2. Promoting the transfer of learning among school staff following an online asynchronous training on dating violence prevention: A mixed-methods study

Abstract

Abstract:The SPARX team responded to the scarcity of training available to school staff concerning dating violence prevention by developing an innovative asynchronous online training program as one facet of their broader initiative. This mixed-methods study aimed to fill the existing gap in the literature regarding the factors influencing the transfer of learning, with two primary objectives. First, it sought to investigate how transfer culture and perceived social support from colleagues and school principals influenced school staff's intention to transfer learning post-online training. Second, it aimed to uncover the barriers and facilitators associated with learning transfer six months post-training. Engaging 101 school staff members, the research utilized an online questionnaire to evaluate their intentions to transfer learning from the SPARX program's School Staff component. Regression analysis was used to identify variables predicting intention to transfer, complemented by a qualitative multiple-case study approach focusing on the experiences of a teacher, school counsellor, and school administrator. Findings highlighted that perceived support from colleagues and opportunities to implement new skills significantly predicted a greater intention to transfer learning. In contrast, mutual support for collective learning predicted a lower intention to transfer. Key barriers included perceptions of sole responsibility for implementation and resource constraints, whereas facilitators encompassed access to human, financial, and material resources. Results underscore the pivotal role of environmental factors in promoting learning transfer within school settings, emphasizing the importance of perceived support from colleagues and advocating for a transfer culture characterized by delegated responsibilities, turnkey activities, and dedicated time for learning transfer.

Abstract

Systemic immune responses caused by chronic hypercholesterolaemia contribute to atherosclerosis initiation, progression and complications1. However, individuals often change their dietary habits over time2, and the effects of an alternating high-fat diet (HFD) on atherosclerosis remain unclear. Here, to address this relevant issue, we developed a protocol using atherosclerosis-prone mice to compare an alternating versus continuous HFD while maintaining similar overall exposure periods. We found that an alternating HFD accelerated atherosclerosis in Ldlr−/−and Apoe−/−mice compared with a continuous HFD. This pro-atherogenic effect of the alternating HFD was also observed in Apoe−/−Rag2−/−mice lacking T, B and natural killer T cells, ruling out the role of the adaptive immune system in the observed phenotype. Discontinuing the HFD in the alternating HFD group downregulated RUNX13, promoting inflammatory signalling in bone marrow myeloid progenitors. After re-exposure to an HFD, these cells produced IL-1β, leading to emergency myelopoiesis and increased neutrophil levels in blood. Neutrophils infiltrated plaques and released neutrophil extracellular traps, exacerbating atherosclerosis. Specific depletion of neutrophils or inhibition of IL-1β pathways abolished emergency myelopoiesis and reversed the pro-atherogenic effects of the alternating HFD. This study highlights the role of IL-1β-dependent neutrophil progenitor reprogramming in accelerated atherosclerosis induced by alternating HFD.

31. Generation of glioblastoma in mice engrafted with human cytomegalovirus-infected astrocytes

Abstract

Mounting evidence is identifying human cytomegalovirus (HCMV) as a potential oncogenic virus. HCMV has been detected in glioblastoma multiforme (GB). Herewith, we present the first experimental evidence for the generation of CMV-Elicited Glioblastoma Cells(CEGBCs) possessing glioblastoma-like traits that lead to the formation of glioblastoma in orthotopically xenografted mice. In addition to the already reported oncogenic HCMV-DB strain, we isolated three HCMV clinical strains from GB tissues that transformed HAs toward CEGBCs and generated spheroids from CEGBCs that resulted in the appearance of glioblastoma-like tumors in xenografted mice. These tumors were nestin-positive mostly in the invasive part surrounded by GFAP-positive reactive astrocytes. The glioblastoma immunohistochemistry phenotype was confirmed by EGFR and cMet gene amplification in the tumor parallel to the detection of HCMV IE and UL69 genes and proteins. Our results fit with an HCMV-induced glioblastoma model of oncogenesis in vivo which will open the door to new therapeutic approaches and assess the anti-HCMV treatment as well as immunotherapy in fighting GB which is characterized by poor prognosis.

32. Mindfulness and posttraumatic growth in childhood sexual abuse and psychological maltreatment survivors

Abstract

Abstract:Many studies focusing on childhood sexual abuse (CSA) may have neglected the consequences related to experiencing other types of childhood maltreatment, such as psychological maltreatment. Moreover, the literature has focused more on negative consequences related to CSA, hence potentially leaving out more adaptive and resilient trajectories. This qualitative study aimed to explore childhood psychological maltreatment and CSA, mindfulness, sexuality, and intimate relationships in adult survivors. The interview data from 19 individuals who experienced CSA and psychological maltreatment were analyzed using conventional content analysis. Four conceptual categories developed: (a) an unsafe family environment that hindered the development of positive experiences and relationships; (b) impaired mindfulness dispositions: the reliving of trauma, the avoidance of suffering, and feeling engulfed; (c) impaired intimacy: instability, codependence, emotional distance, and self-silencing; and (d) mindfulness dispositions and posttraumatic growth (PTG) in participants' intimate lives. These findings highlight that mindfulness, sexuality, and intimate relationships are often poor among survivors of psychological maltreatment and CSA. Yet opportunities to develop mindfulness dispositions appear to enhance PTG in survivors' intimate relationships, significantly contributing to survivors' healing processes.

