2. Whose resilience matters? Addressing issues of scale in supply chain resilience

Abstract

Many existing studies and models of supply chain resilience conceptualize resilience from the perspective of an equilibrium‐seeking focal firm or industry. The term “supply chain resilience” is often discussed as a dimension of firm resilience, as opposed to a standalone concept. In this context, resilience is defined according to how well a firm or industry can respond to a disruption while maintaining existing structures and processes and then “bounce back” to a pre‐disruption equilibrium or “bounce forward” to a new, presumably better, equilibrium in the shortest amount of time with the least cost. In this paper, we argue that equilibrium‐based conceptualizations of supply chain resilience can be misaligned with the notion of a supply chain as complex adaptive system (CAS) that continually adapts and transforms via autonomous actions of the different actors within the system. We offer suggestions on how practitioners and researchers can advance theory and practice by adopting a CASor complexity‐basedperspective on supply chain resilience. Specifically, we address the importance of scale and the challenges associated with mismatched scales.

Abstract

We address how to manage extended supply chains that lie beyond the first‐tier suppliers. The first‐tier suppliers have their suppliers who provide them with goods or services. While lower‐tier suppliers are often not an explicit concern of the focal firm, the firm's performance not only depends on the performance of its first‐tier suppliers, but also on the performance of their suppliers and beyond. These lower‐tier suppliers are much less visible and may not even be known to the focal firm. Our intention is to bring awareness to the importance of these lower‐tier suppliers and capture possible strategies that buying companies may pursue to manage them. Extended supply chains are complex and involve evolving relationships between various buyers and suppliers that are located along different tiers of the chain and across different parts of the globe. We address several topic areas and suggest emerging research focuses that should be considered in coming years.

Abstract

High-dimensional flow cytometry is the gold standard to study the human immune system in large cohorts. However, large sample sizes increase inter-experimental variation because of technical and experimental inaccuracies introduced by batch variability. Our high-throughput sample processing pipeline in combination with 28-color flow cytometry focuses on increased throughput (192 samples/experiment) and high reproducibility. We implemented quality control checkpoints to reduce technical and experimental variation. Finally, we integrated FlowSOM clustering to facilitate automated data analysis and demonstrate the reproducibility of our pipeline in a study with 3,357 samples. We reveal age-associated immune dynamics in 2,300 individuals, signified by decreasing T and B cell subsets with age. In addition, by combining genetic analyses, our approach revealed unique immune signatures associated with a single nucleotide polymorphism (SNP) that abrogates CD45 isoform splicing. In summary, we provide a versatile and reliable high-throughput, flow cytometry-based pipeline for immune discovery and exploration in large cohorts.
