Traditional Chinese medicine plays a significant role in the treatment of the pandemic of coronavirus disease 2019 (COVID-19). Tanreqing Capsule (TRQC) was used in the treatment of COVID-19 patients in the Shanghai Public Health Clinical Center. This study aimed to investigate the clinical efficacy of TRQC in the treatment of COVID-19.

Wireless network optimization is an important link in the beginning of the wireless communication network construction process, which decides the construction of wireless communication network whether scientific. This paper focuses on the method and content of the LTE wireless network optimization, mainly involves in the planning process and key technologies of LTE wireless network. First, this paper discusses LTE network parameters optimization include PCI, Cells, RS, C\N and RSC\RSRP. Then, verify the scheme scientific based on Automatic Cell Planning. Finally, through the network experiment examine the feasibility of the scheme.

Although memory deficits have been clearly documented in patients with human immunodeficiency virus type‐1 (HIV‐1) infection, the physiological basis of this dysfunction is poorly understood. We focused on Tat, a viral protein released from HIV‐1–infected cells and investigated its effect on spatial learning in adult mice. An intracerebroventricular injection of Tat leads to attenuation of spatial learning accompanied by suppression of long‐term potentiation (LTP), the cellular basis of spatial learning, in hippocampal cornu ammonis 1 pyramidal neurons. Tat facilitates extrasynaptic but not synaptic N‐methyl‐D‐aspartate (NMDA) receptor activity. Taken together, these data provide strong evidence that the Tat pathway underlies the development of memory dysfunction in patients with HIV‐1 infection and suggest a causal relationship between Tat, the facilitation of extrasynaptic NMDA receptor activity, inhibition of LTP, and attenuation of spatial learning.

We previously found that the nitric oxide (NO)-cGMP-cGMP-dependent protein kinase (PKG) signaling pathway acts in parallel with the cAMP-cAMP-dependent protein kinase (PKA) pathway to produce protein and RNA synthesis-dependent late-phase long-term potentiation (L-LTP) and cAMP response element-binding protein (CREB) phosphorylation in the CA1 region of mouse hippocampus. We have now investigated the possible involvement of a downstream target of PKG, ryanodine receptors. L-LTP can be induced by either multiple-train tetanization, NO or 8-Br-cGMP paired with one-train tetanization, or the cAMP activator forskolin, and all three types of potentiation are accompanied by an increase in phospho-CREB immunofluorescence in the CA1 cell body area. Both the potentiation and the increase in phospho-CREB immunofluorescence induced by multiple-train tetanization or 8-Br-cGMP paired with one-train tetanization are reduced by prolonged perfusion with ryanodine, which blocks Ca2+release from ryanodine-sensitive Ca2+stores. By contrast, neither the potentiation nor the increase in immunofluorescence induced by forskolin are reduced by depletion of ryanodine and inositol-1,4,5-triphosphate (IP3)-sensitive Ca2+stores. These results suggest that NO, cGMP, and PKG cause release of Ca2+from ryanodine-sensitive stores, which in turn causes phosphorylation of CREB in parallel with PKA during the induction of L-LTP.

Multifunctional Ca2+/calmodulin-dependent protein kinases (CaMKs) are thought to be involved in the induction of long-term potentiation (LTP). In the present study, LTP was induced by theta burst stimulation in the Schaffer collateral area of the stratum radiatum in the hippocampal CA1 region of the rat hippocampus. LTP-induced and control hippocampal slices were studied by Western blot and immunohistochemical analyses using CaMK-I, -II and -IV antibodies. Increased amounts of all three CaMKs were found in LTP-induced hippocampal slices as indicated by Western blot as well as by the density of their immunoreactivity. Our data clearly shows that not only CaMK-II but also CaMK-I and -IV contribute to synaptic plasticity formed in LTP.

We have elucidated the spatial and temporal localization of Cdk5 and p35nck5ain the developing rat postnatal cerebellum. Both proteins were highly expressed in cell bodies of post mitotic and immature neurons. The localization of Cdk5 in cellular compartment was changed from cell body to the axon in development. On the other hand, p35nck5awas always expressed in the cell body throughout cerebellum development. The Cdk5 kinase activity was correlated with the expression of p35nck5arather than that of Cdk5. These results indicate that p35nck5ais a physiological activator of Cdk5 in immature neurons and further suggest that Cdk5 has another function in mature neurons.

To observe the early interventions of traditional Chinese Medicine (TCM) on the conversion time of nucleic acid in patients with coronavirus disease 2019 (COVID-19), and find possible underlying mechanisms of action.

A self-powered microRNA (miRNA) biosensing device is fabricated with high-energy enzymatic biofuel cell (EBFC) and cruciform DNA (cDNA). Sulfur-selenium co-doped graphene/gold nanoparticles (S–Se-GR/AuNPs) is synthesized and used as supporting substrate of biocathode and bioanode. The glucose oxidase (GOD) molecules are bonded to carboxyl-functionalized AuNPs through the condensation reaction between the amino groups in enzyme and carboxyl groups on the AuNPs to form the bioanode. The ultra-thin porous carbon shell/AuNPs-complementary strand of cDNA (UPCS/AuNPs-cDNA) is meticulously designed. The potassium ferricyanides are then inserted in mesoporous UPCS/AuNPs as the biocathode electron acceptor, and the cruciform DNA bioconjugate is synthesized as signal amplifier. When target miRNA is added, the hybridization reaction happens between miRNA and capture probe DNA on the biocathode to open the capture probe DNA chain. Cruciform DNA bioconjugate is immobilized onto the biocathode through base pairing with the capture DNA on the biocathode, which can release electron acceptor [Fe(CN)6]3-, resulting in the dramatically increase of the open circuit voltage of the EBFCs. The self-powered biosensor responds linearly in the miRNA level range of 0.5–10000 fM with a detection limit of 1.5 × 10−16 mol L−1. Detection of miRNA in spiked serum samples is also realized with the self-powered biosensor, demonstrating its great potential in the clinical applications.
