In recent years, numerical simulations have been playing an increasingly important role in the process of casting optimization. The reliability and validity of numerical simulation results are significantly determined by the accuracy setting of the interfacial heat transfer coefficient (IHTC). In this paper, the change law of IHTC between ring type castings and cores under different conditions is systematically investigated by using the Beck inverse algorithm, numerical simulation method and temperature field measurement data with AISI1045 steel as casting material and no-bake furan resin bonded sand as core material. The results show that the change of IHTC with time is in the form of “bimodal” after the cavity is poured, and the value increases rapidly to the maximum after pouring, and then decreases gradually to the minimum, followed by a slow increase and slow decrease process. The accuracy of the calculated IHTC was verified by using Procast software.

The safety assessment of high-level radioactive waste repositories requires a high predictive accuracy for radionuclide diffusion and a comprehensive understanding of the diffusion mechanism. In this study, a through-diffusion method and six machine-learning methods were employed to investigate the diffusion of ReO4-, HCrO4-, and I-in saturated compacted bentonite under different salinities and compacted dry densities. The machine-learning models were trained using two datasets. One dataset contained six input features and 293 instances obtained from the diffusion database system of the Japan Atomic Energy Agency (JAEA-DDB) and 15 publications. The other dataset, comprising 15,000 pseudo-instances, was produced using a multi-porosity model and contained eight input features. The results indicate that the former dataset yielded a higher predictive accuracy than the latter. Light gradient-boosting exhibited a higher prediction accuracy (R2=0.92) and lower error (MSE=0.01) than the other machine-learning algorithms. In addition, Shapley Additive Explanations, Feature Importance, and Partial Dependence Plot analysis results indicate that the rock capacity factor and compacted dry density had the two most significant effects on predicting the effective diffusion coefficient, thereby offering valuable insights.

•Report of 201 culture-confirmed EPTB/HIV co-infection patients in Southwest China.•The immune level of EPTB/HIV co-infection patients was low, and most were in advanced-stage AIDS.•Patients were mainly young males and the site of EPTB was mainly CNS/meningeal and lymphatic, but varied between the sexes.•The most common symptoms were diarrhoea, headache and fever, and high rates of drug resistance were found.•The drug resistance profile in EPTB/HIV co-infection patients was higher than in tuberculous meningitis or EPTB alone.

The thin ribbons and the bulk cylindrical rods with diameters of 2 mm and 10 mm of the Vit1 metallic glass (MG) were prepared by the single roller melt spinning method and the copper mold injection casting method, respectively. The cooling rates of the samples during melt solidification were evaluated. The glass transition behaviors of three groups of MG samples with different solidification cooling rates were studied by differential scanning calorimetry (DSC) at different heating rates. The effects of melt cooling rate on the glass transition kinetic parameters such as apparent activation energy (E) and fragility parameter (m) of the Vit1 MG were studied using the Kissinger and the Vogel-Fulcher-Tammann (VFT) equations. Additionally, the structural relaxation enthalpy (ΔHrel) of three groups of MG samples was quantitatively analyzed by DSC through multi-step temperature rise and fall measurements. Results show that the melt cooling rate (R) has a significant effect on the glass transition kinetics and the structural relaxation of the Vit1 MG. As Rdecreases in the order of magnitude, the glass transition temperature (Tg), E, m, and ΔHrelof the Vit1 MG gradually decreases. Furthermore, in the range of the experimental cooling rates, E, m, and ΔHrelall have an approximately linear relationship with lgR.

Neurofibromatosis type 1 [NF1] is an autosomal dominant genetic disorder affecting multiple organs. NF1 is well known for its various clinical manifestations, including café-au-late macules, Lisch nodules, bone deformity and neurofibromas. However, there is no effective therapy for NF1. Current therapies are aimed at alleviating NF1 clinical symptoms but not curing the disease. By altering pathogenic genes, gene therapy regulates cell activities at the nucleotide level. In this review, we described the structure and functions of neurofibromin domains, including GAP-related domain [GRD], cysteine-serine rich domain [CSRD], leucine-rich domain [LRD] and C-terminal domain [CTD], which respectively alter downstream pathways. By transfecting isolated sequences of these domains, researchers can partially restore normal cell functions in neurofibroma cell lines. Furthermore, recombinant transgene sequences may be designed to encode truncated proteins, which is functional and easy to be packaged into viral vectors. In addition, the treatment effect of gene therapy is also determined by various factors such as the vectors selection, transgene packaging strategies and drug administration. We summarized multiple NF1 gene therapy strategies and discussed their feasibility from multiple angles. Different protein domains alter the function and downstream pathways of neurofibromin.

