Lysine post-translational modification (PTM) is involved in almost all cellular activities, and plays a critical role in protein structure and function. In the past decades, a variety of experimental methods for identifying PTM sites have emerged, which has promoted the development of PTM research. However, the experimental identification method is time-consuming and labor-intensive. Although deep learning has shown great potential in PTM site prediction recently, existing deep learning-based methods have limited effectiveness in predicting species-specific PTM sites. In this article, we propose a semi-supervised domain adaptation method, named Semi-ssPTM, for species-specific lysine PTM site prediction. Semi-ssPTM improves performance by introducing unlabeled PTM data and effectively enhances the generalization ability of species-specific prediction models. In addition, we train and test predictive models on two common lysine PTM acetylation and ubiquitination datasets. The experimental results show that our proposed semi-supervised method can effectively improve the prediction performance of multiple species and is superior to existing lysine PTM site prediction tools. The web server of Semi-ssPTM, as well as all datasets and associated code used in this study, is freely available at (http://bmi.ahu.edu.cn/Semi-ssPTM/). We expect Semi-ssPTM will provide practical guidance and useful insights for PTM site predictions and inspire future bioinformatics research in related fields.

Subcutaneous implantable cardioverter-defibrillator (S-ICD) therapy has become a viable alternative to conventional transvenous ICD implantation. Patients with congenitally corrected transposition of the great arteries (ccTGA) have a high risk of sudden cardiac death due to malignant arrhythmia. The interaction between the S-ICD system and the transvenous pacemaker system is not fully understood. We report a case of S-ICD implantation in a patient with ccTGA and a DDD pacemaker. The patient was a 30-year-old man with a previously placed pacemaker with diagnoses of congenital heart disease, ccTGA (SLL), left atrioventricular valve insufficiency, and third-degree atrioventricular block. He presented with an out-of-hospital cardiac arrest, and an S-ICD was implanted to prevent sudden cardiac death. Defibrillation checks were performed successfully. We tested the compatibility of the DDD pacemaker with the S-ICD and found that there was no interference between them. In conclusion, an S-ICD system is a reasonable and safe option in patients with ccTGA.

ABSTRACTObjectives:Notch1 regulates tumor biology in a complex, context-dependent manner. The roles of Notch1 in tongue cancer are still controversial. The aim of this study is to investigate the roles of Notch1 in tongue cancer.Materials and Methods:The expression of Notch1 was tested between tongue cancer and normal samples by using immunohistochemistry. Tongue cancer cells were transfected with siRNA or plasmid, respectively. Cell proliferation, apoptosis, migration and invasion ability were tested in appropriate ways. The subcutaneous tumor model was established to observe the tumor growth.Results:Notch1 was upregulated in tongue carcinoma tissues and the expression of Notch1 was related with tumor stage and differentiation. Overexpression of Notch1 could increase tongue cancer cells proliferation, invasion and migration. But inhibited the expression of Notch1 could decrease cells proliferation, invasion and migration and promote cell apoptosis in vitroand in vivo.Conclusion:Our results prove that the oncogenic role of Notch1 in tongue cancer and provide the direction of targeted therapy of tongue cancer.

The normal T6 heat treatment process for cast A356 alloy generally requires about 15 h. This long-period procedure increases greatly the manufacturing cost and decreases the productivity in practical production. In this study, a new short-time heat treatment process with only 30 min solution time at 540 °C was developed for the production of motorcycle wheel hubs in order to reduce heat treatment time. Comparisons on microstructure evaluation and mechanical properties, such as tensile strength and ductility, were made between this new fast process and the conventional T6 heat treatment. The results revealed that this new heat treatment process enabled the spheroidization of the eutectic silicon thoroughly, while minimizing the growth of eutectic silicon. The A356 alloy after this new short-time heat treatment shows nearly equal mechanical properties compared with the same alloy heat treated in a normal T6 heat treatment. This investigation makes it possible to significantly improve the efficiency of heat treatment on A356 alloy and, at the same time, improve the mechanical properties of the alloy.

The microfluidic has been applied to the field of bio-medicine, micro fluidic electronic in recent years. In this paper we reported a simple methodology to fabricate PDMS microfluidic channels. The method consists of three main fabrication steps: (1) Direct write the micrometric PCL 2D or 3D pattern by the near-field melt-electrospinning. (2) Cast the PDMS on the printed PCL pattern. (3) Completely remove the embedded sacrificial PCL layer by soaking and flushing with acetone. Uniformity of the morphology of the micro channels and well-alignment could be easily achieved due to include the dimensions achievable using this method. We have also designed and fabricated a few micro fluidic channels. SEM and white-light interferometer images were taken to illustrate the characters of essential parts. This work demonstrated the potential utilization of this technique.
