Our previous studies confirm that exogenous reduced nicotinamide adenine dinucleotide phosphate (NADPH) exerts a neuroprotective effect in animal models of ischemic stroke, and its primary mechanism is related to anti-oxidative stress and improved energy metabolism. However, it is unknown whether nicotinamide adenine dinucleotide (NADH) also plays a neuroprotective role and whether NADPH is superior to NADH against ischemic stroke? In this study we compared the efficacy of NADH, NADPH, and edaravone in ameliorating brain injury and metabolic stress in ischemic stroke. Transient middle cerebral artery occlusion/reperfusion (t-MCAO/R) mouse model and in vitro oxygen glucose deprivation/reoxygenation (OGD/R) model were established. The mice were intravenously administered the optimal dose of NADPH (7.5 mg/kg), NADH (22.5 mg/kg), or edaravone (3 mg/kg) immediately after reperfusion. We showed that the overall efficacy of NADPH in ameliorating ischemic injury was superior to NADH and edaravone. NADPH had a longer therapeutic time window (within 5 h) after reperfusion than NADH and edaravone (within 2 h) for ischemic stroke. In addition, NADPH and edaravone were better in alleviating the brain atrophy, while NADH and NADPH were better in increasing the long-term survival rate. NADPH showed stronger antioxidant effects than NADH and edaravone; but NADH was the best in terms of maintaining energy metabolism. Taken together, this study demonstrates that NADPH exerts better neuroprotective effects against ischemic stroke than NADH and edaravone.

Elasticity and stability play a crucial role in phase change fibers (PCFs), ensuring both comfort and effective thermal management in fabrics. In this study, we present the development of an elastic and shape-stable phase change fiber through coaxial wet spinning. The fiber consists of a paraffin wax (PW) core encapsulated in a thermoplastic polyurethane (TPU) sheath, providing reliable protection against leakage during use. The obtained PCFs exhibit a PW loading capacity of up to 64.9 wt% and a heating enthalpy of 117.0 J/g. Notably, these fibers maintain their morphological and thermal stability even after undergoing 50 cycles of heating and cooling treatments. By integrating the PCFs into a fabric and coating the surface with CNTs, the PCFs-based fabric achieves an outstanding photothermal conversion efficiency of 95.1 % under a light intensity of 250 mW/cm2and an electrothermal conversion efficiency of 73.2 % at a power level of 0.9 W. Furthermore, the fabric, woven with uniformly broken PW core, exhibits excellent tensile resilience in various conditions, including room temperature and during photothermal and electrothermal conversion processes. This study introduces a promising avenue for the development of innovative PCFs, offering significant potential for their application in the realm of personal smart fabrics, particularly in the field of advanced thermal management.

The autoimmune regulator (AIRE) protein is a putative transcription regulator with two plant homeodomain-type zinc fingers, a putative DNA-binding domain (SAND), and four nuclear receptor binding LXXLL motifs. We have shown here thatin vitro, recombinant AIRE can form homodimers and homotetramers that were also detected in thymic protein extracts. Recombinant AIRE also oligomerizes spontaneously upon phosphorylation by cAMP dependent protein kinase A or protein kinase C. Similarly, thymic AIRE protein is phosphorylated at the tyrosine and serine/threonine residues. AIRE dimers and tetramers, but not the monomers, can bind to G-doublets with the ATTGGTTA motif and the TTATTA-box. Competition assays revealed that sequences with one TTATTA motif and two tandem repeats of ATTGGTTA had the highest binding affinity. These findings demonstrate that AIRE is an important DNA binding molecule involved in immune regulation.

Autoimmune polyglandular syndrome type 1 [APS-1] comprises multiple organ-specific autoimmunities such as acquired hypoparathyroidism and autoimmune Addison's disease, and a predisposition to certain infections such as chronic mucocutaneous candidiasis. An APS-1 candidate gene was assigned to chromosome 21q22.3 by linkage analyses in patients with APS-1 from Finland. To examine the influence of ethnic and geographic differences on the location of the candidate gene locus, we studied 24 US patients with APS-1 by microsatellite marker typing, using five microsatellite markers, D21S49, PFKL, D21S171, D21S1903 and CD18, selected from chromosome 21q22.3. By allelic association analyses, the frequencies of allele number 5 for D21S171 and allele number 8 for D21S1903 were significantly higher in the 24 patients with APS-1 than in 33 controls (33/48 vs. 31/66,P=0.0207, Ξ2=5.35; 12/48 vs. 7/66, P=0.0418, Ξ2=4.15 respectively). The frequency of homozygosity for allele number 5 of D21S171 was also significantly higher in the patients than in controls, 15/24 vs. 9/33 (P=0.0078, Ξ2=7.07). Maximum lod scores detected for the five markers in nine families (containing 15 of the patients with APS-1) were: 2.384 for D21S49, 3.144 for PFKL, 3.506 for D21S171, 4.329 for D21S1903, and 1.130 for CD18. These results confirm the linkage of the candidate APS-1 gene to 21q22.3 in US APS-1 patients, and suggest that the candidate gene is located near the D21S1903 marker. The demonstration of the location of the APS-1 candidate gene to 21q22.3 in an out-bred heterogeneous patient population should promote the physical mapping of the responsible gene.

Objective: To study the pharmacological effect of Xiaoke Granule on blood sugar and blood rheological property in the diabetic animals. Methods: Alloxan induced diabetic mice or rats were grouped randomly. The effects of XKG on blood sugar, appetite, capacity of drinking, glucose tolerance, blood lipid and blood rheological property were observed and compared among groups. Results: XKG showed a trend in reducing the appetite and capacity of drinking, increasing the body weight, and significantly inhibiting the increase of blood sugar coused by ectogenic glucose in mice, and could improve the blood lipid and blood rheological property in rats. Conclusion: XKG is effective in reducing serum total cholesterol, lowering the blood viscosity, improving the blood rheological property of alloxan induced diabetic animals. Therefore, it might effective in treating and preventing the occurrence of diabetes mellitus and the complications of blood stasis.