Nowadays, laser is the mainstay treatment for cafe-au-lait macules (CALMs), but no systematic review has been published to demonstrate the overall efficacy and it’s still controversial which type of laser is optimal. Thus, we conduct the meta-analysis to evaluate the effectiveness and side effects of various types of lasers in treating CALMs. Original articles reporting the efficacy and side effects for CALMs in laser treatment were identified in PubMed, EMBASE, and Web of Science from 1983 to April 11, 2023. Using R software and the ‘meta’ package, meta-analysis was conducted for clearance and recurrence for evaluation of efficacy. And the occurrence of hypopigmentation and hyperpigmentation rate was pooled for safety evaluation. We used RoB2 and ROBINS-I tools to assess the risks of bias in RCT studies and non-RCT studies, respectively. The Grading of Recommendations, Assessment, Development and Evaluation system was used to assess the quality of the evidence. Nineteen studies involving 991 patients were included, which had a very low to moderate quality of evidence. The pooled 75% clearance rate was 43.3% (95% CI 31.8–54.7%, I2= 96%), 50% clearance rate was 75% (95% CI 62.2–85.9%, I2= 89%) and the recurrence rate was 13% (95% CI 3.2–26.5%, I2= 88%). The pooled hypopigmentation and hyperpigmentation rates were 1.2% (95% CI 0.3–2.1%, I2= 0%) and 1.2% (95% CI 0.3–2%, I2= 0%), respectively. Subgroup analysis revealed that QS-1064-nm Nd:YAG laser treatment not only achieved more than 75% clearance rate in 50.9% of patients (95% CI 26.9–74.4%, I2= 90%) but also resulted in the lowest hypopigmentation and hyperpigmentation rate of 0.5% (95% CI 0.0–2.5%, I2= 26%) and 0.4% (95% CI 0.0–2.5%, I2= 0%). To draw a conclusion, the laser treatment could reach an overall clearance rate of 50% for 75% of the patients with CALMs, for 43.3% of the patients, the clearance rate could reach 75%. When looking at different wavelength subgroups, QS-1064-nm Nd:YAG laser exhibited the best treatment capability. Laser of all the wavelength subgroups presented acceptable safety regarding of the low occurrence of side effects, namely, hypopigmentation and hyperpigmentation.

The effects of the in-medium nucleon–nucleon (NN) elastic cross section on the observables in heavy ion collisions in the Fermi energy domain are investigated within the framework of the ultrarelativistic quantum molecular dynamics model. The results simulated using medium correction factors of $${\mathcal{F}}=\sigma ^{\mathrm{in-medium}}_\mathrm{NN}/ \sigma ^{\mathrm{free}}_\mathrm{NN}=0.2,~0.3,~0.5,$$ F=σNNin-medium/σNNfree=0.2,0.3,0.5, and the density- and momentum-dependent factor obtained from the FU3FP1 parametrization are compared with the FOPI and INDRA experimental data. It is found that the calculations using the correction factors $${\mathcal{F}} =$$ F= 0.2 and 0.5 reproduce the experimental data (i.e., collective flow and nuclear stopping) at 40 and 150 MeV/nucleon, respectively. Calculations with the FU3FP1 parametrization can best fit these experimental data. These conclusions can be confirmed in both $$^{197}\text{Au}+^{197}\text{Au}$$ 197Au+197Au and $$^{129}\text{Xe}+^{120}\text{Sn}$$ 129Xe+120Sn .

