a very strong increase in pressure. This is particularly clear for the case of foam layered targets, where we also observed a delay of shock breakout and a spatial redistribution of the pressure. The influence of the foam layer doped with high-Z(Au) nanoparticles on the shock dynamics was also studied.

A procedure is presented for the direct and simultaneous determination of ascorbic acid (AA), isoascorbic acid (IA), and uric acid (UA) in human plasma by paired-ion reversed-phase high-performance liquid chromatography. An Ultrasphere ODS (C18) column is used with a pH 5.25 mobile phase containing 0.04M sodium acetate, 0.005M tetrabutylammonium phosphate, and 0.2 mg/mL disodium EDTA. Plasma samples preserved with an equal volume of 10% metaphosphoric acid are diluted 10-fold with mobile phase and filtered through 0.2 micron filters. The injection volume is 10 uL. Detection of AA, IA, and UA is by amperometry using a glassy carbon electrode and Ag/AgCl reference electrode. The applied potential is +0.6 volt and the sensitivity setting is 100 nAmps. As little as 0.25 ng of each component can be detected at this setting and the electrode response is linear over the AA, IA, and UA ranges encountered in human plasma.

Protein kinase activity was detected and assayed directly on polyacrylamide gels after disc electrophoresis of the 100,000 X g supernatant fraction of brown adipose tissue of infant rats. Nine major bands of activity were detected, eight of which could be stimulated by cAMP or inhibited by the cAMP-dependent protein kinase inhibitor protein. This electrophoretic technique revealed heterogeneity in the cAMP-dependent protein kinase activity eluted from DEAE-cellulose by high concentrations of salt, but not in the peak of activity eluted by low concentrations of salt. The catalytic properties and substrate specificities of the kinases in the various bands were studied while the enzymes were still in the gels. The activity in each band differed from each of the others in at least one of these properties. The activities of the protein kinases in brown fat changed as the animals grew, and each band exhibited a distinct and unique developmental pattern. The major changes in kinase activities occurred in the immediate post-parturition period, then at 15 days after birth and at weaning. These developmental stages coincide with the periods during which the tissue undergoes changes in the rate of its proliferation, differentiation, and functional activity.

We report experimental data on the oxidation of steel samples in air by laser irradiation using pulses of different duration generated by a photodissociative iodine laser source. The results indicate an enhancement of the oxidation process under the action of laser radiation as compared with conventional oxidation techniques.

A technique is described for the intermittent collection of blood from the rat tail. By using commonly available equipment, blood samples can easily be obtained from rats without the need for anaesthesia. The development of this technique makes the rat more readily available as an animal model for repeated withdrawals of small blood samples for pharmacokinetic or bioavailability evaluations.

1. The content of carnitine, acylcarnitine and total acid soluble carnitine in brown adipose tissue of rats increases rapidly after birth, attaining a peak on about day 10 and then decreases. Similar changes with age were found for carnitine acetyltransferase activity in mitochondria from brown adipose tissue and heart. The activity of this enzyme in brain and in liver is much smaller, but also increases postnatally. 2. The activity of carnitine palmitoyltransferase in brown adipose tissue, however, decreases after birth then increases later in life. 3. Exposure of 18-day-old rats to the cold for 20 days leads to an increase in carnitine content in brown adipose tissue and raises the activity of carnitine acetyltransferase. The activity of carnitine palmitoyltransferase is not affected by cold adaptation.

Activities of carnitine acetyl- and carnitine palmitoyltransferase in whole homogenates and isolated mitochondria from human fetal heart, liver, brain, skeletal muscle, intestine and placenta were studied. Both enzymes were considerably more active in human fetal liver and heart than in the same organs of fetal rats. Activities in fetal muscle and brain, however, were very low.

Carnitineacetyltransferase (CAT) and carnitinepalmitoyltransferase (CPT) and the carnitine content of white adipose tissue from human newborns and adult volunteers were estimated. The activity of CAT/mg mitochondrial protein did not change with age, while that of CPT was decreased in the adult. Carnitine content of newborn white adipose tissue was about 26 nmol/g wet weight, much lower than in other organs from human fetuses. Isolated white adipose tissue cells from newborn babies reacted to deoxycarnitine addition by a decrease in respiration; no such effect was observed in adult cells. In addition, the enhancing effect of norepinephrine on oxygen consumption of newborn cells could be further elevated by adding l-carnitine and again this was not observed in adult adipocytes.

Correlations of Doppler shifted line shapes with time-of-flight spectra of fast ions has been established for nanosecond-laser pulses. Fast ion energies of different velocity groups have been established in the large interval of Iλ2 = 4 × 1012–5 × 1016 W/cm2 μm2. The obtained scaling relations differ markedly from those reported by Gitomer et al. (1996).

Summary: Free carnitine levels were determined in amniotic fluids between the 10th and 40th week of gestation. They were found to decrease significantly with gestational age. Blood levels of carnitine were lower in pregnant than in non pregnant women. Levels were found to be higher in cord blood than in maternal blood and usually were higher in the umbilical artery than vein. Intra-arterial injection of L-carnitine into a pregnant ewe did not cause a rise in the fetal blood level of carnitine, which, in contrast to human fetal blood, contained less than half the level of carnitine in maternal blood.Speculation: It is suggested that the placenta may play a role in carnitine transport from mother to fetus and that fetal blood and amniotic fluid levels may reflect retention of carnitine by fetal tissues.

We present the results of experiments on the short-term irradiation of a solid material by a laser beam. The data testify to a rise in efficiency of the energy transfer from the laser pulse to a shock wave due to the fast-electron energy transfer. The experiments were performed with massive aluminium targets on the PALS iodine laser, whose pulse duration (0.4 ns) was much shorter than the time of shock decay and crater formation in the target (50–200 ns). The irradiation experiments were carried out using the fundamental laser harmonic (1.315 μm) with an energy of 360 J. The greater part of the experiments were performed for the radiation intensity exceeding 1015W cm-2, which corresponded to the efficient generation of fast electrons under the conditions where the relatively long-wavelength iodine-laser radiation was employed. The irradiation intensity was varied by varying the laser beam radius for a specified pulse energy.