Abstract

Les recommandations de la Haute Autorité de Santé (HAS) et de l’Agence Nationale de Sécurité du Médicament (ANSM) 2014, l’instruction de la Direction Générale des Soins (DGS) de novembre 2021, les recommandations de l’Etablissement Français du Sang (EFS) et les données de la littérature définissent globalement les bonnes pratiques transfusionnelles, mais apportent peu d’éléments quant à la prise en charge immunohématologique et transfusionnelle des patients ayant reçu une allogreffe de Cellules souches hématopoïétiques (CSH). L’objectif de cet atelier était d’harmoniser ces pratiques dans les situations pour lesquelles il n’existe actuellement aucune recommandation. Afin d’anticiper d’éventuelles difficultés transfusionnelles après allogreffe, nous recommandons, avant allogreffe, la réalisation d’un phénotypage érythrocytaire étendu du donneur et d’une recherche d’allo-immunisation anti-HLA chez le receveur. Nous recommandons la réalisation systématique pour les incompatibilités ABO mineures : d’un examen direct à l’antiglobuline entre j8 et j20 et pour les incompatibilités ABO majeures ; d’un titrage des anticorps anti-A/anti-B et d’un chimérisme érythrocytaire à j100. À un an post-allogreffe, nous recommandons la réalisation d’un chimérisme érythrocytaire afin de permettre, le cas échéant, une mise à jour des consignes transfusionnelles (phénotype RH, irradiation des concentrés de globules rouges).

Abstract

This article revisits the little-known history of the humble beginnings of the Catholic mission in the colony of Côte d’Ivoire, from the arrival of the first fathers of the Société des missions africaines de Lyon (SMA) in 1895 to the eve of the First World War. At the confluence of Christian missionary history, imperial or colonial history and African history, it explores the uncertain contribution of missionary Catholicism to the inchoate process of the formation of the Ivorian colony. It focuses on the turbulent relations between colonial and Catholic authorities : their official demeanour and their agreed dissonance, then their marked reversal from 1903-1904 and its consequences. The article also looks at the encounter between French missionaries, priests and nuns, and African subjects and their communities, at the source of a slow movement of Ivorian conversion to Roman Christianity.

Abstract

Le panorama des sarcomes utérins se complexifie de plus en plus avec la description de nouvelles entités associées à des altérations moléculaires récurrentes. Les léiomyosarcomes et les sarcomes utérins indifférenciés sont des sarcomes à génomique complexe. Les sarcomes du stroma endométrial de bas grade et de haut grade, d’autres sarcomes associés aux transcrits de fusion (tels que NTRK, PDGFB, ALK, RET ROS1) et le sarcome utérin déficient en SMARCA4 sont des sarcomes à génomique simple. Le léiomyosarcome est le sarcome utérin le plus fréquent suivi par les sarcomes du stroma endométrial. Il existe trois différents sous-types histologiques de léiomyosarcome (fusiforme, myxoïde, épithélioïde), les léiomyosarcomes myxoïde et épithélioïde étant plus agressifs que le léiomyosarcome fusiforme. La distinction entre un sarcome du stroma endométrial de bas grade et de haut grade est tout d’abord morphologique et immunohistochimique. La mise en évidence des transcrits de fusions aide au diagnostic. Définitivement reconnu comme entité distincte parmi les tumeurs mésenchymateuses gynécologiques, le PECome utérin fait partie des tumeurs rares. Grâce à des algorithmes d’évaluation du risque, les PEComes utérins sont différentiés en borderlineet en malins. Le rhabdomyosarcome embryonnaire du col utérin survient typiquement chez l’enfant, mais peut s’observer également chez la femme adulte. Le rhabdomyosarcome embryonnaire du col utérin est presque toujours DICER1 muté, à la différence de celui du vagin qui est DICER1 sauvage, et de l’adénosarcome qui peut être DICER1 muté mais en moindre fréquence. Parmi les entités émergentes, les sarcomes associés aux transcrits de fusion impliquant les gènes NTRK, ALK, PDGFBbénéficient d’une thérapeutique ciblée. L’intégration des données moléculaires avec l’histologie et la clinique permet de mieux identifier les sarcomes utérins afin de mieux les traiter.