Agropyron cristatum,a wild relative of common wheat (Triticum aestivumL.), provides many desirable genetic resources for wheat improvement, such as tolerance to cold, drought, and disease. To transfer and utilize these desirable genes, in this study, two wheat-A. cristatumderivatives II-13 and II-23 were identified and analyzed. We found that the number of root tip cell chromosomes was 44 in both II-13 and II-23, but there were four and six P genome chromosomes in II-13 and II-23, respectively, based on genomic in situhybridization (GISH). The chromosome configurations of II-13 and II-23 were both 2n=22II by the meiotic analysis of pollen mother cells (PMCs) at metaphase I, indicating that there were two and three pairs of P chromosomes in II-13 and II-23, respectively. Notably, wheat chromosome 7D was absent in derivative line II-13 while II-23 lacked chromosomes 4B and 7A based on SSR analysis combining fluorescence in situhybridization (FISH) analysis with pAs1 and pSc119.2 as probes. Chromosomes 2P and 7P were detected in both II-13 and II-23. Another pair of P genome chromosomes in II-23 was determined to be 4P based on expressed-sequences tags-sequence tagged sites (EST-STS) markers specific to A. cristatumand FISH with probes pAcTRT1 and pAcpCR2. Overall, these results suggest that II-13 was a 7P (7D) substitution line with one pair of additional 2P chromosomes and II-23 was a multiple 4P (4B), 7P (7A) substitution line with one pair of additional 2P chromosomes. Moreover, we obtained six alien disomic addition lines and five alien disomic substitution lines by backcrossing. These new materials will allow desirable genes from A. cristatumto be used in common wheat.

AbstractObjectiveThe objective of this study was to build a database to collect infectious disease information at the scene of a disaster through the use of 128 epidemiological questionnaires and 47 types of options, with rapid acquisition of information regarding infectious disease and rapid questionnaire customization at the scene of disaster relief by use of a personal digital assistant (PDA).MethodsSQL Server 2005 (Microsoft Corp, Redmond, WA) was used to create the option database for the infectious disease investigation, to develop a client application for the PDA, and to deploy the application on the server side. The users accessed the server for data collection and questionnaire customization with the PDA.ResultsA database with a set of comprehensive options was created and an application system was developed for the Android operating system (Google Inc, Mountain View, CA). On this basis, an infectious disease information collection system was built for use at the scene of disaster relief. The creation of an infectious disease information collection system and rapid questionnaire customization through the use of a PDA was achieved.ConclusionsThis system integrated computer technology and mobile communication technology to develop an infectious disease information collection system and to allow for rapid questionnaire customization at the scene of disaster relief. Dressing change is a significant and inevitable process during wound healing. Possible secondary damage caused through dressing removal may impose a great threat on wound recovery, thus resulting in healing delays and ultimately a higher cost of hospitalization. Hence, a non‐contact refreshable dressing with an ease of operation is of great desire, especially for chronic wounds where a long‐term and repeated dressing change would be performed. Herein, an all‐light‐operated hydrogel dressing that would achieve a fast and remote‐controllable dressing change (30 s for gelation/4 min for dissolution upon light irradiation) for chronic wounds is presented. In a diabetic murine model, substantially improved wound healing within 2–3 weeks is observed due to attenuated secondary damage during repeated dressing changes. Moreover, a promising facilitation of the healing processes of epithelialization, collagen deposition, cell proliferation, and inflammatory regulation is also detected, representing a synergistic effect of the photo‐responsive hydrogel dressing for therapeutic efficiency. An in situ on‐demand gelation/dissolution hydrogel dressing is developed for chronic wounds to reduce the secondary damage from repeated dressing changes. The hydrogel dressing enabled fast wound sealing within 30 s and on‐demand dissolution within 4 min, allowing fast and noninvasive dressing application and removal, as well as healing promotion.

In this study, the rapidity distribution, collective flows, and nuclear stopping power in 197Au+197Au collisions at intermediate energies were investigated using the ultrarelativistic quantum molecular dynamics (UrQMD) model with GEMINI++ code. The UrQMD model was adopted to simulate the dynamic evolution of heavy-ion collisions, whereas the GEMINI++ code was used to simulate the decay of primary fragments produced by UrQMD. The calculated results were compared with the INDRA and FOPI experimental data. It was found that the rapidity distribution, collective flows, and nuclear stopping power were affected to a certain extent by the decay of primary fragments, especially at lower beam energies. Furthermore, the experimental data of the collective flows and nuclear stopping power at the investigated beam energies were better reproduced when the sequential decay effect was included.

