AbstractObjectiveChocolate is a popular food that may affect the activity of the autonomic nervous system (ANS). The aim of this study was to determine the effect of a single dose of dark or milk chocolate on ANS cardiac control during rest and mental stress induced by the Stroop test (ST).MethodsHealthy participants, divided into DARK or MILK chocolate groups, ingested corresponding type of chocolate (1 g/kg body weight). They underwent measurement of ANS during relaxation and ST before and 2 h after chocolate consumption. ANS control was assessed by determination of heart rate (HR) and heart rate variability using parameters related to complex autonomic modulation (TP, SDNN) or primary vagal modulation (HFnu, RMSSD).ResultsHR was always increased during ST in both groups. Relaxation HR values after chocolate ingestion were higher only in the DARK chocolate group. During ST, values of TP, SDNN and HFnu decreased before and after chocolate ingestion in the DARK group, but only before chocolate ingestion in the MILK group. RMSSD values decreased during ST before and after chocolate ingestion in both groups. Relaxation TP, RMSSD and HFnu values after chocolate ingestion were lower in the DARK but not in the MILK group.ConclusionThe results suggest that even a single dose of milk chocolate attenuates changes in ANS cardiac control induced by mental stress, whereas a single dose of dark chocolate has an activating effect on the heart viamodification of ANS cardiac control at rest. Different levels of sugars and cocoa biologically active compounds in the two types of chocolate could explain the observed effects.

The sympathetic nervous system participates in the development and progression of several cancer types and this effect is mediated mainly via β-adrenergic signaling. However, the potential of β-adrenergic signaling blockade to prevent cancer development after exposure to carcinogens has not been investigated, yet. Therefore, in our study, we determined the effect of the β-blocker propranolol on the development and progression of mammary cancer induced in female rats by administration of the chemical carcinogen N-methyl-N-nitrosourea (MNU). The propranolol treatment (20 mg/kg body weight) started 12 days after MNU administration and lasted 10 weeks. We found that both saline and propranolol treatment significantly increased gene expression of the catecholamine-synthesizing enzyme tyrosine hydroxylase, indicating that repeated injection of saline or propranolol-induced stress in these two groups. However, compared to the vehicle-treated group, propranolol slightly delayed the development and moderately reduced the incidence of mammary carcinoma in animals. To evaluate the mechanisms mediating the effect of propranolol on the development of MNU-induced cancer, we investigated several parameters of the tumor microenvironment and found that propranolol increased gene expression of Casp3. Our data indicate that propranolol treatment that starts after exposure to carcinogens might represent a new, useful approach for preventing the development of cancer, especially in stressed individuals. However, the potential efficiency of propranolol treatment for preventing cancer development and progression in individuals exposed to carcinogens needs further investigation.

Objectives.Despite extensive research efforts, mechanisms participating on development of Alzheimer’s disease (AD) are covered only partially. Data from the last decades indicate that various stressors, as etiological factors, may play a role of in the AD. Therefore, we investigated the effect of two acute stressors, immobilization (IMO) and lipopolysaccharide (LPS), on the AD-related neuropathology.

Alzheimer’s disease (AD) represents the most prevalent form of dementia in the elderly. However, the pathological mechanisms underlying the development and progression of AD are only partially understood. To date, the accumulated clinical and experimental evidence indicate that the locus coeruleus (LC), the main source of brain’s norepinephrine, represents “the epicenter” of pathology leading to the development of AD. Evidence for this includes observations that neurons of the LC modulate several processes that are altered in brains of AD patients, including synaptic plasticity, neuroinflammation, neuronal metabolism, and blood-brain-barrier permeability. Moreover, the LC undergoes significant degeneration in the brains of AD patients and is considered a source of the prion-like spreading of tau pathology to forebrain structures innervated by the noradrenergic neurons of the LC. Furthermore, lesions of the LC exaggerate AD-related pathology, while augmentation of the brain’s noradrenergic neurotransmission reduces both neuroinflammation and cognitive decline. We hypothesize that better understanding the role of the LC neurons in AD pathogenesis may lead to development of new strategies for the treatment of AD.

Usually, drugs are designed to modulate at cellular level the activity of peripheral tissues and organs affected by pathological processes. Despite unprecedented investment, the number of newly developed drugs remains low. Therefore, potent new drugs that directly influence and restore activity of peripheral tissues and organs altered by somatic diseases are being released into clinical use at the same rate as seen in the past. The brain has long been known to regulate the function of all tissues and organs in the body. This regulatory influence has also been shown to play an important role during pathological conditions, when the brain triggers protective and adaptive reactions modulating the development and progression of somatic diseases affecting peripheral tissues and organs. Importantly, several drugs recently used for the treatment of somatic diseases (e.g., beta-blockers, sartans, statins) also act at the level of the CNS, where they modulate the protective and adaptive reactions of the brain. Pharmacological modulation of brain structures activities may represent the basis for a new approach towards the treatment of various somatic diseases (e.g., cardiovascular and gastrointestinal diseases). Therefore, brain-targeted therapy of somatic diseases may significantly extend the area for development of new drugs.

Both A5 and A7 brainstem noradrenergic cell groups innervate dorsal horns of the spinal cord. Moreover, A5 cell group directly innervates sympathetic preganglionic neurons. Thus, A5 and A7 noradrenergic neurons could modulate the sympathoadrenal system (SAS) activity. We investigated the role of A5 and A7 noradrenergic cell groups in regulation of the SAS activity under control and stressful conditions. We evaluated the effect of electrolytical lesions of A5 or A7 cell groups and also the effect of bilateral brainstem cuts interrupting brainstem pathways on tyrosine hydroxylase gene expression in A5 and A7 areas and on the SAS activity measured by plasma epinephrine and norepinephrine levels. We have found that immobilization stress increases activity of the A5 and A7 brainstem areas and also levels of the gene expression of tyrosine hydroxylase, the rate-limiting catecholamine biosynthetic enzyme. Immobilization of sham-operated and brainstem pathways transected or A5 or A7 lesioned animals induced a similar, highly significant increase in plasma epinephrine and norepinephrine levels in both sham-operated and A5 or A7 destroyed or transected groups. Our data suggest that both A5 and A7 noradrenergic cell groups are activated during immobilization stress. However, transection of brainstem pathways innervating A5 and A7 neurons or lesion of A5 or A7 cell groups is not sufficient enough for changes in immobilization stress-induced activation of the SAS. We suggest that neither A5 and A7 noradrenergic neurons nor the transected brainstem pathways represent structures crucial for an activation of the SAS during immobilization stress.
