Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), mainly obtained from fish, have been implicated in fetal development. Because few studies have examined maternal and umbilical cord blood fatty acid levels and infant body size in Japan with a fish-eating culture, we examined differences in plasma fatty acid levels in pregnant women and infant size at birth. This study is a large birth cohort study of 1476 pairs of Japanese pregnant women and their infants. Maternal blood DHA levels and infant birth weight showed a positive relationship. However, analysis adjusted for gestational age did not reveal correlations. Negative relationships were found between cord blood DHA levels and infant body size, and between the difference in mother-to-child DHA levels and infant body size. Thus, the smaller the birth size, the higher the differences in umbilical cord blood DHA levels and mother-to-child DHA levels when considering gestational age. [ABSTRACT FROM AUTHOR]

In the Great East Japan Earthquake of 11 March 2011, a magnitude 9.0 earthquake and accompanying tsunami struck the Tohoku region of Japan. The tsunami washed away old equipment containing polychlorinated biphenyls (PCBs) stored in the region's factories, and these PCBs may have leaked out of their casings into the marine environment. In this study, we evaluate marine PCB contamination by comparing mussels collected before (in 2005) and after (June 2011) the tsunami. PCB contamination levels were significantly elevated in mussel samples collected after the tsunami in June 2011 (3 months after the tsunami). This indicates that PCBs (specifically, formulation KC-400) leaked out of old equipment swept away by the tsunami and accumulated in mussels. PCBs were estimated to have an environmental half-life (EHL) in mussels of 4 months. Our results show that an earthquake and subsequent tsunami can cause elevated PCB contamination in the marine environment. • The turning point where DHA compositions were exchanged in maternal and umbilical cord blood was 6.6%. • It was observed that if DHA levels in maternal erythrocytes were greater than this turning point, DHA transfer from mother to fetus was through 'bioattenuation'. • If DHA composition in maternal erythrocyte was lower than the turning point, the transfer was under 'biomagnification'. • This corroborates the findings of previous research. Previous reports have shown that the transfer of docosahexaenoic acid (DHA) from mother to fetus during pregnancy is important for development of the child's nervous and visual functions. The amount of DHA passing through the placenta varies depending on the relative DHA compositions of the erythrocytes in the maternal blood and the umbilical cord. Prior research has reported that if the DHA composition of the maternal erythrocytes is over 5.6 g%, DHA in the erythrocytes of the child undergoes bioattenuation, whereas it undergoes biomagnification if the maternal erythrocyte composition is lower than 5.6 g%. The relationship between DHA levels in maternal erythrocytes during pregnancy and in umbilical cord erythrocytes at delivery was assessed in Japanese pregnant women. This study was performed as an adjunct study of the Japan Environment and Children's Study. DHA compositions of maternal erythrocytes at 24–30 weeks of pregnancy and of umbilical cord erythrocytes at delivery were determined in 1368 mother-infant pairs. Median DHA values were 7.41% in the maternal erythrocytes and 6.84% in the umbilical cord erythrocytes, indicating significantly lower levels in the umbilical cord. When DHA composition in maternal erythrocytes was lower than 6.6%, DHA was theoretically higher in umbilical cord erythrocytes than in maternal erythrocytes. Conversely, when DHA composition in maternal erythrocytes was higher than 6.6%, DHA in umbilical cord erythrocytes was theoretically lower than in maternal erythrocytes. We therefore consider that there is a turning point of around 6% in the DHA composition of maternal and umbilical cord blood that is exchanged between mother and fetus: if the composition in the maternal blood is higher, then bioattenuation in DHA transfer from the maternal circulation to the umbilical cord occurs, while if it is lower, then biomagnification occurs. Objective To assess the neurodevelopmental effects of the Great East Japan Earthquake in resident children. Study design The disaster on March 11, 2011, caused severe damage to the Sanriku coastal area, where we had been conducting a birth cohort study since 2003. It occurred in the middle of our 7-year-old examination. Approximately 500 mother-child pairs were compulsorily divided into 2 groups: 123 children finished the examination in the predisaster period, and 289 did in the postdisaster period. The remainder died or moved from that area. At the time of 7-year-old examination, we administered the Wechsler Intelligence Scale for Children-Third Edition and electrocardiography to assess autonomic function. According to the Child Behavior Checklist for ages 2-3 years and the Kaufman Assessment Battery for Children that had been administered at 30 months and 42 months of age, respectively, there were no significant differences in them between the 2 groups. Results Verbal