Inventions of materials to achieve biocompatibility, bioadhesion, and easy manufacturing are the urgent demand for promoting wound healing in clinical treatment. Hyaluronic acid (HA) is probably the ideal candidate for current dressing materials due to its well-known biocompatibility. However, the unavoidable problem for HA dressings is their inherent low adhesiveness to wounds, which severely impairs their treatment efficacy, especially during body movement. Here, we report a one-pot facile fabrication of hybrid double-network polydopamine-HA (PDA-HA) hydrogel with significantly enhanced adhesiveness compared to the HA hydrogel. Besides the easy manufacturing and promoted effectiveness, the PDA-HA hydrogel could be vacuum-dried to form a patch, further benefitting from the convenience for storage and distribution. When applied on the wound, the PDA-HA patch quickly rehydrated by absorbing exudate and stuck tightly to the wound. The applied PDA-HA patches keep the wounds covered for more than 7 days against strenuous exercise. Thus, mouse full-thickness wounds treated with the PDA-HA patches exhibited increased healing rates, where epithelization was finished within 14 days. Moreover, the hydrogel dressing exhibited promoting effects on vascularization and cell proliferation/migration. Together with the easy manufacturing procedure, good adhesion/adaptation, and promotion of wound healing, the PDA-HA patch holds great potential for future clinical translation.

The heterogeneous catalytic oxidation of pyridines to pyridine N-oxides has been studied using tungsten-loaded TiO2as the catalyst and hydrogen peroxide as the green oxidant. The catalysts were synthesized by a simple impregnation technique and characterized by X-ray powder diffraction, Raman spectroscopy, transmission electron microscopy, energy dispersion X-ray spectroscopy, X-ray photoelectron spectroscopy. The catalytic performances of the catalysts were evaluated by the N-oxidation of pyridines with 30 wt% H2O2solution as an environmentally friendly oxidant at room temperature. These processes serve as an efficient method to prepare a variety of pyridine-N-oxides in modest to high yields, and the pyridine N-oxides could be easily separated from the heterogeneous catalytic system. This study will provide a useful strategy for preparation of heterocyclic N-oxides in the mild condition.

Micro electroforming technology is widely used in fabrication of multilayer or moveable metal micro devices. The fabrication of these devices is usually suffered from high internal stress in micro-electroformed layers which seriously restricts the application and development of micro electroforming technology. Therefore, to control the internal stress is very important for improving the quality and performance of micro-electroformed layer. However, published studies on internal stress in the electroforming layer were mostly based on additive-free solution. According to additive solution, the effect of ultrasonic and current density on compressive stress occurring in the electroforming layer is investigated in this paper. The results indicate that the compressive stress keeps increasing with current density within range from 0.2 to 2 A/dm2. Meanwhile, the compressive stress in ultrasonic solution decreases by 73.4 MPa averagely comparing to that in ultrasonic-free solution, and the compressive stress also keeps decreasing with the ultrasonic power which gets the lowest value at 200W. Moreover, the mechanisms of additive-induced compressive stress and ultrasonic relieving compressive stress are discussed. This research work will complement the ultrasonic-stress reduction theory and may contribute to the development of micro electroforming technology.

There are some bottleneck problems in the supervised machine learning and unsupervised machine learning. In view of the current problems, this paper tries to make some meaningful exploration. The main work is as follows: Research on the statistical analysis of factor analysis and latent variable and in some valuable research results of typical machine learning, and some no analysis method and factor analysis of supervised learning or hidden variables method to contact with the typical analysis, summary of the comprehensive characteristics of implicit factor model and to reveal the hiding data structures help and contributions.

Background: Skin and soft tissue expansion is a procedure that stimulates skin regeneration by applying continuous mechanical stretching of normal donor skin for reconstruction purposes. We have reported that topical transplantation of bone marrow‐derived mesenchymal stem cells (MSCs) can accelerate mechanical stretch induced skin regeneration. However, it is unclear how circulating MSCs respond to mechanical stretch in skin tissue. Methods: MSCs from luciferase‐Tg Lewis rats were transplanted into a rat tissue expansion model and tracked in vivo by luminescence imaging. Expression levels of chemokines including macrophage inflammatory protein‐1α, thymus and activation‐regulated chemokine, secondary lymphoid tissue chemokine, cutaneous T‐cell attracting chemokine, and stromal‐derived factor‐1α (SDF‐1α) were elevated in mechanically stretched tissues, as were their related chemokine receptors in MSCs. Chemotactic assays were conducted in vitro and in vivo to assess the impact of chemokine expression on MSC migration. Results: MSC migration was observed in mechanically stretched skin. Mechanical stretching induced temporal upregulation of chemokine expression. Among all the tested chemokines, SDF‐1α showed the most significant increase in stretched skin, suggesting a strong connection to migration of MSCs. The in vitro chemotactic assay showed that conditioned medium from mechanically stretched cells induced MSC migration, which could be blocked with the CXCR4 antagonist AMD3100, as effectively as medium containing 50 ng/ml rat recombinant SDF‐1α. Results from in vivo study also showed that MSC migration to mechanically stretched skin was significantly blocked by AMD3100. Moreover, migrating MSCs expressed differentiation markers, suggesting a contribution of MSCs to skin regeneration through differentiation. Conclusion: Mechanical stretching can upregulate SDF‐1α in skin and recruit circulating MSCs through the SDF‐1α/CXCR4 pathway. StemCells2013;31:2703–2713

With the increase of mining intensity, mining causes significant dynamic disturbance and dynamic disasters, which have exerted a great influence on the safety and high-efficiency of production. This article based on the study of the displacement tendency of the rock masses on or under fractured face of the key layer, deduces the vertical stress calculation formula of the rock mass under the fractured face which indicates that the vertical stress is correlative with the length of fault block in key layer, the depth of key layer, the distance between key layer and mining coal seam, the equivalent elastic modulus of coal stratum underlying the key layer and so on; And then by numerical simulation, it studies the dynamic disturbance effect on front coal-rock masses as the key layer fractures which shows that dynamic disturbance effect caused by high intensity mining is relatively conspicuous on the fractured stratum (the key layer), the stratum under the fractured one and the stratum in the advancing direction of working face; As for the stratum on the fractured one (the key layer),the dynamic disturbance effect becomes less conspicuous as they grow farther away from the fractured one.

Polybenzimidazole (PBI) with a high molecular weight of 69,000 was first synthesized. It was afterwards grafted with benzimidazole pendant groups on the backbones. The acid doped benzimidaozle grafted PBI membranes were investigated and characterized including fuel cell tests at elevated temperatures without humidification. At an acid doping level of 13.1 mol H3PO4 per average molar repeat unit, the PBI membranes with a benzimidazole grafting degree of 10.6% demonstrated a conductivity of 0.15 S cm-1 and a H2-air fuel cell peak power density of 378 mW cm-2 at 180 oC at ambient pressure without humidification.

The goal of the present study was to obtain a comprehensive knowledge about synthesis conditions, structure, thermal behavior and conductivity properties of neodymiumorthophosphates in order to analyzeof use this material in intermediate temperature fuel cells due to their thermal and chemical stability properties.The impedance spectroscopy technique (IS) was used to measure the conductivity. The conductivity of compositesobserved to be lower than conductivity of pure CsH2PO4 but had improvedsufficiently conductivity of pure NdPO4•nH2O.

Based on the wave flume experiments, the response of excess pore water pressure was studied under the regular breaking wave action on a 1:30 slope sandy seabed. The topographic change was observed in the wave broken zone and its causes were analyzed. The results show that the variation gradient of excess pore water pressure are larger in the surface layer and the changing amplitude of variation gradient of excess pore water pressure is bigger in the wave broken zone. The water depth is the most influential factor of excess pore water pressure and the wave height and period has little influence on excess pore water pressure in the same wave steepness range in the wave broken zone. The topographic change is greatly in the wave breaking zone. Excess pore water pressures changing amplitude at the top are significantly higher than it at the bottom. When the seabed scours, the amplitude of the excess pore water pressure increases; and when the sand seabed accumulates, the amplitude of the excess pore water pressure decreases.

The wave breaking forces can exacerbate sediment transport, and lead to erosion of the seabed, coastal deformation and destruction of coastal structures. The experiment is carried out in a wave flume with a 1:30 sloping sandy seabed. A wide range of measurements from the regular wave runs are reported, including time series of wave heights, changes of bed profile. The video records are analysed to measure the time development of the seabed form and the characteristics of the orbital motion of the sand in the wave breaking region. The location and wave height at wave breaking point is measured by experiment. Formation and evolution of sand ripple and sand bar are studied under the breaking waves. It is found that effect of bed surface on wave breaking zone is more significant than wave non-breaking.