IQ, including information, arithmetic, and vocabulary subscores of the Wechsler Intelligence Scale for Children-Third Edition, at 7 years of age was significantly lower in the postdisaster group than in the predisaster group. However, there were no significant differences in performance IQ, full-scale IQ, or autonomic nervous indicators between the 2 groups. Conclusions Since many schools were utilized as primary refuges after the disaster, the deficits in verbal IQ of 7-year-old children may have been due to the interrupted schooling. Mercury (Hg) and other heavy metal profiles, namely, lead (Pb), arsenic (As), cadmium (Cd), and selenium (Se) were investigated in maternal and umbilical cord (cord) red blood cells (RBCs) at parturition in Japanese population. Correlation coefficients of Hg, Pb, As, Cd, and Se between maternal and cord RBCs were 0.91, 0.79, 0.89, 0.31, and 0.76, respectively, and the respective means of cord/maternal RBCs ratios were 1.63, 0.52, 0.62, 0.12, and 1.18. These results indicate that fetal exposure to these metals (excluding Cd) strongly reflected each maternal exposure level. Among these metals, the placental transfer of methylmercury (MeHg) seemed to be extremely high but that of Cd to be limited. Hg showed positive correlations with Se in maternal RBCs but not in cord RBCs, and the Se/Hg molar ratio was lower in the latter, suggesting that the protective effects of Se against MeHg are less expected in fetuses than in mothers. Summary: Accumulating evidence indicates a strong association between obesity and health problems. Childhood obesity increases the risk of adulthood obesity. Although it is reported that the number of obese children in Japan has increased, it is unclear whether the obesity index distribution pattern has changed. To clarify this distribution pattern, we analyzed the changes in the body mass index (BMI) of primary (6th grade, 12 years old) and junior high (3rd year, 15 years old) schoolchildren in the city of Sendai during the period from 1989 to 2003. Our department has accumulated information on the height and body weight of all the schoolchildren in the city of Sendai. BMI was calculated using both the height and body weight data. The change in median BMI for 15 years had a slightly upward trend every year with both gender and school year. Although the BMI distribution showed that there was little or no change over time in the lower (10th and 25th) percentiles, the increases in the 75th and 90th percentiles were more marked than that in the 50th percentile in primary school children. On the other hand, there was only a small increase in BMI in the 3rd year of junior high school. Our results indicated that the changes in BMI distribution were concentrated at the upper end of the distribution in primary school children. The shifts in the value of high percentiles might be due to effect modification of the corresponding exposure by another environmental exposure or genetic predisposition. It is suggested that the factors causing this change influenced only part of this population, and affected individuals might represent a susceptible subpopulation of the exposed children. Secular changes in growth have been well documented in various world populations, with secular increase especially noticeable in the developed countries. Accordingly, we have been monitoring the secular changes in growth status among the 6th year children in primary schools (6thPS, 11-12 years old) and 3rd year children in junior high schools (3rdJHS, 14-15 years old) in the city of Sendai since 1934. After World War II, both primary school children and junior high school students showed marked increases in height and weight up to the early 1970s. Acceleration and the subsequent reduction in the degree of acceleration in growth were observed in 1965-1974 and 1975-1984, respectively, and were followed by reacceleration in 1985-1994. The aim of this study was to assess the growth changes among Sendai schoolchildren in 1994-2003. The period between 1994 and 1999 was characterized by positive trends both in height and weight among schoolchildren. However, the degree of the increases in height and weight was diminished between 1999 and 2003. The linear regression analysis revealed the significant increases in mean weight during the 10-year study period in 6thPS boys and 3rdJHS boys and girls. In contrast, there was no significant increase in mean height in any group. These findings suggest the leveling-off of the mean body height and weight among schoolchildren in Sendai at the end of the 20th century. Mercury in hair is a generally accepted biomarker of methylmercury exposure, and permanent waving has been reported to affect the mercury concentration in hair. We conducted an experimental-field study to examine the changes in the mercury concentration in hair induced by treatments such as permanent waving, straightening and coloring. Hair samples were collected from 19 female subjects enrolled before and after hair treatment by a beautician during each visit to a beauty saloon. A total of 38 pair samples were cut in 1-cm segments from the proximal end up to 10 cm, and then as 2-cm segments up to the distal end thereafter. Each segment was analyzed for total mercury