Abstract: There lies a close relationship between the seabed destruction and the distribution of pore water pressure under the action of breaking wave. The experiments were carried out in a wave flume with a 1:30 sloping sandy seabed to study regular breaking wave induced pore water pressure. A wide range of measurements from the regular wave runs were reported, including time series of wave heights, pore pressures. The video records were analysed to measure the time development of the seabed form and the characteristics of the orbital motion of the sand in the wave breaking region. The pore water pressure in the breaker zone showed the time variation depending on the wave phases including wave breaking and bore propagation. The time-averaged pore water pressure was higher near the seabed surface. The peak values of pore water pressure increase significantly at the breaking point. The direction of pore water pressure difference forces in the breaker zone is of fundamental importance for a correct description of the sediment dynamics. The upwardsdirected pressure differences may increase sand transport by reducing the effective weight of the sediment, thereby increasing the bed form evolution. The seabed configuration changed greatly at the wave breaking zone and a sand bar was generated remarkably. The amplitude of the pore water pressure changed with the seabed surface. The results are to improve the understanding of sand transport mechanisms and seabed responses due to breaking regular waves over a sloping sandy bed.

For the machine tool with complex forming motion, there is not accurate corresponding relationship between the existing criterion on precision design and the machining precision of workpiece. This article proposes a design scheme on machine tool precision based on machining precision of Workpiece. In this scheme, the first work is to complete the precision distribution and inspection from the workpiece to various component parts of technology system and achieve the total output precision of machine tool under the specified machining precision. This article serves YK3610 gear hobber as the example to describe the error model of system and basic application method in this work.

Structures of CaSO4 whisker(A) and CaSO4 whisker(B) were characterized by IR and XRD, and the morphologies were observed by optical microscope. The results showed that the modified CaSO4 whisker(B) has perfect crystal structure, high crystallinity and less defects. Mechanical properties and static thermal stability time(190 ℃) of two composites which were prepared by CaSO4 whisker(A) / PVC and CaSO4 whisker(B) / PVC were tested separately, and the degradation processes were analyzed by TG at 50 ℃-400℃, N2 atmosphere, and the dispersing states of two whiskers in the composites were observed by means of optical microscope and SEM. The research results showed that the tensile strength and elongation at break increased over 3 MPa and 40% separately, and the static thermal stability time was over 90 min of CaSO4 whisker(B) / PVC, the initial degradation time was delayed comparing with CaCO3 / PVC composite. The consistency and interfacial configuration between whisker and PVC resin were perfect, and the toughness and thermal stability of the composite was improved after CaSO4 whisker was modified. The research results indicated that modified CaSO4 whisker has better toughness and thermal stability actions for PVC.

In order to study the law of emergence and its internal attributable causes according to the resources owned by agents that interacts with each other in the complex adaptive system about the supply chain, at the beginning the resources which the agents in the supply chain draw are defined as followings: the resources outside system, the resources in the system, the resources that other else agents own, and the dynamic resources the agent itself own, the multi-compartment agent of resources. The operating points of eight resources owned by agents interact with each other are controlled through eight random numbers, the direction, degree, speed and stability of resources flow during interaction among agents are described respectively by matching capability, matching speed and matching time; Furthermore, the evolution process about the resources among agents interacting is described by rules of GEF(Global Evaluate Function) and LEF(Local Evaluate Function), resources flow model of complex system about the supply chain is established; Eventually, through simulating respectively in the Swarm platform both the proposed model and the simple model that doesn’t cunsder the multi-compartment agent but the linear sum of all resources, it is found that the former describes the change rule of the resource more accurately in the supply chain system. It means that syncretizing eight resources, which interact and inter-constrain in the system, to an indiscerptible one is the attributable cause of the emergence.

The Circle algorithm was proposed for large datasets. The idea of the algorithm is to find a set of vertices that are close to each other and far from other vertices. This algorithm makes use of the connection between clustering aggregation and the problem of correlation clustering. The best deterministic approximation algorithm was provided for the variation of the correlation of clustering problem, and showed how sampling can be used to scale the algorithms for large datasets. An extensive empirical evaluation was given for the usefulness of the problem and the solutions. The results show that this method achieves more than 50% reduction in the running time without sacrificing the quality of the clustering.