concentration by cold-vapor atomic absorption spectrometry. Permanent waving decreased mercury concentration for most of the segments except for the proximal two segments and the 8-9 cm segment from the proximal end. Nevertheless the average mercury concentration of 3-cm segments from the proximal end showed no signifi cant decrease by permanent waving. Since females usually have hair longer than 3 cm, hair samples subjected to permanent waving may give lower mercury exposure estimates when the full-length hair strands are analyzed. However, analyzing the proximal 3-cm segment of hair samples does not give lower mercury exposure estimates. Assuming that hair samples are collected from puerperal women around the time of delivery, the 3-cm segments represent fetal exposure to methylmercury during the third trimester when fetuses are most vulnerable to methylmercury exposure. Therefore, mercury concentrations in the proximal segment of maternal hair collected in the right time can be a good biomarker of fetal methylmercury exposure. [ABSTRACT FROM AUTHOR]

Long-chain polyunsaturated fatty acids (LCPUFAs) required for infant development are produced by Δ6 desaturase (D6D) and Δ5 desaturase (D5D). The D6D index and D5D index are calculated based on their respective precursor/product ratios. The D5D and D6D indices are related to obesity and lifestyle-related diseases. The aim of the present study was to examine the associations of umbilical cord fatty acid profiles, D6D index, and D5D index in appropriate for gestational age (AGA), small for gestational age (SGA), and large for gestational age (LGA) infants. This was a nested case-control study, and the relationship between case and control maternal blood and umbilical cord blood fatty acid compositions was examined. Cases were small for gestational age (SGA; n = 55) and large for gestational age (LGA; n = 149) infants, whereas controls were appropriate for gestational age (AGA; n = 204) infants. Fatty acid profiles in maternal blood and umbilical cord plasma were analyzed by gas-liquid chromatography. The D6D index was calculated as dihomo-γ-linolenic acid (DGLA 20: 3 n-6) / linoleic acid (18: 2 n - 6), and the D5D index was calculated as arachidonic acid (20: 4 n - 6) / DGLA (20: 3 n - 6). Statistical analysis of umbilical cord blood fatty acids was performed with multiple comparisons. SGA infants showed high umbilical cord values for α-linolenic acid and DHA and lower values for DGLA compared to AGA infants. SGA infants showed a higher D5D index but a lower D6D index than AGA infants. LGA infants showed high values for α-linolenic acid and DGLA and lower values for arachidonic acid than AGA infants. LGA infants showed a high D6D index and a low D5D index relative to AGA infants. No significant differences in maternal blood fatty acid profiles, the D6D index, and D5D index desaturase activities were found among the three groups. There were differences in umbilical cord fatty acid profiles and D6D and D5D indices among AGA, SGA, and LGA infants, but further study is needed. [ABSTRACT FROM AUTHOR]

Long-chain polyunsaturated fatty acids (LC-PUFAs) are involved in the fetal growth in utero , and are essential for the development of visual and cognitive functions during infancy. The purpose of this study was to examine the associations of erythrocyte fatty acid compositions with FADS1 gene polymorphism in Japanese mothers and infants. The subjects were 383 mothers who participated in an adjunct birth cohort study of the Japan Environment and Children's Study (JECS). In maternal FADS1 SNP genotypes, the precursor fatty acids composition of the Δ5 desaturase in the maternal blood showed significant differences in levels among the groups, and showed increasing values in the order of TT < TC < CC genotype groups. On the other hand, many product fatty acids levels were significantly reduced in the order of TT > TC > CC genotype groups, and DHA levels were significantly lower in the CC genotype group relative to the other groups. Likewise, the relationship between fetal genotype group and fatty acid composition in cord blood was very similar to the maternal relationship. These results indicate the maternal and fetal blood fatty acid compositions are strongly influenced by the FADS1 genotypes. With respect to the cord blood DHA composition, the levels in the fetal CC genotype group showed a trend toward lower values in the maternal CC genotype group pair (p = 0.066) compared to the maternal TC genotype group pair. However, in the fetal TT and TC genotype groups (p = 0.131, p = 0.729, respectively), the maternal genotype did not have a significant effect. The DHA composition was more influenced by the maternal genotype in the fetal CC genotype group than in the fetal TT and TC genotype groups. It was shown that DHA transport via the placenta from the mother might be promoted in the fetal CC genotype compared to the other fetal genotype groups. In conclusion, differences in the FADS1 SNP genotypes of pregnant women and their children may greatly affect the supply of LC-PUFAs. Further studies on the involvement of the FADS1 polymorphisms and the fetal LC-PUFA levels in the fetal growth and development are warranted. [ABSTRACT FROM AUTHOR]