We proposed the flat-type permanent magnet linear alternator (LA) for free piston linear alternators (FPLAs) instead of the tubular one. Using the finite element method (FEM), we compare these two kinds of LAs. The FEM result shows that the flat-type permanent magnet LA has higher efficiency and larger output specific power than the tubular one, therefore more suitable for FPLAs, and that the alternator design can be optimized with respect to the permanent magnet length as well as the air gap.

Clinical application of mechanical stretching is a reconstructive method for skin repair. Although studies have reported dermal fibroblast heterogeneity, whether stretching affects individual fibroblast subpopulations equally remains unclear. In this study, we show the changes in dermal structure and papillary fibroblast (Fp) in regenerated human skin. Exhausted skin regeneration caused dermal‒epidermal junction flattening, papillary dermis thinning, and an increase in type III collagen-to-type I collagen ratio, with upregulated hallmarks of aging. Well-regenerated skin displayed a notable increase in the Fp population. Consistent changes were observed in the rat expansion model. Moreover, we found that TGFβ1 expression was especially increased in skin showing good regeneration. Activation of the TGFβ1/SMAD2/3 pathway improved exhausted skin regeneration and resulted in increased collagen content and Fp proliferation, whereas pharmacological inhibition of TGFβ1 action impacted well-regenerated skin. Short-term mechanical stretching that promoted skin regeneration enhanced Fp proliferation, extracellular matrix synthesis, and increased TGFβ1 expression, leading to good regeneration. Conversely, long-term stretching induced premature Fp senescence, leading to poor regeneration. This work shows the mechanism of mechanical stretching in well-skin regeneration that enhances Fp proliferation and extracellular matrix synthesis through the TGFβ1/SMAD2/3 pathway and highlights a crucial role of Fps in stretching-induced skin regeneration.

The pedicle paramedian flap is the most common method used to reconstruct the nose, but secondary operations for debulking are required. In addition, this flap is always limited in length, which makes it hard to form the distal part of the nose. In this study, the authors suggest a split forehead flap with an oblique orientation designed to overcome the disadvantages of the paramedian flap.

The RMSD for 66 newly measured nuclei that appeared in AME2020 was also significantly improved. The one-neutron and two-neutron separation energies predicted by these refined models are consistent with several theoretical predictions based on various physical models. In addition, the two-neutron separation energies of several newly measured nuclei (e.g., some isotopes of Ca, Ti, Pm, and Sm) predicted with LightGBM-refined mass models are also in good agreement with the latest experimental data. Conclusions:: LightGBM can be used to refine theoretical nuclear mass models and predict the binding energy of unknown nuclei. Moreover, the correlation between the input characteristic quantities and the output can be interpreted by SHapley additive exPlanations (a popular explainable artificial intelligence tool), which may provide new insights for developing theoretical nuclear mass models.

MEK1/2 inhibitors (MEKi) have recently achieved surprising success in treating unresectable plexiform neurofibromas (PNFs). However, few studies have investigated the mechanisms of MEKi resistance in PNF patients. We determined the efficacy of 6 different MEKi for treating PNFs, explored drug resistance mechanisms and identified potential combination therapies to overcome resistance. By screening drug efficacy among 6 MEKi in human NF1-deficient PNF cell lines, TAK-733 was found reduce PNF cell viability the most. We then cultured the TAK-733 resistant cells and explored potential targets for further treatment. Both high-throughput drug screening and RNA sequencing analyses of MEKi-resistant PNF cells identified cyclin-dependent kinase inhibitors (CDKi) as potential agents for PNFs. Dinaciclib, a CDKi, showed synergistic effects on MEKi-resistant cells. Coadministration of dinaciclib and TAK-733 significantly reduced cell viability, inhibited sphere formation and colony formation. Dinaciclib did not affect MEK signaling but decreased the expression of several prosurvival proteins, including survivin and CDK1, to induce apoptosis and inhibit mitosis. TAK-733/dinaciclib combination therapy induced tumor reduction in PNF patient-derived xenografts mouse models. Therefore, the combination of MEKi and CDKi may be promising for treating inoperable PNFs, especially when drug resistance exists. Our findings provide evidence for future clinical trials with MEKi-resistant PNF patients.
